A PRELIMINARY STUDY ON THE ACUTE TOXICITY OF TARTRAZINE IN RABBIT AND GUINEA PIG

  • 식용색소(食用色素) Tartrazine을 토끼의 경우 $7.5{\sim}50mg/kg$, 몰모트의 경우 $1,000{\sim}2,000mg/$수(首)를 경구(經口) 또는 복막주사(腹膜注射), 정맥주사(靜脈注射)로 투여(投與)하여 그 동물(動物)에 있어서의 색소(色素)의 독성(毒性)을 관찰조사(觀察調査)하였다. 혈청(血淸)은 화학분석(化學分析)하였고 각各 장기(臟器)는 현미경적(的) 관찰(觀察)을 통(通)하여 조사(調査)하였다. 그 결과(結果) 사용(使用)한 양(量)의 색소(色素)로서 대부분(大部分)의 장기(臟器)가 충혈(充血) 또는 출혈(出血)되였고 특(特)히 간장기능장해(肝臟機能障害)를 인정(認定)할 수 있었다.

키토산 부직포의 천연염색 (Natural Dyeing of Chitosan Nonwoven Fabric)

  • Chitin is a derived product from the shell of shrimp or crab. Chitosan, a deacetylated product of chitin, has widely been used in the biomedical sector, food industry, and textile industry. Chitosan exhibits fiber-forming property under certain conditions. Nonwoven fabrics made of chitosan fibers may have diverse applications in the industry. Previous studies have revealed that the dye uptake properties of natural dyestuffs improved by the chitosan pretreatment on the fabric specimens. In this case, fabric specimen is coated with acidic salt form of chitosan, which is different from the pure chitosan, since the coating process employes coating with the acidic solution of the chitosan and subesquent drying. In this study, chitosan nonwoven fabric samples were prepared from chitosan sample having deacetylation degree of $100\%$ and molecular weight of 650,000. Chitosan nonwoven fabrics maintain the form of $-NH_2$ end-group. These in turn exhibit higher dye uptake ability than the fabrics coated with chitosan acidic solutions do.

말똥성게 (Hemicentrotus pulcherrimus)의 수정 및 배 발생률을 이용한 해양배출 폐기물의 독성평가 (Toxicity Assessment of Ocean Dumping Wastes Using Fertilization and Embryo Development Rates in the Sea Urchin (Hemicentrotus pulcherrimus))

  • Toxicity assessment of ocean dumping wastes (dye waste, urban sewage, food waste) were examined in the fertilization and embryo development rates of the Sea Urchin, Hemicentrotus pulcherrimus. Spawning was induced by injecting 1 mL of 0.5 M KCl into coelomic cavity. Males released white or cream-colored sperms and females released yellow or orange-colored eggs. Experiments were began within 30 min the collection of both gametes. The fertilization and embryo development rates test were performed for 10 min and 64 h after fertilization, respectively. The fertilization and embryo development rates in the control condition (not including ocean dumping wastes sludge elutriate) were greater than 90%, but suddenly decreased with increasing of ocean dumping waste sludge elutriate concentrations. The fertilization and normal embryogenesis rates were significantly inhibited in all waste sludge elutriate from dye waste ($EC_{50}$=4.37; $EC_{50}$=1.76), urban sewage ($EC_{50}$=5.79; $EC_{50}$=2.00) and food waste ($EC_{50}$=7.68; $EC_{50}$=2.16), respectively. The NOEC (<3.13) and LOEC (3.13) of fertiliztion and normal embryogenesis rates very similar in all waste sludge elutriate. These results suggest that biological assay using the fertilization and embryo development rates of H. pulcherrimus are very useful test method for the ecological toxicity assessment of ocean dumping wastes.

백색부후균 Coriolus hirsutus LD-1의 리그닌분해효소 활성과 염료탈색에 관한 연구 (Production of Lignin Degrading Enzymes and Decolorization of Dye Compounds by White-rotting Fungi Coriolus hirsutus LD-1)

  • Coriolus hirsutus LD-1 균주의 리그닌분해효소 활성과 몇몇 염료의 탈색능을 조사하였다. 백색부후균인 LD-1 균주는 laccase(16,388.9 U/L)와 manganese- dependent peroxidase (19.81 U/L)는 생산하였으나 lignin peroxidase를 생산하지 않았다. 균주를 염료와 함께 8일간 배양했을때 염료 RBBR과 염료 BB의 탈색율은 각각 70.2%와 98%로 나타났다. Manganese-dependent peroxidase는 8일간 배양 중 효소 활성은 매우 낮은 반면 laccase는 지속적으로 생산되어 대단히 높은 활성을 나타내었다. 백색부후균인 Coriolus hirsutus LD-1에 의한 염료의 탈색은 주로 laccase에 의한 것으로 사료되었다.

Spawning was induced by injecting 1 mL of 0.5 M KCl into coelomic cavity. Males released white or cream-colored sperms and females released yellow or orange-colored eggs. Experiments were began within 30 min after the collection of both gametes. The fertilization and embryo development rates tests were performed for 10 min and 48 h after fertilization, respectively. The fertilization and embryo development rates in the control condition(not including ocean dumping wastes sludge elutriate) were greater than 90%, but markedly decreased with increasing concentrations of ocean dumping waste sludge elutriate. The fertilization and normal embryogenesis rates were significantly inhibited in all waste sludge elutriate from dye waste($EC_{50}$=5.76; $EC_{50}$=4.53), urban sewage($EC_{50}$=9.82; $EC_{50}$=9.67) and food waste($EC_{50}$=3.90; $EC_{50}$=3.27), respectively. The NOEC(>3.13%) and LOEC(3.13%) of fertiliztion and normal embryogenesis rates very similar in all waste sludge elutriate. These results suggest biological assay using the fertilization and embryo development rates of S. nudus are very useful test method for the ecological toxicity assessment of ocean dumping wastes.

한국산(韓國産) 보리의 화학조성(化學組成)에 관(關)한 연구(硏究) II. 대맥품종별(大麥品種別) P, K, Ca, Mg 함량(含量)과 단백질함량(蛋白質含量)과의 관계(關係) (Studies on Chemical Constituents of Barley in Korea II. Relationship between protein content and P, K, Ca, or Mg among barley varieties)

  • 스물두개 품종(品種)의 보리쌀중 단백질함량(蛋白質含量)은 Mg와 P 함량(含量)과 1%에서, K 함량(含量)은 5% 수준(水準)에서 유의성(有意性)이 있으나 Ca와는 유의성(有意性)이 없었다. 색소결합능(色素結合能)(dye binding capacity)과는 Mg 와 P 함량(含量)과만 1%에서 유의상관(有意相關)이 있었다. Mg 및 P 는 단백질함량(蛋白質含量) 식별지표(飾別指標)로 사용(使用)될 가능성(可能性)을 보였다. 이들 4개무기성분함량(個無機成分含量)과 탄수화물(炭水化物)과는 모두 유의성(有意性)없는 부상관(負相關)을 보였다. P,K,Mg 상호간(相互間)에는 1% 수준(水準)에서 유의상관(有意相關)을 보였다. 보리쌀 중 $K_2O$,$P_2O_5$, CaO 및 MgO 의 평균(平均) 함량(含量)은 각기 0. 30, 0. 29, 및 0. 08, 0. 043%였다.

A Portulaca oleracea L. extract promotes insulin secretion via a K+ATP channel dependent pathway in INS-1 pancreatic β-cells

  • BACKGROUND/OBJECTIVE: This study was designed to investigate how a Portulaca oleracea L. extract (POE) stimulates insulin secretion in INS-1 pancreatic ${\beta}-cells$. MATERIALS/METHOD: INS-1 pancreatic ${\beta}-cells$ were incubated in the presence of various glucose concentrations: 1.1 or 5.6, 16.7 mM glucose. The cells were treated with insulin secretagogues or insulin secretion inhibitor for insulin secretion assay using an insulin ELISA kit. In order to quantify intracellular influx of $Ca^{2+}$ caused by POE treatment, the effect of POE on intracellular $Ca^{2+}$ in INS-1 pancreatic ${\beta}-cells$ was examined using Fluo-2 AM dye. RESULTS: POE at 10 to $200{\mu}g/mL$ significantly increased insulin secretion dose-dependently as compared to the control. Experiments at three glucose concentrations (1.1, 5.6, and 16.7 mM) confirmed that POE significantly stimulated insulin secretion on its own as well as in a glucose-dependent manner. POE also exerted synergistic effects on insulin secretion with secretagogues, such as L-alanine, 3-isobutyl-1-methylxanthine, and especially tolbutamide, and at a depolarizing concentration of KCl. The insulin secretion caused by POE was significantly attenuated by treatment with diazoxide, an opener of the $K{^+}_{ATP}$ channel (blocking insulin secretion) and by verapamil (a $Ca^{2+}$ channel blocker). The insulinotropic effect of POE was not observed under $Ca^{2+}$-free conditions in INS-1 pancreatic ${\beta}-cells$. When the cells were preincubated with a $Ca^{2+}$ fluorescent dye, Fluo-2 (acetoxymethyl ester), the cells treated with POE showed changes in fluorescence in red, green, and blue tones, indicating a significant increase in intracellular $Ca^{2+}$, which closely correlated with increases in the levels of insulin secretion. CONCLUSIONS: These findings indicate that POE stimulates insulin secretion via a $K{^+}_{ATP}$ channel-dependent pathway in INS-1 pancreatic ${\beta}-cells$.

한국산(韓國産) 보리의 화학조성(化學組成)에 관(關)한 연구(硏究) -I. 대맥(大麥) 품종별(品種別) 보리쌀의 단백질(蛋白質) 및 탄수화물(炭水化物) 함량(含量)- (Studies on Chemical Constituents of Barley in Korea -I. Varietal Difference in Protein and Carbohydrate Contents of Polished Barley-)

  • 대맥주요(大麥主要)품종 22개의 보리쌀 중 단백질(蛋白質) 및 전탄수화물(全炭水化物) 함량(含量)을 분석(分析)하고 재배현황(栽培現況)의 연간추이(年間趨移)와 관계(關聯) 검토(檢討)하였다. 조단백함량(粗蛋白含量) $(N{\times}5.83)$은 평균(平均) 8.60 최고(最高) 15.39% 최소(最小) 6.06%이었고 약 1/3이 평균치(平均値) 이상(以上)이었다. 조단백(粗蛋白) 함량(含量)은 dye-binding capacity와 유의정상관(有意正相關)$(r=0.82^{**})$을 보였으나 탄수화물(炭水化物)과는 유의성(有意性)없는 부상관(否相關)을 보였다. 경1호(京1號)(15.39%), 관취기(關取埼)(12.42%) 및 영월육각(寧越六角)(11.95%)의 세품종만 고단백품종(高蛋白品種)으로 볼 수 있으나 거의 재배(栽培)되지 않고 있다. 지난 10년간(年間) 대맥(大麥)의 품종별 재배면적비(品種別 栽培面積比)는 거의 변화(變化)가 없으며 조단백함량(粗蛋白含量) 평균치이상(平均値以上)이 품종(品種)이 전대맥재배면적(全大麥栽培面積)의 약 50%를 차지하고 있으며 경남대맥(慶南大麥) 89호(號)는 단백함량(蛋白含量)이 상당히 낮은 데도 불구(不拘)하고 8%의 면적(面積)을 차지하고 있다.

철 함침 낙엽 Biochar에 의한 음이온성 염료의 흡착특성 (Adsorption Characteristics of Anionic Dye by Fe-Decorated Biochar Derived from Fallen Leaves)

  • BACKGROUND: There is a need for a revolutionary method to overcome the problem of biochar, which has relatively low adsorption capacity for existing anion pollutants, along with collectively recycling fallen leaves, a kind of forest by-product. Therefore, the objective of this study was to prepare iron-decorated biochar derived from fallen leaves (Fe-FLB), and to evaluate their adsorption properties to Congo red (CR) as anionic dye. METHODS AND RESULTS: The adsorption properties of CR by fallen leaves biochar (FLB) and Fe-FLB were performed under various conditions such as initial CR concentration, reaction time, pH and dosage with isotherm and kinetic models. In this study, Fe-FLB prepared through iron impregnation and pyrolysis of fallen leaves contained 56.9% carbon and 6.3% iron. Congo red adsorption by FLB and Fe-FLB was well described by Langmuir model and pseudo second order model and the maximum adsorption capacities of FLB and Fe-FLB were 1.1 mg/g and 25.6 mg/g, respectively. In particular, it was found that the adsorption of CR was occurred by chemical adsorption process by the outer boundary layer of Fe-FLB. CONCLUSION: Overall, the production of Fe-FLB using fallen leaves and using it as an anion adsorbent is considered to be a way to overcome the problem of biochar with relatively low anion adsorption in addition to the reduction effect of waste.

Development of Liposome Immunoassay for Salmonella spp. using Immunomagnetic Separation and Immunoliposome

  • The ability to detect Salmonella spp. is essential in the prevention of foodborne illness. This study examined a Salmonella spp. detection method involving the application of immunomagnetic separation and immunoliposomes (IMS/IL) encapsulating sulforhodamine B (SRB), a fluorescent dye. A quantitative assay was conducted by measuring the fluorescence intensity of SRB that was produced from an immunomagnetic bead-Salmonella spp.-immunoliposome complex. The results indicated detection limits of $2.7{\times}10^{5}$ and $5.2{\times}10^{3}$ CFU/ml for Salmonella enterica subsp. enterica serovar Enteritidis (S. Enteritidis) and Salmonella enterka subsp. enterka serovar Typhimurium (S. Typhimurium), respectivley. The signal/noise ratio was improved by using 4% skim milk as a wash solution rather than 2% BSA. In addition, higher fluorescence intensity was obtained by increasing the liposome size. Compared with the conventional plating method, which takes 3-4 days for the isolation and identification of Salmonella spp., the total assay time of to h only including 6 h of culture enrichment was necessary for the Salmonella detection by IMS/IL. These results indicate that the IMS/ IL has great potential as an alternative rapid method for Salmonella detection.
