Facile Preparation of Water Dispersible Red Fluorescent Organic Nanoparticles for Cell Imaging

  • Red fluorescent organic nanopaticles (FONs) based on a diarylacrylonitrile derivative conjugated molecule were facilely prepared by surfactant modification. Such red FONs showed excellent water solubility and biocompatibility, making them promising for cell imaging applications.

Synthesis of dimeric fluorescent TSPO ligand for detection of glioma

  • TSPO, an 18-kDa translocator protein, is a peripheral-type benzodiazepine receptor that has been associated to a variety of biological activities such as apoptosis, steroidogenesis, and cell proliferation. Because TSPO overexpression has been found in various forms of cancer, it has recently become one of the most appealing biological targets for cancer therapies and detection. In order to create new optical imaging agents for improved diagnostics, we synthesized a novel dimeric fluorescent TSPO ligand based on PRB28 structure and SCy5.5. Following the preparation of the novel TSPO ligand, in vivo and ex vivo imaging tests were performed to examine the tumor uptake characteristics of the fluorescent TSPO ligand in a glioma animal model, and it was found that novel TSPO ligand was accumulated in glioma. These results suggested that novel dimeric fluorescent TSPO ligand will be applied to detect glioma.

Prefluorescent-Dye-Induced Fluorescent Imaging based on Polymeric Photobase Generators

  • Copolymers containing oxime-urethane groups, which led to the formation of pendant amino groups photochemically, were applied to a fluorescent image-recording material through the reaction with fluorescamine, a prefluorescent dye for amino groups. This imaging method permits the fluorescent image to be erased or restored by treatment with base or acid. Copolymers containing phthalimide carbamate groups were applied to a bicolor fluorescent imaging material through the consecutive reaction with fluorescamine and rhodamine. A various colored fluorescent micropattern (green, red or red-yellow) was observed through the changes of excitation wavelength by using a conforcal microscope.

  • Fluorescent-magnetic nanoclusters were synthesized for biomedical applications. The nanoclusters consisted of superparamagnetic core-nanoclusters, highly fluorescent shell of nanocrystals, and lipid A. Magnetic cores were used for both magnetic resonance imaging (MRI) and cell separation. Fluorescent shell was used for optical imaging. The lipid-A-loaded nanoclusters were up-taken by dendritic cells via phagocytosis, which successfully activated dendritic cells. The dendritic cells were migrated to lymph nodes and spleen of mice. The results showed that our novel nanoclusters can play a role as an efficient optical and magnetic imaging, a cell separating and a pathogen mimetic agent at the same time. Additionally, synthesis of wavelength conversion nanowires will be discussed, which may be used as an optical nanoprobe in biological studies.

A Rapid and Convenient Method for in Vivo Fluorescent Imaging of Protoscolices of Echinococcus multilocularis

  • Human and animal alveolar echinococcosis (AE) are important helminth infections endemic in wide areas of the Northern hemisphere. Monitoring Echinococcus multilocularis viability and spread using real-time fluorescent imaging in vivo provides a fast method to evaluate the load of parasite. Here, we generated a kind of fluorescent protoscolices in vivo imaging model and utilized this model to assess the activity against E. multilocularis protoscolices of metformin (Met). Results indicated that JC-1 tagged E. multilocularis can be reliably and confidently used to monitor protoscolices in vitro and in vivo. The availability of this transient in vivo fluorescent imaging of E. multilocularis protoscolices constitutes an important step toward the long term bio-imaging research of the AE-infected mouse models. In addition, this will be of great interest for further research on infection strategies and development of drugs and vaccines against E. multilocularis and other cestodes.

  • Intravital video microscopy permits the observation of microcirculatory blood flow. This often requires fluorescent probes to visualize structures and dynamic processes that cannot be observed with conventional bright-field microscopy. Conventional light microscopes do not allow for simultaneous bright-field and fluorescent imaging. Moreover, in conventional microscopes, only one type of fluorescent label can be observed. This study introduces multispectral intravital video microscopy, which combines bright-field and fluorescence microscopy in a standard light microscope. The technique enables simultaneous real-time observation of fluorescently-labeled structures in relation to their direct physical surroundings. The advancement provides context for the orientation, movement, and function of labeled structures in the microcirculation.

Synthesis of a Novel Near-Infrared Fluorescent Dye: Applications for Fluorescence Imaging in Living Cells and Animals

  • Fluorescence imaging is considered as one of the most powerful techniques for monitoring biomolecule activities in living systems. Near-infrared (NIR) light is advantageous for minimum photodamage, deep tissue penetration, and minimum background autofluorescence interference. Herein, we have developed a new NIR fluorescent dye, namely, RB-1, based on the Rhodamine B scaffold. RB-1 exhibits excellent photophysical properties including large absorption extinction coefficients, high fluorescence quantum yields, and high photostability. In particular, RB-1 displays both absorption and emission in the NIR region of the "biological window" (650-900 nm) for imaging in biological samples. RB-1 shows absorption maximum at 614 nm (500-725 nm) and emission maximum at 712 nm (650-825 nm) in ethanol, which is superior to those of traditional rhodamine B in the selected spectral region. Furthermore, applications of RB-1 for fluorescence imaging in living cells and small animals were investigated using confocal fluorescence microscopy and in vivo imaging system with a high signal-to-noise ratio (SNR = 10.1).

  • Monoamine oxidases catalyze the oxidative deamination of dietary amines and amine neurotransmitters, and assist in maintaining the homeostasis of the amine neurotransmitters in the brain. Dysfunctions of these enzymes can cause neurological and behavioral disorders including Parkinson's and Alzheimer's diseases. To understand their physiological roles, efficient assay methods for monoamine oxidases are essential. Reviewed in this Perspective are the recent progress in the development of fluorescent probes for monoamine oxidases and their applications to enzyme assays in cells and tissues. It is evident that still there is strong need for a fluorescent probe with desirable substrate selectivity and photophysical properties to challenge the much unsolved issues associated with the enzymes and the diseases.

형광 X선 CT에서 촬상 시간의 단축화 알고리즘 (Algorithm to Shorten Imaging Time in Fluorescent X-ray Computed Tomogrpahy)

  • 방사광을 이용한 형강 X선 CT 이미징의 고속화를 위한 촬상 시스템과 처리 알고리즘을 검토하였다. 본 논문에서 사용된 촬상시스템은 고계수율 영역에서 안정된 동작을 한 전자 시스템으로 불감시간이 약 6%로 감소되고 계측시간의 경우도 1 점 당 3초로 단축되었다. 또한 재구성 알고리즘의 효율화를 증명하였고, 메모리와 계산량을 약 1/100로 감소시켰다. 물리적 phantom으로 그 정량성을 확인하였고, 시험관내의 갑상선의 화상으로부터 요드 분포를 추정하였다. 이러한 결과는 생체내에서 형광 X선 CT 계측의 실현 가능성을 보여준 것이다.

피부병변의 정량적 평가를 위한 디지털 컬러 영상 시스템 (Digital Color Imaging Systems for Quantitative Evaluation of Skin Lesions)

  • In this paper, we introduce a digital cross-polarization and fluorescent color imaging system for quantitative evaluation of skin lesions. We describe the characterization of the imaging systems and the quantitative image analysis methods to show the feasibility for quantitative evaluation of skin lesions. The polarization color image was used to compute erythema and melanin index image which are useful for quantitative evaluation of pigmentation and vascular skin lesions, respectively. The fluorescent color image was used to quantitatively evaluate "sebum" and "vitiligo". In quantitative evaluation of various skin lesions, we confirmed the clinical efficacy of the imaging systems for dermatological applications. Finally, we sure that the imaging systems can be utilized as important assistant tools for the evaluation of skin lesions by providing reproducible quantitative result for widely distributed skin lesions.
