Change of fluorescence in ambers according to artificial aging (인공열화에 따른 호박(amber)의 형광특성 변화)

  • Ambers are composed of polymer molecules which contain aromatic moieties such as benzene, naphthalene, phenanthrene and anthracene. They emit fluorescence when irradiated with ultraviolet light, which was used for confirming an amber. The fluorescence of amber, however, tends to decrease as the surface of amber is weathered with light, heat, oxygen for a long time. In this study, the reliability of confirming amber with its fluorescence by measuring the changes of fluorescence after artificial aging. Aging factors were UV light (${\lambda}$=340 nm), oxygen with heat (100%, $90^{\circ}C$) and heat ($90^{\circ}C$) and aging time was for 5, 15, 30 and 60 days, respectively. In the excitation and emission spectra of amber, the intensity decreased and the maximal wavelength was shifted to longer wavelength with artificial aging time. Especially, there was a drastic decrease in the intensity of spectra to 1.7% of initial value after 60 days aging under oxygen with heat. Only in Colombian amber there showed an increase of fluorescence intensity for a certain aging time, which could be explained by the production of aromatic ring in the presence of light and heat. Conclusively, the fluorescence can be lessened by the natural weathering with light, heat and oxygen and it is not accurate to recognize amber just with UV irradiation method.

Cure Monitoring of Am Epoxy-Anhydride System by Means of Fluorescence Spectroscopy (형광분석기를 이용한 에폭시-산무수물계의 경화 모니터링)

  • In the present study the cure behavior of diglycidyl ether of bisphenol-A(DGEBA) using an anhydride-based hardener in the presence of N,N-dimethyl benzyl amine (BDMA) or 1-cyanoethyl-2-ethyl-4-methyl imidazole (2E4MZ-CN) as an accelerator has been monitored and interpreted from the viewpoint of photophysical properties by means of fluorescence spectroscopy. To do this, 1,3-bis-(1-pyrene)propane (BPP) was well incorporated in the epoxy resin system by mechanical blending. The BPP probe, which is very sensitive to conformational change of the molecule influenced by the surrounding medium, successfully formed intramolecular excimer fluorescence. It is susceptible to the micro-viscosity or local viscosity and molecular mobility according to the epoxy cure. The cure behavior was explained with monomer fluorescence intensity ($I_{M}$ ), excimer fluorescence intensity ($I_{E}$ ) and $I_{M}$ /$I_{E}$ ratio as a function of cure time, cure temperature and accelerator. The present work agreed with the previous report on the cure behavior of an epoxy-anhydride system studied using DSC or torsion pendulum method. This study also suggests that the use of fluorescence technique may provide information on cure behavior of a thermosetting resin in a low temperature region, which has not been well interpreted by other analytical methods.

  • Recently, as an aging society progresses, a lot of interest in health and diagnosis is increasing, As the field of various bio-imaging systems for guided surgery capable of accurate diagnosis has emerged as important, a Fluorescence imaging system capable of accurate measurement and real-time confirmation has emerged as an important field. Fluorescence images currently being used are mainly in the NIR-I band, but many studies are in progress in the NIR-II band in order to improve resolution and confirm fluorescence deeply and accurately. In this paper, the difference between NIR-I and NIR-II, optical characteristics, and SBR (signal to background ration) of a fluorescent imaging system, was investigated using the finite element (FEM) method. After confirming, it was confirmed that the SBR was 16.2 times higher in the NIR-II area than in the NIR-I by making the skin phantom and measuring the fluorescence. It is confirmed that the enhancement in SBR of the Fluorescence imaging system is more effective in the NIR-II region than in the NIR-I region and expected to be used in application fields such as guided surgery, bio-sensor and also device which can detect the defect of optical devices.

Research Method of Fatty Acids Transfer between Phospholipid Model Membranes (인지질 모델막에서의 지방산 이동에 관한 연구 방법)

  • Direct measurement of the kinetics of free fatty acid transfer between phospholipid model membrane is technically limited by the rapid nature of the transfer process. Separation of membrane-bound fatty acid by centrifugation has shown that although the equilibrium distribution of free fatty acid is determined by this method, fatty acid transfer occurs too rapidly for accurate kinetic measurements. Recently fluorescence resonance energy transfer(FRET) assay has been developed to examine transfer of fatty acids between membranes. Donor membranes which has fluorescent fatty acid, anthroyloxy fatty acid(AOFA), is mixed with acceptor membranes which has non-interchangeable fluorescent quencher, nitrobenzo-xadiazol(NBD), using stopped flow apparatus. As the fluorescent fatty acids transfer from donor membrane to acceptor membrane, fluorescence intensity would be decreased and the rate and degree of fatty acid transfer can be analyzed. Fatty acid transfer between micelles is more complicated because of bile salt. Therefore in experiments with micelles, fluorescence self quenching assay is used. At high concentrations, a fluorophore tends to quench its own fluorescence causing a reduction in fluorescence intensity. Donor micelles contained self quenching concentrations of fluorophore and acceptor micelles had no fluorophore. Upon mixing of donor and acceptor micelles, the rate of transfer of the fluorophore from the donor to the acceptor was measured by monitoring the release in self quenching when its concentration in donor decreased over time.

  • The purpose of this study was to develop a practical caries activity test using laser fluorescence. The subjects of study were 30 children of $7\sim10$ years old. Fluorescence from initial carious lesion of teeth illuminated by an argon laser(488nm) was observed through barrier alter. For evaluation of accuracy and propriety of this method fer caries activity test, teeth with initial caries lesion on buccal or labial surface of children was examined with visual inspection and laser fluorescence. Visual examination for the dDfFtT and the $Cariescreen^{(R)}$ test were also done. The results from the present study can be summarized as follows: 1. Laser fluorescence test could differentiate initial caries lesions more easily than visiual inspection. 3. There was highest correlation(= 0.73) between laser fluorescent test and $Cariescreen^{(R)}$ test and. And also apparent correlation(= 0.66) exists between laser fluorescent test and caries experience measured by dDfFtT.

Effects of Sulfite on Chl Fluorescence in Barley(Hordeum vulgare L.) Seedlings in Light and Dark

  • The effect of sulfite on barley seedlings was investigated through Chl content, the electron transport activity of the photosystem, and Chl fluorescence. Barley leaves were harvested every 12 hrs during greening periods, and were then treated with a sulfite solution in either light or dark conditions. In both cases, the Chl content decreased in comparison with the control at any greening period. After sulfite treatment in the light, the activity of PS I decreased slightly, yet that of PSII showed a decrease of about 15%. The values of Fv, qP and qE decreased, however, the value of ql increased compared with the control. In addition, the value of qE decreased in leaves greened more than 12 hrs compared with that of the control. This indicates that the photosynthetic complex involved in energy dependent fluorescence quenching is undeveloped in a 12 hrs greened leaf, accordingly, it was a hardly affected by sulfite. After sulfite treatment in the dark, the activities of PSII and PSI decreased slightly, there was a small change in the value of Fv, qP decreased, and qE and the ratio of qNP/q increased in comparison with the control. As a result, PSII and PSI were not inhibited, however, the redox of QA was inhibited, and the excited energy was lost through the nonphotochemical pathway. The effects of sulfite in light or dark conditions were not considerably different with the Chl fluorescence quenching analysis method. In both light and dark conditions, the value of qP significantly decreased with sulfite compared to that of the control. This implies that the redox of QA was inhibited by sulfite in both light and dark contions.

Development of Monoenergetic Photon Source in the Energe Range below 100 keV by the X-ray Fluorescence Method (형광 X 선을 이용한 100 KeV 이하의 에너지 영역에서의 단색 Photon 선원개발에 관한 연구)

  • The development of mono energetic photon sources using $K_{\alpha}$ fluorescence X-ray of pure material was carried out in the energy range below 100 keV. The monoenergetic photons are very useful in the calibration of the radiation measuring instruments and can be produced as the $K_{\alpha}$ fluorescence X-ray by irradiating the bremsstrahlung to the thin pure metal foils called ‘radiators’. In this experiment, several radiators such as $_{47}Ag,\;_{50}Sn,\;_{68}Er,\;_{70}Yb,\;and\;_{82}Pb$ provide the wide monoenergetic photon energy ranging from 20 keV to 80 keV. By the spectrometry with HpGe LEPS, spectral purity factors which measure the monochrometicity for the $K_{\alpha}$ fluorescence X-ray, were determined as $0.64{\sim}0.94$. Dosimetry for the purpose of the determination of the exposure rate with a 600cc thin window ionization chamber, which was calibrated by the standard free-air ionization chamber, was performed. Exposure rates ranging $8.3{\sim}232.5mR/h$ was obtained according to the $K_{\alpha}$ fluorescence X-ray energy for each radiator.

Identification of Spilled Oils in the Marine Environment by Fluorescence Fingerprints and Library Search System (해양유출유의 형광지문에 의한 식별연구)

  • Multi-spectral analyses of excitation and emission fluorescence was applied to spilled crude oils in characterization of their specific fluorescence patterns which is called oil fingerprints. In the present study, oil fingerprints of 33 crude, 4 fuel and 2 other oils were analyzed to establish data base library search system. Cluster analysis showed that crude oils could be classified into two large groups according to their fluorescence characteristics. In simulated experiments, all the spilled sources was identifiable by the present library search system. In the natural environment this system could identify the exact source of weathered crude oil slicks upton 10 days. The present study shows that the fluorescence fingerprinting method with the library search system is reliable and superior to toutine GC/HPLC analyses in identification of the source of weathered spilled oils in the marine environment.

  • Two-dimensional (2D) spectrofluorometer is often used to monitor various fermentation processes. The change in fluorescence intensities resulting from various combinations of excitation and emission wavelengths is investigated by using a spectra subtraction technique. But it has a limited capacity to classify the entire fluorescence spectra gathered during fermentations and to extract some useful information from the data. This study shows that the self-organizing map (SOM) is a useful and interpretative method for classification of the entire gamut of fluorescence spectral data and selection of some combinations of excitation and emission wavelengths, which have useful fluorometric information. Some results such as normalized weights and variances indicate that the SOM network is capable of interpreting the fermentation processes of S. cerevisiae and recombinant E. coli monitored by a 2D spectrofluorometer.

DEVELOPMENT OF HIGH SENSITIVE MODEL OF CARIES ACTIVITY TEST FOR EARLY DIAGNOSIS OF DENTAL CARIES (치아우식증의 조기진단을 위한 고감도 우식활성검사 모형개발)

  • The purpose of this study is to develop the system which convert the optical difference of teeth texture between intact enamel and incipient caries lesion into shade difference by laser fluorescence and to develop new and simple caries activity test using laser fluorescence. The experimental design of this study consists of three parts. In first part, a new method for the in vitro assessment of changes in initial enamel caries lesion of Bovine teeth using laser fluorescence is tested. In second part, in vivo assessment undertaken. Number of teeth which showed incipient carious lesion on buccal surface examined by laser fluorescence was compared with the caries activity test of $Cariescreen^{(R)}$ test and other oral environmental test of dDfFtT. In third part, new caries activity test measured by laser fluorescence was developed on the basis of above results and evaluated the sensitivity, specificity, and diagnostic power. Optical density measured by laser fluorescence was increased as increasing the depth of incipient carious lesion and showed high correlation$(\gamma=0.7015)$ with lesion depth. Optical density showed direct proportion to lesion depth. Linear equation was obtained between the optical density and the lesion depth by regression analysis. The result of caries activity test with laser fluorescence showed high correlation with those of $Cariescreen^{(R)}$ test and dDfFtT examination. Caries activity test with laser fluorescence showed 48% of sensitivity, 52% of specificity, and 45% of diagnostic power on the basis of dDfFtT examination, and also showed 48% of sensitivity, 51% of specificity, and 36% of diagnostic power on the basis of $Cariescreen^{(R)}$ test. In regard above result, caries activity test with laser fluorescence considered to be reliable for caries activity test compared with other oral environmental test. and it was also considered to be practical because it would be simple, inexpensive, and time saving method.
