Survey on the Green house Flower Soil Chemicophysical Properties and Amount of Fertilizers and Soil Amendment Applications (시설화훼(施設花卉) 재배지(栽培地) 토양(土壤)의 이화학성(理化學性)과 화학비료(化學肥料) 및 토양개량제(土壤改良制) 시용량(施用量) 조사(調査))

  • This study was conducted to obtain basic information for soil improvement in flower crop cultivating greenhouse soil through survey on the chemical and physical properties of greenhouse soils. Total of 85 Flowcultivating farms were surveyed and analysis was done on the soil characteristics, amounts of chemical fertilizer and soil amendmentuse. The result are as follows: In soil properties of flower cultivating greenhousees, silt clay loam was 51%and 68% of the surveyed soils had good drainage condition. Ground water table was over 90-120cm which was optimum range for flower cultivation. Flower cultivating farms had problem with accumulation of fertility. Nitrate nitrogen was accumulated in Gypsophila paniculate farms and available phosphorus, and exchangeable postassium were significantly higher in greenhouse soils about 2 times than in open field soil. Application amount of chemical feltilizers in greenhouses were nitrate 211,phosphorus 135, and potassium 132kg/ha, respectively. Amount of organic matter used in greenhouse were high in order of cattle manure> compost> organic fertilizer> poultry manure> swine manure and their application amounts were69, 103, 32, 20, and 43 MT/ha, respectively.

  • In Korea, Brassica species planted in summer remain in rosette and do not flower. Seed production of vernalized rape seedlings grown in summer was evaluated to establish method of generation shortening and genetically pure seed production without isolation of seed fields. When vernalized rape seedlings of different maturity groups were transplanted in summer, all plants bolted and flowered, and average days from sowing to bolting and flowering was 37 and 58. respectively. It took 85days for early maturity group, 88 days for medium maturity group, and 108 days for late maturity group to mature. Seed yield of vernalized plants grown in summer was 8.8 gr per plant and 53kg per l0a which was 23% of seed yield of rape planted in autumn. However, enough seeds were produced to test oil composition and cake and to plant for a next generation. If seedlings are raised, they can be transplanted in more than 1, 000 times of original seed field, indicating that genetically pure seed can be produced without caging seed fields with nets and isolation of seed fields since other Brassica species do not flower in summer.

Evaluation of Agronomic Characteristics and Fatty Acid Composition of Flax Germplasms (아마 유전자원의 농업적 특성 및 지방산 조성 평가)

  • In this study, agronomic characteristics and fatty acid composition of 121 flax (Linum usitatissimum L.) germplasms were analyzed. The flax germplasms had a fairly short growth period of 75-95 days. The plant height at maturity varied from 52 to 124 cm. The flower color varied from white to lilac and even purple. Early flowering flax germplasms had white flowers and were resistant to lodging. In early flowering flax germplasms, all agronomic characteristics, including growth period, plant height, branch number per plant, stem diameter, number of capsules per raceme, seed number per capsule, 1000-seed weight, and seed yield, were decreased. The average fatty acid composition of 121 flax germplasms comprised palmitic acid (5.3%), stearic acid (4.3%), oleic acid (29.7%), linoleic acid (13.1%) and linolenic acid (46.4%). The total content of unsaturated fatty acids ranged from 84~94% with an average of 88%. Among the fatty acids, linolenic acid (omega-3) content was the highest with a range of 36~54%. In five selected flax germplasms (Hollandia 1803, Red son, C & F Res Br 1767, Wiera, and Ireland 1657) with high linolenic acid (${\geq}51%$), total lipid and protein contents were 30~36% and 5.4~6.9%, respectively. Overall, flax can practically be adoptable into cropping systems in South Korea and utilized as a scenery crop, since flax has a short cultivation period with diverse flower colors.

Antifungal and Antioxidant Activities of Extracts from Leaves and Flowers of Camellia japonica L. (동백나무 잎과 꽃 추출물의 항미생물 활성 및 항산화 효과)

  • This research was performed to investigate the possibilities of industrial usage of camellia (Camellia japonica L.) by examining the antioxidant and antimicrobial effects of methanol extract with different sections. Content of total phenolics, DPPH radical scavenging activities and antibacterial activity of young leaf, mature leaf, flower bud, flower, bark, and seed of camellia were compared in vitro experimental models. Total phenolics was contained the higher in young leaf (74.62 mg), flower bud (65.02 mg) and flower (62.42 mg) but less than 20.95 mg per 100 g of dry weight in other parts of Camellia japonica L. And effects of antioxidant measured by DPPH radical scavenger activity ($RC_{50}$, reduce concentration 50%), was shown higher $7.16{\sim}18.14\;{\mu}g/m{\ell}$ in methanol extract of young leaf, flower bud and flower than $61.23\;{\mu}g/m{\ell}$ of BHT as a chemical oxidant. Also, the antimicrobial activity of Camellia japonica L. extracts determined using a paper disc method against food-borne pathogen and food spoilage bacteria, the young leaves extracts showed the most active antimicrobial activity against 7 kinds of harmful microorganisms. Flower bud extracts showed the highest antibacterial activity against P. aeruginosa and Enterobacter spp. C1036. In addition, the minimum inhibitory concentration (MIC) of young leaf extract against B. subtillis,S. fradiae,S. aureus,E. coli,P. aeruginosa, Enterobacter spp. C1036, and S. typhimurium were revealed 1 to 15 ${\mu}g/m{\ell}$. As a result, antimicrobial activity of camellia extracts was shown higher gram positive bacteria than gram negative bacteria.

Inhibitory Effects of Methanol Extracts from Korean Medicinal Plants against HIV-1 Protease Activity

  • Korean medicinal plants were screened for their inhibitory activity against HIV-1 protease. The inhibitory activity of protease was determined by incubating the extracts in reaction mixtures containing protease and substrate $His-Lys-Ala-Arg-Val-Leu-(p-NO_{2}-Phe)-Glu-Ala-Nle-Ser-NH_{2}$ to perform proteolytic cleavage reactions. In this study the twenty six extracts from medicinal plants were investigated. Of the extracts tested, the extracts from the stem of Morus alba. exhibited the strongest activity with inhibition of 81% at a concentration of $100{\mu}g/ml$. The extracts of the flower of Saxjfraga stolonifera, and stems of Euonymus japonica and Castanea crenata showed appreciable inhibitory activity (>50%) against HIV-1 protease at same concentration.

  • To obtain information on the status of vegetative and reproductive growth of Fritillariae rhizoma-bulb were sown in 5 levels-weight (1. 5 gram, 3gram. .5gram, 7gram, and 9gram). Results observed were as follows; A clear plus correlations were shown between the germination status, plant height and other agronomic characteristics and the weight of rhizoma-bulb. As the weight of rhizoma-bulb was increasing, also the number of flower and yields were increased.

Screening of Plant Extracts with Cholinesterase Inhibition Activity (콜린 에스테라제 저해효과 보유 식물 추출물 탐색)

  • This study was conducted to select candidates from plant resources for the purpose of improving or treating Alzheimer's disease, a type of dementia. One hundred and eighty-four plant extracts at a final concentration of $100{\mu}g/ml$ were screened to determine their capacity to inhibit acetylcholinesterase (AChE) by in vitro assay. From this AChE assay, seven plant extracts - including methanol ext. and water ext. of Phellaodendron amurense Rupr. (bark), methanol ext. of Nelumbo nucifera Gaertn (stamen/ovary), methanol ext. of Persicaria tinctoria H. GROSS (flower), methanol ext. of Coptis chinensis (rhizome), ethanol ext. of Cinnamomum cassia Blume(bark) and ethanol ext. of Carthamus tinctorius L. (fruit) - showed effective inhibition activity ranging from 18.7% to 63.1%. The selected extracts were testified their inhibition activities on AChE and BuChE (butyrylcholinesterase) at concentrations of 25, 50, 100, $200{\mu}g/ml$. In the AChE assay, five extracts including methanol ext. of Nelumbo nucifera Gaertn. (stamen/ovary), methanol ext. of Persicaria tinctoria H. GROSS (flower), methanol ext. of Coptis chinensis (rhizome), methanol ext. and water ext. of Phellaodendron amurense Rupr. (bark) showed inhibition activity of 15.0%~73.5%, 19.5%~63.5%, 81.6%~58.5%, 69.9%~80.5%, and 54.8%~78.3%, respectively, at concentrations of 25, 50, 100, $200{\mu}g/ml$. In the BuChE assay, the extracts of Nelumbo nucifera Gaertn. (stamen/ovary), Persicaria tinctoria H. GROSS (flower), and Coptis chinensis (rhizome) showed inhibitory capacities of 58.9~81.6%, 45.8%~72.4%, and 33.1%~55.4% at concentrations of 25, 50, 100, $200{\mu}g/ml$, respectively. In conclusion, it is suggested that Nelumbo nucifera Gaertn. (stamen/ovary), Persicaria tinctoria H. GROSS, Coptis chinensis (rhizome) and Phellaodendron amurense Rupr. (bark) could be selected as candidate materials for improving or treating Alzheimer's disease on the basis of further study.

A New Perilla Cultivar for Edible Seed 'Anyu' with Early Maturity and High Oil Content (성숙이 빠르고 기름함량이 높은 종실용 들깨 신품종 '안유')

  • 'Anyu', a perilla (Perilla frutescens (L.) Britton) cultivar for edible seed was developed by the Department of Functional Crop Science, National Institute of Crop Science, RDA in 2008. It was originated from the cross between a pedigree of YCPL1824 as a female and SF9225-3-1 as a male parent in 1998. 'Anyu' can be characterized by purple flower color and brown grain color. 'Anyu' showed semi-dwarft type with 120 cm height, and has lodging tolerance. Maturing date of 'Anyu' was September 24, which is 15 days faster than that of 'Saeyeopsil'. The early maturity of this new cultivar showed a great advantage to various planting system with other crops. This new cultivar has high oil content (45%) and high linolenic acid in the fatty acid composition. The yield potential of 'Anyu' was about 0.9 ton/1ha in the regional yield trial.

  • We investigated the structure of floral organs and possibility of seed-set to breed a variety in ginger Zingiber officinale Rosc. Floral bud was formed from collected domestic Seosan var, and foregin Thailand var, the number of florets per bud were 8 and 10 in Seosan and Thailand var, respectively, Flowering time ranged from 18 to 25 August irregularly at 4-5 pm. The flower has the long styled with fiber hairs on top of stigma and connected-two anthers. Pollens were mixed of circular and ellips shape and its extine was two layer structure. Callus formation from anther explants was effective with compact and embryogenic on N$_{6}$ medium supplemented 2 mg/l of NAA(NCM). Plant regeneration was on the MS medium with BA of 1-2 mg/l from 40 days old callus after transferred callus medium.m.

Effect of Genotype and Explant on Somatic Embryogenesis and Acclimatization of Acanthopanax senticosus (가시오갈피의 수집종과 배양조직에 따른 체세포배발생 및 재분화 식물체의 순화)

  • Callus induction and embryogenesis were studied in three different genotypes of Acanthopanax senticosus, to develop a protocol for somatic embryogenesis and acclimatization. Young leaf, stem, node, petiole, peduncle, flower and root explants were collected from 3-year old trees of A. senticosus accessions (Korea, Russia and Japan). Callus was obtained from all cultured explants but showed the higher rate of callus formation in flower cultured. For the three A. senticosus accessions, callus was well formd on MS media containing 2mg/ l of 2,4-D and 2mg/ l of TDZ, 4mg/ l of 2,4-D and 1mg/ l of TDZ than other treatments. For three A. senticosus accessions, when callus transferred to MS medium with 2,4-D, embryogenic cell formed. For A. senticosus accessions Korea, embryogenic cells were obtained on callus induced from petiole, stem, node and root explants, and induction rate was lower than 3%. 200mg of embryogenic callus was transferred to MS free liquid medium and somatic embryos of heart stage were obtained after 45days of culture. When somatic embryo of germination stage were transferred to solid medium, most of the embryos were regenerated into plantlets on 1/4 MS medium. Normal plants with both shoots and roots were transferred to greenhouse soil and were successfully acclimatized.
