Optical Detection of Red Blood Cell Aggregation under vibration (진동장에서의 적혈구 응집성을 측정하는 광학적 방법)

  • Aggregability of red blood cells (RBCs) was determined by a laser backscattering light analysis in a microfluidic channel. Available techniques for RBC aggregation often adopt a rotational Couette-flow using bob-and-cup system for disaggregating RBCs, which causes the system to be complex and expensive. A disposable microfluidic channel and vibration generating mechanism were used in the proposed new detection system for RBC aggregation. Prior to measurement, RBC aggregates in a blood sample were completely disaggregated by applying vibration-induced shear. With the present apparatus, the aggregation indexes of RBCs can be easily measured with small quantities of blood sample. The measurements with the present aggregometer were compared with those of LORCA and showed a strong correlation between them. The aggregability of the defibrinogenated blood RBCs is markedly lower than that of the normal RBCs. The noble feature of this design is the vibration-induced disaggregation mechanism, which enables to incorporate disposable element that holds the blood sample.

  • In this study, a cancer treatment by accumulating and aggregating ferromagnetic particles in newborn blood vessels was examined. It is necessary for this treatment to control dispersion-aggregation property of ferromagnetic particles. Ferromagnetic particles required in this method disperse at low magnetic field, aggregate at high magnetic field and maintain the aggregation even after removal of the magnetic field. In order to control the dispersion-aggregation property, the surface of magnetite particles was modified with higher fatty acids having different lengths. As a result, we succeeded to prepare propionic acid-modified magnetite particles that form irreversible aggregation by magnetic field. The model experiments simulating newborn blood vessels showed that these particles can block the flow by the magnetic field, and the blockage was maintained after removal of the magnetic field.

  • The modem large electric power system has made power system analysis much more complex and difficult. For effective analysis of the power system, model reduction and aggregation is required. In this paper, a new relation index to identify the coherency among the generators is presented and also a new aggregation method is presented to aggregate the coherent generators in the large scale power system while matching the power flow. In order to demonstrate the effects of this aggregation method, it is applied to the IEEE 39 bus test power system and to the 2006 summer power system of Korea Electric Power Corporation, and the simulation results are compared with those of the original system.

Effects of Sambutoxin on the Rabbit Platelet Aggregation (Sambutoxin이 토끼의 혈소판 응집에 미치는 영향)

  • Sambutoxin, a newly purified mycotoxin in Koea, caused hemorrhage in the stomach and intestine of rats. To elucidate the mechanism of hemorrhage, effects of sambutoxin on rabbit platelet aggregation were investigated. First of all, the effects of sambutoxin on the platelet aggregation response and ATP release from platelet by various appregating factors were investigated. And then the role of $Ca^{2+}$ on the platelet aggregation was investigated by flow cytometer. Finally, morphological effect of sambutoxin on platelet ultrastructure was examined by transmission electron microscope. Sambutoxin inhibited aggregation induced by ADP, collagen, thrombin, and arachidonic acid and decreased platelet activating factor-induced disaggregation time in a dose dependent manner. Sambutoxin also decreased thrombin and arachidonic acid-induced ATP release, but increased all factors induced $Ca^{2+}$ release. Sambutoxin showed severe ultrastructural changes of platelet such as appearance of disorganization debri of cellular organelle in intercellular space. Our results indicate that sambutoxin inhibitis rabbit platelet aggregation, and it may be party due to the decrease of ATP release. However, it is not clear whether the antiaggregating effect of sambutoxin is related to $Ca^{2+}$ increase.

Effects of Shiitake Mushroom on Anti-platelet Aggregation and Anti-thrombotic (표고버섯 추출물의 항혈소판 응집 및 항혈전 효과)

  • In in vitro study, the anti-platelet aggregation effect of Shiitake mushroom extract was examined by measuring the collagen induced platelet aggregation and the DPPH radical scavenging. In in vitro study, anti-thrombotic effect of Shiitake mushroom extract was examined using the carotid artery thrombosis rat model. Carotid artery thrombosis rat model was made by 35% $FeCl_3$ treatment. After that, we investigate thrombus weight and blood flow. In platelet aggregation test, the extract significantly inhibited platelet aggregation in a concentration dependent manner(p<.001). Also, extract increased DPPH radical scavenging activity in a concentration dependent manner. Extract significantly inhibited thrombus weight to compare with control group. And blood passage time were shorter in the Shiitake mushroom extract supplemented groups than in control group. These results provide experimental evidence that Shiitake mushroom can be used to prevent platelet aggregation and thrombosis, then could apply the clinical diseases such as cardiovascular disease, and so on.

Effects of Bangpoongsan on the Cardiovascular System in the Experimental Animals (방풍산(防風散)이 실험동물(實驗動物)의 심혈관계(心血管系)에 미치는 영향(影響))

  • The present experiments were designed to investigate the effects of BangPoongSan on the cardiovascular system in the experimental Animals. And thus the change of blood pressure, auricular blood flow, artery contraction, death rate, platelet aggregation repression, plasma coagulation factor activity, plasma antithrombin activity, whole blood viscosity and plasma viscosity were studied. The result were summarized as the followings: 1. BangPoongSan dropped the blood pressure in the spontaneous hypertensive rat. 2. The drug increased the auricular blood flow in rabbit. 3. The drug relaxed the artery contraction by pretreated norepinephrine in white rat. 4. The drug inhibited the death rate of mouse which was led to thromboembolism by serotonin and collagen. 5. The drug inhibited the platelet aggregation in rat. 6. The drug prolonged the prothrombin time and activated partial thromboplastin time on the test of plasma coagulation factor activity in rat, but was not valuable. 7. The drug presented the antithrombin activity in rat. 8. The drug reduced the whole blood viscosity and plasma viscosity in rat, but the latter was not valuable. According to the results, Bangpoongsan increased the blood flow and dropped the blood pressure by dilatation of blood vessel smooth muscle. And the drug presented the antithrombin acivity, inhibited the platelet aggregation and reduced blood viscosity. Therefore these effects are assumed to improve the cardiovascular circulation disorder and prevent thrombosis.

A Study on the Optimal Aggregation Interval for Travel Time Estimation on the Rural Arterial Interrupted Traffic flow (지방부 간선도로 단속류 통행시간 추정을 위한 적정 집락간격 결정에 관한 연구)

  • In this paper, we conduct the research about optimal aggregation interval of travel time data on interrupted traffic flow and verify the reliability of AVI collected data by using car plate matching method in RTMS for systematic collection and analysis of link travel time data on interrupted traffic flow rural arterial. We perform Kolmosorov-Smirnov test on AVT collected sample data and on entire population data, and conclude that the sample data does not represent pure random sampling and hence includes sample collection error. We suggest that additional review is necessary to investigate the effectiveness of AVI collected sample data as link representative data. We also develop statistical model by applying two estimation techniques namely point estimation and interval estimation for calculating optimal aggregation interval. We have implemented our model and determine that point estimate is preferable over interval estimate for exactly selecting and deciding optimal aggregation interval. Our final conclusion is that 5-minute aggregation interval is optimal to estimate travel time in RTMS, as is currently being used our investigation is based on AVI data collected from Yang-ji to Yong-in $42^{nd}$ National road.

Effect of aggregation on shear and elongational flow properties of acrylic thickeners

  • The effect of intermolecular aggregation induced by hydrophobic and electrostatic interactions on shear and elongational flow properties of aqueous acrylic thickener solutions is discussed. Complex shear modulus is determined at frequencies up to $10^4$ rad/s employing oscillatory squeeze flow. Extensional flow behavior is characterized using Capillary Break-up Extensional Rheometry. Aqueous solutions of poly(acrylic acid)(PAA)/poly(vinylpyrrolidone-co-vinylimidazole) (PVP-VI) mixtures exhibit unusual rheological properties described here for the first time. Zero-shear viscosity of the mixtures increases with decreasing pH and can exceed that of the pure polymers in solution by more than two orders of magnitude. This is attributed to the formation of complexes induced by electrostatic interactions in the pH range, where both polymers are oppositely charged. PAA/PVP-VI mixtures are compared to the commercial thickener Sterocoll FD (BASF SE), which is a statistical co-polymer including (meth) acrylic acid and ethylacrylate (EA) forming aggregates in solution due to "sticky" contacts among hydrophobic EA-sequences. PAA/PVP-VI complexes are less compact and more deformable than the hydrophobic Sterocoll FD aggregates. Solutions of PAA/PVP-VI exhibit a higher zero-shear viscosity even at lower molecular weight of the aggregates, but are strongly shear-thinning in contrast to the weakly shear-thinning solutions of Sterocoll FD. The higher ratio of characteristic relaxation times in shear and elongation determined for PAA/PVP-VI compared to Sterocoll FD solutions reflects, that the charge-induced complexes provide a much stronger resistance to extensional flow than the aggregates formed by hydrophobic interactions. This is most likely due to a break-up of the latter in extensional flow, while there is no evidence for a break-up of complexes for PAA/PVP-VI mixtures. These flexible aggregates are more suitable for the stabilization of thin filaments in extensional flows.

The Effect of Transverse Vibration on Red Blood Cell Aggregation and Blood Viscosity

  • The present study investigated the effect of transverse vibration on the hemorheological characteristics of blood using a newly designed pressure-scanning capillary viscometer. As vibration was applied, aggregated blood cells (rouleaux) were disaggregated. The range of vibration frequency and amplitude are $0{\sim}100\;Hz$ and $0{\sim}0.8\;mm$, respectively for a capillary diameter 0.84 mm. As vibration increased, blood viscosity initially increased and tended to decrease. In order to delineate the unexpected results, the present study proposed two counteracting mechanisms of vibration related with red blood cell (RBC) aggregation affecting hemo-rheological properties. One is the reduction of RBC aggregation due to vibration causing an increase of blood viscosity. The other is forced cell migration due to the transverse vibration, which in turn forms a cell-free layer near the tube wall and causes a decrease of flow resistance.

Impacts of Surface Roughness Integration Using Remote Sensing Data: Concentration of Flood Flow Variation (원격탐사자료를 활용한 지표면 조도계수 통합의 영향: 홍수유출 변화를 중심으로)

  • A physical-based aggregation method was suggested to estimate surface roughness, which adequately represents the spatial heterogeneity of vegetation factors, from land cover property obtained from the remote sensing data. For the sensitivity analysis of surface roughness, the peak flow, peak time, and total volume were simulated by the NWS-PC. Effects of surface roughness estimated by three different integration methods (predominant, arithmetic mean, and aggregation approach) on the conceptual rainfall-runoff model parameters was analyzed. In the preliminary sensitivity test to surface roughness, the peak time had 10% variation and total volume had 2% variation. The peak time increased with surface roughness. A physical-based aggregation method was better than the existing method in the Soyanggang Dam basin for the results of STDEV, RMSE, NSE, and PME, but difference between them were small. The parameters related on the total baseflow were changed significantly with change of the surface roughness.
