To investigate flowering related genes in winter-type oilseed rape (Brassica napus L. cv. Tammi), differentially expressed genes were isolated from leaves of the plant after low temperature treatment which is requirements for floral induction. As a result of suppression subtractive hybridization (SSH), 288 clones were randomly selected from SSH library. Using reverse Northern blot analysis, 150 of 288 clones were identified to be differentially expressed. Out of these 150 clones, 45 clones showed very high identities with the known genes. Four clones showed very high identities over 90% with metallothionein-like gene that is related to flowering-induced genes. Of these 4 clones, the cDNA clone, rfs-13, revealed high identity with meotallothionein-like protein in Arabidopsis thaliana (98%) and Brassica compestris (89%). Furthermore, gene expressed in immature flower stages was confirmed by Northern blot analysis.
To establish the system of In vitro plant regeneration, the floral bud and leaf explants of Sedum rotundifolium were cultured on the MS media supplemented with different concentration of 2,4-D, NAA, and BA. The callus induction was more effective in the floral explants than the leaf explants, and was the best on MS medium containing 1.0 or 2.0 mg/L 2,4-D and 1.0 mg/L BA. The highest numbers of shoots were regenerated when callus were cultured on MS medium containing 2.0 mg/L 2,4-D and 1.0 mg/L BA for 8 weeks. The normal root formation from shoot was effective on the MS medium containing IAA alone. The regenerated plantlets were transferred to the pot and acclimatized successfully.
Flowering time (or heading date) is controlled by intrinsic genetic programs in response to environmental cues, such as photoperiod and temperature. Rice, a facultative short-day (SD) plant, flowers early in SD and late in long-day (LD) conditions. Casein kinases (CKs) generally act as positive regulators in many signaling pathways in plants. In rice, Heading date 6 (Hd6) and Hd16 encode $CK2{\alpha}$ and CKI, respectively, and mainly function to delay flowering time. Additionally, the major LD-dependent floral repressors Hd2/Oryza sativa Pseudo-Response Regulator 37 (OsPRR37;hereafter PRR37) and Ghd7 also confer strong photoperiod sensitivity. In floral induction, Hd16 acts upstream of Ghd7 and CKI interacts with and phosphorylates Ghd7. In addition, Hd6 and Hd16 also act upstream of Hd2. However, whether CKI and $CK2{\alpha}$ directly regulate the function of PRR37 remains unclear. Here, we use in vitro pull-down and in vivo bimolecular fluorescence complementation assays to show that CKI and $CK2{\alpha}$ interact with PRR37. We further use in vitro kinase assays to show that CKI and $CK2{\alpha}$ phosphorylate different regions of PRR37. Our results indicate that direct posttranslational modification of PRR37 mediates the genetic interactions between these two protein kinases and PRR37. The significance of CK-mediated phosphorylation for PRR37 and Ghd7 function is discussed.
The influences of night break by costly artificial light sources were investigated on the photo-morphogenesis and growth of leafy perilla (Perilla ocymoides L.). The irradiation of red, blue, and three-colored light for night break significantly increased the stem length and stem diameter compared to dark. Three-colored light gave the highest fresh and dry weight of stem, followed by red and blue light. Floral induction was suppressed up to 100 days after the night break, by red and three-colored light, but the plants grown under the dark or treated with blue light showed 85% and 31% flowering rate, respectively. The time needed for floral induction after night break was 60 days in dark and 80 days in blue light. The number of leaf, leaf area, and fresh weight per plant were the highest in red and three-colored light night break, followed by blue light and dark. The photosynthetic rate observed 80 days after night break was the highest in red light, followed by blue and three-colored light. A low light compensation point of $20\;{\mu}mol\;m^{-2}{\cdot}s^{-1}$ was observed in three-colored light, while red and blue light tended to show higher measurements.
BACKGROUND: This experiment was conducted to investigate the effects of red light on inhibition of flowering and vegetative growth of perilla (Perilla Frutescens. L). METHODS AND RESULTS: To determine red light intensity for inhibiting floral induction of perilla 6h light plus daylength extension (17:00-23:00) with three different intensity of red lights 0.046, 0.114 and $0.177{\mu}mol/m^2/s$ were treated respectively, and control plants were grown under 11(06:00-17:00)/13(17:00-06:00)h light/dark environment. Red(660nm) and far-red(730nm) light were irradiated for night break treatment subsequently to investigate photoreversible flowering response of perilla 'Manchu'. The flowering was inhibited by night break with red light, but sequential far-red light induced floral induction of perilla. Perilla not flowered by red light intensity over $0.177{\mu}mol/m^2/s$. Red light of $0.2{\mu}mol/m^2/s$ was irradiated for 6 hours (20:00-02:00) with LEDs device in plastic house. Perilla not flowered and continued the vegetative growth by red light treatment and the plant length, number of leaves, fresh weight, and leaf area of perilla were increased by 3%, 7%, 21%, and 19%, respectively, compared to incandescent control. CONCLUSION: These results showed that red(660nm) light for daylength extension could be used to control flowering and to enhance production of perilla leaf.
The effects of vernalization and darkness on flower initiation were reviewed by using naked barley varieties. The dark effects were pronounced on flower induction even at 25$^{\circ}C$. There were no differences in flowering responses among varieties. The effects of vernalization and darkness combined with 25$^{\circ}C$were 100% and 60% respectively which showed a significant difference. When the alternation of darkness with light was taken as first and last the flowering rate was 92 percent. When it was intercalated by light treatments the flowering rate was 91 per cent. Thus the effects of vernalization and darkness seems to differ.

Seedlings and grafts of Betula pendula and Betula platyphylla var. japonica were grown under partially controlled environments in a greenhouse and in a plastic-greenhouse installed inside a laboratory. Plant growth conditions such as photoperiod, temperature, nutrient supply were partially controlled to enhance the vegetative and reproductive growth of the birch seedlings and grafts. By the treatments twenty and seventy one percents of the seedlings, respectively, for the Betula pendula and Betula platyphylla var. japonica developed visible floral organs between 250 to 508 days after seeding. By the same treatments eighty and fifty three percents of the grafts, respectively, for Betula pendula and Betula platyphylla var. japonica developed visible male catkins between 51 to 497 days and female catkins between 365 to 396 days after grafting. Breeding cycle of birch species can be reduced to a great extent by the induction of precocious flowering at early stages of seedling and graft development.
This study aimed to get the basic data on the breeding of good varieties in Hypericum patulum Thunberg. The optimum materials, concentration and soaking time were examined to identify the effective approach to induce tetraploid plant by colchicine treatment to cultivate the varieties. For the seed germination rate of seed by colchicine treatment, the higher colchicine concentration was and the longer soaking time was, the more the germination rate decreased. While individuals were germinated in 16 test groups except control group (no treatment group), all the plants were diploid and no tetraploid was induced. For the plant regeneration rate by colchicine treatment on the explant of Hypericum patulum Thunberg that was under in vitro culture, the higher the colchicine concentration increased, the ress the regeneration rate. While total 147 individuals were regenerated in all treatment, when the explant was soaking treatment in more than 0.05% for over 6 hours, tetraploid could be obtained. In the soaking treatment of 0.05% for over 6 hours, tetraploid could be obtained. In particular, for the soaking treatment in 0.05% for 12 hours, 8 tetraploids were induced, which was about 47.1% of the number of plant regenerated. In accordance with the observation on doubling of DNA contents in leaf in order to identify polyploidy, the peak DNA content of G1 phase was 94.5 for diploid and 192.5 for tetraploid. It confirmed doubling of DNA content. Furthermore, the number of chloroplasts per guard cell depending on polyploid was around 10 in diploid and 17 to 19 in tetraploid, which were around 1.7 to 1.9 times as much as diploid.
A cDNA clone for a salicylic acid-induced gene in Chinese cabbage (Brassica rapa subsp. pekinensis) was isolated and characterized. The cabbage gene encoding a protein of 392 amino acids contained a tandem array of two thiJ-like sequences. ThiJ is a thiamin biosynthesis enzyme that catalyzes the phosphorylation of hydroxymethylpyrimidine (HMP) to HMP monophosphate. Although the cabbage gene shows a similarity to bacterial thiJ genes, it also shares a similarity with the human DJ-1, a multifunctional protein that is involved in transcription regulation, male fertility, and parkinsonism. The cabbage thiJ-like gene is strongly induced by salicylic acid and a nonhost pathogen, Pseudomonas syringae pv. tomato, which elicits a hypersensitive response in Chinese cabbage. Treatment of the cabbage leaves with BTH, methyl jasmonate, or ethephon showed that the cabbage thiJ-like gene expression is also strongly induced by BTH, but not by methyl jasmonate or ethylene. This indicates that the cabbage gene is activated via a salicylic acid-dependent signaling pathway. Examination of the tissue-specific expression revealed that the induction of the cabbage gene expression by BTH occurs in the leaf, stem, and floral tissues but not in the root.
