Genetic Variability of Flammulina velutipes Monosporous Isolates (팽나무버섯(Flammulina velutipes) 단포자 분리주의 유전적 변이성)

  • The characteristics of monosporous isolates of winter mushroom [Flammulina velutipes (Curt. ex Fr.)] were investigated to obtain useful breeding materials. Within monokaryons from cultivars which have a white colored fruitbody showed narrow genetic variation, while domestic strains which have a brown colored fruitbody showed wide variation. The mating type of the white strains was A1A2B1B2 genotype, but that of the domestic brown strains were A3A4B3B4. In intra-crossing, the genetic stability of dikaryons mated by monokaryons from white strain was less than that of parents. While in brown strain, dikaryons with high yield and color variation were obtained.

Contents of Nucleic Acids(Nucleosides and Mono-Nucleotides) in Extracts of Pleurotus ostreatus, Agaricus bisporus and Flammulina velutipes (느타리버섯, 양송이버섯, 팽이버섯 추출물의 핵산 관련 물질 함량 분석)

  • Mushrooms(Pleurotus ostreatus, Agaricus bisporus and Flammulina velutipes) are popular food sources in Korea, and have been reported as therapeutic foods, useful for preventing various diseases. In this study we researched HPLC conditions for the determination of nucleic acids in extracts of the three type of mushrooms. The method for nucleic acids analysis of mushrooms was developed using HPLC with UV detection. To determine the nucleic acids, mushroom extracts were extracted in hot water at $90^{\circ}C$ by reflux extraction for 1 hr. Then, the extracts were hydrolyzed by enzymes RP-1G and 50000G. The HPLC conditions were simple, rapid, and sensitive, and were applicable for the analysis of 4 nucleosides(cytidine, uridine, guanosine and inosine) and 3 mono-nucleotides(5'-CMP, 5'-UMP, and 5'-IMP) in the mushrooms. The nucleic acids in the mushrooms were cytidine, guanosine, inosine, uridine, 5'-CMP, 5'-IMP, and 5'-UMP. The analysis results for total nucleic acids in the mushroom extracts(Pleurotus ostreatus, Agaricus bisporus, and Flammulina velutipes) indicated levels of 25.28, 27.75, and 19.87 mg/g, respectively. In conclusion, this method can be used successfully for qualitative and quantitative analysis of nucleic acids in Pleurotus ostreatus, Agaricus bisporus, and Flammulina velutipes.

Effect of Natural Oil on the Mycelial Growth of Flammulina velutipes (팽이버섯 균사체 생장에 미치는 Natural Oil의 영향)

  • Vegetable oils supplemented to the basal medicum stimulated mycelial growth of Flammulina velutipes. The mycelial yield was increased 3.5 folds by addition of 3% (v/v) ricebran oil. Maximum mycelial yield (18.2mg/ml) was obtained by addition of 3.0% ricebran oil with 1.0% $CaCl_2$ to the basal medium. There was no significant difference between the liquid and solid spawn in the yield of sporophores.

  • White and brown strains of Flammulina velutipes were inter-crossed. All $F_1$ showed light-brown fruitbody, suggesting that a gene for the brown fruitbody was incompletely dominant against the white one. And backcross experiment showed that more than two genes were involved in color determination. To isolate a molecular marker linked to fruitbody color, a set of primers was designed from a sequence of clones derived by a bulked segregant analysis. These markers showed a specific band which co-segregated with brown fruitbody forming strains.

  • The production of lac case by the funguson various media was studied. The characteristics of the enzyme were also studied regarding to the optimum pH, stability, Km value, and inactivation. The maximum activity of laccase reached the 40 days of incubation and the barley straw extract appeared to be a strong inducer for laccase. The enzyme showed stability at wide range of pH with optimum pH of 6.6. Temperature stability of the enzyme was high. Laccase was not inactivated by the organic solvents used for the precipitation. The enzyme, how­ever, was completely inactivated by trichloroacetic acid and sodium azide.

  • Flammulina velutipes was transformed efficiently by Agrobacterium-mediated transformation system. The transformation frequency was about 16% with the gill tissues of the fungal fruiting body. Southern hybridization and genetic analysis suggest that the introduced DNA was inserted onto different locations of the fungal genome, and inherited stably to the next generation via basidiospores. Transformation or gene tagging with Agrobacterium T-DNA based vector should be useful for wide ranges of genetic or molecular biological studies of the mushroom.

Effects of Addition of Cathamus tinctorius, Coptis japonica and Asarum sieboldii to Sawdust substrate on the Growth and Development of Winter Mushroom, Flammulina velutipes (홍화, 황련, 세신 배지첨가제가 팽이버섯 생육에 미치는 영향)

  • Cathamus tinctorius, Coptis japonica and Asarum sieboldii were tested as substrate for the production of Flammulina velutipes. Among the C. tinctoris , C. japonica and A. sieboldii , C. tinctoris was the best substrate for the production of fruitbody. The effects of addition of C. tinctoris to sawdust substrate resulted in the increased mycelial growth on inoculum culture, 3.1% in ratio of fully culture and shorted one day in culture period. C. tinctoris was decreased 6.1% in ratio of fully culture, 11.0% in ratio of fruitbody productive culm. The addition of C.tinctoris, C.japonica to sawdust substrate increased 134.6%, 114.1% on the yield of the mushroom fruitbody respectively . But A. sieboldii decreased the mycelial growth and pineheading ratio delayed the production of fruitbody.

Genome-Wide Comparison of Carbohydrate-Active Enzymes (CAZymes) Repertoire of Flammulina ononidis

  • Whole-genome sequencing of Flammulina ononidis, a wood-rotting basidiomycete, was performed to identify genes associated with carbohydrate-active enzymes (CAZymes). A total of 12,586 gene structures with an average length of 2009 bp were predicted by the AUGUSTUS tool from a total 35,524,258 bp length of de novo genome assembly (49.76% GC). Orthologous analysis with other fungal species revealed that 7051 groups contained at least one F. ononidis gene. In addition, 11,252 (89.5%) of 12,586 genes for F. ononidis proteins had orthologs among the Dikarya, and F. ononidis contained 8 species-specific genes, of which 5 genes were paralogous. CAZyme prediction revealed 524 CAZyme genes, including 228 for glycoside hydrolases, 21 for polysaccharide lyases, 87 for glycosyltransferases, 61 for carbohydrate esterases, 87 with auxiliary activities, and 40 for carbohydrate-binding modules in the F. ononidis genome. This genome information including CAZyme repertoire will be useful to understand lignocellulolytic machinery of this white rot fungus F. ononidis.

A Phylogenetic Relationship between Foreign and Korean Strains of Flammulina velutipes Identified by rDNA-ITS Sequence Analysis (Flammulina velutipe의 국내 균주와 외래 균주 간의 ITS region을 이용한 계통학적 유연관계 분석)

  • This study was carried out to investigate the genetic relationship of Flammulina velutipes with other species. The ribosomal DNA cluster containing 4 rRNA genes from F. velutipes 4154 were sequenced. The length of the rDNA cluster sequence was estimated at 7,403 bp long and consisted of 1,806 bp of SSU rDNA, 245 bp of ITS 1 region, 159 bp of 5.8S rDNA, 308 bp of ITS 2 region, 3,402 bp of LSU rDNA, 1,400 bp of IGS 1 region, and 83 bp of 5S rDNA. The F. velutipes 4154 genes were contained in the rDNA cluster of F. velutipes in the order of SSU rDNA - ITS 1 - 5.8S rDNA - ITS 2 - LSU rDNA - IGS 1 - 5S rDNA. The phylogenetic relationships of 20 strains of Tricholomataceae and Physalacriaceae were analyzed by conducting distance analysis using the Neighbor-joining (NJ) method. The 20 strains used in this study were divided into three groups and the strains of the genus Flammulina were related very closely to strains of Physalacria bambusae.

Applicability of Spent Mushroom Media as Horticultural Nursery Media (버섯재배 후 탈병배지의 원예용 상토재료 이용성 검토)

  • This study was carried out to investigate applicability of Spent Mushroom Media(SMM) as horticultural nursery media. After the mushroom has been harvested, the SMM contains a lot of organic material, different microorganism and high density of mushroom hypha. The pH, phosphate and exchangeable cation concentrations of SMM of Flammulina velutipes were higher than those of any other treatment. The CEC and $NH_4-N$ were the highest in SMM of bottle-cultivated oyster mushroom (Pleurotus ostreatus). Bacteria and fungi showed the highest density in SMM of Flammulina velutipes. Most dominant bacteria were Microbacterium sp., Rhodococcus sp. and Agrobacterium sp. in SMM of Flammulina velutipes and Bacillus sp., Pseudomonas sp., Curtobacterium sp. and Microbacterium sp. in that of Pleurotus eryngii. The SMM contained high density of mushroom hypha that inhibited germination of seed and growth of young seedlings. Therefore, composting process of the SMM is indispensible to decline of vitality of mushroom hypha. The SMM of Flammulina velutipes with 0~30% vermiculite showed high germination rate in red pepper and chinese cabbage seeds. SMM of Pleurotus eryngii with 20% vermiculite showed 100% germination rate in red pepper seeds, but chinese cabbage seeds nearly failed to germinate with 30% vermiculite. The growth of red pepper was increased according to increasing mixture ratio of vermiculite. Accordingly, we concluded that SMM of Flammulina velutipes contained 0~30% of vermiculite can be used to horticultural growth bed for red pepper.
