Changes in Microbial and Chemical Composition and Sensory Characteristics of Fermented Soybean Paste, Chungkukjang, by High Dose Gamma Irradiation $(10{\sim}120\;kGy)$ (고선량$(10{\sim}120\;kGy)$ 감마선 조사 청국장의 미생물 변화와 화학적 성분과 관능적 특성 변화)

  • Changes in the number of microorganisms, chemical composition, and sensory quality of chungkukjang by gamma-irradiation up to 120 kGy were investigated. The total viable cell counts in chungkukjang irradiated with 10 kGy decreased from $10^9\;CFU/g$ of the control to $10^5\;CFU/g$. Proximate chemical composition and pH of chungkukjang were not much affected by irradiation. Acidity of chungkukjang decreased by irradiation with over 20 kGy. Fatty acid contents of chungkukjang irradiated under 20 kGy were the same as those of the nonirradiated ones. Polyunsaturated fatty acids content, decreased at over 40 kGy. The color and odor of chungkukjang were not significantly affected by irradiation up to 20 kGy. Unpalatable odor (p<0.01), off-flavor (p<0.001), and fish-odor (p<0.001) increased, and brown intensity (p<0.05) and acceptability (p<0.001) decreased at over 20 kGy. Results indicate 20 kGy is sufficient to decrease the microbial count of chungkukjang and prevent decrease in the content of polyunsaturated fatty acids.

Development of a Food Frequency Questionnaire for the Korea National Health and Nutrition Examination Survey: Data from the Fourth Korea National Health and Nutrition Examination Survey (KNHANES IV) (국민건강영양조사 식품섭취빈도조사지 개발: 국민건강영양조사 제4기 (2007-2009) 자료 이용)

  • The objective of this study was to develop a semi-quantitative food frequency questionnaire (FFQ) for an adult population for use in the Korea National Health and Nutrition Examination Survey (KNHANES). The 24-hour recall data for 2,939 subjects aged 19 years and over from the fourth KNHANES first year (2007) were used to extract the items included in the questionnaire. The FFQ items were developed by selection of major dishes based on the cumulative contribution rate, between-person variability based on the cumulative R2 of energy and 14 nutrients (carbohydrate, protein, fat, crude fiber, calcium, phosphorous, iron, sodium, potassium, vitamin A, thiamin, riboflavin, niacin, and vitamin C) and the number of consumers of each dish. In addition, the FFQ items were revised with analysis of 24-hour recall data of the fourth KNHANES second and third year (2008, 2009). Finally, 112 items were included in the FFQ and grouped as follows: rice (5 items), noodles and dumplings (6), breads and rice cakes (8), soups and stews (12), soybeans, eggs, meat and fish (23), vegetables, seaweed and potatoes (27), milk and dairy products (4), fruits (13), beverages (5), snacks (6) and alcoholic beverages (3). The food items of FFQ accounted for an average of 87.0% of energy and 14 nutrient intakes and also accounted for 81.7% of the between-person variability. The frequency range of the FFQ items was classified into nine categories (never or seldom, once per month, 2-3 times per month, once per week, 2-4 times per week, 5-6 times per week, once per day, twice per day and three times per day) and the portion size was divided into three categories (small, medium and large). We expect that this developed dish-based FFQ could be used in assessment of longterm dietary intakes of Korean adults.

  • We examined the immune response in flounder, Paralichthys olivaceus, with immunization of formalin killed Edwardsiella tarda as an antigen. The ELISPOT-assay (enzyme-linked immunospot assay) was optimized technically and applied to count the number of total and specific antibody secreting cells (TASC and SASC) in lymphocytes of different lymphatic organs. Incubation of lymphocytes on 96 well plate for more than 2.5hrs came out enough time in ELISPOT-assay for counting the antibody secreting cells in the anterior kidney and spleen. However, too much of plate-coated antigen or rabbit anti-flounder immunoglobulin for SASC or TASC counting, respectively, was appeared to decrease the sensitivity of the assay system. Specificity of the system was also confirmed by the absence of TASC in lymphocytes treated with cycloheximide to prevent protein synthesis. The peak numbers of SASC appeared at wk 3 post immunization after that there was a sharp decrease and reached to almost zero at wk 7. In the spleen and kidney, the timing and numbers of SASC on peak response were concurrent without preferential organ distribution. The specific antibody level in the sera increased rapidly between wk 2 and 3 after immunization, i.e. like the specific cellular response found with ELISPOT-assay on that period, However, the remained high level of specific serum antibody from wk 5 after immunization until the end of experiment was clearly distinguishable from the kinetics of SASC response decreased sharply.

Distribution and Characterization of Vibrio vulnificus Isolated in Coastal Areas of Chungcheongnam-do Province (충청남도 해안지역의 비브리오 패혈증균 분포 및 분리주의 특성 분석)

  • Background: Vibrio vulnificus has been frequently detected in seawater, fish, and shellfish mainly in the coastal areas of Chungcheongnam-do Province. Objectives: This study was conducted to investigate the analyzed biochemical properties, genetic characteristics, and distribution of Vibrio vulnificus isolated from environmental sources in coastal areas of Chungcheongnam-do Province from 2019 to 2020. Methods: A total of 1,510 samples were obtained from six different sites in Chungcheongnam-do Province. Isolated strains from the samples were identified by a VITEK 2 system and MALDI-TOF. Antibiotic susceptibility testing for 85 isolates was done by microdilution minimum inhibitory concentration methods, and 11 isolates were analyzed for 16s rRNA sequences in multiple alignments. Results: Among the 1,510 samples taken during the investigation period, 306 strains were isolated and the detection rate of V. vulnificus was 20.3%. One hundred eighty-eight strains (24.6%) from seawater and 118 strains (15.8%) from mud flats were isolated. It was mainly detected in July (17.3%), August (36.5%), and September (28.8%), and the proportion was 82.0%. Based on the CLSI-recommended breakpoints, V. vulnificus isolates were all susceptible to amoxicillin/clavulanic acid, amikacin, gentamicin, ciprofloxacin and trimethoprim/sulfamethoxazole. However, nonsusceptible isolates to ampicillin, ampicillin/sulbactam, cefazolin, cefoxitin, imipenem, tetracycline and chloramphenicol were identified. In the analysis of the nucleotide sequences for 16s rRNA of V. vulnificus isolates, it was confirmed that mutations frequently occurred between nucleotide number 922 and 952, and 98.2% to 100% nucleotide identities between isolates was verified. Conclusions: The results of this study can be used as a basic data for infection control and prevention of Vibrio vulnificus infection by describing the distribution and characteristics of Vibrio vulnificus strains isolated in coastal areas of Chungcheongnam-do Province.

The Validation Study of Normality Distribution of Aquatic Toxicity Data for Statistical Analysis (수생태 독성자료의 정규성 분포 특성 확인을 통해 통계분석 시 분포 특성 적용에 대한 타당성 확인 연구)

  • Objectives: According to the central limit theorem, the samples in population might be considered to follow normal distribution if a large number of samples are available. Once we assume that toxicity dataset follow normal distribution, we can treat and process data statistically to calculate genus or species mean value with standard deviation. However, little is known and only limited studies are conducted to investigate whether toxicity dataset follows normal distribution or not. Therefore, the purpose of study is to evaluate the generally accepted normality hypothesis of aquatic toxicity dataset Methods: We selected the 8 chemicals, which consist of 4 organic and 4 inorganic chemical compounds considering data availability for the development of species sensitivity distribution. Toxicity data were collected at the US EPA ECOTOX Knowledgebase by simple search with target chemicals. Toxicity data were re-arranged to a proper format based on the endpoint and test duration, where we conducted normality test according to the Shapiro-Wilk test. Also we investigated the degree of normality by simple log transformation of toxicity data Results: Despite of the central limit theorem, only one large dataset (n>25) follow normal distribution out of 25 large dataset. By log transforming, more 7 large dataset show normality. As a result of normality test on small dataset (n<25), log transformation of toxicity value generally increases normality. Both organic and inorganic chemicals show normality growth for 26 species and 30 species, respectively. Those 56 species shows normality growth by log transformation in the taxonomic groups such as amphibian (1), crustacean (21), fish (22), insect (5), rotifer (2), and worm (5). In contrast, mollusca shows normality decrease at 1 species out of 23 that originally show normality. Conclusions: The normality of large toxicity dataset was not always satisfactory to the central limit theorem. Normality of those data could be improved through log transformation. Therefore, care should be taken when using toxicity data to induce, for example, mean value for risk assessment.

Changes in Habitats of Fish and Amphibian Due to Erosion Control Dam Constructed in a Mountain Stream, Gongju, Chungchoengnamdo (충청남도 공주시 소재 산지계류 내 시공된 사방댐에 의한 어류 및 양서류 서식의 변화)

  • The aim of this study was to analyze the factors that affect the habitat of fishes and amphibians in a mountain stream that is part of an experimental forest at Kongju National University, Gongju, Chungcheongnam, Korea, and examine the differences in the presence of fishes and amphibians in the stream before and after construction of an erosion control dam. The results showed that the factors that affect the presence of fishes are pH, electrical conductivity (EC), dissolved oxygen (DO), flow velocity, and step-pool number, and that the factors that affect the presence of amphibians are monthly rainfall, pH, EC, DO, and crown density. Of these factors, pH, EC, flow velocity, and monthly rainfall were significantly different before and after dam construction; however, the differences among the other three factors from dam construction, except EC, might not have been enough to affect the presence of fishes and amphibians. Our results suggest that the difference in the frequency of fishes and amphibians surveyed before and after dam construction in the upper and lower stream sections were not statistically significant. One exception to this was the presence of amphibians in the lower stream section during and after dam construction, which could have been the result of a large amount of sediment produced by excavation that led to high EC.

Egg Development and Early Life History of the Korean Endemic Sand Spine Loach, Cobitis nalbanti (Pisces: Cobitidae) (한국고유종 점줄종개 Cobitis nalbanti의 난발생 및 초기생활사)

  • Egg development and early life history of the Korean endemic sand spine loach, Cobitis nalbanti, were investigated in the present study. Adult fish were sampled using spoon nets at the Yeongsan River in Seongam-ri, Bukha-myeon, Jangseong-gun, Jeollanam-do, Korea, June 2011. Eggs were obtained after injecting Ovarprim into females. Eggs were then artificially fertilized using the dry method in the laboratory. Mature eggs were transparent and slightly adhesive with light yellowish coloring, measured $0.99{\pm}0.03mm$ ($mean{\pm}SD$) in diameter. And number of spawned eggs were $1,527{\pm}410$ per individual. Hatching (50%) of the embryo occurred 52 hours after fertilization a water temperature of $25^{\circ}C$, and the average newly hatched larvae size was about $4.2{\pm}0.22mm$ in total length. At fifth day after hatching, the larval total length reached $6.0{\pm}0.34mm$ on average and their yolk sac had been completely absorbed. At fifteen day after hatching, larva entered the juvenile stage and reached $10.8{\pm}0.45mm$ in total length. At 100th day after hatching, the formation of Gambetta's zone of four dotted line was complete and juveniles were similar in general appearance to adults, and they averaged $41.1{\pm}2.95mm$ in total length.

Partial or complete replacement of fishmeal with fermented soybean meal on growth performance, fecal composition, and meat quality in broilers

  • The current study was aimed to examine the effect of partial or complete replacement of fishmeal (FM) with fermented soybean meal (FSBM) on growth performance, fecal composition, and meat quality in broiler chickens. A total number of 240 one-day-old broiler chicks were randomly allotted into four dietary treatments with six replications and ten birds per one pen. Dietary treatments were followed as; 1) Diet incorporated with 4% FM without FSBM (Control), 2) Diet incorporated with 3% FM and 2% FSBM (FSBM2), 3) Diet incorporated with 2% FM and 3% FSBM (FSBM3) and 4) Diet incorporated with 4% FSBM without FM (FSBM4). Body weight and feed intake were recorded weekly for 35 days of the experimental period. Moreover, fecal samples were collected to evaluate moisture, ash, nitrogen, calcium and phosphorus content on day 21 post-hatch. On day 35, two birds were sacrificed from each pen to measure meat quality parameters and visceral organ weights. Results revealed that, no dietary treatment effect (p > 0.05) was observed either in both body weight or average daily gain of broilers within the entire experimental period while broilers fed FSBM2 increased (p < 0.05) average daily feed intake by 10.07% whereas FSBM4 improved (p < 0.05) feed efficiency ratio by 8.45% compared to birds fed other dietary treatments on day 7 post-hatch. Besides, birds fed FSBM3 obtained the improved (p < 0.05) feed conversion ratio over the birds fed control diet by 7.51% from hatch to day 35 post-hatch (1.60 vs. 1.73). Nevertheless, no difference (p > 0.05) was detected on visceral organ weight, proximate composition and physicochemical characteristics of meat while broilers offered FSBM4 obtained the lowest (p < 0.05) calcium and phosphorous in faces (2.27% and 1.21% respectively) over those offered control feed and other FSBM treatments. In conclusion, FSBM would be a better replacement for ousting FM partially or completely in broiler diet as it did not impair the growth performance and meat quality while reducing the calcium and phosphorous excretion in broilers for 35 days post-hatch.

Spawning Period Characteristics and Early Life History of the Eight Barbel Loach, Lefua costata (Pisces: Balitoridae) (쌀미꾸리(Lefua costata)의 산란기 특징 및 초기생활사)

  • This study was conducted to elucidate spawning period characteristics and early life development of eight barbel loach, Lefua costata (Balitoridae) at the Jusucheon stream, Okgye-myeon, Gangneung-si, Gangwon-do, Korea between January and December in 2018. The spawning period was estimated to be from May to August, considering the change in the gonad-somatic index, the appearance of young fry, and frequency distribution in egg diameter. It was a multi-spawning type. The gender ratio was 1:0.79 with 1,117 females and 879 males collected. The egg size was 0.24-0.93 mm, with mature and immature eggs found during the spawning period. The size of mature eggs was 0.71±0.02 mm, and the average number of fecundity was 1,786±818 (n=31). Observation of the egg development showed that the fertilized eggs were the sticky, gray, segregated, and demersal type with 0.76±0.03 mm in diameter. The hatching of the embryos began at about 34 hours (hatching rate 50%) after fertilization underwater temperature of 25℃. The average length of the newly hatched pre-larvae was 2.7±0.11 mm. The average length of pre-larvae at 4 days after hatching was 4.5±0.16 mm, and the yolk sac was completely absorbed and entered the post-larvae stage. At 20 days after hatching, the average length of post-larvae was 11.5±0.67 mm, and their fin rays were formed before they transitioned to the juvenile stage. At 100 days after hatching, the average length reached 49.8±2.60 mm, and the appearance and the lateral sideband patterns were similar to those of the adult fish.

A Study on the Characteristics of Aquatic Insect Fauna in Differently Disrupted Ponds Located in Gungdaeoreum, Jeju (제주도 궁대오름 일대 교란정도가 다른 연못의 수서곤충 분포 특성에 관한 연구)

  • This study was conducted to understand the distribution characteristics of aquatic insects in ecologically different ponds in terms of the disruption. We investigated the fauna of aquatic insects in three artificial ponds (pond 1, 2 and 3) and two natural ponds (pond 4 and 5) located within 1 km each other around Gungdaeoreum in Jeju Island, from March 2018 to June 2020. A total of 50 species belonging to 15 families were found in the surveyed ponds: total 850 individuals with 14 species in 4 families of the order Odonata, total 4,391 individuals with 14 species in 6 families of the order Hemiptera, and total 2,014 individuals with 22 species in 4 families of the order Coleoptera. In overall, total abundance and species numbers were relatively higher than those of artificial pond in natural ponds in which animal and plant ecosystems were well established. In the case of artificial ponds, the number of individuals and species recovered rapidly when reconstituted by introducing aquatic plants, etc. (Pond 1). The nymphs of Odonata were observed largely in ponds without natural enemies such as large fish, and where adults could freely access without interception by artificial structure. Phytophagous Corixidae of the order Hemiptera were abundant, and Haliplidae populations of the order Coleoptera were affected by the distribution of the plants. Accordingly, the major factors affecting aquatic insect abundance were identified as the presence of refuges such as the topography and aquatic plants and presence of predators. Species of the order, Odonata were vulnerable based on these factors. Our results can be useful as basic information for the restoration of wetlands and construction of artificial wetlands or for conservation of species diversity in the future.
