C-type lectins are crucial for pathogen recognition, innate immunity, and cell-cell interactions. In this study, a C-type lectin gene was cloned from the rock bream. The full-length RbCTL cDNA was 729 bp with a 429 bp ORF encoding a 164-residue protein. The deduced amino acid sequence of RbCTL had all of the conserved features crucial for its fundamental structure, including the four cysteine residues involved in sulfide bridge formation and potential $Ca^2+$/carbohydrate-binding sites. RbCTL contains a signal peptide one single carbohydrate recognition domain. It showed 29.4% similarity to the C-type lectin of rainbow trout. RbCTL mRNA was predominately expressed in gill and head-kidney tissue and expressed less in peripheral blood leukocytes, trunk-kidney, spleen, liver, intestine and muscle. Expression of RbCTL was differentially upregulated in rock bream stimulated with LPS, Con A/PMA and poly I:C.
Natural-killer-cell-enhancing factor (NKEF) belongs to the newly defined peroxiredoxin (Prx) family. It was originally isolated from human erythroid cells. The black rockfish NKEF cDNA was identified through the expressed sequence tag (EST) analysis of PBLs libraries. The full-length NKEF cDNA was 1433 bp long and contained an open reading frame (ORF) of 594 bp that encoded 198 amino-acid residues. The 5' UTR had a length of 39 bp, and the 3’UTR 800 bp. The deduced amino-acid sequence of the black rockfish had a density 93.4, 92.9, 87.8, 85.8, 84.8, 83.8, 80.3, 79.7, 77.2, and 75.2% that of the pufferfish, olive flounder, channel catfish, zebrafish, chicken, common carp, Myotis lucifugus, cattle, human PrxI, rat PrxI, human NKEF-A, and Xenopus tropicalis, respectively. The NKEF gene was expressed in all the tissues of the black rockfish. The RT-PCR indicated that the NKEF transcripts were predominantly in the spleen and gill, less dominantly in the PBLs, head kidney, trunk kidney, and liver, and least in the intestine and muscles. This is the first report on the existence of the NKEF-A gene in black rockfish.
White spot disease by Cryptocaryon irritans broke out among cultured river puffer (Takifugu obscurus) cultured in sea water in Oct. 2001 and May 2004. Many white spots were easily observed on the gills with the naked eyes. The parasites attached on the gills at the base of the gill lamellae. The trophonts penetrated the interlamellar epithelium. Epithelial cell proliferation in interlamellar spaces occurred. PAS- positive mucous cells were prominent in the proliferated epithelium.
The study was performed to investigate changes in hematological responses and antioxidative enzyme activities (superoxide dismutase, SOD; catalase, CAT) of Japanese eel Anguilla japonica following exposure to 0 (control), 2.33, 4.60, 6,64 and 8.78 mM nitrite-N in fresh water for 48 h. Hematological parameters such as plasma nitrite, electrolytes, cortisol, glucose, glutamate oxaloacetate transaminase (GOT), glutamate pyruvate transaminase (GPT), hemoglobin (Hb), methemoglobin (metHb) and NADH-methemoglobin reductase (NMR) were measured. Plasma nitrite, cortisol, metHb and NMR increased directly with increasing ambient nitrite concentration, while Hb content showed a progressive decline. Levels of plasma potassium, GOT and GPT of the eel exposed to 6.64 mM ambient nitrite were significantly higher than the control fish. The activity of SOD and CAT in plasma, gill and liver of the eel following exposure to nitrite were augmented by increasing ambient nitrite. Levels of plasma nitrite, metHb, NMR, cortisol, glucose and antioxidative enzyme activities of the eel exposed to 2.33 mM ambient nitrite were significantly higher than the control fish. This study suggested that the eel acutely exposed to elevated ambient nitrite causes nitrite-induced stress responses, changes in antioxidative enzyme activities and hematological parameters.

This study aimed to examine the role of two aquaporin isoforms (AQP3 and AQP8) in response to the hyperosmotic challenge of transitioning from freshwater (FW) to seawater (SW) during parr and smoltification (smolt) using the rainbow trout, Oncorhynchus mykiss. We examined the changes in the expression of AQPs mRNAs in the gills and intestine of the parr and smolt stages of rainbow trout transferred from FW to SW using quantitative real-time PCR in an osmotically changing environment [FW, SW, and recombinant AQP3 (rAQP3) injection at two dosage rates]. Correspondingly, AQPs were greater during smoltification than during parr stages in the rainbow trout. Plasma osmolality and gill $Na^+/K^+$-ATPase activity increased when the fish were exposed to SW, but these parameters decreased when the fish were exposed to SW following treatment with rAQP3 during the transition to seawater. Our results suggest that AQPs play an important role in water absorbing mechanisms associated with multiple AQP isoforms in a hyperosmotic environment.
Monogeneans(Phylum Platyhelminthes) have been known as common parasites onto the skin and gills of cultured freshwater fishes. Infestation with these parasites result in a great loss in aquaculture industry. Some classification studies on these parasites have mostly been conducted in Japan but rarely in Korea. For the purpose of classifying monogenean parasites infesting two Korean freshwater fishes, carp(Cyprinus carpio) and loach(Misgurnus anguillicaudatus), samples captured from March 1998 to April 2000 were examined. Here we report that three different species of Monogeneans, Diplozoon nipponicum, Dactylogyrus extensus from carp and Gyrodactylus paralatus from loach, respectively. Of these the latter two parasites have never been identified in Korea.
In April and June 2014, mortalities of goldfish occurred at Korea. The principal signs included pale gills, severely enlarged and softened spleen and kidney and red liver. Moreover, the histopathological characteristics observed mainly in hematopoietic tissues of kidney, gill lamellae and intestine. The predominant histopathological changes were severe necrosis of hematopoietic tissue. In addition, nucleus exhibiting peripherally displaced chromatin were particularly abundant in the kidney of affected fish. The histological examination led to hypothesize the implication of a virus in the mortality. The hematopoietic necrosis herpesvirus (Cyprinid herpesvirus 2, CyHV-2) DNA polymerase gene successfully detected in DNA extracted from kidney and spleen of affected fish using PCR assay and showed 100% identity with already deposited CyHV-2 DNA polymerase gene in NCBI. Artificial infection trials using affected tissues filtrates gave cumulative mortalities of 30% for virus injected goldfish. In the present study, CyHV-2 was detected and identified as the causative pathogen of the epizootic in goldfish in Korea.

White spot syndrome virus (WSSV) is a prevalent and virulent pathogen affecting cultured whiteleg shrimp (Litopenaeus vannamei) in Korea. In this study, seven monoclonal antibodies (mAbs) (10A12, 16C3, 17G4, 21G5, 22C4, 23B6 and 24G6) were produced by using purified WSSV. The reactivity of these mAbs was analysed by Western blot (WB), indirect immunofluorescence (IIF), and lateral flow immunochromatographic assay (LFIA). WB analysis demonstrated that three mAbs (17G4, 22C4, and 23B6) reacted specifically to VP28 with an approximate molecular weight of 24 kDa, mAb 16C3 reacted with approximately 17 kDa. IIF analysis demonstrated specific fluorescence signals on gill tissues of WSSV-infected shrimp, with five mAbs (10A12, 16C3, 22C4, 23B6, and 24G6), pleopods from WSSV-infected shrimp were used for LFIA, where, two mAbs (21G5 and 22C4) exhibited positive reaction. In conclusion, it can be inferred that the mAbs usage and specificity depends on the nature of assay used for diagnosis.

This study evaluated the risk of single and combined exposure to microplastics in zebrafish (Danio rerio) through biomarkers, such as survival rate, excretion rate, and histological alterations of organ systems. The experimental groups were the control(Cont.), single microplastics exposure group(MPs, 1.83%/fish total weight(g)), the copper group(Cu, 21.6 ㎍ L-1), and a group with combined exposure to MPs and copper (MPs*Cu). The experiment was conducted with individual exposure (7 days) for MP excretion rate analysis and group exposure (14 days) for biomarker analysis. The daily excretion rate of MPs tended to decrease in a time-dependent manner. The copper concentration in the body was not significantly different between single and combined copper exposure. The degeneration of mucous cells in the skin, capillary dilation of the gill lamella, increased intestinal mucous, hepatocyte hypertrophy, and the degeneration of glomeruli and renal tubules were observed in all exposure groups. These histological alterations showed the highest tendency in the MPs*Cu group. In this study, the changes in biomarkers were attributed to the single effect of copper or the combined effect of copper and MPs rather than being solely influenced by MPs.
This study was to evaluate the ecological stream health through the Multimetric Fish Assessment Index (MFAI) along with fish fauna analysis based on the tolerance and trophic guilds at Miho stream in 2008 and 2009. Also, we analysed total mercury concentration in fish tissues to examine heavy metal contamination. Total sampled fish were 40 species and 2,557 individuals and Zacco platypus was the most dominant with 35% relative abundance. It was sampled with 11.4% RA for Korean endemic species (10 species 291 individuals) less than average RA 39.3% for the Geum river watersheds. According to the tolarance guild analysis, tolerant species was more dominant with 58.9% RA (15 species, 1,507 individuals) than sensitive species with 6.6% RA. Trophic guild analysis also suggested that omnivores were more dominant (60.5% RA) than insectivores (31.5% RA). Riffle-benthic species was also sampled with 7.7% RA. Ecological stream health based on the MFAI were averaged 25.3 (n=3) with fair-poor condition in 2008 and also 26.3 (n=3) with fair condition in 2009, just slightly increased than 2008. Qualitative habitat evaluation index was averaged 134 (n=3) with fair condition but most of sites had sediment accumulation that reflected substrate degradations proceeding. From the result of total mercury accumulation in fish tissues, kidney and liver tissues showed the highest but the lowest for gill tissues. Overall mercury concentration were not exceed the national standards by Korean Federation of Drug and Administration (KFDA). Consequently, our result could correspond with the characteristics of Miho stream where point sources such industrial complexes and wastewater treatment plant widely distributed around the stream along the gradient of up and downstream.
