어병균 Vibrio anguillarum 생육 저해 인자를 생산하는 Bacillus amyloliquefaciens H41의 분리 (Isolation of Bacillus amyloliquefaciens H41 Producing Growth Inhibition Factor against Vibrio anguillarum)

  • 양식 산업의 문제점인 어병균을 방제하기 위한 생물 전구체의 개발을 목적으로 해산어의 내장 추출물에서 어병균인 Vibrio anguillarum의 생육을 저해하는 새로운 균주를 분리, 동정하여 Bacillus amyloliquefaciens H41으로 명명하고 생육특성 및 생육저해인자 생성 조건을 검토하였다. 분리 균의 생육 최적 조건은 기본배지로 1% peptone, 질소원으로 1.5% yeast extract, 탄소원으로 1% sucrose, 0.5% NaCl, 금속이온으로는 0.05% $MgSO_4{\cdot}7H_20$이었으며 pH는 7.0-8.0, 온도는 $28-35^{\circ}C$에서 20시간의 진탕배양이었다. V. anguillarum 생육 저해물질은 B. amyloliquefaciens H41의 배양 상등 액에서 볼 수 있었는데 paper-disk 법으로 투명환의 생성여부로 확인하였다. 분리 균에 의한 생육저해 물질 생산 최적 조건은 1% peptone, 1.5% yeast extract, 1% NaCl, 0.05% $MgSO_4{\cdot}7H_20$, 1%의 sucrose이었으며 최적 pH는 7.5, 최적온도는 $35^{\circ}C$이었다. 분리 균은 최적 배양조건하에서 저해물질을 배양 후 16시간부터 24시간대에 가장 높은 활성을 나타내었다. 저해 활성반응은 분리 균에서는 저해 활성을 나타내었으나 B. amyloliquefaciens KCTC1724 표준균주에는 저해활성을 나타나지 않는 대조를 보였다. 또한 분리 균에 의해 생산된 저해물질은 V. anguillarum 에는 생육 억제 효과를 나타내었으나 다른 인체 병원성 Vibrio에는 전혀 영향을 미치지 않는 것으로 나타나 균주 선택성이 특이적이었다.

영어후계자육성총론 (The Theory of Fisherman's Successor-Cultivation)

  • The fisherman has been playing the roles of forming the fisheries village and surviving the fisheries production. Therefore, we should understand the nature of the fisherman's successor-cultivation which has been performed in order to maintain equlibrium between the fisheries economy and the national economy. Thesedays, fisherman's successor-cultivation mainly has been undertaken by the government in order to facilitate forming the economic foundation of fisherman's successor who has intended to choose "fisheries" as his own occupation in the future. From the above-mentioned, we find the meaning of the undertaking as both securing the foundation of living permanently and having the worth-while life of a professional fisherman by the survival and growth of his own business. And then, choosing the "fisheries" as an occupation, the successor should be satisfied with the requisites of occupation: 1) maintaining one's living, 2) working out one's individuality, 3) fulfilling one's social role-through engaging in the fisheries. Therefore, in order to have these requisites satisfied, I intend to suggest the followings : Firstly to maintain one's living, the environment of fisheies village should be kept in good condition to facilitate the successor's economic performance. Secondly to work out one's individuality, the fisheries' educational system should be adjusted to improve fisherman's ability. 1 hope to institute "the course of regional fisheries management" in fisheries high school and junior vocational collage, and "the school of fisheries management" for the learning of advanced fisheries knowledge. Thirdly to fulfill one's role, the opportunity actively participating in decision-making of the village's affair should be prepared more sufficiently, and the present fishing-centered fisheries system should be reformed into the fish-farming-centered fisheries system. To do this, the long-perspective vision is necessary. These three suggestions are come out from the macro-view point. Meanwhile the fisherman as an individual to improve his business is from the micro-view point. That is, the public aid, financial and legal, should be prepared. The above-mentioned suggestions are necessary to equalize fisherman's social status with other professional's especially from the result of improving the successor's economic force. Therefore, the principle of cultivating the fisherman's successor should be the principle of social equilibrium. be the principle of social equilibrium.

충남 지역 고등학생의 식습관 및 급식 만족도에 관한 연구 (Research on the Dietary Habits and Satisfaction of School Meals of High School Students in Chungnam Area)

  • This study was conducted via survey of 300 enrolled male and female high school students in the Chungnam area. The survey focused on students' dietary habits, changes in dietary habits based on the school lunch menu, and satisfaction of school lunches, in order to identify new methods for changing dietary habits and improving satisfaction of school lunches. Regarding the general characteristics of those surveyed, most were 17 years old, and of the same gender. Ninety-nine percent of those surveyed lived in farming areas, and the majority of students' parents graduated from high school and are self-employed. Research on dietary habits according to gender found that male students ate more regularly than did female students, with a difference of (p<0.001) between the two genders. Regarding the degree of which students ate only what they want, differences of (p<0.05) were found between the genders. The types of snacks frequently consumed were found to be flour-based snacks for the male students, whereas the females mostly consumed sweet foods and bread snacks. Research on satisfaction of school lunches according to gender also showed a difference of (p<0.5). Specifically, 48.7% of male students responded 'average', whereas only 45.3% of female students responded in the same way. The reason for dissatisfaction of school lunches were quality and nutrition for the male students (30%) and taste for the female students (60.4%), with a significant difference according to gender (p<0.05). Regarding the types of food most left over, both female and male students answered fish, bean curd, and soy products.

  • 발파에 의한 소음 진동의 물리적 크기 이외에 영향을 미치는 대상의 응답조건, 주변 환경에 적응상태 및 주관적 감정, 정신적 상태 등에 따라 크게 좌우되는 특성을 가지고 있어 명확히 피해영향을 판단하는 것은 매우 어려운 일이다. 특히 어류의 피해영향에 대해서는 실험을 통한 연구자체도 어렵고, 적용할 수 있는 제시기준이 모호하여 정확한 평가방법은 물론 실제 소음 진동 노출정도의 예측에 많은 어려움이 있다. 국내의 경우 수중소음 피해인정 기준을 140 dB re $1{\mu}Pa$, 수중 배경음과의 차가 어류에 미치는 피해기준은 20 dB re $1{\mu}Pa$ 이상으로 규정하고 있으나, 이는 연속음에 대한 것으로서 충격음인 발파음에 대한 평가로서는 적합하지 않다. 이 연구에서는 시험발파를 통해 바위갯지렁이 양식장 주변에서 발파 시 진동속도와 수중소음의 관계를 예측해보았다. 그리고 수중소음 피해인정기준에서 배경소음과의 차에 대한 의견을 제시해 보고자 한다.

Effect of adult population density on egg production in the black soldier fly, Hermetia illucens (Diptera: Stratiomyidae)

  • The black soldier fly is economically important because its prepupae are used as feed for many animals, including fish and swine. In Korea, black-soldier-fly farms have attempted to increase annual breeding and mass egg production for use in animal feed, as well as the decomposition of organic waste. Such efforts require an understanding of optimal mating and oviposition techniques. Specifically, adult densities and cage size may both improve the efficiency of mass egg production. Our study used four sizes of nylon cages ($1.0{\times}1.0{\times}2.5m$, $1.5{\times}1.5{\times}2.5m$, $2.0{\times}2.0{\times}2.5m$, $2.5{\times}2.5{\times}2.5m$) and three density treatments (4 kg, 8 kg, and 10 kg of pupae) to investigate optimal habitat size and adult density. We found that cage size (independent of density) did not significantly influence female fecundity (number of egg clutches and egg weight), whereas higher densities increased egg number and weight regardless of cage size. Thus, we recommend manipulating adult density to enhance productivity in commercial black-soldier-fly farming. However, we also propose further detailed research to develop methods that account for seasonal changes and environmental conditions, as climatic variables (temperature, sunlight) likely influence female fecundity as well.

양식 생김의 양륙 작업 개선에 관한 연구 (A study on the improvement of loading and unloading work in laver aquaculture industry)

  • In this study, an automatic system for improving the working environment and increasing production efficiency of a laver aquaculture industry in Korea was developed by combining a hydraulic control system and a load cell in a current landing work of the laver. The improved gathering laver system allowed the automatic gathering process of the laver in the sea with the hydraulic control system connected to a cutting machine of the laver on the operating ship, which has been used for gathering the laver semi-automatically in a form of the traditional farming method. The transporting process of the laver from an operating ship to the land was improved as follows. A frame installed on the operating ship and the bag nets were designed and made to hold about 1,000 kg of the laver inside. The bag nets contain the laver on the improved operating ship were tied in knots and hooked on a crane using a load cell. The weight is measured immediately by lifting the bag nets through the load cell system. Weight information is communicated to the fishermen and successful bidders through the application. The advantages of the improved system can help fishermen to fish by improving their working environment and increasing production efficiency. The field survey to improve the landing operation of the laver aquaculture was conducted in Gangjin, Goheung, Shinan, Wando, Jindo, and Haenam in South Jeonnam Province. A total of 10 sites including Gunsan in Jeonbuk Province, Daebu Island in Ansan City, Jebu Island in Hwaseong City in Gyeonggi Province, and Seocheon in Chungnam Province were searched to collect data. Prototypes of the system were tested at the auction house of laver located in Goheung, where laver collection using hydraulic control and landing using road cell could be improved.

농업인 대상의 식생활 교육자료 개발 (Development of Educational Material on Nutrition for Farmers)

  • The farmers' nutritional condition can not be compared with other citizens according to the Korean National Health & Nutrition Examination Survey, and based on other literature reviews. These farmers have either less access to educational materials or have no idea and do not understand the importance of nutritional education. The purpose of this study is to develop an appropriate reading material on nutrition to educate the farmers. To do this, the first step was to gather sufficient available materials for the review of related literature. Second, a survey was conducted to assess the needs of farmers and extension workers for educational materials on nutrition. The results showed that the farmers preferred the following topics: Menu Planning or Menu for Health' (62.1%), Dietary Assessment (49.7%), Dietary Guideline for Farmers (35.7%), and Cooking Method and Recipe (32.6%). They also preferred the booklet type (62.7%). These materials basically contained literature review, lesson plan, and needs assessment. The booklets contained in detail: 1) Dietary assessment - nutritional assessment, and assessment of meal 2) Dietary guidelines for farmers - reduce salt intakes, eat calcium-rich foods, ideal body weight, reduce alcohol-drinking and smoking, tips on eating soybean, eat meat and fish, tips on eating fruits, eat shellfish and seaweeds, and tips on eating dietary fibers, and 3) Plan a meal - menu planning and sample menu by farming patterns. Finally, the text of the final material was improved by the graphic designer using Quark Express 3.3, Photoshop 6.0, Illustrator 10.0, and Painter 8.0. The booklet has 46 pages and divided into three chapters. This contains simple and affordable recipes and provides some helpful tips for dietary and changing lifestyles. This booklet is made available for farmers and other groups interested in nutrition education.

앙상블 학습을 이용한 적조 발생 예측의 성능향상 (Enhancing of Red Tide Blooms Prediction using Ensemble Train)

  • 적조란 유해조류의 일시적인 대 번식으로 바다를 적색으로 변화시키며 양식장의 어패류를 집단 폐사 시킬 뿐 아니라 연안환경 및 바다 생태계에 악영향을 미치는 자연 현상이다. 적조에 의한 양식어업의 피해는 매년 발생하고 있으며 매년 적조방제에 많은 비용을 소비하고 있다. 이 때문에 적조 발생을 미리 예측할 수 있으면 적조에 대한 피해 및 방재 비용을 최소화 시킬수 있다. 본 논문은 앙상블 학습은 이용한 적조발생 예측 방법을 제안한다. 제안방법은 앙상블 학습의 bagging과 boosting 방법을 이용하여서 적조를 예측의 성능을 향상시킨다. 실험결과 제안방법은 단일 분류기에 비하여서 더 좋은 적조 발생 예측 성능을 보였다.

  • The infection status of domestic farmed eels Anguilla japonica, Anguilla bicolor and Anguilla marmorata with Japanese eel endothelial cell-infecting virus (JEECV) and anguillid herpesvirus 1 (AnHV) was examined at the major eel farming areas in Korea. These viruses were detected in all areas examined, regardless of the eel species or age. Any farm with a history of viral infection in adult fish confirmed the infection to be transmitted to stocked fry within 3 to 5 months. It is proposed that both viruses are horizontally transmitted within a given farm. The primary symptoms and histopathological lesions produced by the two viral infections are similar, making it difficult to distinguish the two diseases through clinical symptoms. Both viruses displayed 100% detection in the gills, suggesting that the gills are an optimal tissue for JEECV and AnHV monitoring. This study concluded that JEECV and AnHV were prevalent on eel farms across the country and caused very high mortality when the two viruses co-infected fry. Additional studies, including experimental infections, are needed to clearly understand the pathogenicity of each virus and the risk of co-infection.

Oxytetracycline의 투여방법에 따른 넙치(Paralichthys olivaceus) 체내의 잔류 특성 (Effect of Oxytetracycline Injection an the Body of Paralichthys Olivaceus)

  • 어류 질병은 매년 대규모 해산물 양식 및 소비량에 따라 증가하고 있으며, 이를 예방 및 치료하기 위해 항생제인 OTC (Oxytetracycline)를 사용하고 있다. 하지만 이는 부적절한 사용 및 오남용을 하게 되면 인체 내 통증을 유발시킬 수 있다고 알려져 있다. 따라서 본 연구에서는 간이시험법 및 HPLC 분석과 함께 OTC의 약욕 처리 및 경구 투여에 따른 넙치의 피부, 간, 근육에 관해 연구하였다. 약욕처리구는 물 1톤당 25 g의 농도로 1시간 1회 실시 하였고, OTC의 투여 농도를 어체중 kg 당 62.5 mg을 사료에 혼합시켜 7일간 경구 투여하였다. 미생물학적간이 검사법에서 약욕 처리구는 피부 직후에서만 양성을 보였고 경구 투여구에서는 77일째 양성으로 조사되었다. 그리고 근육은 14일째까지 양성을 보였다. HPLC 분석에서는 약욕 처리구인 간에서는 13일째, 피부는 37일째 잔해가 남은 것을 확인할 수 있었고 근육에서는 발견하지 못했다. 경구 투여구인 피부에서는 높은 농도(1.07 mg/kg)로 나타났으며, 간에서는 소량(0.56 mg/kg)을 나타냈으며 42일째의 근육에서는 남아 있지 않았다. 종합적으로 어류 양식장에서 사용 되는 OTC의 휴약 기간을 40일로 준수하면 OTC는 거의 잔류하지 않고 우리 몸에 무해하나 영양물질의 중간대사 또는 저장, 해독 등의 중요한 역할을 하는 간과 어체의 몸의 표면을 보호하고 감각작용을 하는 역할을 하는 피부는 근육에 남아 있는 OTC의 잔류량보다 몇 배의 높은 수치를 나타냈다. 하지만 이는 이전 연구에 따르면 OTC의 잔류량은 조리 온도에 따라 감소될 것으로 추측 된다.
