Effects of Synchronization of Carbohydrate and Protein Supply in Total Mixed Ration with Korean Rice Wine Residue on Ruminal Fermentation, Nitrogen Metabolism and Microbial Protein Synthesis in Holstein Steers

  • Three Holstein steers in the growing phase, each with a ruminal cannula, were used to test the hypothesis that the synchronization of the hourly rate of carbohydrate and nitrogen (N) released in the rumen would increase the amount of retained nitrogen for growth and thus improve the efficiency of microbial protein synthesis (EMPS). In Experiment 1, in situ degradability coefficients of carbohydrate and N in feeds including Korean rice wine residue (RWR) were determined. In Experiment 2, three total mixed ration (TMR) diets having different rates of carbohydrate and N release in the rumen were formulated using the in situ degradability of the feeds. All diets were made to contain similar contents of crude protein (CP) and neutral detergent fiber (NDF) but varied in their hourly pattern of nutrient release. The synchrony index of the three TMRs was 0.51 (LS), 0.77 (MS) and 0.95 (HS), respectively. The diets were fed at a restricted level (2% of the animal's body weight) in a $3{\times}3$ Latin-square design. Synchronizing the hourly supply of energy and N in the rumen did not significantly alter the digestibility of dry matter, organic matter, crude protein, NDF or acid detergent fiber (ADF) (p>0.05). The ruminal $NH_3$-N content of the LS group at three hours after feeding was significantly higher (p<0.05) than that of the other groups; however, the mean values of ruminal $NH_3$-N, pH and VFA concentration among the three groups were not significantly different (p>0.05). In addition, the purine derivative (PD) excretion in urine and microbial-N production (MN) among the three groups were not significantly different (p>0.05). In conclusion, synchronizing dietary energy and N supply to the rumen did not have a major effect on nutrient digestion or microbial protein synthesis (MPS) in Holstein steers.

Evaluation of Structural Carbohydrates Losses and Digestibility in Alfalfa and Orchardgrass during Ensiling 1

  • The evaluation of structural carbohydrate losses and its effect on silages digestibility in alfalfa (Medicago sativa L.) and orchardgrass (Dactylis glomerata L.) was studied during 5, 21 and 56 days ensiling. About 70 and 60 kg fresh matter of the two forages were ensiled in 9 silos of 120 L capacity. The digestion trials were conducted in two phases using the two grasses in two $4{\times}4$ Latin square design according to the four treatments being the grass and the three silages. There were no differences in the DM and CP contents resulting from 5 to 56 days ensiling in both forages. The water-soluble carbohydrates (WSC), hemicelllose, pectin, and energy were slightly reduced and appeared lower in 56 days silage. The ether extract and cellulose contents slightly increased as the ensiling process advanced in the two species. Hemicellulose losses of 29 and 41 g/kg DM were obtained in alfalfa and orchardgrass, respectively, 56 days after ensiling. While the cellulose losses in both species were very little, compared to that for hemicellulose, the pectin losses, 56 days after ensiling were 15 and 12 g/kg DM in alfalfa and orchardgras respectively. The total structural carbohydrates lost (ie., hemicellulose + cellulose + pectin) in g/kg DM of fresh material forage ensiled, is about four fifths the amount lost by WSC, in alfalfa and about two thirds, in orchardgrass, by 21 days ensiling after the activity of microorganism terminated, indicating that appreciable amount was used as substrate for silage fermentation. Ensiling alfalfa and orchardgrass for 0, 5, 21 and 56 days maintained a decreasing trend of 83.8, 82.5, 79.3 and 78.9% digestibility in alfalfa and 80.5, 77.0, 77.1 and 76.4% digestibility in orchardgrass. While the digestibility of cellulose and ether extract increased in silage in both species, the digestible energy values in silage were reduced from 2.6 to 2.3 and 2.9 to 2.7 Mcal/kg DM respectively in alfalfa and orchard during 5-56 days ensiling.

Nutrient Utilization of Broiler Litter and Bakery By-product Ration in Sheep (육계분-제과부산물 발효 완전혼합사료(TMR)의 면양 체내에서의 영양소 이용성 평가)

  • This study was conducted to determine the effect of feeding a total mixed ration(TMR) of broiler litter(BL) and bakery by-product(BB) with additional BL or rice straw incorporated at 10% of dietary DM as a roughage source on behavior pattern, nutrient intake, digestibility, digestible nutrient intake, ruminal and blood parameters, and N balance of sheep. All the treatment diets were formulated to be isoenergetic[total digestible nutrients(TDN) 66.9%]. Compared with the conventional formulated feed - rice straw feeding system(control), feeding TMR with BL(T1) or rice straw(T2) at 10% of dietary DM resulted in reduced eating, ruminating and total chewing time(P<0.05), similar DM intake, low(P<0.05) digestible DM, OM, fiber and total nutrients intake, low(P<0.05) nutrients digestibilities except EE, similar ruminal characteristics(pH, VFA concentrations and ratios, efficiency of carbohydrate fermentation, NH3-N), and favorable N digestion and retention. There were no differences in the above parameters between T1 and T2 with the exception of increased(P<0.05) eating, ruminating and total chewing time for T2. These results suggested that a TMR of BL and BB with or without rice straw may replace the conventional formulated feed and rice straw in ruminant diets successfully and furthermore feeding the TMR with rice straw made sheep behavior pattern more favorable.

A Study on Practical Applications of Environmental Education related to Food Waste Collected from Elementary School Foodservices(I) (초등학교 급식에서 배출되는 음식물쓰레기의 환경교육적 활용(I) - 음식물쓰레기 현황 및 환경교육적 활용 분석을 중심으로 -)

  • This study was conducted as a preliminary step to establish a model for the practical application of environmental education related to food wastes collected from elementary school foodservices. Methods of treatment and present recycling status of food wastes in elementary schools in Kyonggido were surveyed. The consciousness of dieticians who have been serving for elementary school foodservices was also analyzed through the survey questions concerning environmental education for the reduction and recycling of food wastes. The results derived from this survey were as follows: The major portion of food wastes from elementary school foodservices was constituted with vegetables and soup, and an average amount of food wastes per day was highest in June and July. Therefore environmental education related to reduction of wastes was needed in the first semester of school terms, and the possible reduction of food wastes achieved through the proper planning of school foodservice menu was also needed. In most cases food wastes were collected and treated by animal growing farmers or composted by machines in school. In operating composting machines microorganism inoculant was mostly used but dieticians pointed out the problem of a nasty odor, insects, and high energy consumption. This situation means it has not been operated efficiently and suggests an efficiency problem of machine composting in elementary schools because composting itself is based on the aerobic digestion and high temperature fermentation which kills insects and harmful microorganisms. Elementary school dieticians in Kyonggido were aware that food wastes cause main pollution problem, and that food wastes are valuable resources which can be recycled, and recycling of food wastes is inevitable. But more than half of the schools surveyed have not been reused food wastes in school, so a proper model for recycling and reuse of food wastes in school grounds was thought to be needed. Environmental education programs related to food wastes have not been peformed in more than half of the schools surveyed. It was concluded that the following three plans will be helpful to reduce school food wastes. First, environmental education should be enforced, second, teachers' and dieticians' intensive teaching concerning food wastes should be needed, and finally establishment of a model for recycling and reuse of food wastes in school grounds and its application to environmental education would offer a valuable field experience to school students.

A study on inspection methods for waste treatment facilities(I): Derivation of impact factor and mass·energy balance in waste treatment facilities (폐기물처리시설의 세부검사방법 마련연구(I): 공정별 주요인자 도출 및 물질·에너지수지 산정)

  • Despite the continuous installation and regular inspection of waste treatment facilities, complaints about excessive incineration and illegal dumping stench continue to occur at on-site treatment facilities. In addition, field surveys were conducted on the waste treatment facilities currently in operation (6 type) to understand the waste treatment process for each field, to grasp the main operating factors applied to the inspection. In addition, we calculated the material·energy balance for each main process and confirmed the proper operation of the waste disposal facility. As a result of the site survey, in the case of heat treatment facilities such as incineration, cement kilns, and incineration heat recovery facilities, the main factors are maintenance of the temperature of the incinerator required for incineration and treatment of the generated air pollutants, and in the case of landfill facilities Retaining wall stability, closed landfill leachate and emission control emerged as major factors. In the case of sterilization and crushing facilities, the most important factor is whether or not sterilization is possible (apobacterium inspection).In the case of food distribution waste treatment facilities, retention time and odor control during fermentation (digestion, decomposed) are major factors. Calculation results of material balance and energy resin for each waste treatment facility In the case of incineration facilities, it was confirmed that the amount of flooring materials generated is about 14 % and the amount of scattering materials is about 3 % of the amount of waste input, and that the facility is being operated properly. In addition, among foodwaste facilities, in the case of an anaerobic digestion facility, the amount of biogas generated relative to the amount of inflow is about 17 %, and the biogas conversion efficiency is about 81 %, in the case of composting facility, about 11 % composting of the inflow waste was produced, and it was comfirmend that all were properly operated. As a result, in order to improve the inspection method for waste treatment facilities, it is necessary not only to accumulate quantitative standards for detailed inspection methods, but also to collect operational data for one year at the time of regular inspections of each facility, Grasping the flow and judging whether or not the treatment facility is properly operated. It is then determined that the operation and management efficiency of the treatment facility will increase.

Purification, and Biochemical and Biophysical Characterization of Cellobiohydrolase I from Trichoderma harzianum IOC 3844

  • Because of its elevated cellulolytic activity, the filamentous fungus Trichoderma harzianum has a considerable potential in biomass hydrolysis applications. Trichoderma harzianum cellobiohydrolase I (ThCBHI), an exoglucanase, is an important enzyme in the process of cellulose degradation. Here, we report an easy single-step ion-exchange chromatographic method for purification of ThCBHI and its initial biophysical and biochemical characterization. The ThCBHI produced by induction with microcrystalline cellulose under submerged fermentation was purified on DEAE-Sephadex A-50 media and its identity was confirmed by mass spectrometry. The ThCBHI biochemical characterization showed that the protein has a molecular mass of 66 kDa and pI of 5.23. As confirmed by smallangle X-ray scattering (SAXS), both full-length ThCBHI and its catalytic core domain (CCD) obtained by digestion with papain are monomeric in solution. Secondary structure analysis of ThCBHI by circular dichroism revealed ${\alpha}$- helices and ${\beta}$-strands contents in the 28% and 38% range, respectively. The intrinsic fluorescence emission maximum of 337 nm was accounted for as different degrees of exposure of ThCBHI tryptophan residues to water. Moreover, ThCBHI displayed maximum activity at pH 5.0 and temperature of $50^{\circ}C$ with specific activities against Avicel and p-nitrophenyl-${\beta}$-D-cellobioside of 1.25 U/mg and 1.53 U/mg, respectively.

Quality Characteristics of Korean Traditional Wine Using Seolgaengbyeo for Brewing Rice (양조전용 설갱벼로 제조한 전통주의 품질 특성)

  • This study was conducted to compare the quality characteristics of Korean traditional wine fermented from different rice cultivars. Two kinds of rice cultivars, Seolgaengbyeo and Chucheongbyeo, were used. The hardness values of Seolgaengbyeo and Chucheongbyeo were 2126.6 g/sec and 4056.1 g/sec. The alkali digestion value (ADV) and amylose contents of Seolgaengbyeo and Chucheongbyeo were 6.6 and 6.4, 19.8 and 19.3%, respectively. Amylogram results were not significantly different between Seolgaengbyeo and Chucheongbyeo using a rapid visco analyzer (RVA). After fermentation for 7 days, the alcohol contents of the fermented wines ranged from 17.34 to 17.53%. The pH, total acidity, and glucose contents of the wines fermented with Seolgaengbyeo and Chucheongbyeo were 3.63 and 3.64, 0.17 and 0.17 mg/mL, 3.22 and 3.65 mg/mL, respectively. Finally, sensory evaluation of wine fermented with Seolgaengbyeo showed better taste than that of wine made with Chucheongbyeo.

Application of Statistical Analysis for Optimization of Organic Wastes Acidogenesis (유기성 폐기물의 산발효 최적화를 위한 통계학적 분석 방안의 적용)

  • This study shows how to find out optimum co-substrate conditions and continuous operating parameters for maximum acidification of three different organic wastes - livestock wastewater, sewage sludge and food waste. Design of experiments and statistical analysis were revealed as appropriate optimization schemes in this study. Analyses of data obtained from batch tests demonstrated the optimum substrate mixing ratio, which was determined by maximum total volatile fatty acids(TVFA) increase and soluble chemical oxygen demand(SCOD) increase simultaneously. Suggested optimum mixing ratio of livestock wastewater, sewage sludge and food waste was 0.4 : 1.0 : 1.1 based on COD, respectively. Response surface methodology(RSM) contributed to find out optimum operating parameter - hydraulic retention time(HRT) and substrate concentration - for the semi-continuous acidogenic fermentation of mixed organic wastes. The optimum condition for maximum TVFA increase was 2 days of HRT and 29,237 mg COD/L. Empirical equations obtained through regression analysis could predict that TVFA increase would be 73%. To confirm the validity of the statistical experimental strategies, a confirmation experiment was conducted under the obtained optimum conditions, and relative error between theoretical and experimental results was within 4%. This result reflects that using statistical and RSM technique can be effectively used for the optimization of real waste treatment processes.

  • Effect of shapes (noodle, grain and brick types) and strains (Aspergillus oryzae and Aspergillus sojae) of Meju (microorganism inoculated soybean substrate for fermentation) on the quality of soy sauce was investigated. Generally, the highest protease activity was found in the noodle type-Meju inoculated Asp. sojae during Meju preparation and soy sauce brewing, however, the lowest value was noted in brick type. Meju inoculated Asp. oryzae. Similar tendency was found on the contents of total solid, total nitrogen, amino-nitrogen, nitrogen digestion yield and amino-nitrogen ratio during soy sauce brewing. No effect was shown on the reducing sugar content, alcohol formation, pH, buffer action and salt content according to different Meju types during soy sauce brewing. Organoleptic tests on the color, taste and flavor of soy sauce after 3 months brewing with various Meju scored in the order of noodle type-Asp. sojae soy sauce (best quality), noodle type-Asp, orzae soy sauce, brick type-Asp. sojae soy sauce and brick type-Asp. oryzae-soy sauce (worst quality).

  • Casein is considered to be the main source of protein in milk; therefore, many studies have been conducted to identify casein-derived bioactive peptides and their physiological effects. Casein is inactive within the parent protein but can be liberated by various proteases and enzymatic hydrolysis during microbial fermentation and gastrointestinal digestion. Once absorbed, casein exhibits different bioavailabilities in the body. Specifically, casein-derived peptides function as angiotensin converting enzyme (ACE) inhibitor in the cardiovascular system; thus, they are expected to reduce and prevent hypertension. Additionally, casein-derived peptides behave as opioid-like peptides in the nervous system, which impacts relaxation. These peptides are also expected to modulate various aspects of immune functions. Finally, caseinophosphopeptide (CPP) and glycomacropeptide (GMP) may exhibit a number of nutritional effects such as the absorption of calcium, iron or zinc. Many studies have been conducted to evaluate casein-derived peptides due to their multifunctional properties and the results of these studies have contributed to the development of a wide variety of functional dairy products. The purpose of this paper was to review the generation of bioactive peptides, their absorption and metabolism, and their specific bioactive effects.
