An extensive investigation was carried out to describe the kinetics of cell growth, substrate consumption, and product formation in the batch fermentation using starch as substrate. Evaluation of intrinsic kinetic parameters was carried out using a best-fit unstructured model. A nonlinear regression technique was applied for computational purpose. The Andrew's model showed a comparatively better $R^2$ value among all tested models. The values of specific growth rate (${\mu}_{max}$), saturation constant ($K_S$), inhibition constant ($K_I$), and $Y_{X/S}$ were found to be 0.109 $h^{-1}$, 11.1 g/l, 0.012 g/l, and 1.003, respectively. The Leudeking-Piret model was used to study the product formation kinetics and the process was found to be growth-associated. The growth-associated constant (${\alpha}$) for protease production was sensitive to substrate concentration. Its value was fairly constant up to a substrate concentration of 30.8 g/l, and then decreased.
Three male Gayal, two years of age and with a mean live weight of $203{\pm}26$ kg, and three adult Yunnan Yellow Cattle, with a mean live weight of $338{\pm}18$ kg were fed a ration of pelleted lucerne hay and used to collect rumen fluid for in vitro measurements of digestibilities and gas production from fermentation of a range of forages. The forages were: bamboo stems, bamboo twigs, bamboo leaves, rice straw, barley straw, annual ryegrass hay, smooth vetch hay and pelleted lucerne hay. There were significant (p<0.05) effects of the source of rumen fluid on in vitro dry matter digestibility (IVDMD) and gas production during fermentation of forage. For the roughage of lowest quality (bamboo stems and rice straw), gas production during fermentation was higher (p<0.05) in the presence of rumen fluid from Gayal than Yunnan Yellow Cattle. Differences for these parameters were found for the better quality roughages with gas production being enhanced in the presence of rumen fluid from Yunnan Yellow Cattle. Moreover, the IVDMD of investigated roughages was significantly higher (p<0.05) in Gayal than Yunnan Yellow Cattle. The results offer an explanation for the positive live weight gains recorded for Gayal foraging in their natural environment where the normal diet consists of poor quality roughages.

Four male crossbred beef steers about 2 years old were used in a 4$\{times}$4 Latin square design to investigate the effect of pelleted sugarcane tops on voluntary feed intake, rumen fermentation and digestibility of nutrients. Experimental treatments were; Control (dried-chopped sugarcane tops (DCST)); PS1 (Pelleted sugarcane tops at 1 cm of diameter); PS2 (Pelleted sugarcane tops at 2 cm of diameter) and PS3 (Pelleted sugarcane tops at 3 cm of diameter). Roughage intake and total dry matter intake were 1.59, 1.62, 1.61, 1.63% BW and 2.09, 2.12, 2.11 and 2.13% BW in control, PS1, PS2 and PS3 treatments, respectively (p<0.05). Digestibility of DM, OM and CP were similar in control and PS3 treatment but there was significant difference (p<0.05) between control and PS1, PS2 treatments. Digestibility of neutral detergent fiber (NDF) and acid detergent fiber (ADF) were 52.89, 50.01, 50.05 and 50.56% and 41.91, 39.96, 39.91 and 39.69% in control, PS1, PS2 and PS3, respectively (p<0.05). Total volatile fatty acids concentrations in rumen contents was 67.68, 65.93, 66.15 and 66.67 mM in control, PS1, PS2 and PS3, respectively (p<0.05). Even though, concentrations of acetate and butyrate (%) were significant different (p<0.05) but concentration of propionate (%) was not affected by treatments (p>0.05). Rumen pH, ammonia nitrogen and plasma urea nitrogen were significantly different (p<0.05) among treatments. From this experiment, it was found that dried-chopped sugarcane tops increased digestibility of nutrients whereas pelleted sugarcane tops increased feed intake in beef cattle. However, pelleted sugarcane tops at 3 cm of diameter did similar result in digestibility and rumen parameters with DCST. Therefore, it could be concluded that pelleting sugarcane top is an alternative way to improve the quality of sugarcane tops for use as ruminant roughage source.
A commercially important alkaline protease, produced by Bacillus sp. RRM1 isolated from the red seaweed Kappaphycus alvarezii (Doty) Doty ex Silva, was first recognized and characterized in the present study. Identification of the isolated bacterium was done using both biochemical characterization as well as 16S rRNA gene sequencing. The bacterial strain, Bacillus sp. RRM1, produced a high level of protease using easily available, inexpensive agricultural residues solid-state fermentation (SSF). Among them, wheat bran was found to be the best substrate. Influences of process parameters such as moistening agents, moisture level, temperature, inoculum concentration, and co-carbon and co-nitrogen sources on the fermentation were also evaluated. Under optimized conditions, maximum protease production (i.e., 2081 U/g) was obtained from wheat bran, which is about 2-fold greater than the initial conditions. The protease enzyme was stable over a temperature range of 30-$60^{\circ}C$ and pH 6-12, with maximum activity at $50^{\circ}C$ and pH 9.0. Whereas the metal ions $Na^+$, $Ca^{2+}$, and $K^+$ enhanced the activity of the enzyme, others such as $Hg^{2+}$, $Cu^{2+}$, $Fe^{2+}$, $Co^{2+}$, and $Zn^{2+}$ had rendered negative effects. The activity of the enzyme was inhibited by EDTA and enhanced by $Cu^{2+}$ ions, thus indicating the nature of the enzyme as a metalloprotease. The enzyme showed extreme stability and activity even in the presence of detergents, surfactants, and organic solvents. Moreover, the present findings opened new vistas in the utilization of wheat bran, a cheap, abundantly available, and effective waste as a substrate for SSF.

Eight crossbred (75% Holstein Friesian) cows in mid-lactation were randomly assigned to a switchback design with a 2x2 factorial arrangement to evaluate two nonstructural carbohydrate (NSC) sources (corn meal and cassava chips) with different rumen degradability and used at two levels of NSC (55 vs. 75%) with protein source (supplied by urea in the concentrate mix). The treatments were 1) Low degradable low level of corn (55%) 2) Low degradable high level of corn (75%) 3) High degradable low level of cassava (55%) and 4) High degradable high level of cassava (75%). The cows were offered the treatment concentrate at a ratio to milk yield at 1:2. Urea-treated rice straw was offered ad libitum as the roughage and supplement with 1 kg/hd/d cassava hay. The results revealed that total DM intake, BW and digestion coefficients of DM were not affected by either level or source of energy. Rumen fermentation parameters; NH3-N, blood urea nitrogen and milk urea nitrogen were unaffected by source of energy, but were dramatically increased by level of NSC. Rumen microorganism populations were not affected (p>0.05) by source of energy, but fungal zoospores were greater for cassava-based concentrate than corn-based concentrate. Milk production and milk composition were not affected significantly by diets containing either source or level of NSC, however concentrate than corn-based concentrate averaging (4.4 and 4.2, respectively). Likewise, income over feed, as estimated from 3.5% FCM, was higher on cassava-based concentrate than corn-based concentrate averaging (54.0 and 51.4 US$/mo, respectively). These results indicate that feeding diets containing either cassava-based diets and/or a higher of oncentrates up to 75% of DM with NPN (supplied by urea up to 4.5% of DM) can be used in dairy rations without altering rumen ecology or animal performance compared with corn-based concentrate.
Objective: This study was carried out to determine the effects of elevated dietary cation and anion difference (DCAD) on dry matter intake (DMI) and ruminal fermentation pattern in lactating dairy goats under tropical conditions. Methods: Ten dairy goats were divided into two groups of five animals each. The groups received diets at different DCAD levels, either a control diet (22.81 mEq/100 g dry matter [DM], DCAD-23) or a DCAD-39 diet (39.08 mEq/100 g DM, DCAD-39). After parturition, DMI and water intake were recorded daily. Ruminal fluid and urine were collected, and nutrient digestibility measurements were carried out at 8th weeks postpartum (PP-8). Blood samples were collected at PP-4 and PP-8 to measure plasma leptin. Results: Dry matter intake/body weight (DMI/BW) at PP-8 of the animals fed the DCAD-39 diet was significantly higher than those fed with DCAD-23 diet (p<0.05). Animals fed with DCAD-39 consumed more water than those fed DCAD-23 over 24 h, particularly at night (p<0.05). Ruminal pH, acetate concentration, and urinary allantoin excretion increased with the DCAD-39 diet, whereas ruminal butyrate concentration was lower with the DCAD-39 diet. On the other hand, other ruminal parameters, such as total volatile fatty acid concentration, propionate molar proportion and acetate/propionate average ratio, were not affected by increased DCAD supplementation. Apparent digestibility was improved by increased DCAD supplementation. Plasma leptin concentration was higher with DCAD supplementation. Conclusion: When feeding goats with DCAD-39 under tropical conditions, an increase in DMI was associated with improved apparent digestibility of nutrients, ruminal fermentation and microbial protein synthesis. An increase in plasma leptin concentration could not explain the effect of high DCAD on DMI.
lactic acid production from cellulose by simultaneous saccharification and fermentation(SSF) was studied. The SSF using cellulase enzyme Cytolase CL and Lactobacillus delbrueckii was strongly inhibited by the end product(lactic acid). An ion-exchange resin(RA-400) was used for in-situ product removal during SSF. The sorption capacity of the resin was 200mg/g-resin. The simple SSF and the extractive SSF resulted in lactic acid concentrations of 30.4g/L and 32.0g/L, respectively, at the initial substrate concentration of 50g/L. A model was developed to simulate the extractive SSF. The lactic acid conversion for the initial substrate of 100g/L was estimated to be improved from 60% to 09% by in-situ product removal. The experimentally determined kinectic parameters were pH dependent, and fitted as empirical expressions to establish their values at different pH's. Lactic acid productivity was predicted to be maximum at pH 4.5-5.0.
Changes in pH and acidity were investigated in kimchi containing different amounts of added onion and/or pear during fermentation at $10^{\circ}C$. As the fermentation period increase, the pH and sugar contents of the onion and/or pear kimchi were higher compared to the control kimchi. However, the acidity of the kimchi containing onion and/or pear was lower than that of the control. To enhance the acceptance of kimchi, the optimal conditions for the addition of onion and pear were evaluated with the central composite design, ANOVA, and response surface methodology. According to variations in the mixing ratio of onion and pear, smell, taste and overall acceptability differed significantly, but the appearance and texture were not different. The optimal levels of onion and pear, predicted on the basis of each corresponding sensory parameters for kimchi, were $4.55{\sim}5.00%$ and $0{\sim}0.30%$ for smell, 2.61% and 2.38% for taste and 2.78% and 2.57% for overall acceptability, respectively.
To investigate the effects of sago palm pith (SPP) substitution of corn in the diets on intake, digestibility, rumen fermentation characteristics, nitrogen balance and microbial N Supply, five ruminally fistulated Southern indigenous cattle (mean initial BW = 226${\pm}$5 kg) were randomly assigned to a 5${\times}$5 Latin Square Design to receive five diets, $T_1$ = concentrate with 0% SPP, $T_2$ = 25% SPP, $T_3$ = 50% SPP, $T_4$ = 75% SPP and $T_5$ = 100% SPP, of dietary dry matter, respectively. Plicatulum hay (PH) was offered ad libitum as the roughage. A metabolism trial lasted for 21 days during which liveweight changes and feed intakes were measured. Based on this experiment, there were no significant differences (p>0.05) among treatments groups regarding total DM intake (OMI, NDFI and ADFI) and digestion coefficients of nutrients (DM, OM, CP, NDF and ADF), while total DM intake (% BW) was significantly (p<0.05) higher as higher levels of SPP were incorporated into diets. Rumen parameters (ruminal temperature, pH, glucose, packed cell volume, volatile fatty acid and rumen microorganism populations) were similar among treatments (p>0.05), whereas $NH_3-N$, blood urea nitrogen and molar proportion of propionate concentrations were significantly (p<0.05) higher as higher levels of SPP were incorporated into diets. The amount of N absorption, N retention and microbial protein synthesis were similar among treatments. These results indicate that SPP can be included in diets for Southern indigenous cattle to supply up to 100% of supplemental corn when fed with PH without negative impact on animal performance and it was a good approach in exploiting the use of local feed resources for beef cattle production.

In a balanced incomplete block design, two dry Holstein cows were used to investigate the effect of $\beta$ 1-4 galactooligosaccharides (GOS) supplementation on nitrogen (N) utilization, rumen fermentation and microbial N supply in the rumen. During the experiment, cows were fed four diets: orchardgrass (Dactylis glomerata L.) silage (OS), OS with GOS supplementation (OSG), OS mixed with alfalfa (Medicago sativa L.) silage (MS) and MS with GOS supplementation (MSG). GOS was supplemented at 2% of dry matter intake. Diets were fed at maintenance level of protein and energy. Results showed that N digestion was affected by silage and interaction of silage and GOS supplementation. Cows fed OSG had the highest N digested (p<0.05) followed by MS, OS and MSG. Supplementation of GOS to OS or MS diets tended to improve N utilization through reducing the N losses on dairy cows. There was no effect of GOS supplementation on rumen fermentation parameters (i.e. pH, $NH_3$-N and total VFA) at 1 h and 6 h after feeding. Compared to cows fed MS, cows fed OS silage had higher (p<0.05) allantoin excretion (80.8 vs. 67.1 mmol/d) and higher (p<0.05) total purine derivatives excretion (92.9 vs. 78.5 mmol/d). The microbial N supply in cows fed OSG was higher (p<0.05) than those fed OS, MS and MSG.
