Characterization of Endoglucanase (F-I-III) Purified from Trichoderma sp. C-4 (새로운 섬유소분해 균주 Trichoderma sp. C-4에서 분리한 Endoglucanase (F-I-III)에 대한 연구)

  • One of the endoglucanases, F-I-III, was purified from the culture filtrate of T. sp. C-4 through procedures including chromatography on Sephacryl S-200, DEAE-Sepharose A-50, and Chromatofocusing on Mono-P (FPLC). The molecular weight of the enzyme was determined to be about 56,000 Da by SDS-PAGE, and pI of 4.9 by analytical isoelectric focusing. F-I-III showed the highest enzyme activity at $55^{\circ}C$, and the pH optimum of the enzyme was 5.0. There was no loss of activity when the enzyme was incubated at $50^{\circ}C$ for 24 hours. The specific activity of the enzyme F-I-III toward the CMC was 315.4 U/mg. The Km value for $PNPG_2$ of F-I-III was 2.69 mM. N-terminal sequence of F-I-III was analyzed to be QPGTSTPEVHPKKLTTYK. It showed $95\%$ of homology to that of EGI from T. reesei. The presence of some metal ions (1 mM) had only a little effect on CMCase activity. The treatment of the reducing agents resulted in the increase of endoglucanase activity.

Effects of occlusal load on the stress distribution of four cavity configurations of noncarious cervical lesions: A three-dimensional finite element analysis study (네 가지 형태의 비우식성 치경부 병소의 3차원 유한요소법적 응력분석)

  • The objective of this study was to investigate the effect of excessive occlusal loading on stress distribution on four type of cervical lesion, using a three dimensional finite element analysis (3D FEA). The extracted maxillary second premolar was scanned serially with Micro-CT. The 3D images were processed by 3D-DOCTOR. ANSYS was used to mesh and analyze 3D FE model. Four different lesion configurations representative of the various types observed clinically for teeth were studied. A static point load of 500N was applied to the buccal and lingual cusp (Load A and B). The principal stresses in lesion apex, and vertical sectioned margin of cervical wall were analyzed. The results were as follows 1. The patterns of stress distribution were similar but the magnitude was different in four types of lesion 2. The peak stress was observed at mesial corner and also stresses concentrated at lesion apex. 3. The compressive stress under load A and the tensile stress under load B were dominant stress. 4. Under the load, lesion can be increased and harmful to tooth structure unless restored.

  • The objective of this study was to investigate the effects of various occlusal loads on the stress distribution of the buccal cervical region of a normal maxillary second premolar, using a three dimensional fnite element analysis (3D FEA). After 3D FE modeling of maxillary second premolar, a static load of 500N of three load cases was applied. Stress analysis was performed using ANSYS (Swanson Analysis Systems, Inc., Houston, USA). The maximum principal stresses and minimum principal stresses were sampled at thirteen nodal points in the buccal cervical enamel for each four horizontal planes, 1.0 mm above CEJ, 0.5 mm above CEJ, CEJ, 0.5 mm under CEJ. The results were as follows 1. The peak stress was seen at the cervical enamel surface of the mesiobuccal line angle area, asymmetrically. 2. The values of compressive stresses were within the range of the failure stress of enamel. But the values of tensile stresses exceeded the range of the failure stress of enamel. 3. The tensile stresses from the perpendicular load at the buccal incline of palatal cusp may be shown to be the primary etiological factors of the NCCLs.

The Surface Characteristic Changes of Hydroxyapatite Coated Ti Disc When Immersed in NaCl Solution (NaCl 수용액에 담근 Hydroxyapatite 코팅된 타이타늄 시편의 표면 변화)

  • In previous studies, methods for enhancing cellular response on the Hydroxyapatite coated implant surface were described. In this study, the changes of surface characteristics such as surface roughness, contact angle, surface energy and surface morphology were observed when Hydroxyapatite coated Ti discs were immersed in NaCl solution for various time. Hydroxyapatite coated Ti discs were immersed in 0.9% NaCl solution for 7, 14 and 21 days at $37^{\circ}C$. The control group comprises dry identical discs not immersed in a solution. (n=3) All discs were dried in air completely and the surface roughness was measured using confocal laser scanning microscopy(CLSM). Static contact angle was recorded by video contact angle analyzer after dropping distilled water on the surface. The surface energy was calculated from contact angles of the three liquids. Surface was observed using a field emission-scanning electron microscope(FE-SEM). As a result, the surface roughness of immersed Hydroxyapatite coated Ti discs increased significantly and the contact angle decreased comparing with control group discs. The surface energy of immersed discs increased except for discs immersed for 14 days.

Petrochemical Study on the Cretaceous Volcanic Rocks in Kyeongsang Basin, Korea: Possibility of Magma Heterogeneity (경상분지 백악기 화산암류에 대한 암석화학적 고찰: 이원성 마그마의 가능성)

  • The Creataceous volcanic rocks distributed in the southeastern part (Kyeongsang basin) of Korea peninsula are composed of basalt, basaltic andesite, andesite, dacite and rhyolite. The variation of major elements show that contents of MgO, CaO, $FeO^T$, $Al_2O_3$, $TiO_2$ and $P_2O_5$ decrease with increasing of $SiO_2$, but $K_2O$ contents are increased slightly, $Na_2O$ widely dispersed. We can show slightly inflection point and low frequency of dacites in range between 63-65 wt.% $SiO_2$, while continuous trend exit in variation diagram. Variation trends in Harker diagrams for the major, minor, trace and REEs suggest that the BAV (basaltic to andesitic volcanics) and DRV (dacitic to rhyolitic volcanics) are not related to a simple crystal fractionation process. In the regime of under 65 wt. % in silica content, fractionation of olivine and clinopyroxene is predominant, while that of plagioclase happens strongly higher than 65 wt.% (e.g., $SiO_2$, vs. Eu and Sr, MgO vs. $Al_2O_3$ and CaO). The latter means low-pressure fractional crystallization for DRV. On the discriminant diagram, DRV are located in more mature environment than BAV. The $(Ce/Sm)_N$ vs. CeN digram shows that these two classes cannot be related to crystal fractionation. If they had been produced by fractionation, although they plotted in a slightly elongate cluster along the same horizontal trend, DRV should lie to the right of these primitive compositions. These diagrams clearly rule out a simple fractionation throughout from BAV to DRV. BAV had been influenced greatly subductiong slab as shown by K/Yb vs. Ta/Yb. We suggest that BAV primitive magma generated higher degree of partial melting than DRV primitive magma. LILE (K, Ba, $Rb{\pm}Th$) enriched characteristics as shown in BAV are inherited from subducting slab fluids and/or higher degree of partial melting of mantle material. However, lower degree of partial melting of mantle relative to BA V and contamination at high-level magma reservoir caused LILE enrichment to DRV.

  • The physical and chemical properties of cover soils of 10 waste landfill sites in Kyonggi-Do area, where social circumstances at present forces to consider the reuse of landfill, were investigated to provide the informations of soil environment which are necessary to establish the appropriate ecological restoration plan of waste landfills. The pH and electrical conductivity of soils were higher in landfills sites than in reference sites (area around landfill sites), indicating the salt accumulation in surface soil. However, total-N and organic matter contents were lower in landfills sites than in reference sites. In landfill sites, the total-N and plant available-P contents were less than 0.15% and 20mg/kg, respectively. Exchangeable cations (K, Ca, Mg and Na) and heavy metal (Cd, Cr, Cu, Fe, Mn, Ni, Pb and Zn) contents varied between the landfill sites, but were higher in landfills sites than in reference sites. The major exchangeable cation of soil was Ca. Heavy metal contents were much lower than the critical concentration which phytotoxicity is considered to be possible and the standard for agricultural land of Korean Soil Environmental Preservation Act. Therefore, the proper soil management plan to increase the soil fertility is recommended for the ecological restoration of landfill using natural or artificial vegetation.

A Case of Nasal Cryptococcosis in a Domestic Shorthair Cat (코리안 쇼트헤어 고양이에서 발생한 크립토코쿠스 감염증 의심 1증례)

  • An 8-year-old spayed female domestic shorthair cat was presented with a chief complaint of chronic nasal discharge and dyspnea. Physical examination revealed pyohemorrhagic nasal discharge, inspiratory dyspnea and stertor, and an enlarged right mandibular lymph node. Abnormalities of blood works and serum chemistry included mildly increased hematocrit, and globulin concentration. Serologic tests for FeLV and FIV, and a panel of polymerase chain reaction tests for Chlamydophila felis, Feline Calicivirus, Herpesvirus, Bordetella, Mycoplasma felis, and H1N1 influenza was all negative. Only radiographic finding showed increasing soft tissue density in the right nasal cavity and computed tomography disclosed soft tissue/fluid opacification in the right nasal cavity, paranasal sinus, and pharyinx along with slight deviation to the right of the osseous nasal septum. Focal lysis of ventral nasal septum was also suspected in CT scan. Cytological evaluation of fine needle aspirate smears of the enlarged mandibular lymph nodes revealed numerous fungal yeasts having variably thick capsule both extracellularly and intracellularly with low numbers of macrophages. Some yeasts showed narrow based budding, which was a consistent finding with Cryptococcus organisms. Serum protein electrophoresis was a polyclonal consistent with chronic infection and serum was submitted for a fungal serology panel test. In serologic tests Cryptococcus antigen titer was 1 : 32,768. In vitro culture was unsuccessful. Treatment was initiated with administration of fluconazole, clindamycin, and tocopherol. Clinical signs resolved within 3 days after the initial treatment. The cat was discharged and scheduled for periodic evaluation and continued therapy, but was lost to follow-up thereafter.

Thermal Recovery Behaviors of Neutron Irradiated Mn-Mo-Ni Low Alloy Steel (중성자에 조사된 Mn-Mo-Ni 저합금강의 열처리 회복거동)

  • The recovery activation energy, the order of reaction and the recovery rate constant were detemined by isochronal and isothermal annealing treatment to investigate the recovery behaviors of neutron irradiated Mn-Mo-Ni low alloy steels$(fluence: 2.3\times10^{19}ncm^{-2}, 553K, E\geq1.0 MeV)$. Vickers microhardness tests were conducted to trace the recovery behavior after heat treatments. The results were analyzed in terms of recovery stages, behavior of responsible defects and recovery kinetics. It was shown that recovery occurred through two annealing stages(stage I : 703-753K, stage n : 813-873K) with re$\infty$very activation energies of 2.5 eV and 2.93 eV for each stage I and n, respectively. From the comparison of unirradiated and irradiated isochronal anneal curves, a radiation anneal hardening(RAH) peak was identified at around 813K. Most of recovery have occurred during about 120 min irrespective of isothermal annealing temperatures of 743K and 833K. Recovery rate constants were determined to be $3.4\times10^{-4}min^{-1} and 7.1\times10^{-4}min^{-1}$ for stage I and II, respectively. The order of reaction was about 2 for both recovery stages. Comparing the obtained data with those of previously reported results on neutron irradiated Mn- Mo- Ni steels, the thermal recovery be­havior of the present material seems to occur by the dissociation of point defect clusters formed during irradiation, and by the recombination process of self-interstitials and vacancies from dissociated vacancy clusters.

Fabrication of Photocatalyst Glass Beads Coated with TiO2 Thin Film by a Layer-by-Layer Process (LBL법에 의해 TiO2막이 코팅된 광촉매 글라스 비드 제조)

  • $TiO_2$ thin films consisting of positively charged poly(diallyldimethylammonium chloride)(PDDA) and negatively charged titanium(IV) bis(ammonium lactato) dihydroxide(TALH) were successfully fabricated on glass beads by a layer-by-layer(LBL) self-assembly method. The glass beads used here showed a positive charge in an acid range and negative charge in an alkaline range. The glass beads coated with the coating sequence of(PDDA/TALH)n showed a change in the surface morphology as a function of the number of bilayers. When the number of bilayers(n) of the(PDDA/TALH) thin film was 20, Ti element was observed on the surface of the coated glass beads. The thin films coated onto the glass beads had a main peak of the (101) crystal face and were highly crystallized with XRD diffraction peaks of anatase-type $TiO_2$ according to an XRD analysis. In addition, the $TiO_2$ thin films showed photocatalytic properties such that they could decompose a methyl orange solution under illumination with UV light. As the number of bilayers of the(PDDA/TALH) thin film increased, the photocatalytic property of the $TiO_2$-coated glass beads increased with the increase in the thin film thickness. The surface morphologies and optical properties of glass beads coated with $TiO_2$ thin films with different coating numbers were measured by field emission scanning electron microscopy(FE-SEM), X-ray diffraction(XRD) and by UV-Vis spectrophotometry(UV-vis).

Environmental Geochemistry and Heavy Matel Contamination of Ground and Surface Water, Soil and Sediment at the Kongjujuil Mine Creek, Korea (공주제일광산 수계에 분포하는 지하수, 지표수, 토양 및 퇴적물의 환경지구화학적 특성과 중금속 오염)

  • Enviromental geochemisty and heary metal contamination at the Kongjueil mine creek were underaken on the basis of physicohemical properties and mineralogy for various kinds of water (surface, mine and ground water),soil, precipitate and sediment collected of April and December in 1998. Hydrgeochemical composition of the water samples are characterized by relatively significant enricant of Ca+Na, alkiali ions $NO_3$ and Cl inground and surfore water, wheras the mine waters are relatively eneripheral water of the mining creek have the characteristics of the (Ca+Mg)-$(HCO_3+SO_4)$type. The pH of the mine water is high acidity (3.24)and high EC (613$\mu$S/cm)compared with those of surface and ground water. The range of $\delta$D and $\delta^{18}O$ values (relative to SMOW) in the waters are shpwn in -50.2 to -61.6% and -7.0 to -8.6$\textperthousand$(d value=5.8 to 8.7). Using computer program, saturation index of albite, calcite, dolomite in mine water are nearly saturated. The gibbiste, kaolinite and smectite are superaturated in the surface and ground water, respectively. Calculated water-mineral reaction and stabilities suggest that weathing of silicate minerals may be stable kaolinite owing to the continuous water-rock reaction. Geochemical modeling showed that mostly toxic heavy metals may exist larfely in the from of metal-sulfate $(MSO_4\;^2)$and free metal $(M^{2+})$ in nmine water. These metals in the ground and surface water could be formed of $CO_3$ and OH complex ions. The average enrichment indices of water samples are 2.72 of the groundwater, 2.26 of the surface water and 14.15 of the acid mine water, normalizing by surface water composition at the non-mining creek, repectively. Characteristics of some major, minor and rate earth elements (Al/Na, K/Na, V/Ni, Cr/V, Ni/Co, La/Ce, Th/Yb, $La_N/Yb_N$, Co/Th, La/Sc and Sc/Th) in soil and sediment are revealed a narrow range and homogeneous compositions may be explained by acidic to intermediate igneous rocks. And these suggested that sediment source of host granitic gneiss colud be due to rocks of high grade metamorphism originated by sedimentary rocks. Maximum concentrations of environmentally toxic elements in sediment and soil are Fe=53.80 wt.% As=660, Cd=4, Cr=175, Cu=158, Mn=1010, Pb=2933, Sb=4 and Zn=3740 ppm, and extremely high concentrations are found are found in the subsurface soil near the ore dump and precipitates. Normalizing by composition of host granitic gneiss, the average enerichment indices are 3.72 of the sediments, 3.48 of the soils, 10.40 of the precipitates of acid mine drainage and 6.25 of the soils near the main adit. The level of enerichment was very severe in mining drainage sediments, while it was not so great in the soils. mineral composition of soil and sediment near the mining area were partly variable being composed of quartz, mica, feldspar, chlorite, vermiculite, bethierin and clay minerals. reddish variable being composed of quartz, mica, feldspar, chlorite, vermiculite, bethierin and clay minerals. Reddish brown precipitation mineral in the acid mine drainage identifies by schwertmanite. From the separated mineralgy, soil and sediment are composed of some pyrite, arsenopyite, chalcopyrite, sphalerite, galena, malachite, goethite and various kinds of hydroxied minerals.
