Morphological Characterization of Fagopyrum esculentum Germplasm for Rutin and Quercetin Contents

  • Buckwheat is well-known crop for containing a high contents of flavonoids that are effective in vascular disease. The current study was performed to estimate the influence of morphological characterization of Fagopyrum esculentum (ES) germplasm for seed's two major flavonoids contents: rutin and quercetin. We found that the red stem color, pale green leaf color, arrowhead leaf shape, white flower color, pale brown seed coat color, and egg-shaped seed were significantly associated with 77%, 56.7%, 83.7%, 98.7%, 70.8% and 74.5% germplasm, respectively. Overall, the rutin contents of ES germplasm ranged from 0.30 to 47.86 mg/100g dry weight (DW) and the quercetin contents ranged from 0 to 1.22 mg/100g DW. The rutin contents of germplasm possessing red stem color, pale green leaf color, arrowhead leaves, white flower color, pale brown seed coat color and egg-shaped seed ranged from 7.22 to 47.86 mg/100g DW. However, the quercetin contents of germplasm with red stem color and pale brown seed coat color ranged from 0 to 1.15 mg/100g DW, with pale green leaves ranged from 0 to 0.96 mg/100g, with arrowhead leaves and white flower ranged from 0 to 1.22 mg/100g and with egg-shaped seed ranged from 0.32 to 1.22 mg/100g DW. In PCA analysis, the first three principal components (PCs) showed Eigen value more than 1 and accounted for 51.70% of variation. For both higher contents of rutin and quercetin, the morphological evaluation in ES shows a tendency of red stem color, arrowhead leaves, pale green leaf color, white flower color, pale brown seed coat color and egg-shaped seed. From this information, we can assume the rutin and quercetin contents by the morphological characteristics of the germplasm. And It could be useful in improving the rutin and quercetin contents and selecting proper resources for cultivation in existing buckwheat cultivars.

Optimization of Extraction Conditions to Obtain Functional Components from Buckwheat (Fagopyrum esculentum M.) Sprouts, using Response Surface Methodology (반응표면분석법에 의한 메밀(Fagopyrum esculentum M.) 새싹 기능성분의 추출 조건 최적화)

  • Response surface methodology (RSM) was used to optimize extraction conditions for functional components of buckwheat (Fagopyrum esculentum). A central composite design was applied to investigate the effects of three independent variables, namelyextraction temperature (X1), extraction time (X2), and ethanol concentration (X3), on responses including extraction yield (Y1), total phenolic content in the extract (Y2), $\alpha$-glucosidase inhibition activity (Y3), and acetylcholine esterase (ACE) inhibition activity (Y4). Data were analyzed using an expert design strategy and statistical software. The maximum yield was 24.95% (w/w) at $55.75^{\circ}C$ extraction temperature, 8.75 hextraction time, and 15.65% (v/v) ethanol. The maximum total phenolic yield was 222.45 mg/100 g under the conditions of $28.11^{\circ}C$ extraction temperature, 8.65 h extraction time, and 81.72% (v/v) ethanol. The maximum $\alpha$-glucosidase inhibition activity was 85.38% at $9.62^{\circ}C$, 7.86 h, and 57.58% (v/v) ethanol. The maximum ACE inhibition activity was 86.91% under extraction conditions of $10.12^{\circ}C$, 4.86 h, and 44.44% (v/v) ethanol. Based on superimposition of a four-dimensional RSM with respect to levels of total phenolics, $\alpha$-glucosidase inhibition activity, and ACE inhibition activity, obtained under various extraction conditions, the optimum ranges of conditions were an extraction temperature of $0-70^{\circ}C$, an extraction time of 2-8 h, and an ethanol concentration of 30-80% (v/v).

Antioxidative, Antimicrobial and Cytotoxic activities of Fagopyrum esculentum $M{\ddot{o}}ench$ Extract in Germinated Seeds (발아 메밀 추출물의 항산화.항균활성 및 세포독성)

  • This research was conducted to investigate the possibilities of usage of germinated-buckwheat (Fagopyrum esculentum $M{\ddot{o}}ench$) by examining antioxidative, antimicrobial and cytotoxic effects of extracts from different germinated root length of buckwheat. Antioxidant activity $(RC_{50})$ was shown higher in extracts of non-germinated seed $(50.41\;{\mu}g/mL)$ and root length 10 mm $(80.57\;{\mu}g/mL)$, 2 mm $(93.77\;{\mu}g/mL)$, 5 mm $(107.09\;{\mu}g/mL)$ than BHT $(163.96\;{\mu}g/mL)$ as a synthetic antioxidant. In antimicrobial activity, non-germinated and germinated seeds were formed inhibitory zone against S. aureus $(4{\sim}10\;mm)$, P. aeruginosa $(2{\sim}9\;mm)$ at the concentrations of $10{\sim}40\;mg/mL$ but B. subtilis, E. coli and S. typhimurium were not apparent antimicrobial activity. Extracts of germinated seed also decreased their antimicrobial activity compared to non-germinated seed extract. In addition, the growth of Calu-6 was inhibited of both 5 mm root length germinated and non-germinated seeds $(800\;{\mu}g/mL)$ as 95.12% and 87.15%, respectively, but these did not show any influence on cytotoxic effect against MCF-7 and Caco-2 cell lines. Extracts of 2 mm and 5 mm germinated seeds were also inhibited against Calu-6 and SNU-601 cell lines.

Diversity of Arbuscular Mycorrhizal Fungi in Arable and Natural Soils in Korea

  • The diversity of arbuscular mycorrhizal fungi (AM) was investigated in cultivated and natural fold sites or chungbuk, Korea. soils were collected from rhizosphere or Sorghum bicolor, Fagopyrum esculentum and Glycine max in cultivated sites, and of Miscanthus sinensis, Glycine soja and Lespedeza cuneata in natural sites. Total 20 species of Glomalean fungi were found in this study. Species richness, species diversity and density of AM fungi were significantly lower in the arable sites. While only AM fungal spores belong to Glomus and Acaulospora were found in arable fold sites, more diverse fungal spores including Gigasporaceae were found in natural grasslands. AM fungal spore composition did not significantly differ among crop plant species. Results suggest that the agricultural practices were significantly influenced on AM fungal community structures and mycorrhizal developments.

Effects of Some Heavy Metals(Al, Cd, Hg, and Pb) on ATP Content in Plant Leaves (식물엽의 ATP함량에 미치는 중금속(Al, Cd, Hg 및 Pb)의 영향)

  • The present study was carried out to estimate the comparative effects of hydroponic heavy metals (Al, Cd, Hg, and Pb) on ATP content in plant leaves grown with Hoagland solution under green house condition. The two plants, kidneybean(Phaseolus vulgaris L.) and buckwheat(Fagopyrum esculentum M nch), showed similar inhibitory effect of heavy metals on ATP content in order of Hg, Cd, Pb, and Al. But the overall inhibitory effect was greater in kidneybean than in buckwheat. The affinity of heavy metals, in vitro, toward the enzyme (luciferin-luciferase) is in order of Hg, Al, Cd, and Pb, similar to that toward ATP. The results showed that the inhibitory effect of heavy metals on ATP hydrolysis is mainly due to the coordination of heavy metals with enzyme than ATP.

  • Changes in the w-amylase activity and free sugar contents were Investigated during buckwheat (Fagopyrum esculentum Moench) germination at 1$0^{\circ}C$ for 7 days. The a-amylase activity in ungerminated seeds was 1.66 U. It increased for the 1st day of germination, but then decreased until 3rd day, and thereafter Increased. The free sugar contents In ungerminated seeds were as follows. The maltose, fructose, glucose and rhamnose were 1.81mg%, 0.42mg%, 7.71mg%, 6.80mg% on dry weight basis, respectively. The maltose and fructose contents decreased in the initial stage of germination, but then gradually increased. The glucose contents decreased for the 3rd day, but sharply Increased afterwords. The rhamnose contents decreased until 1 day, and then there was no significant change for the 6 days.

Changes in Physico-chemical Properties and Mineral Contents during Buckwheat Germination (메밀 발아 중 물리화학적 특성과 무기질 함량의 변화)

  • To provide the effective application scheme and basic information of buckwheat (Fagopyrum esculentum Moench), buckwheat was germinated at 10$^{\circ}C$ for 7 days and 100 kernels weight, germination rate, root length, chemical composition and mineral contents were examined at 24 hour Intervals. During the germination period, the 100 kernels weight increased approximately 0.3g per day. The germination rate increased sharply after 2 days and the root length increased greatly after 4 days. The crude protein contents increased with germination time, whereas the carbohydrate contents decreased. The crude ash and fat contents did not differ significantly during the germination period. The Ca contents Increased for the 4th day of gemination, but gradually decreased afterwords. The Na contents increased in the initial stage of germination, but then gradually decreased. However, there were no significant change In the Mg, K. Fe. Mn and Zn contents.

Anti-oxidant and Anti-inflammatory Effects of Rutin and Its Metabolites

  • Rutin is one of the major flavonoids found in buckwheat (Fagopyrum esculentum Moench). While rutin is already known to exhibit anti-oxidative, anti-inflammatory, and anti-carcinogenic activities. However, the health beneficial function of rutin metabolites is not well understood. In DPPH radical scavenging assays, the present study found that 3,4-dihydroxyphenyl acetic acid had the highest total anti-oxidant activity, followed by 3,4-dihydroxyphenylacetic acid, rutin, homovanillic acid, and 3-hydroxyphenyl acetic acid. Further, 3,4-dihydroxyphenylacetic acid strongly reduced LPS-induced IL-6 production in RAW 264.7 cells, compared with other metabolites. Therefore, these results suggest that rutin metabolites have potential to be utilized as food ingredients with anti-oxidant and anti-inflammatory activities.

STUDIES ON THE DIMORPHISM OF THE PERSICARIA SENTICOSA NAKAI (Persicaria senticosa Nakai의 Dimorphism에 관한 연구)

  • HARN, Chang Yawl (Chonnam U. Kwangju, Korea): Siudies on the dimorpism of the Perisicaria senticosa Nakai-Kor. jour. Bot. 3(1) 16-25 During his researches regarding the morphological and physiological properties of Polygonecenae, the author has found that the species, Persicaria senticosa, aiso, besides the heterostylous plants of Polygonum family, Fagopyrum esculentum, and Persicaria japonica which was recently verified by the author as dimorphic, shows the typical floral structure of heterostylism, the description of which is not found in taxonomical works. Further research on this species have revealed that this plant, despite pressessing characteristic structural dimorphism, does not exhibit even the slight signs of heterostlylous properties physiologically. This is a deviation from the usual behavior of normal heterostylous plants. What is interesting is the fact that the physiological behavior of this species is quite contrary to that of P. japonica which is considered to be the most highly specialized dimorphic plant. Thus it is assurred that if some species of this family had taken a heterostylic form in its course of development from autogamy to allogamy, P. seoticosa would be the least differentiated type of dimorphic forms among the three heterostylous plants, including buckwheat, of this family. The results obtained in this experiment are summarized as follows: 1) P. senticosa has two forms of flower, one, long style-short stamened; the other, short style-long stamened. Not only conspicuous is this primary difference, but the secondary difference, such as pollen grain size, is noticeable between long and short styled individuals, thus expressing structurally the definite trait of a dimorphic plant. 2) Structural alteration of floral parts towards dimorphism has preceeded far less in comparison with those of the P. japonica and F. esculentum. 3) Elaborate studies on fertility reveal that this species does not show the slightest sign of the physiological characteristics of dimorphic plants. In other words, regardless of the modes of combinations, legitimate and illegitimate, fertilization and fruit setting flourish unimpaired. 4) Growth of pollen tubes apparently parallels the results in the fertility, tubes reaching ovary approximately 30 minutes after pollination both in legitimate and illegitimate combinations. Pollen tube penetration appears to be comparatively rapid. 5) A slight difference in the growth of pollen tube seems to exist between legitimate and illegitimate combinations, legitimate union giving slightly faster tube penetration. 6) In the present experiment it was clarified that P. senticosa, known to possess one form of flower in taxonomy, is in reality dimorphic plant having two of flowers. Although this species is definitely heterostylous in floral structure, physiological evidence and pollen tube behavior show that the differentiation of this plant toward the dimorphism has apparently proceeded slightly except for some parts of floral organ. In ordinary heterostylous plants it is a matter of common occurrence that when illegitimately cmbined, there is poor or no fertility, Contrary to the universal property of heterostylous plants, no difference is observed in the fertility and pollen tube growth between the legitimate and illegitimate combinations in the case of P. senticosa. Compared to the P. japonica and F. esculentum, which are supposed to have undergone high degree of dimorphic differentiation, it is an unavoidable conclusion that P. senticosa has not yet developed as a heterostyle plant except for some of its floral parts. If P. japonica is assumed to be the most differentiated type of hetenostylous plant, thenthe P. senticosa would be regarded as the primitive, retaining still the self-fertile nature so common to the Polygonum genus. In nature, however, this plant has a better chance to be pollinated legitimately owing to the two forms of flowers than to be pollinated illegitimately.
