HDDM, a formula consisting of seven herbs, had anti-diabetic but no immunomodulatory activities in multiple low doses of streptozotocin-treated female of B6C3F1 mice

  • The objectives of this study were to determine the effect of herb formula HDDM, a modification of Huangdan decoction that has been shown to be effective in the treatment of glomerulonephritis and chronic renal failure, on the blood glucose levels in multiple low doses (MLD; 50 mg/kg for five consecutive days) of streptozotocin (STZ)-treated female B6C3F1 mice. Initial studies were performed to compare diabetes induction in five strains (e.g., B6C3F1, NOD, CD-1, C3H/HeN and C57BL/6) of mice by MLD-STZ, and immune changes following the treatment. The results suggested that the order of susceptibility to diabetes induction was NOD $\approx$ CD-1 > B6C3F1 $\approx$ C3H > C57BL/6. Furthermore, STZ modulation of T cell development, differentiation and activation might play a role in diabetes induction by MLD-STZ treatment. MLD-STZ-induced diabetes in female B6C3F1 mice was moderate, which allowed the evaluation of drug-induced protection or exacerbation of diabetes to be performed. As such, modulation of blood glucose by HDDM, which consisted of Da Huang (Radix Et Rhizoma Rhei), Huang Qi (Radix Astragali Seu Hedysari), Dan Shen (Radix Salviae Miltiorrhizae), Yin Yang Huo (Herba Epimedii), Yi Yi Ren (Semen Coicis or Coix lacryma-jobi), Mai Dong (Radix Ophiopogonis) and Shan Zhu Yu (Fructus Corni), was evaluated in MLD-STZ-treated female B6C3F1 mice. The results suggested that HDDM could lower the blood glucose levels, but it had no immunomodulatory activities. Additionally, HDDM-treated mice exhibited improved glucose tolerance. In conclusion, these studies have suggested that MLD-STZ-induced diabetes in female B6C3F1 mice is a useful model to evaluate drug modulation of diabetes, and that the herb formula HDDM possesses anti-diabetic effects.

Histological Analysis of Reproductive System in Low-Dose Nonylphenol-treated F1 Female Mice

  • Previously, we reported adverse effects of low-dose nonylphenol (NP) exposure on the reproductive parameters of F1 female mice. In the present study we further investigated the pathohistological effect of NP exposure on the reproductive organs in F1 female mice. NP exposures were continuously conducted from parental pre-mating period until the postnatal day (PND) 33 of F1 offspring for vaginal examination. Mice were sacrificed on PND 30 and the reproductive tissue weights were measured. The initial (at PND 21) body weights of the NP-50 group animals were significantly lower than those of control group animals, and the weight deficit were recovered when the terminal (PND 33) body weights were measured. Early vaginal opening was found in NP group animals (p<0.05). Pathohistological studies revealed that NP-treated F1 animals showed prominent increase in the ovarian follicle numbers (p<0.01), and decrease in the diameter of uterine myometrium (p<0.01), and increase in the diameter of luminal epithelium (p<0.05). The present study demonstrated that the subchronic low-dose NP exposure induced early beginning of puberty and pathohistological abnormalities in ovary and uterus of F1 mice. Further studies are needed to achieve a better understanding on the action mechanism of NP in pubertal onset and to find a way to avoid a hazardous situation provoked by NP exposure.

ENHANCEMENT OF NORMAL AND NEOPLASTIC MAMMARY GROWTH BY CROSSBREEDING BETWEEN STRAINS OF FEMALE AND MALE MICE WITH HIGH MAMMARY GROWTH POTENTIALS

  • Based on our previous results that among 4 strains of mice SHN and GR/A showed the highest mammary growth potentials in females and males, respectively. Effects of crossbreeding on normal and neoplastic mammary growth were studied in $(SHN\;{\times}\;GR/A)F_1$ virgin female mice. $F_1$ mice were higher than the parental strains in the end-bud formation and the ductal growth of mammary glands at 60 days of age and at tumorous age, respectively. While there was little difference between $F_1$ and both parental strains in the onset age of the first mammary tumors, mammary tumorigenic potential was apparently higher in the former than in the latters. This would be the first report that demonstrated directly the contribution of mammary growth potential of males to that of female offspring.

  • The smear preparations of the semen from Korean native bull and the tissue preparations of the organs from male and female mice were performed by fluorescent staining method. More than 600 spermatozoa per straw from two semen straw groups and more than 300 cells per mouse organ from two mice per sex were observed and then the ratio of spermatozoa bearing B-body and the cells bearing F-body were assessed, respectively. 1. The ratios of spermatozoa bearing B-body in semen of Korean native bull were $37.3{\pm}3.1%$. 2. The ratios of cells bearing F-body in the organs of mice were $63.5{\pm}4.5%$ in male tissues and $7.5{\pm}3.2%$ in female tissues. 3. The organs with higher appearance frequency of F-body were ordered as brain, kidney, stomach, lung, testis, liver, small intestine, spleen and pancreas in male mice and pancreas, small intestine, liver, brain, kidney, lung, spleen and stomach in female mice.

The Inheritance of Jumping Activity in Reciprocal Cross of Two Subspecies of Mice

  • The objective of this study was to describe the nature of the inheritance of jumping as a behavioral trait and to analyze quantitatively the jumping height as a measure of vigor in two subspecies of mice. Two subspecies of mice, Yonakuni wild mouse (Y) and $CF_{{\sharp}1}$ laboratory mouse (C), were used as the parental types. Reciprocal mating between these two subspecies was made to produce subsequently the first and second generations. The first generation was $F_1$ (YC) resulting from $Y\;male{\times}C\;female$, and $F_1{^\prime}$ (CY) from $C\;male{\times}Y\;female$. The second generation $F_2$ (YCYC) was from mating $F_1{\times}F_1$ and $F_2{^\prime}$ (CYCY) from $F_1{^\prime}{\times}F_1{^\prime}$. Individuals were treated with a set of direct current shock apparatus at six weeks of age to evoke jumping. The results showed that the ratio between jumping and non jumping mice (J: NJ) for C was 0%:100% (0:1), which means that all C did not jump throughout the experiment, whereas Y was 68%:32% (2:1); and the $F_1$ and $F_2$ showed 65%:35% (2:1) and 51%:49% (1:1), respectively. All $F_1{^\prime}$ and $F_2{^\prime}$ individuals jumped as indicated by the ratio 100%:0% (1:0) for both these two genetic groups. Of the jumped mice, average height of the first three jumping observed for pooled sexes in Y, $F_1$, $F_2$, $F_1{^\prime}$ and $F_2{^\prime}$ were 19.3 cm, 19.3 cm, 18.0 cm, 19.9 cm and 16.4 cm, respectively. The distribution of jumping height showed a tendency to be a normal distribution. The jumping activity and jumping height may be affected by some major genes and polygenes, respectively.

  • The transgenic mice were produced by microinjection of human growth hormone gene fused with mouse metallothionein Ⅰ promoter. They were mated with momal mice by backcross or brother-sister mating. The reproduction efficiencies of female and male n the FO transgenic mice were 17.6%(3/17 mice) and 31.2%(5/16 mice), respectively, and were very lower than that in normal mice(85.7% and 100%, respectively). Interestingly, a few of female transgenic mice were fertile which was different from the previous reports. Of 6 fertile transgenic mice, 2 mice were identified as mosaic type by the reduced frequency of genetic transmission to successive generation below Mendelian levle and the enhanced copy numbers of transgene in progeny mice compared with the transgenic parent. In the group of F1, F2, F3 transgenic mice the reproduction efficiencies of males were gradually improved, whereas females were absolutely infertile. It was consequently shown that the transgenic mice expressing human growth hormone gene were frequently infertile, but the genotypic and phenotypic characteristics of the fertile transgenic mice were normally passed on to the progeny through herm line. Therefore it must be considered wheter or not the products of foreign DNA introduced into animals will detrimentally affect their physiological aspects.

Repopulation of autophagy-deficient stromal cells with autophagy-intact cells after repeated breeding in uterine mesenchyme-specific Atg7 knockout mice

  • Objective: Autophagy is highly active in ovariectomized mice experiencing hormone deprivation, especially in the uterine mesenchyme. Autophagy is responsible for the turnover of vasoactive factors in the uterus, which was demonstrated in anti-Müllerian hormone receptor type 2 receptor (Amhr2)-Cre-driven autophagy-related gene 7 (Atg7) knockout (Amhr-Cre/Atg7f/f mice). In that study, we uncovered a striking difference in the amount of sequestosome 1 (SQSTM1) accumulation between virgin mice and breeder mice with the same genotype. Herein, we aimed to determine whether repeated breeding changed the composition of mesenchymal cell populations in the uterine stroma. Methods: All female mice used in this study were of the same genotype. Atg7 was deleted by Amhr2 promoter-driven Cre recombinase in the uterine stroma and myometrium, except for a triangular stromal region on the mesometrial side. Amhr-Cre/Atg7f/f female mice were divided into two groups: virgin mice with no mating history and aged between 11 and 12 months, and breeder mice with at least 6-month breeding cycles with multiple pregnancies and aged around 12 months. The uteri were used for Western blotting and immunofluorescence staining. Results: SQSTM1 accumulation, representing Atg7 deletion and halted autophagy, was much higher in virgin mice than in breeders. Breeders showed reduced accumulation of several vasoconstrictive factors, which are potential autophagy targets, in the uterus, suggesting that the uterine stroma was repopulated with autophagy-intact cells during repeated pregnancies. Conclusion: Multiple pregnancies seem to have improved the uterine environment by replacing autophagy-deficient cells with autophagy-intact cells, providing evidence of cell mixing.

High-level Expression and Characterization of the Human Interleukin-10 in the Milk of Transgenic Mice

  • Interleukin-10 (IL-10) is a homodimeric protein with a wide spectrum of anti-inflammatory and immune activities. It inhibits cytokine production and expression of immune surface molecules in various cell types. The transgenic mice carrying the human IL-10 gene in conjunction with the bovine $\beta$-casein promoter produced the human IL-10 in milk during lactation. Transgenic mice were generated using a standard method as described previously. To screen transgenic mice, PCR was carried out using chromosomal DNA extracted from tail or toe tissues with a primer set. In this study, stability of germ line transmission and expression of IL-10 gene integrated into host chromosome were monitored up to generation F15 of a transgenic line. When female mouse of generation F9 was crossbred with normal male, generation F9 to F15 mice showed similar transmission rates (66.0$\pm$20.13%, 61.5$\pm$16.66%, 41.1$\pm$8.40%, 40.7$\pm$20.34%, 61.3$\pm$10.75%, 49.2$\pm$18.82%, and 43.8$\pm$25.91%, respectively), implying that the IL-10 gene can be transmitted stably up to long term generation in the transgenic mice. For ELISA analysis, IL-10 expression levels were determined with an hIL-10 ELISA and a mIL-10 ELISA kit in accordance with the supplier's protocol. Expression levels of human IL-10 from milk of generation F9 to F13 mice were 3.6$\pm$1.20 mg/ml, 4.2$\pm$0.93 mg/ml, 5.7$\pm$1.46 mg/ml, 6.3$\pm$3.46 mg/ml, and 6.8$\pm$4.52 mg/ml, respectively. These expression levels are higher than in generation F1 (1.6 mg/ml) mice. We concluded that transgenic mice faithfully passed the transgene on their progeny and successively secreted target proteins into their milk through several generations, although there was a little fluctuation in the transmission frequency and expression level between the generations.

Expression and Characterization of the Human Lactoferrin in the Milk of Transgenic Mice

  • Human lactoferrin (hLF) is an 80 kDa iron-binding glycoprotein that is expressed in high concentration in milk and in lesser amount in the secondary or specific granules of neutrophils and in plasma, LF is classically considered to be related to the binding, transport, and storage of iron. The transgenic mice carrying the human hLF gene in conjunction with the bovine $\beta$-casein promoter produced the human hLF in their milk during lactation. To screen transgenic mice, PCR was carried out using chromosomal DNA extracted from tail or toe tissues. In this study, stability of germ line transmission and expression of hLF were monitored up to generation Fl7 of a transgenic line. When female mouse of generation F9 was crossbred with normal male, generation F9 to Fl7 mice showed similar transmission rates ($66.0 \pm 12.57%, 42.0 \pm 14.98%, 72.2 \pm 25.45%, 50.0 \pm 16.70%, 65.7 \pm 6.45%, 48.6 \pm 14.65%, 54 1 \pm 18 11%, 57.8 \pm 16.16% and 48.6 \pm 20.66$, respectively), implying that the hLF gene can be transmitted stably up to long term generation in the transgenic mice For ELISA analysis, hLF expression levels were determined with an hLF ELISA kit in accordance with the supplier's protocol. Expression levels of human hLF from milk of generation F9 to Fl3 mice were $ 3.2 \pm 0.69 mg/ml, 3.1 \pm 0.81 mg/ml, 4.6 \pm 1.38 mg/ml, 3.1 \pm 0.42 mg/ml, and 4.5 \pm 1,48 mg/ml$, respectively. These expression levels were lower than that of founder (6.6 mg/$m\ell$) mouse. We concluded that transgenic mice faithfully passed the transgene on their progeny and successively secreted target proteins into their milk through several generations.

  • AEY의 acetone 추출물이 mouse liver microsome의 산화에 미치는 영향을 조사하였다. Mouse liver microsome에 함유된 단백질 AEY 처리가 CEY 처리구에 비해 상대적으로 7~48%정도가 증가하였다. 지방산 분석 결과, CEY 처리에 비해 AEY 처리구에서 stearic acid를 포함하는 포화지방산의 함량비가 상대적으로 높았으며, oleic acid, linoleic acid 등을 함유하는 불포화지방산의 함량비가 현저히 감소하였다. AEY 처리구는 F $e^{2+}$가 관여하는 Asc/F $e^{2+}$, NADPH/F $e^{+2}$ 를 산화유도물질로 사용한 경우에 TC와 비슷하거나 보다 강력한 항산화 효과를 보였다. ABIN이나 CuOOH가 산화유도물질로 작용할 시에는 AEY처리에 의해 유의성있는 항산화 효과가 나타나지 않았다.
