Fates of water and salts in non-aqueous solvents for directional solvent extraction desalination: Effects of chemical structures of the solvents

  • Non-aqueous solvents (NASs) are generally known to be barely miscible, and reactive with polar compounds, such as water. However, water can interact with some NASs, which can be used as a new means for water recovery from saline water. This study explored the fate of water and salt in NAS, when saline water is mixed with NAS. Three amine solvents were selected as NAS. They had the same molecular formula, but were differentiated by their molecular structures, as follows: 1) NAS 'A' having the hydrophilic group ($NH_2$) at the end of the straight carbon chain, 2) NAS 'B' with symmetrical structure and having the hydrophilic group (NH) at the middle of the straight carbon chain, 3) NAS 'C' having the hydrophilic group ($NH_2$) at the end of the straight carbon chain but possessing a hydrophobic ethyl branch in the middle of the structure. In batch experiments, 0.5 M NaCl water was blended with NASs, and then water and salt content in the NAS were individually measured. Water absorption efficiencies by NAS 'B' and 'C' were 3.8 and 10.7%, respectively. However, salt rejection efficiency was 98.9% and 58.2%, respectively. NAS 'A' exhibited a higher water absorption efficiency of 35.6%, despite a worse salt rejection efficiency of 24.7%. Molecular dynamic (MD) simulation showed the different interactions of water and salts with each NAS. NAS 'A' formed lattice structured clusters, with the hydrophilic group located outside, and captured a large numbers of water molecules, together with salt ions, inside the cluster pockets. NAS 'B' formed a planar-shaped cluster, where only some water molecules, but no salt ions, migrated to the NAS cluster. NAS 'C', with an ethyl group branch, formed a cluster shaped similarly to that of 'B'; however, the boundary surface of the cluster looked higher than that of 'C', due to the branch structure in solvent. The MD simulation was helpful for understanding the experimental results for water absorption and salt rejection, by demonstrating the various interactions between water molecules and the salts, with the different NAS types.

Extraction and Purification of Polysaccharides from Phellinus linteus Mycelia (상황버섯의 균사체로부터 다당류의 추출과 정제)

  • To maximize the obtained polysaccharides from Phellinus linteus mycelia, effects of extraction and purification conditions including dialysis time (8, 16, 24, 48 h) on an amount of the polysaccharides were investigated. As extraction temperature ($50{\sim}95^{\circ}C$), ratio of solvent volume to the dry weight of mycelia (10, 15, 20, 25 ml/g), extraction time(2, 4, 6, 8 h), final concentration of ethanol (70, 75, 80, 85%), and aging time (1, 4, 8, 16 h) increased, an amount of the polysaccharides was increased. An increase in precipitation time up to 24 h increased an amount of the polysaccharides but a further increase in precipitation time after 24 h did not changed largely an amount of the polysaccharides. Three precipitation solvents (ethanol, methanol, acetone) were tested. An amount of the polysaccharides was increased in order to acetone, ethanol, and methanol. On the optimal extraction condition, extraction temperature, ratio of water volume to the dry weight of mycelia, and extraction time were $95^{\circ}C$, 25 ml/g, and 8 h, respectively.

Antioxidant activities of phenolic compounds from Ziziphus jujuba Mill extract using subcritical water (대추 아임계수 추출물의 폴리페놀 함량 및 산화방지 평가)

  • Subcritical water extraction (SWE) is an eco-friendly extraction method that uses only purified water as a solvent under high temperature and high pressure conditions. In this study, total phenolics, and antioxidant activity were evaluated in Ziziphus jujuba Mill extracts from subcritical water obtained by varying the extraction temperature (110-190℃) and extraction time (1-20 min). Total phenolics was maximized with extraction at 190℃ for 15 min (67.79±3.45 mg gallic acid equivalent/g jujube). The 2,2-diphenyl-1-picrylhydrazyl free radical scavenging activity (48.84±4.74%) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) free radical scavenging activity (84.75±1.15%) were maximal at extraction conditions of 190℃, for 20 min. All jujube extracts prepared using SWE had higher total phenolics and antioxidant activities than extracts prepared using organic solvent extraction (60℃, 120 min), including methanol and ethanol. SWE could be an excellent alternative to organic solvents for extracting phenolics and antioxidant compounds.

Effects of Extractants on the Characteristic of Soluble Dietary Fiber from Apple Pomace (추출용매가 수용성 사과박 식이섬유의 특성에 미치는 영향)

  • The dietary fiber was extracted from apple pomace with various concentrations of acidic, alkaline solvents and water in $24.4{\sim}29.3%$ yield. The extractions of water-soluble dietary fiber with acidic(6.85%) and alkaline solvent(6.55%) were more effective than the water extraction(6.0%). The higher extraction yield was obtained with increased concentration of acidic or alkaline solvents. The content of galacturonic acid was higher in the order, 0.01 M HCl(77.5%)>0.05 M HCl(76.8%)>$H_2O(76.1%)$>0.05 M NaOH(73.8%)> 0.01 M NaOH(69.0%) and the extraction with acidic solvent showed the highest pectin yield. The color value increased by extraction with solvent in higher concentration. And at equal concentration, the color value of alkaline extract was higher than that of acidic exctract. There was no relationship between the color value and the extraction yield of dietary fiber. In the investigation on the rheological properties, the shear stress increased in proportion to galacturonic acid contents. While water-soluble dietary fiber extracted with acid and water represented Newtonian rheological properties, the alkaline extract was inclined to have converting property from pseudoplastic to Bingham pseudoplastic with increment of alkaline concentration. In the studies on the time-dependence of apparent viscosity, the thixotropic property with hysteresis loop was observed in alkaline extract.

Antioxidant Activities of Ostrich Fern by Different Extraction Methods and Solvents (추출방법 및 용매에 따른 청나래고사리의 항산화 활성)

  • This study was performed to investigate effective extract conditions in fronds of the Ostrich fern (Matteuccia struthiopteris) to increase antioxidant compound contents and antioxidant capacity. Powder (1 g) of lyophilizated fronds were mixed with 3 kinds of solvents (MeOH, 80% EtOH and water). Extractions were carried out using not only immersion (room temp.), heating ($60^{\circ}C$) and stirring (200rpm) for 6 hr, but also through sonication in a 42 kHz ultrasonic bath for 15, 30 and 45 min. Extracts were filtrated and measured for contents of soluble solids (SS), total polyphenols (TP; tannic acid as a standard) and total flavonoids (TF; Naringin as a standard). Antioxidant activity was expressed as $RC_{50}$ for DPPH and ABTS radical scavenging. SS (0.317 $g{\cdot}g^{-1}$ db), TP (70.90 $mg{\cdot}g^{-1}$ db) and TF (41.53 $mg{\cdot}g^{-1}$ db) contents reached their highest levels when 30 minute sonication extraction with 80% EtOH was performed, and the highest DPPH and ABTS scavenging activity was observed in the same extraction conditions ($RC_{50}$=0.14 $mg{\cdot}ml^{-1}$ and 0.09 $mg{\cdot}ml^{-1}$, respectively). From the present investigation, it can be concluded that fronds of the ostrich fern can be used as a natural material for antioxidants, and sonication for 15-30 min with 80% EtOH is an ideal extraction method for increasing their antioxidant effects and saving extraction time.

  • In order to detect poisonous or physiologically active constituents in the higher fungi growing in Korea, thirtyeight species and three varieties were collected in Kyung Ki Province and subjected to systematic extraction with two kinds of organic solvents. The extracts wre examined for alkaloid by adding alkaloid reagents. Of these, seven species and one variety gave positive confirmatory tests.

  • Allergen-removed-extract was produced from Rush verniciflua by two phase methods. Phase one was high temperature treatment of Rush verniciflua tree to get allergen-removed-extract. Phase two was extraction of solution from phase one product using water or organic solvents. The solutions from above method show high antioxidant activity, anticancer activity, and improvement in lung function, but did not contain urushiol family compounds.

  • The inhibitory effect of cacao bean husk (CBH) extract on glucosyltransferase(GTasc) from Streptococcus mutans B13 was investigated. Water solube extract from CBH showeda sarong inhibitory effect (88-89%) on GTase from Streptococcus mutans Bl3. GTase inhibitors from sequential extraction by hot water or water-methanol had the strongest inhibition. Sources, fermentation, and types of solvents and fumigation processes did not influence the effect. These active compounds proved to be polyphones through acid hydrolytic analysis of the precipitates by ammonium sulfate or ethanol and proteinase K. It was also confirmed by additional column chromatography of Sephadex G-50, Sephadex LH-20 and DEAE-Sephdex A-50.

Comparative Studies on Methods of Extracting Polyacetylene Compounds from White Ginseng (인삼 Polyacetylene 성분의 추출방법 비교연구)

  • The amounts of panaxynol and panaxydol, which are major polyacetylone compounds In white ginseng were determined by capillary-GC (FID), and the extraction efficiencies when using varictus extraction solvents (petroleum ether, dichloromethane, ether, ethyl acetate, acetone, acetonitrilr and methanol) and various extraction methods (shaking, Soxhlet and reflux) were compared . The GC column was SPB-1 fused silica capillary (0.25 mm id x30 m, Supelco), and the column oven temperature was programmed to rise from $200^{\circ}C$ to $300^{\circ}C$ at the rate of $4^{\circ}C$ per minute. The extraction efficiencies for panaxynol and panaxydol according to extraction souvtints were the highest in methanol and decreased in the order of dichloromethane, acetone, ether, ethyl acetate, acetonitrile and petroleum ether. The extracttion efficiencies for panaxynol and panaxydol according to extraction methods were the highest for reflux and the lowest for shaking, and those with Soxhlet were almost equal to those for reflux. The analytical amounts of panaxynol and panaxydol obtained by reflux with methanol %mere 4.2 and 6.4 mg/g, respectittely in white ginseng.

Safety Profile Assessment and Identification of Volatile Compounds of Krill Eupausia superba Oil and Residues Using Different Extraction Methods

  • In this study, Krill Eupausia superba oil was extracted using different solvents and supercritical carbon dioxide (SC-$CO_2$). During SC-$CO_2$ extraction, the pressure was set at 40 MPa and temperatures ranged from $40^{\circ}C$ to $55^{\circ}C$. We examined the differences in volatile compounds and safety profiles among extraction methods. Volatile compounds were determined using the thermal desorption system integrated with gas chromatography-mass spectrometry (GC-MS). Heavy metal content was analyzed by inductively coupled plasma mass spectrometry (ICP-MS). According to our results 10 volatile compounds were identified in krill sample. After SC-$CO_2$ extraction of oil, the concentrations of volatile compounds decreased, but increased after solvent extraction. In krill, heavy metal concentrations remained within the permissible limit. Moreover, Zn and Fe which have health benefits were detected at high concentrations. During a 90 days storage period at different temperatures, microbial activity was found to be lowest in SC-$CO_2$ extracted residues. Thus, the quality of krill oil and the residues obtained using SC-$CO_2$ extraction was higher and the oil was safer than those obtained using conventional solvent extraction. These results can be applied to the food industry to maintain high quality krill products.
