This study was to investigate the effect of flavonoid treatment on in vitro development of bovine somatic cell nuclear transfer (SCNT) embryos, and their pregnancy and delivery rate after embryo transfer into recipient. In experiment 1, to optimize the flavonoid concentration, parthenogenetic day 2 ($\geq$ 2-cell) embryos were cultured in 0 (control), 1, 10 and $20\;{\mu}M$ flavonoid for 6 days. In the results, in vitro development rate was the highest in $10\;{\mu}M$ flavonoid group (57.1%) among treatment groups (control, 49.5%; $1\;{\mu}M$, 54.2%; $20\;{\mu}M$, 37.5%), and numbers of total and ICM cells were significantly (p<0.05) higher in $10\;{\mu}M$ flavonoid group than other groups. We found that $10\;{\mu}M$ flavonoid treatment can significantly (p<0.05) decrease the apoptotic index and derive high expression of anti-oxidant, anti-apoptotic, cell growth and development marker genes such as Mn-SOD, Survivin, Bax inhibitor, Glut-5, In-tau, compared to control group. In experiment 2, to produce the cloned Jeju Black Cattle, beef quality index grade 1 bull somatic cells were transferred into enucleated bovine MII oocytes and reconstructed embryos were cultured in $10\;{\mu}M$ flavonoid added medium. When the in vitro produced day 7 or 8 SCNT blastocysts were transferred into a number of recipients, $10\;{\mu}M$ flavonoid treatment group presented higher pregnancy rate (10.2%, 6/59) than control group (5.9%, 2/34). Total three cloned Jeju Black calves were born. Also, two cloned calves in $10\;{\mu}M$ flavonoid group were born and both were all healthy at present, while the one cloned calf born in control group was dead one month after birth. In addition, when the result of short tandem repeat marker analysis of each cloned calf was investigated, microsatellite loci of 11 numbers matched genotype between donor cell and cloned calf tissue. These results demonstrated that the flavonoid addition in culture medium may have beneficial effects on in vitro and in vivo developmental capacity of SCNT embryos and pregnancy rate.
Embryoid bodies (EBs) generated from human embryonic stem cells (hESCs) include spontaneously induced endodermal lineage cells (ELCs). Activin-A plays important roles in the endoderm differentiation of hESCs. Despite studies on the generation of ELCs from hESCs with treatment of Actvin-A, it was unclear for localization and pattern of ELCs by Activin-A during differentiation of hESCs. Accordingly in this study, we knew that Actvin-A increased the cystic EBs formation, including the highly enriched AFP (endoderm lineage specific marker)-expressing cells in the surface of cystic EBs. To induce the EBs formation from undifferentiated hESCs, cells were transferred onto petri-dish and cultured in suspension condition without bFGF removed hESC media (EB media) for 3 days. Next to investigate the effect of Activin-A, EBs were subsequently cultured in EB media supplement with 100 ng/ml Activin-A for 3 days. After 5~7 days of Activin-A treatment, cystic EBs began to appear which increased in numbers reaching ~60% of initially formed EBs over 5 days. Endoderm lineage marker, AFP were highly expressed and specifically localized at the surface region of cystic EBs comparison with normal EBs. We next attached the cystic EBs onto gelatin-coated plates and cultured for 5 days. In the results of real-time PCR and immunocytochemistry analysis, AFP-expressing cells migrated and localized at the outgrowth region of attached cystic EBs. To obtain the AFP-expressing cells of the outgrowth region, we manually isolated by using micro-dissection and cultured them. These cells strongly express AFP over 70% of isolated cells post re-plating. Here, we first showed an expression pattern of specifically localized ELCs by Activin-A during differentiation of hESCs. From this observation, we could highly purified ELCs from undifferentiated hESCs. Taken together, our system will provide a novel and efficient option to generate ELCs from hESCs.
본 연구는 생태체험 프로그램 활동을 실시함으로써 유아들에게 기쁨과 추억을 만들어 주고 스스로 정서지능에 대한 변화를 알아보는데 목적이 있다. 연구대상은 충청도에 소재하는 어린이집 만 5세반 중 두 학급의 어린이 40 명을 대상으로 실험집단과 통제집단으로 나누어 남, 녀 각각 20명씩으로 구성하였으며, 연구대상인 실험집단 유아들은 생태체험활동의 프로그램을 따라 정한 요일에 5명씩 그룹을 나눠서 관찰하고, 체험하며 실험을 진행하였다. 분석결과 생태체험 프로그램 활동은 전반적인 유아정서지능과 하위요인별 자기정서의 이용, 타인정서의 인식 및 배려, 자기정서의 인식 및 표현, 감정의 조절 및 충동억제, 교사와의 관계, 또래와의 관계에 있어 통제집단에 비해 실험집단의 경우 상대적으로 높은 증가효과를 나타내었다. 이는 자연과의 유기적인 관계형성과 생태 중심의 체험 탐구수업이 실내교육에서 느낄 수 없는 자연 세계의 이해와 자연친화적 태도, 생태위기 인식, 사회적 협력과 더불어 함께 생활하는 태도 기반 형성을 제공한다는 점에서 유아들의 정서지능 향상에서 유용한 프로그램임을 입증시켜 준 것이라 할 수 있다.
It has been reported that CLA decreases fat deposition in vivo and in vitro experiments. Among CLA isomers, c9t11 and t10c12 have been shown to exert active biological activities. For example, t10c12 reduces body weight and increases lean body mass, whereas, c9t11 has little effect on body fattness. However, the underlying molecular mechanism for the anti-obesity action of CLA isomers are not well understood. The purpose of this study was to examine the effects of t10c12 and c9t11 on lipid accumulation, cell proliferation, cell death and the expression levels of Ucp genes which are proposed as targets for anti-obesity in 3T3-L1 preadipocytes. Isomers of CLA at 50$\mu$M were added into preadipocyte differentiation medium for 3, 6 and 9days. Control cells received only the vehicle in the differentiation medium. Cytochemical analyses for lipid accumulation, cell proliferation and apotosis were carried out to compare lipidogenesis and cellular activity. RT-PCR analysis of GAPDH, Ucp 2,3 and 4 were also performed to find any modulatory effects of CLA isomers on the metabolic genes. Lipid accumulation indicated by Oil Red-O staining was inhibited in CLA isomers as compared to the control. T10c12 isomer showed less lipidogenesis than c9t11 did. A decrease occurred in CLA isomers as shown by BrdU incorporation. Apotosis has occured at higher level in t10c12 when compared to that of t9c11. Ucp 2, 3 and 4 genes were also upregulated in CLA isomers. T10c12 showed higher level of Ucp gene expressions than the c9t11 did. The biological activities of CLA isomers were also found to be different during differentiation of 3T3-L1 preadipocytes, suggesting that different isomers may be active in certain stage of lipidogenesis. The results indicate that both c9t11 and t10c12 CLA isomers decrease lipidogenesis, inhibit cell proliferation, increase cell death and upregulate in Ucp gene expressions during 3T3-L1 preadipocyte differentiation. T10c12 isomer was more effective than c9t11 in overall anti-obesity activity.
Objective : Hyaluronidase (HAse), a degrading enzyme of hyaluronic acid (HA), is highly expressed in patients with malignant glioma. The purpose of this study was to verify whether HAse is related to the invasion of glioma cells. We also investigated if glioma cells with higher mobility in 2-dimensioal (2-D) method have also higher mobility at 3-dimensional (3-D) environment. Methods : Malignant glioma cell lines (U87MG, U251MG, U343MG-A, and U373MG) were used, and their HAse expressions were evaluated by HA zymography. The migration ability was evaluated by simple scratch technique. The invasiveness of each cell lines was evaluated by Matrigel invasion assay and HA hydrogel invasion assay. In HA hydrogel invasion assay, colonies larger than $150\;{\mu}m$ were regarded as positive ones and counted. Statistical analysis of migration ability and invasion properties of each cell lines was performed using t-test. Results : In scratch test to examine migration ability of each cell lines, U87MG cells were most motile than others, and U343MG-A least motile. The HAse was expressed in U251MG and U343MG-A cell lines. However, U87MG and U373MG cell lines did not express HAse activity. In Matrigel invasion assay, the cell lines expressing HAse (U251MG and U343MG-A) were more invasive in the presence of HA than HAse deficient cell lines (U87MG and U373MG). In HA hydrogel invasion assay, the HAse-expressing cell lines formed colonies more invasively than HAse-deficient ones. Conclusion : Malignant Glioma cells expressing HAse were more invasive than HAse-deficient ones in 3-dimensional environment. Therefore, it might be suggested that invasion of malignant gliomas is suppressed by inhibition of HAse expression or HA secretion. Additionally, the ability of 2-D migration and 3-D invasion might not be always coincident to each other in malignant glioma cells.
본 연구는 인간 유래 유방 암세포 MDA-MB-231를 대상으로 CA에 의한 세포사멸, 세포 이동 및 침윤 효과를 조사하였다. 암세포의 증식 억제 효과는 CA 농도 의존적으로 증식률이 감소하는 것을 확인하였다. CA에 의한 apoptosis 양성 세포를 확인하기 위해 DAPI stain를 진행한 결과, CA 농도 의존적으로 죽은 세포를 확인하였다. MDA-MB-231 세포에서 CA에 의한apoptosis marker 단백질 발현 증가와 ROS production증가를 확인할 수 있었다. 또한 CA에 의한 MDA-MB-231의 세포 이동률이 유의적으로 감소하는 것을 확인하였다. 마지막으로 세포의 이동과 전이 능력 또한 CA를 처리한 군에서 통계적으로 감소하는 것을 확인하였다. 본 연구 결과를 통해서 CA의 암세포 증식률 억제, 세포사멸 증가, 그리고 세포 이동 및 전이 억제 효과가 나타나는 것을 확인했으며, 이 결과를 통해서 향후 유방암에 대한 항암제로 개발될 수 있는 가능성을 가지고 있는 것으로 사료된다.
본 연구의 목적은 영유아교사의 정서표현성, 놀이교수효능감, 유아-교사 상호작용 간의 관계를 분석하고 정서표현성, 유아-교사 상호작용에 미치는 영향에 대해 알아보고자 한다. 이를 위하여 본 연구에서는 전라북도에 소재하고 있는 유아교육기관 13개소에 근무하고 있는 영유아교사 272명을 대상으로 설문 조사를 하였다. 본 연구에 사용 된 연구도구는 정서표현성 척도, 놀이교수 효능감 척도, 유아-교사 상호작용 척도이며 본 연구에 맞게 수정, 보완하여 사용하였다. 본 연구의 결과를 분석하기 위하여 SPSS 22.0 프로그램을 이용하였으며, 빈도와 백분율, Pearson의 적률상관관계, 중다회귀분석을 통해 자료를 분석하였다. 연구결과 첫째, 영유아교사의 정서표현성 중 긍정적 정서표현성, 놀이교수효능감, 유아-교사 상호작용 간에는 정적인 상관관계가 있는 것으로 나타났다. 그러나 정서표현성 중 부정적 정서표현성은 부적상관관계가 나타났다. 둘째 영유아교사의 정서표현성, 놀이교수효능감이 유아-교사 상호작용에 미치는 영향을 살펴 본 결과 모두 통계적으로 유의한 영향을 미치는 것으로 나타났다. 결론적으로 영유아교사의 정서표현성, 놀이교수 효능감, 유아-교사 상호작용 간에는 유의미한 상관이 있었으며, 두 변인 모두 유아-교사 상호작용에 영향을 미치는 요인임을 확인하였다는 점에서 본 연구의 의의를 찾을 수 있다.
Basic leucine zipper(bZIP) 단백질은 식물의 성장과 발달 그리고 스트레스 반응을 조절하는 전사인자이다. 본 연구에서는 현사시나무(Populus alba ${\times}$ P. glandulosa)에서 SE3 그룹에 속하는 bZIP인 PagbZIP1 유전자를 분리하여 구조와 발현 특성 등을 조사하였다. 현사시나무의 PagbZIP1 유전자는 844개의 염기쌍으로 이루어져 있으며, 144개의 아미노산으로 구성되는 예상 분자량 16.6 kDa의 단백질을 암호화한다. PagbZIP1은 보존영역인 basic domain과 leucine zipper domain을 가지고 있으며, 현사시나무의 염색체에 2 copy가 존재하는 것으로 나타났다. PagbZIP1은 현사시나무의 뿌리와 배양세포에서 특이적으로 발현하며, 배양세포의 생장주기에서는 정지기에 높게 발현하였다. 또한 건조와 염 그리고 저온 스트레스 뿐 아니라 식물호르몬인 ABA 처리에 의해서도 발현이 유도되어, ABA를 경유한 신호전달경로를 따라 유전자 발현을 조절하는 것으로 판단되었다. 본 연구결과는 PagbZIP1 유전자의 도입과 발현조절을 통한 바이오매스 증진 및 스트레스 내성 나무의 개발에 도움을 줄 것으로 생각된다.
온라인 분석 처리(OLAP, On-Line Analytical Processing)에서는 다양한 분석을 효과적으로 처리하기 위해, 다차원 구조의 데이타를 열에 차원 애트리뷰트의 값이 표시되는 넓은 형태의 수평 테이블로 표현한다. 관계형 테이블들은 보통 애트리뷰트의 개수에 제한이 있으므로 이러한 수평 테이블을 직접저장하기 어렵고, 저장하더라도 많은 수의 널(null) 간을 갖는 희박(sparse) 테이블이 되기 쉽다. 따라서 관계 데이타베이스에서는 이러한 수평 테이블을 차원의 이름을 열로 갖는 수직 테이블로 바꾸어 저장할 수 있다. 이렇게 할 경우, 수평 뷰 테이블에 대한 OLAP 질의를 저장된 수직 테이블에 대한 질의로 변환하는 작업이 필요하다. 본 논문에서는 전통적인 관계 대수 연산자들 외에 최근 DBMS 버전들이 제공하는 PIVOT 연산자를 사용하여 수평적인 뷰의 질의를 효율적으로 변환하는 방법을 제안하였다. 이를 위해 PIVOT 연산과 동등한 관계 대수식을 만들고 이를 증명하였으며, 이 PIVOT 연산을 사용하여 수평적인 테이블에 대한 질의를 수직적인 테이블에 대한 질의로 변환하는 방법을 제시하였다. 또한 실험을 통해 제안한 변환 방법이 기존 방법에 비해 성능이 더욱 우수함을 보였다.
Nitric oxide(NO) play an important role in normal and pathophysiological cells including as a messenger molecule, neurotransmitter, microbiocidal agent, or dilator of blood vessels and artheriosclerosis, hypertension, myocardial infarction, respectively. To investigate that Ondamtang in the potential contribution of the levels of nitric oxide generated by endothelial nitric oxide synthase (eNOS) and the mechanisms of protection against L-NAME, human ECV304 cells, which normally do not express eNOS, were expressed by L-NAME. L-NAME stimulated rat or cells were found to be resistant to injury and delayed death following the Ondam-tang. Inhibition of nitric oxide synthesis abolished the protective effect against L-NAME, thrombin and collagen exposure. Interestingly, such effects have bee observed during stimulation with agents such as KCl on L-NAME mediate rats, were damaged by the NOS inhibitor NG-nitro-L-arginine methyl ester (L-NAME). Cardiovascular diseases is one of the blood vessels and renin-angiotensin system dynfunction. So we studied on herbal medicine that have a relation of vessels endothelium necrosis. In Oriental Medicine, Ondam-tang has been used for disease in relation to cardiovascular system. We studied on the protection and inhibitory effects of cardiovascular diseases in L-NAME induced rat or ECV304 cell lines through the Cell morphological pattern, Tunel assay, LDH activity, heart rate, blood pressure and immunohistochemistric analysis by Ondam-tang. As the result of this study, In group, the anti-apoptosis and necrosis in the cardiovascular system have a potential capacity for prevented, protected and treating the diseases of cardiovascular system, against the necrosis of rat and ECV304 cells with eNOS and calpain expression by L-NAME is promoted.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
