Functional Characterization of the ${\alpha}$- and ${\beta}$-Subunits of a Group II Chaperonin from Aeropyrum pernix K1

  • We isolated and functionally characterized the ${\alpha}$- and ${\beta}$-subunits (ApCpnA and ApCpnB) of a chaperonin from Aeropyrum pernix K1. The constructed vectors pET3d-ApCpnA and pET21a-ApCpnB were transformed into E. coli Rosetta (DE3), BL21 (DE3), or CodonPlus (DE3) cells. The expression of ApCpnA (60.7 kDa) and ApCpnB (61.2 kDa) was confirmed by SDS-PAGE analysis. Recombinant ApCpnA and ApCpnB were purified by heat-shock treatment and anion-exchange chromatography. ApCpnA and ApCpnB were able to hydrolyze not only ATP, but also CTP, GTP, and UTP, albeit with different efficacies. Purified ApCpnA and ApCpnB showed the highest ATPase, CTPase, UTPase, and GTPase activities at $80^{\circ}C$. Furthermore, the addition of ApCpnA and ApCpnB effectively protected citrate synthase (CS) and alcohol dehydrogenase (ADH) from thermal aggregation and inactivation at $43^{\circ}C$ and $50^{\circ}C$, respectively. In particular, the addition of ATP or CTP to ApCpnA and ApCpnB resulted in the most effective prevention of thermal aggregation and inactivation of CS and ADH. The ATPase activity of the two chaperonin subunits was dependent on the salt concentration. Among the ions we examined, potassium ions were the most effective at enhancing the ATP hydrolysis activity of ApCpnA and ApCpnB.

포도전정가지 추출물이 UVB로 유도된 결합 조직 손상에 미치는 피부 보호 효과 (Skin Protection Effect of Grape Pruning Stem Extract on UVB-induced Connective Tissue Injury)

  • 본 연구는 포도수확 후 버려지는 가지를 이용한 포도전정가지 추출물(Grape Pruning Stem Extracts, GPSE)에 함유된 polyphenol류 중 항산화, 항염증, 항암 등의 효과가 있다고 알려진 rutin, procyanidin B3, quercetin, kaempferol의 함량을 분석하고, UVB로 유도된 HR-1 mice의 손상된 피부에 대한 교원섬유, 탄력섬유 파괴억제 효과를 측정하여 기능성 화장품 소재로서의 응용 가능성을 확인하고자 하였다. 포도전정가지에서 polyphenol 성분을 80% EtOH로 추출하고 여과하여 농축한 후, 동결건조하여 $-20^{\circ}C$에 보관하면서 사용하였다. GPSE의 유효성분 함량은 HPLC를 사용하여 분석하였다. 피부손상을 유도하기 위해 UVB를 HR-1 hairless mice에 조사하였고, 손상된 피부에 교원섬유 파괴억제 효과를 보기 위해 Masson's trichrome stain, 탄력섬유 파괴 억제 효과를 보기 위해 Verhoeff's elastic fiber stain, MMP-9 단백 발현을 보기 위해 면역조직화학적 염색을 실시하였다. 본 연구에 사용된 포도전정가지 시료 53 kg에서 EtOH 분획 추출물 2.34 kg을 추출하여 4.42%의 수율 결과를 얻었다. 유효성분 분석결과, procyanidin B3, 0.28 mg/g, rutin 12.81 mg/g, quercetin 0.51 mg/g 및 kaempferol 8.24 mg/g로 나타났다. GPSE와 serum base를 혼합 도포한 그룹에서 UVB에 의한 교원섬유와 탄력섬유를 분해하는 MMP-9 단백발현 결과, 대조군 대비 평균 약 90% 감소 억제하는 효과를 볼 수 있었다. 교원섬유 파괴억제 효과는 모든 농도에서 평균 24%의 억제율과, GPSE와 serum base의 혼합물 농도 2,000 mg/kg 에서 43%로 가장 우수한 교원섬유 파괴억제효과가 나타났으며, 탄력섬유파괴 억제 효과는 모든 농도에서 평균 12%의 억제율과 GPSE와 serum base의 혼합물 농도 1,000 mg/kg에서 35%로 가장 우수한 탄력섬유 파괴 억제 효과가 있는 것으로 나타났다. 이 결과는 GPSE가 진피층의 세포 외 기질에 UVB 조사에 의해 파괴되는 교원섬유, 탄력섬유의 파괴를 억제하는 작용을 한다는 것을 알 수 있었다. 이상의 결과로 포도전정가지로부터 rutin, kaempferol, quercetin, procyanidin B3 등의 폴리페놀성 화합물이 화장품 기초 원료인 serum base에 함유토록 제조된 GPSE 는 교원섬유 파괴 억제, 탄력섬유파괴 억제효과 및 피부손상 기능개선 효과의 결과를 얻을 수 있었다. 본 연구의 의의는 지역포도농가에서 수확 후 버려지는 포도전정가지에서 폴리페놀류와 같은 유효성분을 확보하여 화장품 원료로서의 활용 가치가 높을 것으로 기대된다.

점토에 대한 2개 염기성 염료 RB5와 GB4의 경쟁 흡착 (Competitive Adsorption of Two Basic Dyes RB5 and GB4 on a Local Clay)

  • 알제리에서 출토되는 소듐-교환된 Maghnia 점에 대한 2개 염기성 염료인 RB5와 BG4의 흡착평형을 조사한 결과, 최대 흡착능은 각기 465.13 와 469.90 mg/g 으로 나타났다. ADMI 방법을 사용한 BR5 and BG4의 동시 흡착 실험에서 3개 다른 초기 농도비R (R=$C_{(BR5)}/C_{(BG4)}$) 2.5/1, 1/1, 1/2.5 을 사용하였다. 염료혼합물로부터 등온흡착 여부를 조사하여 경쟁 흡착인지를 알수 있다. 점토 표면 위 흡착 활성자리를 위한 BR5 and BG4 간 매우 강한 상호작용(R = 1/1)이 관찰되었다. RB 5 와 BG 4의 흡착능의 비율 R' (R'=$Qe_{(mixture)}/Qe_{(single)}$)은 초기 농도 비율 R (R=$C_{(BR5)}/C_{(BG4)}$)이 2.5/1, 1/1 and 1/2.5 인 경우에 대해 각기 0.86, 0.74 and 0.84 로 얻어졌다. Langmuir 와 Freundlich 모델이 각기 염료 뿐만 아니라 염료 혼합물의 흡착 거동에 잘 들어 맞음을 알 수 있었다.

만형자(Vitex trifolia L.)로부터 항균활성물질의 분리 및 구조결정 (Isolation and Identification Antifungal Compounds from Vitex trifolia L.)

  • 합성농약의 단점을 보완하기 위하여 무공해, 무독성의 환경 친화적인 농약이 요구되고 있다. 특히, 식물 유래 천연물이 각광받고 있으며 이러한 관점에서 선행 실험 결과 강한 항균활성을 나타낸 만형자(Vitex trifolia L.)로부터 항균활성물질을 분리하여 기기분석을 통해 화학구조를 결정하고, 항균활성 검정을 실시하였다. 만형자를 80% aqueous MeOH로 추출한 후 n-hexane, EtOAc, n-BuOH 및 $H_2O$ 등 4개의 층으로 분획하였다. 이들 분획물을 이용하여 2000 ppm 수준으로 6가지 식물병원균에 대해 in vivo 항균활성을 검정한 결과, n-hexane 분획물이 벼 도열병균에 대해 88%, EtOAc 분획물이 벼 도열병균에 대해 85%, 밀 붉은녹병에 대해 87%의 방제효과를 나타내었다. Silica gel column, Sephadex LH-20 column chromatography, preparative HPLC를 사용하여 단일 화합물로 분리한 다음, ESI-MS, $^1H$-NMR, $^{13}C$-NMR의 기기분석 결과를 해석하여 agnuside, chrysosplenol B, artemetin로 구조를 동정하였다. Chrysosplenol B는 만형자로부터 처음 분리되었다. 분리한 물질들의 in vitro 및 in vivo 항균활성에 대한 연구를 진행하고 있다.

브로콜리 새싹 용매 추출물의 항산화 및 면역조절 활성 (Anti-oxidative and Immunomodulating Activities of Solvent Extracts from Broccoli (Brassica oleracea) Sprouts)

  • In order to examine the functionality of broccoli sprout (Brassica oleracea, BS), solvent extracts were prepared and their anti-oxidative and immunomodulating activities were compared with those of broccoli (B). EtOH extracts (E) were potently higher than hot-water extracts (HW) in the antioxidant contents and radical scavenging activity. In particular, the total polyphenolic contents in addition to ABTS and DPPH radical scavenging activity were significantly higher in EtOH extract of broccoli sprout (BS-E; 9.15 mg GAE/g, 4.52 mg AEAC/g, and 1.14 mg AEAC/g) compared with that of broccoli (B-E; 7.83 mg GAE/g, 3.63 mg AEAC/g, and 0.97 mg/AEAC/g). Whereas, total flavonoid content was significantly higher in B-E (1.60 mg QE/g) than BS-E (1.43 mg QE/g). Anti-inflammatory activity was investigated using LPS-induced cell line model at a concentration of $10{\sim}100{\mu}g/mL$, in which all solvent extracts of both broccoli sprouts and broccoli were not toxic to RAW 264.7 cell lines. In anti-inflammatory activity of broccoli sprouts, EtOH extracts also showed significantly more potent activity than hot-water extracts in all sample concentrations tested. In addition, BS-E ($100{\mu}g/mL$) inhibited nitric oxide (NO) and IL-6 production to 60.9% and 68.9% compared with the LPS inflammation group (without extracts), whereas B-E inhibited 49.6% and 54.9%. On the other hand, in immunostimulating activity by splenocytes and macrophages, hot-water extract showed significantly higher activity than EtOH extract. Especially, BS-HW stimulated the splenocyte proliferation (1.2-fold against saline group) and IFN-${\gamma}$ production (264.39 pg/mL) at $100{\mu}g/mL$, and the production of IL-6 (1.33-fold), IL-12 (1.09-fold) and TNF-${\alpha}$ (1.49-fold) from macrophages was also significantly enhanced over broccoli. In conclusion, broccoli sprouts showed more potent anti-oxidative, anti-inflammatory and immunostimulating activity than broccoli, suggesting the possibility of using broccoli sprouts as functional food materials.

소나무와 해송 화분의 화학적 성분과 파쇄 조건에 관한 연구 (Chemical Components and Pulverization Conditions of the Pollens of Pinus densiflora S. et Z. and Pinus thunbergii Parl)

  • 소나무와 해송 화분에 대하여 화학적 성분을 분석하고 화분 파쇄 최적 조건을 구명하였던 바 다음과 같은 결과를 얻었다. (1)소나무 화분의 일반성분 함량은 수분 9.8%, 조회분 2.3%, 조단백질 11.9%, 조지방 2.9%, 조섬유 7.7%, 당질 65.4%이었으며, 해송 화분은 수분 10.5%, 조회분 2.2%, 조단백질 13.5%, 조지방 3.5%, 조섬유 8.5%, 당질 61.8%로서 일반곡류보다 높게 나타났다. (2)소나무와 해송 화분에서 각각 18종의 아미노산이 확인되었으며, 특히 10종의 필수아미노산을 포함하고 있어 영양원으로서 가치가 높은 것으로 나타났다. (3)소나무 화분은 비탄민 A 7.9㎎, B1 8.7㎎, B2 19.2㎎, C 31.5㎎, E 2.0㎎, 해송 화분은 비타민 A 8.5㎎, B1 9.4㎎, B2 15.7㎎, C 35.3㎎, E 1.8㎎ 함유하고 있었으며, 특히 비타민 C가 풍부하였다. (4)소나무 화분립은 5,000rpm으로 40분간, 해송 화분립은 5,000rpm으로 50분간 파쇄함으로써 100% 화분 파쇄가 가능하였다.

쥐눈이콩-노루궁뎅이버섯 균사체 발효물의 생리활성 (Physiological Activity of the Fermented Small Black Soybean (Rhynchosia volubilis) with a Solid State Culture of the Bearded Tooth Mushroom (Hericium erinaceum) Mycelia)

  • To enhance the physiological activity of the Rhynchosia volubilis (RV), R. volubilis (RVHE-A) and R. volubilis-added herbal powder (RVHE-B) were fermented with a solid state culture of Hericium erinaceum mycelia (HE). The total isoflavone contents of the non-fermented RV-A ($489.9{\mu}g/g$) and RV-B ($571.1{\mu}g/g$) were remarkably increased in fermented RVHE-A ($1,836.4{\mu}g/g$) and RVHE-B ($1,276.7{\mu}g/g$). In particular, aglycone isoflavones such as daidzein and genistein were significantly higher in the RVHE-A than any other sample. When hot-water (HW) and EtOH extracts (E) were fractionated from the RV and RVHE, both extracts from the RVHE-A were higher than those from the RV-A in total polyphenol and flavonoid contents. However, the RVHE-B-HW showed a lower polyphenol and flavonoid content level than did RV-B-HW. RVHE-A-HW and -E also had more potent ABTS radical scavenging activity than any extract from the non-fermented RV and other ferments (RVHE-B). In the meanwhile, RVHE-A-HW potently stimulated the production of macrophage activation-related cytokines such as $TNF-{\alpha}$, IL-6 and IL-12 ($841.7{\pm}71.3pg/mL$, $3.9{\pm}0.1ng/mL$, $179.3{\pm}30.2pg/mL$) from peritoneal macrophage more than RV-A-HW ($92.5{\pm}1.5pg/mL$, $0.1{\pm}0.0ng/mL$, $37.4{\pm}5.4pg/mL$) as well as RVHE-B-HW ($557.0{\pm}21.3pg/mL$, $1.8{\pm}0.0ng/mL$, $90.0{\pm}10.0pg/mL$). However, all the EtOH extracts did not show significant activity. In addition, the RVHE-A-HW showed a significantly higher intestinal immune system modulating activity through Peyer's patch and GM-CSF production than did any other extract from RV and RVHE-B. In conclusion, these results suggest that the fermented R. volubilis with H. erinaceum mycelia possesses a possible use as an industrial application as functional food or material.

Isolation and Quantitative Analysis of Chemical Constituents in Codonopsis lanceolata

  • Codonopsis lanceolata has numerous chemical constituents that includes polyphenols, saponins, tannins, triterpene, alkaloids, and steroids. The extract of C. lanceolata was partitioned with Haxane, CH2Cl2, EtOAc, n-BuOH and MeOH. The determination of structure for lancemaside G, lancemaside B, lancemaside A were based on physicochemical and HPLC chromatogram data, including NMR and HR-MS. In addition, tangshenoside I and lobetyolin were identified in the material separation process for the extract of C. lanceolata, and content analysis was performed using HPLC. The compounds were confirmed as lancemaside G, lancemaside B, lancemaside A, tangshenoside I, and lobetyolin.

HDDM, a formula consisting of seven herbs, had anti-diabetic but no immunomodulatory activities in multiple low doses of streptozotocin-treated female of B6C3F1 mice

  • The objectives of this study were to determine the effect of herb formula HDDM, a modification of Huangdan decoction that has been shown to be effective in the treatment of glomerulonephritis and chronic renal failure, on the blood glucose levels in multiple low doses (MLD; 50 mg/kg for five consecutive days) of streptozotocin (STZ)-treated female B6C3F1 mice. Initial studies were performed to compare diabetes induction in five strains (e.g., B6C3F1, NOD, CD-1, C3H/HeN and C57BL/6) of mice by MLD-STZ, and immune changes following the treatment. The results suggested that the order of susceptibility to diabetes induction was NOD $\approx$ CD-1 > B6C3F1 $\approx$ C3H > C57BL/6. Furthermore, STZ modulation of T cell development, differentiation and activation might play a role in diabetes induction by MLD-STZ treatment. MLD-STZ-induced diabetes in female B6C3F1 mice was moderate, which allowed the evaluation of drug-induced protection or exacerbation of diabetes to be performed. As such, modulation of blood glucose by HDDM, which consisted of Da Huang (Radix Et Rhizoma Rhei), Huang Qi (Radix Astragali Seu Hedysari), Dan Shen (Radix Salviae Miltiorrhizae), Yin Yang Huo (Herba Epimedii), Yi Yi Ren (Semen Coicis or Coix lacryma-jobi), Mai Dong (Radix Ophiopogonis) and Shan Zhu Yu (Fructus Corni), was evaluated in MLD-STZ-treated female B6C3F1 mice. The results suggested that HDDM could lower the blood glucose levels, but it had no immunomodulatory activities. Additionally, HDDM-treated mice exhibited improved glucose tolerance. In conclusion, these studies have suggested that MLD-STZ-induced diabetes in female B6C3F1 mice is a useful model to evaluate drug modulation of diabetes, and that the herb formula HDDM possesses anti-diabetic effects.

Numerical simulation of the thermoelectric behavior of CNTs/CFRP aircraft composite laminates

  • The present paper focuses on the development of a model for simulating the thermoelectric behavior of CNTs/CFRP Organic Matrix Composite (OMC) laminates for aeronautical applications. The model is developed within the framework of the thermodynamics of irreversible processes and implemented into commercial ABAQUS Finite Element software and validated by comparison with experimental thermoelectric tests on two types of composites materials, namely Type A with Carbon Nanotubes (CNT) and Type B without CNT. A simplified model, neglecting heat conduction, is also developed for simplifying the identification process. The model is then applied for FEM numerical simulation of the thermoelectric response of aircraft panel structures subjected to electrical loads, in order to discuss the potential danger coming from electrical solicitations. The structural simulations are performed on quasi-isotropic stacking sequences (QI) $[45/-45/90/0]_s$ using composite materials of type A and type B and compared with those obtained on plates made of metallic material (aluminum). For both tested cases-transit of electric current of intermediate intensity (9A) and electrical loading on panels made of composite material-higher heating intensity is observed in composites materials with respect to the corresponding metallic ones.
