인삼이 흰쥐의 성 행위에 미치는 영향 (Influence of Panax Ginseng upon mating Behavior of Male Rats)

  • The influence, upon male rat's mating behavior, of Korean Panax Ginseng administered for 3 and 5 days was investigated by direct behavioral observation and also by counting the number of copulation plugs the animals deposited. Four쇼-four male albino rats were used. Part of the animals received ginseng for 3 days (ginseng-3 day group, N= 12) or for 5 days(ginseng-5 day group, H=10), while the remaining animals received saline for 3 days (saline-3 day group, N=12) or for 5 days (saline-S day group, N=10). Each animal belonging to the 2 ginseng groups received subcutaneously 0.5 ml/100 g body weight of ginseng alcohol extract solution (4 mg of the ethyl alcohol extract in 1 ml of physiological saline), and each rat belonging to the 2 saline groups received the same amount of saline per day. During the dark period of the light-dark cycle on the next day following the last drug administration, a female rat in the artificial estrus was introduced to each male and the mating behavior was observed for 45 minutes. The observation session was divided into two parts and, in the early part which terminated with the first ejaculation and succeeding intromission, following behavioral measures were taken: mounting latency, intromission latency, inter-intromission period, ejaculatory latency(time from the first intromission until the first ejaculation), occurrence of mounting with intromission, occurrence of mounting without intromission, and postejaculatory interval. Behavioral measures taken in the later part of the session after the first ejaculation were: occurrence of mounting with intromission, occurrence of mounting without intromission, and occurrence of ejaculation. Immediately after the behavioral observation session the experiment turned to measure, for 10 days, the number of copulation plug which each pair of rats deposited. Following results were obtained: 1. After several mountings mounting with intromission, males of the 2 ginseng groups finished the first ejaculation significantly earlier than the corresponding 2 saline groups did. 2. The postejaculatory latency was significantly reduced in the ginseng-5 day group compare with the value of the saline-5 day group and also compared with the value of the ginseng-3 day group. 3. The 2 ginseng groups ejaculated significantly more often in 45 minutes' observation session than the corresponding 2 saline groups did. 4. The number of copulation plug deposited in 10 days by the animals of the 2 ginseng groups. significantly exceeded the number deposited by the corresponding 2 saline group animals. The animals of the ginseng-5 day group deposited copulation plugs significantly more than the animals of the ginseng-3 day group did. It is inferred from the above results that the ginseng facilitates mating behavior of male rats, and that the degree of facilitation may be influenced by the duration of drug administration.

Monitoring the Reproductive Status of Dairy Cows by Urinary Pregnanediol Glucuronide

  • This study was undertaken with the aim to establish a reliable radioimmunoassay (RIA) system for urinary pregnanediol glucuronide (PdG) and to employ it for monitoring the reproductive status of dairy cows. Urine and blood samples were collected from the Holstein cows both pregnant and non-pregnant. The samples were then investigated for evaluating the relationship between progesterone ($P_{4}$) in blood and PdG in urine adjusted with or without urinary creatinine basis. Biweekly urine collection was employed for three cows in estrous and those artificially inseminated, while urine from pregnant cows was collected on a monthly basis. P_{4}$ and PdG levels were measured by enzymeimmunoassay (EIA) and RIA techniques, respectively. Our results indicated the sensitivity of PdG for RIA being 35 pg/tube and the recovery rate of 100%. Urinary creatinine concentrations also fluctuated within a day, but change at midday was not noteworthy. Regardless of the time of urination the change in concentrations of PdG was relatively smaller and did not vary significantly. The urinary PdG concentration showed periodic changes as that with serum P_{4}$ levels during the cow's estrus cycle. The correlation coefficient rose when creatinine level in urine was adjusted but the change was also not significant. The concentrations of PdG during the luteal phase were detected between 8.2 and 17.4 ng/ml, three to five times higher than that in the follicular phase. The concentration of PdG from pregnant cows (21 days after conception) was three to four times higher than in the nonpregnant cows. Our finding suggests that the determination of urinary PdG could be reliably employed for early pregnancy detection. The urinary PdG level continued to raise until 30 days pre-partum while the concentration reached its peak at 30 ng/ml, after which it started to fall 18 to 30 days before parturition and finally fell to its nadir value one week after parturition. As the correlation coefficient between the urinary PdG and serum P_{4}$ was higher than that corrected by urinary creatinine it can be suggested that the adjustment is not needed. The concentrations of urinary PdG could be maintained stably for 2 days in urine samples stored at room temperature and extended to 8 days when the samples were pretreated by boiling for 30 minutes. In conclusion urinary PdG concentration even without the need for creatinine basis adjustment can be used directly for monitoring the reproductive status of dairy cows.

한우에서 신체충실지수(BCS)가 다배란 처리 시 체내 수정란 생산과 수태율에 미치는 영향 (Effect of Body Condition Score (BCS) on In Vivo Embryo Production and Pregnancy Rate Following Superovulation in Hanwoo)

  • Body condition score (BCS) is a useful management tool for distinguishing differences in nutritional needs of cows in the herd. Although it is not always possible to quantify the nutrient content of the feed supplied to the donor cow, the nutritional status can be determined by the BCS. The objective of this study was to evaluate in vivo embryo production, return to estrous of donor and pregnancy rate of recipients following BCS in Hanwoo superovulation. Sixty nine Hanwoo donor cows were flushed on day 7 of estrus cycle with same FSH and artificial insemination by the same technicians. Embryos were recovered on 7 days after the third insemination by flushing the uterus with Embryo Collection Medium. The results obtained were as follows: No differences were observed in the efficiency of superovulation rates regardless of BCS ($${\leq_-}2.0$$, 2.5 to 3.0, and $${\geq_-}3.5$$). The mean number of total embryos was each $5.20{\pm}0.86$, $11.56{\pm}1.04$, and $6.23{\pm}1.07$. The mean number of transferable embryo from $${\leq_-}2.0$$, 2.5 to 3.0, and $${\geq_-}3.5$$ of BCS was $2.60{\pm}0.87$, $7.94{\pm}0.89$, and $4.75{\pm}1.32$, respectively (p<0.05). Return to estrous regardless of donor BCS was no difference. The pregnancy rates of recipient were BCS $${\leq_-}2.0$$ 11.76%, 2.5 to 3.0 40.79%, and $${\geq_-}3.5$$ 11.11%, following transfer of fresh embryos produced in vivo, respectively. These results indicate that if the Hanwoo with BCS 2.5 to 3.3 are used for donor and recipient, the embryo production and the conception rate will be greater.

Relationship between Transferable Embryos and Major Metabolite Concentrations in Holstein Donor Cows

  • This research was investigated the relationship, in high-producing Holstein donor cows, between the number of the transferable embryos and the blood serum concentrations of Blood Urea Nitrogen (BUN), glucose and cholesterol, which affect the nutritional state of cows. CIDRs were inserted into the vaginas of twenty two heads of Holstein cows, regardless of estrous cycle. Superovulation was induced using folliclar stimulating hormone (FSH). For artificial insemination, donor cows were injected with $PGF_{2{\alpha}}$ and estrus was checked about 48 hours after the injection. Then they were treated with 4 straws of semen 3 times, with 12-hour intervals. Embryos were collected by a non-surgical method 7 days after the first artificial insemination. The total numbers of ova collected from 3 experimental groups whose blood BUN concentrations were <10 mg/dl, 11~18 mg/dl and ${\geq}19$ mg/dl were 8.9, 12.5 and 19.0, respectively; whereas the numbers of transferable embryos were 5.8 + 1.9, 7.9 + 2.8 and 5.2 + 1.4, respectively. When glucose concentration was <60 mg/dl, the total number of collected ova was 9.9, which was smaller than when the concentration was 60~70 mg/dl or ${\geq}70$ mg/dl. When glucose concentration was 60~70 mg/dl, the number of transferable embryos was 7.1 + 2.4, which was slightly larger than the numbers 6.4 + 2.1 and 6.1 + 1.7 that were obtained when the concentrations were <60 mg/dl and ${\geq}70$ mg/dl, respectively ; however, the differences were not significant (p>0.05). When cholesterol concentrations were <150 mg/dl, 150~200 mg/dl and ${\geq}200$ mg/dl, the total numbers of collected ova were 11.2, 11.3 and 8.6, respectively. Whereas the numbers of transferable embryos were 7.1 + 2.1, 7.3 + 1.9 and 5.6 + 1.3, respectively ; however, the differences were again not significant (p>0.05). The result of this research showed no significant difference in ovum recovery rate and the number of transferable embryos according to major metabolite concentrations in high-producing Holstein donor cows. However, it is considered that the failure of maintaining proper nutritional status would cause the fall in in vivo embryo productivity.

자궁평활근의 Carbachol 및 Oxytocin 수축에 있어서의 세포내 $Ca^{2+}$ 동원 (Intracellular $Ca^{2+}$ Movement in Contraction Induced by Carbachol and Oxytocin in Rat Myometrium)

  • The properties of cytosolic $Ca^{2+}$ level$([Ca^{2+}]_i)$ movement of high KCl, carbachol and oxytocin were examined with myometrium isolated from non-pregnant rat(estrus cycle). High concentration of KCl$({\leq}23.3mM)$ induced rhythmic increases in $[Ca^{2+}]_i$ and muscle contraction. However, sustained $[Ca^{2+}]_i$ and contracion were obtained at higher KCl concentration $({\geq}30.3mM)$ The rhythmic and sustained contraction closely associated with changes in $[Ca^{2+}]_i$ induced by high KCl. Carbachol $(3{\sim}30{\mu}M$ generated rhythmic increases with tonic component in $[Ca^{2+}]_i$ and muscle contraction. Myometrial contraction stimulated by carbachol was also closely correlated with change in $[Ca^{2+}]_i$. And the $[Ca^{2+}]_i/contraction$ relationships were similar when muscle strips were stimulated by high KCl and carbachol. Maximal concentration of carbachol $(10{\mu}M)$ and oxytocin(100 nM) increased $[Ca^{2+}]_i$ and contraction which were slightly greater than that of high KCl in non-pregnant myometrium, respectively. However, the $[Ca^{2+}]_i$ and contraction were strongly inhibited by verapamil $(10{\mu}M)$, a 1-type $Ca^{2+}$ channel blocker, as in the case of high KCl. Additionally, although carbachol further increased $[Ca^{2+}]_i$ and contraction induced by high KCl, these changes also strongly inhibited by application of verapamil. These results suggest that uterotonic agents, carbachol and oxytocin, induced contraction by increase in $[Ca^{2+}]_i$ through $Ca^{2+}$ influx than by a regulation of $Ca^{2+}-sensitization$ in non-pregnant myometrium.

액상정액을 이용한 인공수정시 품종, 계절, 인공수정 횟수 및 정자농도가 번식성적에 미치는 영향 (Effects of Breeds, Insemination Time, Breeding Season, Sperm Concentration on Reproductive Performance of Sows Inseminated by Liquid Boar Semen)

  • 돼지에서 액상정액을 이용한 인공수정이 번식성적에 미치는 영향을 구명하기 위하여 축산기술연구소 종축개량부 (충남, 성환) 돼지 인공수정센터에서 사육중인 인공수정용 종모돈을 이용하여 1995년부터 2000년까지 액상정액을 이용한 인공수정시 종모돈의 품종, 인공수정 횟수, 계절 및 정자농도가 번식성적에 마치는 영향을 조사하여 돼지 인공수정시 번식성적 향상과 실용화에 기여하고자 본 연구를 실시하였다. 액상정액 인공수정시 정액을 생산한 종모돈의 품종 (Landrace, Yorkshire, Duroc)간 번식성적은 유의차가 인정되지 않았다. 발정당 2회 또는 3회 인공수정 시에도 번식성적에 차이가 없었다. 인공수정을 실시한 계절별로 번식성적을 조사하였으나 계절간 차이점은 발견할 수 없었다. 액상정액 인공수정시 1회 주입정자의 최저농도를 구명코자 활력 70% 이상인 정자수를 기준으로 80 ml 병당 정자농도를 3.0, 2.5, 2.0 및 $1.5{\times}10^9$로 조절하여 인공수정한 결과 수태 율, 분만 율, 총산자수 및 생존산자수에서 통계적인 유의차가 인정되지 않아 1회 주입정자농도는 운동성 정자 기준으로 $1.5{\times}10^9/80ml$ 이상으로 조절하면 인공수정에 실용적으로 사용할 수 있음을 보였다.

과배란 처리에 있어 성감별 정액을 이용한 한우 체내 수정란의 생산 효율 (Efficiency of In Vivo Embryo Production following Superovulation with Sex-soted Semen in Hanwoo (Korean Native Cattle))

  • Sexed semen is commonly used for the production of calves of the desired gender. Gender selection is important in animal production industries. For example, female cattle are required for the dairy industry while males are preferred in the beef cattle industry. The present study was to assess the in vivo embryo production efficiency using the semen separated according to sex during superovulation in Hanwoo. Seventy Hanwoo donor cows were flushed on day 7 of estrus cycle with same FSH and artificial insemination by the same technicians. Embryos were recovered on 7 days after the third insemination by flushing the uterus with embryo collection medium. KPN semen straws used artificial insemination contained 20 million sperm (total number 60 million per donor). Sex-sorted semen straws contained 4 million sperm (total number 12 million per donor). The results obtained were as follows: No differences were observed in the efficiency of superovulation rates on KPN semen 87%, and sexed semen 100%, respectively. The mean numbers of total embryos are each $12.58{\pm}8.31$ and $13.25{\pm}7.86$. The mean numbers of transferable embryos, sexed semen were significantly lower than KPN semen ($3.75{\pm}1.98$ vs. $8.23{\pm}6.07$, P<0.05). The rates of unfertilized embryos from superovulation using sexed semen were significantly higher than KPN semen (50% vs. 15%, P<0.05). The rate of degenerated 2-cell embryos from sexed and KPN semen was 60.87% and 11.11%, respectively (p<0.05). In conclusion, these results indicate that superovulation using sexed semen was useful, but efficient embryo production was important to reducing the damage caused by the Flowcytometer-based sperm sorting procedure.

The Use of Radioimmunoassay to Moniter Reproductive Status of Cheju Native Cattle and the Effect of Su, pp.ementary Feeding on Reproduction 1. Body Weight Changes, Breeding Performances and Progesterone Levels from Weaning until First Calving

  • Studies were conducted to investigate the relationship between hormone leveles and nutritional levels for improving performance of Cheju native cattle. In June 1984 a trial was initiated using 8 Cheju native calves after weaning, fed at two su, pp.ementary feeding levels (NRC 100% and 70%). The body weight, breedng performence, change in progesterone level during pregnancy and estrus cycle were evaluated. Mean body weight at 6 months of age was 155kg when fed 100% NRC ration but it was only 137kg when heifers received the 70% NRC ration. At 10, 15 and 20 months of age the body weight was 66, 160 and 115kg, respectively, showing that heifers fed the standard ration gained weight rapidly (P<0.01). Average size of the lefe ovary in the standard group was 2.1${\times}$1.6cm and right ovary was 2.6${\times}$1.8cm. However in the restricted feeding group the ovaries were found to be smaller. Diameter of graffian follicles showed a similar tendency to ovarian size in the two groups. The first oestrus in the standard feeding group a, pp.ared at 14.6 months when body weight was 265kg. Age at first calving was on average 28.9 months at a body weight of 436kg. On the other hand when heifers were fed the restricted ration the first oestrus a, pp.ared at 23.0 months at a body weight of 250kg. Average age at first calving was 38.9 months which was 10 months later than the average in the standard feeding group (P<0.01). In standard feeding group the progesterone level was 2.0ng/ml at two weeks after pregnancy and gradully increased up to 4 weeks and peaked at 18 weeks. This peak (6.4-6.5ng/ml) was maintained up to 24 weeks when progesterone level decreased until it reached 2.1ng/ml at the end of pregnancy. In the restricted group progesterone level up to 16 weeks followed a similar pattern to the standard group but there was a tendency in the restricted group to have lower progesterone levels(P<0.01). The standard and srstricted groups showed similar patterns of progesterone concentration during the oestrus cylce. There were no statistically significant differences in progesterone levels between standard and restricted groups but there was variation between induvidual animals.

RIA 및 CIA에 의한 혈장과 우유내 Progesterone 측정 (Measurement of Progesterone in Plasma and Milk by RIA and CIA)

  • 본 연구는 국립종축원과 경기도 남양주시의 개인 농장으로 부터 얻은 혈장과 유즙은 발정전 5일부터 시작 하여 2일 간격으호 30일간 채취하여 RIA와 CIA로 혈장, 전유 그리고 탈지유내의 progesterone을 측정 하여 발정주기중의 progesterone 농도 변화를 조사하였으며, 아울러 RIA와 대체할 수 있는 비방사성 면역 분석법인 CIA방법의 적용가능성을 조사하기 위하여 탈지유내 progesterone을 CIA와 RIA로 옹시에 측정하였던 바 다음과 같은 결과를 얻었다. 1. 혈장내 progesterone 수준은 발정전 8일까지 낮은 수준을 유지하다가 그후 급격히 증가하여 발정전 14일에 최고수준인 6.3ng/ml에 도달하였다. 그후 감소 하기 시작하여 발정전 16일에는 3.6ng/ml, 18일에는 1.5 ng/ml, 20일에는 2.2 ng/ml로서 최고수준을 나타냈다. 2. 임산우의 전유내 progesterone 농도는 발정일에 0.1 ng/ml에서 점차 증가하기 시작하여 발정후 10일 부터는 11ng/ml ~ 21ng/ml 까지의 높은 수준을 계속 유지하였다. 3 비임산우의 경우 전유내 progesterone농도는 발정일에 1.5 ng/ml 로서 낮은 수준을 나타냈으며 발정후 6일부터는 갑자기 증가하기 시작하며 발정후 16일까지 17.8~20.0 ng/ml까지의 높은 수준을 유지하다가 18일 이후부터는 1.4 ng/ml로서 낮은 수준을 유지하였다. 4. 탈지유내 progesterone수준은 임산우의 경우 발정개시때에 130-490pg/ml로서 낫은 수준을 유지하다가 발정 10일 이후에는 990-1650 pg/ml 까지의 높은 수준을 계속해서 유지했다. 5. 비임산부의 경우 탈지유내 progesterone 농도는 발정일에 160 pg/ml 로서 낮은 수준을 유지하다가발정후 8일부터 16일까지 1050-1300 pg/ml 까지 높은 수준을 오르내렸다 그후 탈지유내 progesterone수준을 240~450 pg/ml의 낮은 수준으로 나타났다. 6. 탈지유내 progesterone 농도를 CIA 와 RIA 방법으로 동시에 측정하여 상간관계플 조사하였던 바 좋은 상간관계(r=0.914)를 보였다.

젖소 과배란 처리시 PEG(Polyethylene Glycol) 처리가 수정란 생산에 미치는 영향 (Effects of PEG on Embryo Production in Superovulated Hostein Cows)

  • 본 연구는 젖소에 있어서 다량의 수정란을 생산하여, 고능력의 유전인자를 조기에 확산할 수 있도록, 젖소에 있어서 PEG를 이용하여 과배란 처리기법 확립을 위해 실시하였으며, 그 결과는 다음과 같다. 1. 성선자극호르몬의 처리 방법에 따른 황체반응은 Folltropin 단독처리시 $5.8\%$, Folltropin+EB는 $20.6\%$, Folltropin+PEG는 $24.0\%$로 나타났으며, 유의적인 차이를 보였다(p<0.01). 2. 성선자극호르몬의 처리 방법에 따른 이식 가능 수정란 수에 미치는 영향은 Folltropin V 단독처리시 3.6개, Folltropin +EB 처리시 3.3개, Folltropin +PEG 8,000 처리시 2.8개이었으며, 유의적인 차이를 나타내지 않았다(p<0.01). 3. 성선자극호르몬의 처리방법에 따른 퇴행란과 미수정 란 Folltropin+EB와 Folltropin+PEG 처리구에서 Folltropin 단독처리보다 많이 회수되었으며,배반포 수정란의 수에 있어서는 처리간에 유의적인 차이를 보이지 않았다. 이상의 결과는 임의의 발정주기에 CIDR 삽입후 Folltropin의 1일 2회 4일간의 분할 투여법 뿐만 아니라 Folltropin을 PEG로 용해하여 1회 투여법을 이용한 과배란 처리기법도 젖소 공란우에서 수정란의 생산이 가능함을 보여준다.
