마이크로파 조사에 의한 세포 미세구조의 고정 효과 (Fixation of Cellular Ultrastructure by the Microwave Irradiation)

  • The microwave fixator has recently been introduced in morphological research. The present study was carried out to investigate the ultrastructural effects of microwave fixation of rat brain. kidney, liver and skeletal muscle tissues. The results are as follows: In the case of microwave fixed cerebrum. the cytoplasmic processes of neurons and the various membranous organelles such as nuclear envelope, mitochondria, rough endoplasmic reticulum and Golgi apparatus were well preserved, The myelin sheath wrapping neuronal axon was prominent. Microwave fixed hepatocytes showed the microvilli on the free surface of bile canaliculus, the evident nucleolar components, and typical organelles. In nephron, ultrastructures of glomerulus and Bowman's capsule were preserved, and also tubular wall were structurally observed. Among the skeletal muscle cells, plentiful collagen fibers were appeared, myofibrils and mitochondria were typically observed. In conclusion, the microwave fixation procedures result in an good preservation of the tissues and would be time- and reagent-saving.

Random Amplitude Variability of Seismic Ground Motions and Implications for the Physical Modeling of Spatial Coherency

  • An initial approach for the identification of physical causes underlying the spatial coherency of seismic ground motions it presented. The approach relies on the observation that amplitude and phase variability of seismic data recorded over extended areas around the amplitude and phase of a common, coherent component are correlated. It suffices then to examine the physical causes for the amplitude variability in the seismic motions, in order to recognize the causes for the phase variability and, consequently, the spatial coherency. In this study, the effect of randomness in the shear wave velocity at a site on the amplitude variability of the surface motions mi investigated by means of simulations. The amplitude variability of the simulated motions around the amplitude of the common component is contained within envelope functions, the shape of which suggests, on a preliminary basis, the trend of the decay of coherency with frequency.

  • Advances in the technology of computational design are giving architects and engineers the opportunity to analyze buildings with complex geometries. This study explores the optimization and automation process using the parametric design method, and uses digital tools to achieve surface representation and panelization for curved shaped office buildings. In this paper, we propose parametric algorithms of dimensional and geometric constraints using the Knowledge-ware scripts embedded in Gehry Technologies' Digital Project. The knowledge-based design methods proposed in this study can be used to systemize the knowledge possessed by experts in the form of data. Such knowledge is required to promote collaboration between designers and engineers in the process of CAD/CAE/CAM. The aim of this study is to integrate the process into design, which establishes an integrated process. This integration enables two-way feedback between design and construction data by combining the methods used in designing, engineering, and construction.

  • Glossy architectural concrete means high qulity concrete which develops marble-like gloss on the surface of concrete. In spite of many benefits of architectural concrete to R.C. structure such as appearance of natural stone, saving of envelope materials and reduction of building weight, the failure of in-situ application of architectural concrete by the absence of knowledge on the physical properties and form work for it can often be found in Korea. This study is to offer the basic materials on the mix proportion, vibrating, curing and treatment of concrete result from the mock up test and in-situ application of architectural concrete to embody high quality architectural concrete in Korea.

The D/H ratio of N2H+ in the inner envelope of YSOs

  • Recent interferometric observations toward embedded protostellar systems show a clear offset between the emission peaks of N2D+ and N2H+. However, the chemical model that con-siders solely freeze-out and desorption from the dust grains could not reproduce the ob-servations. This difference between two spe-cies in the depletion zone might be caused by the reduction of the deuterated molecules, due to the reactions on grain surfaces. We present that the abundance offset between N2H+ and N2D+ can be explained if the chemi-cal model includes the surface chemistry, especially for the deuterated.

MCC 모델 및 Hvorslev-MCC 모델의 비교 연구 (Study on MCC and Hvorslev-MCC Models)

  • 본 연구에서는 한계상태이론에 기초하며 비교적 적은 매개변수를 활용하도록 최근에 개발된 Hvorslev-MCC 모델 및 전통적 MCC 모델의 거동예측 능력을 엄격하게 수행된 삼축압축시험 결과와 비교하여 고찰하였다. 모델예측 결과의 차이는 과압밀점토 시료에서 주로 발생하였다. Hvorslev-MCC 모델은 과압밀점토 시료의 첨두강도를 매우 정확히 예측하였으며, 이는 한계상태 건조 측의 항복면에 단순 타원이 아닌 Hvorslev 면을 활용하였기 때문이다.

Expression of Newer Outer Membrane Proteins (OMPs) Induced by Cephalosporins and Quinolone Group of Antibiotics in Klebsiella pneumoniae

  • Effect of antibiotics belonging to three different groups, including third generation cephalosporins, aminoglycosides, and quinolones, on the outer membrane protein (OMP) profile of Klebsiella pneumoniae was examined. It was found that a new OMP (porins) of 40 kDa molecular mass was expressed in Klebsiella pneumoniae, when grown in the presence of ceftazidime, whereas new proteins with 30 kDa and 22 kDa masses were detected in the presence of ofloxacin. The immunoblot analysis showed that the new proteins of 40 kDa and 30 kDa molecular masses were expressed on the outer envelope, when being exposed to antibiotics ceftazidime and ofloxacin, respectively. This finding is important, as the outer surface comes in contact with the immune system, and therefore may have a bearing on the outcome of the disease.

  • The envelope glycoprotein E2 of hepatitis C virus (HCV) binds to various cell surface receptors for viral infection. We performed biopanning against this protein and selected peptides from phage display peptide libraries. Two short peptides, pep7-1 and pep12-1, were selected and their ability to inhibit the infection process was investigated. When pep7-1 was present, the infectivity of HCV particles in cell culture was notably decreased. This decrease was demonstrated by Western blot analysis, immunofluorescence assay, and reverse transcription PCR assay. However, pep12-1 showed little inhibitory effect on HCV infection.

Effect of rotation on the evolution of Population III protostars

  • To figure out the effect of rotation on the final mass of Pop III stars, 1D stellar evolution simulations of the evolution of mass-accreting protostars are performed, with zero metalicity and high constant mass accretion rates. The protostar reaches the Keplerian rotation very soon after the onset of mass accretion, but it may continue mass accretion via angular momentum transport induced by viscous stress or magnetic field. However, as the accreting star evolves, the envelope expands rapidly when the total mass reaches $5{\sim}6M_{\odot}$ and the corresponding Eddington factor sharply increases. Strong radiative pressure with rotation imposes different criteria for breakup at the stellar surface, and the so-called 'critical rotation (${\Omega}{\Gamma}$-limit)' is reached. As a result mass accretion rate has to be significantly lowered. This implies that characteristic masses of Pop III stars would be significantly lowered than the previous expectation.

  • This paper concerns on modal analysis of mechanical structures by using a continuous scanning laser Doppler vibrometer. In modal analysis the Hilbert transform based approach is superior to the Fourier transform based approach because of its fine accuracy and its flexible experimental settings. In this paper the Hilbert transform based approach is extended to measure area mode shape data of a structure by simply modifying the scanning pattern ranging the entire surface of the structure. The effectiveness of this proposed method is illustrated along with results of numerical simulation for a rectangular plate.
