Studies on the Procedures of Accelerating Generation Advancement in Wheat and Barley Breeding IV. Advancement of Two Generations of Wheat Materials a Year at Suweon by Growing a Summer Generation (맥류의 세대촉진방법에 관한 연구 IV. 수원지역에서 소맥 1년 2기작 세대촉진재배)

  • To establish a generation acceleration technique, two crops a year at field condition of Suweon, 10 varieties different in their spring growth habit were tested with 6 different seeding time after harvesting. These materials were harvested on June 10, 19, 79 and tested for their seed production ability at varions seeding time from July I I to August 15 with a week interval. An immatured seed germination technique and green vernalization methods were applied in cycling generations and the results obtained were summarized as follows. 1. In summer growing, seedlings establishment after transplanting was better in earlier transplanting. 2. Heading time was remarkably enhanced by earlier transplanting. Considering the results of two years early or mid of July was the suitable time to plant the second summer crop. 3. Those varieties of spring growth habit expressed little variations in plant height among the varieties. In 1978 which is referred as warm year produced plant height as tall as 8-16cm and poor crop but produced good crop with 25-65cm plant height in 1979. 4. No definit tendency in the length of spike was. observed among the cultivars but longer spike was found in winter wheat compared to the spring. 5. Number of spikes per plant was ranged from 1 to 3 regardless the transplanting time in 1979. However, more spikes per plant were produced in early or mid of July transplanting and those varieties of higher growth habit than V did not produces any spikes. 6. Higher number of grains per spike was found at earlier transplanted varieties. Therefore, it is concluded that those materials of I-IV growth habit with mid or early July transplanting would be suitable in practical sense considering their ability of seed production. 7. Two-year results indicated that wheat crop can not tolerate the temperature level higher than average 32$^{\circ}C$ C at Suweon. In this regard, the cultivation schedule was established assuming average temperature condition like the year of 1979 which was possible to grow wheats.

Growth of Intestinal Bacteria and Intestinal Inflammation of Sprout Extract from Common Buckwheat and Tartary Buckwheat (일반메밀과 쓴메밀의 새싹 추출물의 장내 유익균 증식 및 염증조절 효능 평가)

  • We aimed to assess the potential growth-promoting effects of buckwheat sprout on intestinal bacteria and their anti-inflammation effects in a cellular model of intestinal inflammation. The growth of Bifidobacterium longum ssp. infantis BT1 was enhanced with the addition of the sprout extract of tartary buckwheat. Further, in the inflammatory model cells cultured with Raw 264.7 cells were treated with buckwheat sprout including each 10 probiotics before the addition of lipopolysaccharide (LPS) to induce inflammation in Raw 264.7 cells. Buckwheat sprout in both Bifidobacterium longum ssp. infantis BT1 and Lacticaseibacillus paracasei LPC5 significantly reduced the production of NO and PGE2. The above results indicate that buckwheat sprout extract which contains with various physiologically active substances such as rutin, quercetin, and choline is effective in suppressing NO and PGE2 production, which are inflammation-related indicators. The present study suggests that buckwheat sprout could induce positive effects on the intestinal beneficial bacteria and in anti-inflammation.

Seasonal Variations of Particle Fluxes in the Northeastern Pacific (북동태평양 심해에서 관측된 퇴적물 입자 플럭스의 계절적 변동)

  • Particle fluxes were measured with a time-series sediment trap from July 2003 to June 2005 at the St. KOMO(KOMO; Korea Deep-Sea Environmental Study Long-Term Monitoring Station, $10^{\circ}30'N,\;131^{\circ}20'W$) in the northeastern Pacific. Total mass fluxes at a depth of 4,960 m showed distinct seasonal variations with high values in the winter(December-February) and spring(March-May) and low values in the summer(June-August) and fall(September-November). Biogenic origin fluxes also displayed distinct seasonal variations similar to total mass fluxes. Particularly, calcium carbonate fluxes in winter and spring were more than two times greater than those in summer and fall. The prominent seasonal variations of total mass and biogenic fluxes were closely related with the seasonal changes of primary production in the surface waters; in winter and spring, primary production increased due to the enhanced supply of nutrients below the surface mixed layer by strong wind and less stratification, whereas it decreased as a result of the less supply of nutrient by reduced wind speed and strong stratification in summer and fall. The seasonal variations of total mass and biogenic fluxes in this study were higher than the differences of total mass and biogenic fluxes caused by the environmental changes such as El $Ni\tilde{n}o$ and La $Ni\tilde{n}a$ events in the previous studies. In order to understand the effects of El $Ni\tilde{n}o$ and La $Ni\tilde{n}a$ on the particle flux, therefore, the seasonal variation of particle flux in the northeastern equatorial Pacific needs to be well defined.

  • Farming of the fleshy shrimp Fenneropenaeus chinensis which is a major cultured species in the west coast of South Korea, has been suffered :trom mass mortality due to disease epizootics including viruses. Since the Pacific white shrimp Litopenaeus vannamei was introduced to Korea in 2003, farming of this species has rapidly increased for years, occupying 62.5% of total cultured shrimp production in 2007. However the studies on L. vannamei culture methods for shrimp farming situations in Korea are very limited. Nursery culture of shrimp larvae has some advantages including increased survival, improved feed efficiencies, enhanced growth performance and reduced grow-out period. In this study, L. vannamei postlarvae (${PL_3}-{PL_{10}}$) with a density of $3,750-9,090/m^3$ were cultured in four raceways under limited water exchange condition for 35 days. Survival was the highest (93.6%) in tank stocked with $4,090/m^3$ and was the lowest in tank with $9,090/m^3$ (58.1 %). Mean body weight at harvest ranged from 0.071 to 0.108 g, and FCR was 0.59-0.70 in all tanks. Concentration of total ammonia nitrogen was increased up to 20 ppm on day 10 in all tanks and thereafter gradually decreased by the third week of culture. Nitrite-nitrogen was rapidly increased from the third week, representing bio-floc condition by developed nitrifying bacterial community. Of the present nursery system some modification of structure and consideration for commercial scale are needed in order to be implemented to shrimp farmers.

Division of Soil Properties in Reclaimed Land of the Mangyeong and Dongjin River Basin and Their Agricultural Engineering Management (만경강과 동진강 유역 간척농경지 토양특성 구분과 농공학적 관리 대책)

  • The physical and chemical properties of soil in the Mangyeong and Dongjin river basin had been investigated in order to establish the most optimum soil improvement plan on the reclaimed land. The total soil area by reclamation in Saemangeum basin is 113,971 ha. The classification by the distribution of soil series and soil texture is as following. 13 soil series including Chonnam, Buyong and Chonbuk series are period-unknown areas. Regarding the soil texture, they are fine silty ~ clayey very fine. From 1920s to 1960s, Mangyeong, Gwanghwal and Chonbuk series had coarse silty textured soil. After the 1970s, Mangyeong, Gwanghwal, Munpo, Yeompo, Poseung, Gapo and Hasa series have more sandy soil ~ moderately coarse loamy textured soil. Regarding the chemical properties, the concentrations of EC, Exch. $K^+$, $Mg^{2+}$, $Na^+$ and pH are high regardless of the time of reclamation. On the other hand, organic matter (OM) of top soil were 3.3~16.1 g $kg^{-1}$. The organic matter contents were very low though the soil had been farmed for a long time. Furthermore, the deep soil had almost no organic matter with 5.6~1.1 g $kg^{-1}$. The reason is believed that there had not been any movement of OM and clay because pressure or induced pans had been formed by large agricultural machineries and poor vertical drain. Regarding the forming of illuvial horizon (B layer) which tells the development extent of soil, only in the Hwapo reclaimed area where rice had been cultivated for past 90 years, Fe and Mn from top soil are deposited at underground 20~30 cm with 7~8 cm thickness by the movement of clay. It is believed that it had been possible because the earthiness is silty clay loam soil with relatively high content of clay. The soils are soil with concern of damage from sea water, soil on flimsy ground and sandy soil. Therefore, soil improvement for stable crop production can be expected; if the water table would be lowered by subsurface drainage, the water permeability would be enhanced by gypsum and organic matter, and the sandy soil would be replaced by red soil with high content of clay.

  • For as to world, the concern about the environment problem is enhanced than any other time in the past because of being 21 century. And the environment problem is highlighted as the world-wide issue. The time of the environment problem intimidates the alive of the mankind and presence of an earth over the time. It becomes the essentiality not being selection in the personal living or the economical viewpoint now to prepare for the climatic modification. As to the company management, the green growth period which it excludes the environment management considering an environment, cannot carry on the company the continued management comes. That is, in the change center of the management paradigm, there is the environment management. Nearly, the greenhouse gas which the publication industry is the environmental toxic material like all industries is generated. The greenhouse gas is ejected in the process of running the manufacturing process and print shop of the various kinds material used as the raw material of the book. Particularly, the tree felling for getting the material of the paper is known to reach the direct influence on the global warming. This study does according to an object it considers and organizes the environment parameter based on this kind of fact as to the publication industry. And it is determined as the reference which is used as the basic materials preparing the case that carbon exhaust right transaction(CAP and TRADE) drawing are enforced in all industries and is sustainable the management of the publication industry and reduces the environmental risk among the company many risk management elements and plans and enforces the publication related policy that there is a value. In the printing publication industry, this study tried to inquire into elements discharging the environmental pollutant or the greenhouse gas. Additionally, in the printed publication production process, it tried to inquire into the effort for an environment-friendly and necessity at the printing paper and the printers ink, regarded as the element discharging the greenhouse gas all kinds of the printing materials, operation of the print shop and all kinds of the machines and recycle process, and etc. These considerations make these industrial field employees aware of the significance about a conservation and environmental protection. They try to give a help in the subsequent study producing quantitatively each environmental parameter emission of green house gas. This makes the calculation of the relative $CO_2$ output reproached ultimately possible. Meanwhile, in a sense, many research protects and improving an environment in connection with the contents of research at the printing publication industrial field is in progress. There will be the voluntary human face that it has to protect an environment but this can not do by the outside factor according to all kinds of environment related law and regulation. Anyway, because of acting on company management as the factor of oppression, the increase of this environment-related correspondence cost could know that the research that the environment loading relates with a procurement and development, environment management system introduction, quality control standard, including, normalizing including a material, and etc. through the part of the effort to reduce the cost low was actively in progress. As to the green growth era, as follows, this paper prescribed the subject and alternative of the print publication industry. It is surrounded by the firstly new digital environment and the generation of the subject. And secondly the printing industry is caused by the point of time when the green growth leaves by the topic which is largest in the global industry and it increases. The printing publication industry has to prepare the bridgehead for the environment-friendly green growth as the alternative for this resolution with first. The support blown in each industry becomes the obligation not being selection. Prestek in which the print publishing was exposed to spend many energies and which is known as the practice of the sustainable print publishing insisted that it mentioned importance of the green printing through the white pages in 2008 and a company had to be the green growth comprised through the environment-friendly activity. Inhibition of Quorum Sensing and Biofilm Formation by Synthetic Quorum Signal Analogues in Pseudomonas aeruginosa (합성된 쿼럼 신호 유사 물질에 의한 녹농균 쿼럼 센싱 및 생물막 형성의 제어)

  • Pseudomonas aeruginosa is an opportunistic pathogen that causes various infections on urinary track, cornea, respiratory track, and burn wound site, and mainly relies on quorum sensing (QS) for its virulence. To control the infectivity of P. aeruginosa, we previously synthesized the structural analogues of a major QS signal, N-3-oxododecanoyl homoserine lactone (3OC12-HSL) to use as a QS inhibitor. Two of them (5b and 5f) had been confirmed to have an inhibitory effect on LasR, a major QS signal receptor of P. aeruginosa in the screening by the recombinant Escherichia coli reporter. To further evaluate these compounds, we tested their efficacy to control the QS and virulence of P. aeruginosa. Unlike the result from E. coli reporter, both 5b and 5f failed to affect the LasR activity in P. aeruginosa, but instead they selectively affected the activity of QscR, another 3OC12-HSL receptor of P. aeruginosa. Interestingly, their effect on QscR was complex and opposite to what we obtained with E. coli system. Both 5b and 5f enhanced the QscR activity at the low concentration range (< 10 ${\mu}m$), but high concentration of 5f (${\approx}$1 mM) strongly inhibited QscR. While 5b and 5f didn't affect the production of proteases, the key virulence factor, they significantly reduced the biofilm formation that is important in mediating chronic infections. Especially, 5f inhibited the initial attachment of P. aeruginosa, rather than the biofilm maturation. Based on our results, we suggest that 5f can be applied for an anti-biofilm agent without increasing virulence of P. aeruginosa.

Socheongja and Socheong 2 Extracts Suppress Lipopolysaccharide-induced Inflammation and Oxidative Stress in RAW 264.7 Macrophages through Activating Nrf2/HO-1 Signaling and Suppressing MAPKs Pathway (RAW 264.7 대식세포에서 Nrf2/HO-1 신호 전달계 활성화와 MAPKs 경로 억제를 통한 소청자와 소청2호의 LPS 매개 염증성 및 산화적 스트레스 반응의 억제)

  • Inflammatory response and oxidative stress play critical roles in the development and progression of many human diseases. Therefore, a great deal of attention has been focused on finding functional materials that can control inflammation and oxidative stress simultaneously. The purpose of this study was to investigate the effects of Socheongja and Socheong 2, Korean black seed coat soybean varieties, on the inflammatory and oxidative stress induced by lipopolysaccharide (LPS) in RAW 264.7 macrophages. Our data indicated that the extracts of Socheongja (SCJ) and Socheong 2 (SC2) significantly suppressed LPS-induced production of nitrite oxide (NO) and prostaglandin $E_2$, key pro-inflammatory mediators, by suppressing the expression of inducible NO synthase and cyclooxygenase-2. It was also found that SCJ and SC2 reduced the LPS-induced secretion of pro-inflammatory cytokines, such as tumor necrosis $factor-{\alpha}$ and $interleukin-1{\beta}$, which was concomitant with a decrease in the protein levels. In addition, SCJ and SC2 markedly diminished LPS-stimulated intracellular reactive oxygen species accumulation, and effectively enhanced nuclear factor erythroid 2-related factor 2 (Nrf2) and heme oxygenase (HO)-1 expression. Furthermore, LPS-induced activation of mitogen-activated protein kinases (MAPKs) was abrogated by SCJ and SC2. Taken together, these data suggest that SCJ and SC2 may offer protective roles against LPS-induced inflammatory and oxidative responses in RAW 264.7 macrophages through attenuating MAPKs pathway, and these effects are mediated, at least in part, through activating Nrf2/HO-1 pathway. Given these results, we propose that SCJ and SC2 have therapeutic potential in the treatment of inflammatory and oxidative disorders caused by over-activation of macrophages.

Biological Activity of Oenothera Biennis Seed Extracts (달맞이꽃 종자 추출물의 생리활성)

  • In the current study, comparisons of Oenothera Biennis seed extracts with water, ethanol, methanol, and 70% ethanol in their total polyphenolics contents, anti-oxidant, anti-neurotoxicity, anti-cancer, and immune-modulatory activities were investigated. Compared with other extracts, those concentrations of total phenolics and flavonoids were the highest in MeOH extract (31.90 mg GAE/g and 20.66 mg QE/g). The radical scavenging and reducing power activities were dose-dependently increased by treatment of O. Biennis seed water, EtOH, MeOH, and 70% EtOH extracts. Furthermore, pretreatment of water, EtOH, and MeOH extracts significantly reduced glutamate-induced cytotoxicity in HT22 hipocampal neuron cells. In the case of cancer cells, MeOH extracts showed lower $IC_{50}$ values in HepG2 ($74.21{\mu}g/ml$), A549 ($188.24{\mu}g/ml$), MCF-7 ($186.42{\mu}g/ml$), and B16 ($101.80{\mu}g/ml$) than other extracts, where those water ($101.96{\mu}g/ml$) and EtOH ($788.39{\mu}g/ml$) extracts showed the lowest $IC_{50}$ activity in HT-29 and PC-3 cells, respectively. O. Biennis seed extracts did not show any cytotoxicity in RAW 264.7 macrophages at the concentration of $1-10{\mu}g/ml$, whereas 70% EtOH extract dose-dependently enhanced nitric oxide (NO) production in RAW 264.7 cells. Overall, we evaluated that various bioactive potentials of O. Biennis seed extracts which would relate with phenolic compounds abundance, thus these can be useful to future developments as functional food ingredients and natural medicines.

Pharmacological Activities of Coffee Roasted from Fermented Green Coffee Beans with Fungal Mycelia in Solid-state Culture (진균류 균사체의 고체발효 커피생두로부터 조제한 원두커피의 생리활성)

  • Green coffee beans (CB, Indonesian Mandheling) were fermented with three kinds of mushrooms (Phellinus linteus, PL; Hericium erinaceum, HE; Ganoderma lucidum, GL) or two kinds of mycelia from molds (Monascus purpureus, MP; Monascus ruber, MR) using solid-state culture to enhance physiological activity. After the roasting of fermented green coffee beans, roasted coffees were extracted with a hot-water decoction or 95% ethanol reflux. Yields from hot water extracts (HW, 17.7~25.3%) were higher than those from ethanolic extracts (EE, 9.5~12.2%). Hot-water extracts of roasted coffees from green coffee beans fermented with two molds (MP-CB-HW and MR-CB-HW) showed higher total polyphenols, flavonoids, and DPPH free radical scavenging activity than roasted coffees from non-fermented (CB-HW) or fermented green coffee beans with the three mycelia from mushrooms. MR-CB-HW also had the most potent macrophage stimulating and mitogenic activity (1.32 and 1.40-fold of CB-HW, respectively). In addition, MP-CB-EE and MR-CB-EE did not show any cytotoxicity to the RAW 264.7 cell at a concentration of $100{\mu}g/mL$, and these extracts significantly inhibited nitric oxide (NO) production from the LPS-stimulated RAW 264.7 cell line (38.6 and 37.0% of the LPS-treated group). Meanwhile, the chlorogenic acid concentrations of MP-CB-HW or MR-CB-HW highly increased (to 76.21 or $76.73{\mu}g/mL$, respectively), but caffeine concentrations were not affected by solid-state fermentation. In conclusion, the physiological activities of roasted coffees were enhanced by the solid-state culture of green coffee beans with M. purpureus or M. ruber, suggesting that these roasted coffees could possibly serve industrial applications as functional coffee beverages.
