EFFECT OF TRYPSIN-DIGESTED BOVINE GROWTH HORMONE ON WHOLE-BODY PROTEIN SYNTHESIS IN VITRO IN CHICKEN EMBRYOS

  • The effect of bovine growth hormone digested with trypsin on whole-body protein synthesis in vitro of chicken embryos was investigated by using a whole-embryo culture system. Bovine growth hormone at 5.3 and 530 ng/ml was digested partially and completely with trypsin for 4 min and 18 h, respectively. After culturing chicken embryos with a synthetic medium containing $L-[4-^3H]$ pheylalanine, whole-embryo protein synthesis was determined from the ratio of specific radioactivities of free and protein-bound pheylalanine. Whole-embryo protein synthesis of the control group cultured with no bovine growth hormone was $49.5{\pm}2.2%/d$. There was no significant interaction between digestion time and the concentration of trypsin-digested bovine growth hormone. Tryptic digestion of bovine growth hormone increased fractional synthesis rates of whole-body protein compared to the 0-min groups, and there was no significant difference between the 4-min and 18-h groups. The higher concentration (530 ng/ml) of trypsin-digested bovine growth hormone was more effective in enhancing whole-embryo protein synthesis than the lower concentration (5.3 ng/ml).

Factors influencing somatic embryo maturation, high frequency germination and plantlet formation in Terminalia chebula Retz.

  • The factors influencing somatic embryo maturation, high frequency somatic embryo germination, and plantlet formation were studied in Terminalia chebula Retz. Maturation of somatic embryo were influenced by a number of factors such as in vitro culture passage, concentrations of sucrose, levels of abscisic acid (ABA), basal media and media additive combinations. Maximum frequency of somatic embryo maturation ($57.22{\pm}2.02$), was obtained on MS medium supplemented with 50 g/l sucrose. Different factors such as strengths of MS nutrients, plant growth regulators, media additives and their combinations controlling somatic embryo germination and plantlet formation were studied. High frequency of germination and plantlet formation ($58.80{\pm}1.47$) were achieved by subsequent subculture of mature somatic embryos on MS medium containing 30 g/l sucrose and 0.5 mg/l benzyl-adenine (BA). However, although duration of in vitro passage of the callus tissue was critical, contribution of the combinations of plant growth regulators and media additives showed nugatory effect on somatic embryo maturation and germination as evident from variable responses.

  • 인삼종자의 내과지는 배생장을 현저히 억제시켰다. 이 억제작용은 내과지에 의한 수분흡수 때문이 아니라 기계적인 것으로 추측되어졌다. 수산화나트리움 2.5% 용액에 인참종자를 10분 내지 30분 침적시키므로써 배생장을 촉진시킬 수 있었으나 황산처리는 인삼종자의 배생장에 영향을 주지 못했다.

  • Objective: To evaluate incidence of microbial growth from the tip of the embryo transfer catheter after embryo transfer in relation to clinical pregnancy rate following in-vitro fertilization and embryo transfer. Method: This study was performed prospectively at the time of transcervical embryo transfer following conventional in-vitro fertilization and intracytoplasmic sperm injection procedures. Sixty three patients were enrolled in this study. Microbiological cultures were performed on endocervical swabs and embryo transfer catheter tips. Results: Positive microbial growths were observed from endocervical swabs in 45 (71.4%) women and from catheter tips in 30 (47.6%) women. There was no statistically significant difference seen in the mean number of oocytes fertilized or number and grade of embryos transferred between the group of patients without growth and the group of patients with positive microbial growth from catheter tips. The clinical pregnancy rate were 30.3% in the group of patients without growth and 13.3% in the group with positive microbial growth from catheter tips. This difference in clinical pregnancy rates was statistically significant. Conclusion: Our finding is that microbial contamination at embryo transfer may influence implantation rates. The major questions arising from our finding are whether eradication of endocervical micro-organisms is possible and whether their eradication will improve implantation rates.

Role of Temperature and Gibberellic Acid in Stimulating Embryo Elongation and Radicle Emergence of Hepatica asiatica Nakai Seeds

  • This research was performed to determine the temperature requirements for embryo growth and radicle emergence of Hepatica asiatica Nakai, a perennial herb native to Korea. Seed viability, embryo growth, and radicle emergence were monitored in seeds exposed to various temperatures (10, 15, $20^{\circ}C$ and $30{\rightarrow}15^{\circ}C$). Laboratory experiments at various temperatures revealed that (1) embryo elongation occurred effectively between 10 and $15^{\circ}C$; (2) radicle emergence occurred only at $15^{\circ}C$; (3) a warm stratification (2-8 weeks at $30^{\circ}C$) was not required for embryo elongation and radicle emergence, and led to inhibition of radicle emergence; (4) application of gibberellic acid ($GA_3$) promoted embryo growth, but not radicle emergence. These results suggested that H. asiatica seeds have two separate mechanisms to overcome dormancy, either by $GA_3$ (morphological dormancy) or temperature (physiological dormancy), and warm stratification is not involved in breaking radicle dormancy. These findings indicate that H. asiatica seeds have morphophysiological dormancy.

Changes in Metabolites and Embryo Growth during Seeds Priming in Tobacco

  • Some metabolites and embryo growth of primed tobacco (Nicotiana tabacum L. cv. ‘KFI09’) seeds were observed during priming. The seeds were primed at 15 and $25^{\circ}C$ for 1, 2, 3, 5, 10 and 15 days in a -0.8 MPa polyethylene glycol 6000 (PEG) solution. The time to 50% seed germination (T$_{50}$) was greatly reduced when the seeds were primed at $25^{\circ}C$ when compared with 15$^{\circ}C$. The $\alpha$-amylase activity and sugars and amino acid contents in the seeds primed at $25^{\circ}C$ greatly increased, while $\alpha$-amylase activity was similar, and sugar and amino acid contents increased slightly in the seeds primed at 15$^{\circ}C$. When the seeds were primed at $25^{\circ}C$, growth of the embryo which was enclosed by endosperm was detected, while the endosperm became thinner as the priming duration was extended.d.

  • This experiment was carried out to develop the model system for mass production of biomedical and nutritional proteins (human proteins) through mamraary gland of the transgenic cattle produced by gene manipulation and embryological technologies. Human growth hormone gene fused with rat $\beta$-casein gene promoter was microinjected into pronuclei of one cell bovine embryos produced by in vitro fertilization. After microinjection, embryos were cultured in vitro for 6 or 7 days. Twenty embryos reaching to blastocysts were transferred to 10 beef recipients, each receiving two embryos. Recipients were diagnosed for pregnancy by rectal palpation at 76 days after embryo transfer. One of them was pregnant to term and produced a female calf weighing 21 kg at 280 days following embryo transfer. DNA was extracted from umbilical cord tissue and blood of calf born for confirming gene insertion. As determined by Southern hybridization, the transgene was not found.

Propagation by In Vitro Zygotic Embryos Cultures of the Quercus myrsinifolia

  • Zygotic embryo culture was performed to propagate evergreen oak, Quercus myrsinifolia, which has recalcitrant seeds and is difficult to propagate by cuttings. Zygotic embryos appeared in WPM medium after 14 days, and after 56 days, they developed into complete plants with cotyledons and roots. The medium suitable for zygotic embryo culture was 1/4 WPM medium, showing a shoot growth of 2.43 cm and root growth of 8.7 cm after 8 weeks of culture. As a result of investigating the effect of GA3 on the growth of plants germinated from zygotic embryos through GA3 treatment, the best growth was shown in 0.5 mg/l GA3 treatment. The in vitro rooting and growth of IBA-treated zygotic embryo-derived plants were good in the 0.5 mg/l IBA treatment and rooting and shoot growth were not observed at higher concentrations. And the callus induction rate also increased as the concentration of IBA increased. Plants grown in vitro were transferred to a plastic pot containing artificial soil and acclimatized in a greenhouse for about 4 weeks, resulting in more than 90% survival. As a result of this study, the zygotic embryo culture method was confirmed to be effective for mass propagation of Q. myrsinifolia. The results of this study are expected to contribute significantly to the mass propagation of elite Q. myrsinifolia.
