Radioimmunoassay of High-Density Lipoprotein in Japanese Quail Serum (메추리혈청 리포단백질의 방사면역분석법)

  • The development of radioimmunoassay (RIA) for high-density lipoprotein (HDL) in Japanese quail serum will contribute to the screening of drugs acting on cholesterol transport. We have developed a double antibody RIA method for J. quail HDL. The first antibody was raised in rabbit by immunization of HDL isolated by the dextrane sulfate-$Mn^{#}$ precipitation method. For the preparation of raclioiodinated antigen, HDL was further purified by combination of electrophoretic procedure. Using the second antibody raised in goat by rabbit IgG, we have furnished the RIA method for HDL. It showed high specificity and sensitivity of working assay range, 0.1-33.mu.g HDL/tube. There was no correlation between the radioimmunoassay of HDL and the enzyme assay of HDL-cholesterol.

  • Acrylamide gel electrophoretic pattern of 13 wild soybean (Glycine ussuriensis) was compared with that of G. gracilis and G. max var. Gwanggyo. Average Protein content (50%) of wild soybean was greater than that of C. gracilis (46%) and Gwanggyo (45%). Grain weight of wild soybean was one third of G. gracilis and one ninth of Gwanggyo. Electrophoresis of wild soybean protein showed total 16 different bands and three of which (Rm 0.09, 0.59 and 0.84) were specific and did not appeared in 86 var. of G. max which showed four specific bands (Rm 0.35, 0.45, 0.50 and 0.77) of total 17 bands. G. glacilis had all bands of Gwanggyo and two bands (Rm 0.53 and 0.59), one of which (Rm 0.59) was specific for wild soybean indicating that G. gracilis is middle type. Of 16 protein bands the third band (32%), the first band (28%) and the 5th band (13%) were main bands. Electrophoretic pattern could be sorted qualitatively into 4 groups, semiquantitatively into 6 groups and 2 or 4 groups depending on reference pattern by correlation or pattern similarity method. All sorting methods separated a wild soybean from Sogri mountain into a group and except that there were no similarity among methods but correlation methods seems more reasonable. Protein content was no relation with electrophoretic pattern but positively correlated with percent contribution of first band at 5% level suggesting that the first band may have a important role for protein synthesis.

  • Carbon nanotubes (CNTs) were grown on conical tip substrates by using various methods such as electrophoretic deposition, dip-coating, and spray. The scanning electron microscope measurement showed that the spray method ascertained the most uniform deposition of CNTs. The CNT-emitter that was fabricated by the spray method revealed the lowest turn on voltage of electron emission and the highest emission current. In addition, the spray-produced CNT emitter showed the most stable long-term emission characteristics.

Property Improvement of YBCO Thick films by EPD with Addition of PEG (PEG 첨가에 의한 YBCO 전착후막의 특성 향상)

  • The electrophoretic deposition method using the suspension solution with additives under the electric potential was applied for the fabrication of YBCO superconductor wire. This method was able to simplify the fabrication facilities, and produce an uniform and dense thick film. To improve the critical current density of deposited films, the additive PEGs(Poly Ethylene Glycole) with the molecular weight of 600, 1000 and 3400 were used as chemical binders for the suspension solution. The organic additive (PEG) showed better effects to the properties of YBCO superconductor wire. The PEG improved the adhesion between superconductor particles and suppressed the crack on the surface, which enhanced the surface uniformity and density of YBCO deposited film. It was found that acetone suspension solution showed better deposition properties than the others. The samples fabricated in the solution with the additive, 8 vol.% of 1% PEG(1000), showed the highest critical current density measured as 2300∼2400 A/$\textrm{cm}^2$ at 77 K, 0 T.

  • The progress in the field of electronic materials has been especially significant for applications involving a range of electrical properties. Its importance is increasing with the increasing demand for integrated circuits. The sol-gel technique has been used for many years, and the metal alkoxides have featured prominently as source materials. The method consist of making a homogeneous solution of the component metal alkoxides in a suitable solvent, usually the parent alcohol; and then causing the hydrolysis under controlled conditions to produce a gel containing the hydrated metal oxide. The gel is then dried, and fired to produce a ceramic or glassy material at a temperature much lower than that required by the conventional melting process. This project consists of important theoretical considerations, processing techniques and applications related to electrophoresis derived thin films. In the electrophoretic process a metal alkoxide solution is gelled through hydrolysis-polymerization and converted the gel thin layer to an oxide by heating at relatively low temperatures.

Problems and Solutions of Zymography Techniques (자이모그라피 기술의 문제점과 해결)

  • Enzymes are widely used in industrial applications such as detergents, food, feed production, pharmaceuticals and medical applications and major contributors to clean industrial products and processes. To screen, identify, and characterize the enzymes the zymography techniques are routinely used. The zymography technique is a simple, sensitive, and quantifiable technique that is widely used to detect functional enzymes following electrophoretic separation in sodium dodecyl sulfate (SDS)-polyacrylamide gels. The method is a versatile two-stage technique involving protein separation by electrophoresis followed by the detection of enzyme activity in polyacrylamide gels under non-reducing conditions. It is based on SDS-polyacrylamide gel (PAG) copolymerization with substrates, which are degraded by the hydrolytic enzymes restored in enzyme reaction buffer after the electrophoretic separation. Any kind of biological sample can be applied and analyzed on zymography, including culture supernatants of microbes, plants extracts, blood, tissue culture fluids, enzymes in foods extracts and metaproteome. The advantage of zymography is that it is possible to directly detect the protein with activity on the electrophoretic gel as well as confirm the activity at the nanogram level. Thus, this zymography technology can be applied in various fields. However, these advantages are rather disadvantageous and can often lead to experimental errors. In this review, the advantages, disadvantages, and problem solving of zymography technique are described.

Interspecific Relationships of the Genus Leucophenga(Drosophiliae) by Electrophoretic Analysis of Protein (단백질 전기영동 분석에 의한 Leucophenga 속 (Drosophilidae) 초파리의 종간 유연관계)

  • The interspecific relationships of the genus Leucophenga, L.concilia, L.maculata, L.orientalis , and L.quinquenmaculipennis were investigated by soluble proteins analysis. the electrophoretic banding patterns of the four species analyzed by SDS-PAGE were scanned by densitormeter. The soluble protein patters of L.maculata and L.concilia by SDS-PAGE appeared similarly and on the other hand, those of L.quinquemaculipennis had a different forms from three other species. The genetic distance among the four species of TDE analysis was calculated by formula of the Aquadro and Avis(1981) . The genetic distance between L.maculata and L.concilia was 0.393 , the lowest of all and between L.maculata and L.quinquemaculipennis was 0.496, the highest of all. The dendrogram was established using UPGMA method based on genetic distance obtained by TDE analysis. Clustered patterns as follows ; first, L.maculata and L.concilia were clustered, and L.orientalis and L. quinquemaculipennis followed the firstone in order. In consequence, it seems that the interspecific relationship of L.quinquemaculipennis was farther from the other three species, while that of L.maculata and L.concilia was closer than that of L.orientalis.

Estimation of the $T_4$ Binding Capacity of Serum Thyroxine Binding Globulin (혈중(血中) Thyroxine-결합(結合)-globulin(TBG)의 $T_4$ 결합능(結合能) 측정(測定)에 관한 고찰(考察))

  • The most commonly used methods for determining thyroxine binding globulin(TBG) concentration as the total thyroxine-binding capacity utilize electrophoretic seperation of serum. Although technically simple, the electrophoretic method is time consuming and is limited in the number of samples which can be run in a single assay. The author presented a single $T_4$ load ion exchange resin method as an approach to simplify the technique as with clinical practicability and results were analyzed. For construction of the standard curves, serum mixtures were diluted with barbital buffer.which effectively blocked $T_4$-binding to TBPA. For each serum dilution, a constant amount of $T_4-^{125}I$ and increments of unlabelled $T_4$ were added. After incubation in water bath, resin beads were dispensed to the samples which binded all $T_4$ not bound to TBG. The radioactivity in the supernatant was counted in the gamma scintillation counter. Each standard curve was plotted from the percent counts in the supernatant and total $T_4$ in each tube. Unknown samples were diluted to 1:40 and ran at a single $T_4$ loading concentration, and the TBG capacity of the samples was able to be read on the standard isobars. The following results were obtained. 1) Mean and standard deviation for TBG capacity in normal population was $28.6{\pm}5.09{\mu}g\;T_4/100ml$. 2) $24.9{\pm}3.87{\mu}g\;T_4/100ml$ in hyperthyroidism showed low TBG capacity comparing to normal population.(p<0.025) 3) $31.0{\pm}2.40{\mu}g\;T_4/100ml$ in hypothyroidism showed high TBG capacity tendency comparing to normal population. 4) Reversed correlationship existed between TBG capacity and $T_3$ resin uptake(r=-0.624), TBG capacity and serum $T_4$ value (r=-0.859), and TBG capacity and free thyroxine index(r=-0.623). The author assumes that this method of assay is considerably simpler in instrumentation and technique than any other assays traditionally being used, and seems to be more practical for routine clinical laboratory use.

  • Changes in the concentration of total protein, albumin, ${\alpha}_1-,{\alpha}_2-,{\beta}-,$ and ${\gamma}-globulin$ in Serum from 138 healthy, normal pregnant woman were studied by the method of cellulose-acetate electrophoresis. 1. The concentration of total serum protein decreased gradually during the first 7 month, and showed a tendency to increase thereafter. 2. The concentration of serum albumin showed a steady continuous fall untill term. 3. During pregnancy,${\alpha}_1$ and ${\beta}-globulin$ value rose, ${\gamma}-globulin$ value fell and ${\alpha}_2-globulin$ value showed no significant change.

  • When total cellular DNA was isolated from Porphyra tenera by ultracentrifugation on Hoechst dye/CsCl gradients method, plasmid like DNA's were concentrated at the upper band which were characterized with a A+T rich organelle DNA's in the CsCl gradients. Based on their electrophoretic migration in different concentration of agarose gel, buffer system, and electric power etc. and the results of restriction digestion, the plasmid like DNA's were concluded to have circular conformation. This is the first report of putative circular plasmid DNA from the P. tenera, which is a autonomously replicating plasmid existing with a high copy number plasmid in the cell. The minimum size of this plasmid estimated by restriction endonuclease digestion was appeared to be 2.5kb in size.
