Development of Ultra-High Pressure Capillary Reverse-Phase Liquid Chromatography/Tandem Mass Spectrometry for High-Sensitive and High-Throughput Proteomics

  • Recently mass spectrometry and separation methods such as liquid chromatography have become major tools in the field of proteomics. In this report, we describe in detail our efforts to develop ultra-high pressure capillary reverse-phase liquid chromatography (cRPLC) and its online coupling to a mass spectrometer by a nanoelectrospray (nanoESI) interface. The RPLC system is constructed in house to deliver LC solvents at the pressure up to 20,000 psig, which is four times higher than conventional RPLC systems. The high operation pressure allows the efficient use of packed micro-capillary columns (50, 75 and 150 ${\mu}$m i.d., up to 1.5 m long). We will discuss the effect of column diameter on the sensitivity of cRPLC/MS/MS experiments and the utility of the developed technique for proteome analysis by its application in the analysis of proteome samples having different levels of complexity.

Chemical Structural Effects of Polyimides on the Alignment and Electro-optical Properties of Liquid Crystal Cells

  • The nature of the nematic liquid crystal (LC) alignment induced by the rubbed polyimide (PI) alignment layers (ALs) and the electro-optical (EO) properties of the LC cells are expected to depend on the chemical and physical features of the PI. By employing five pyromellitic dianhydride (PMDA)-type PIs having different functionalities, we have studied the effects of the PI's structure and chemistry on the alignment characteristics and the cell's EO properties. Increasing the flexibility of the PI increases the pretilt angle and tends to improve the alignment stability. On the other hand, the rigid, fluorinated PI displays poor stability for LCs and induces a less stable/uniform LC alignment and, subsequently, a small pre tilt angle. It also transpired that fluorination of the PI deteriorated the voltage-transmittance characteristics and the voltage holding ratio; increasing the flexibility of the PI structure improves these EO properties. The finding that the qualitative trends for the PI's functionalities are similar for both the alignment and EO properties suggests that the EO properties are closely related to the alignment characteristics, which are determined by short-range interactions between LC and PI molecules.

Monitoring of the Transfer of Tetrachloroaurate(III) Ions by Thin-layer Electrochemistry and Electrochemical Deposition of Metallic Gold over a Graphite Electrode

  • This study demonstrates the electrochemical conversion of the synthetic procedure of monolayer-protected clusters using a thin toluene layer over an edge plane pyrolytic graphite electrode. A thin toluene layer with a thickness of 0.31 mm was coated over the electrode and an immiscible liquid/liquid water/toluene interface was introduced. The transfer of the tetrachloroaurate ($AuCl_4^-$) ions into the toluene layer interposed between the aqueous solution and the electrode surface was electrochemically monitored. The $AuCl_4^-$ ions initially could not move through into the toluene layer, showing no reduction wave, but, in the presence of the phase transfer reagent, tetraoctylammonium bromide (TOABr), a cathodic wave at 0.23 V vs. Ag/AgCl was observed, indicating the reduction of the transferred $AuCl_4^-$ ions in the toluene layer. In the presence of dodecanethiol together with TOABr, a self-assembled monolayer was formed over the electro-deposited metallic gold surface. The E-SEM image of the surface indicates the formation of a highly porous metallic gold surface, rather than individual nanoparticles, over the EPG electrode.

Dispersity and Electro-Conductivity of PU Grafted MWCNT/PU Composite via Simple Blending Method (블렌딩을 이용한 폴리우레탄 그라프트 다중벽 탄소나노튜브/폴리우레탄 복합체의 전기 전도성 및 분산 특성)

  • The PU-g-MWNTs/PU film was synthesized by simple blending method to fabricate composites which have excellent mechanical and electrical properties. PU-g-MWNTs based composite revealed much enhanced dispersity than pristine MWNTs composite because of interfacial interaction related with interfacial compatibility between polymer matrix and PU on the MWNTs surface. The electro-conductivity of composite was measured as a function of PU-g-MWNTs concentration. The results were correlated with percolation threshold theory. As a result, the critical concentration and exponent of electro-conductivity behavior was equal to 0.78 wt% and 0.945.

Growth of Graphene on Electro-polished Copper Foil by Thermal CVD

  • The continuous monolayer graphene was synthesized on electro-polished copper foil. Electro-polishing sticks off the coating layer of copper foil, which prevents the continuous graphene growth. The quality of continuous graphene is dependent on roughness of copper foil. Copper foil roughness could be controlled by changing polishing condition. The effects of working voltage (4-6 V) and time (30-70 sec) for electro-polishing were systematically examined. The change of surface roughness was checked with AFM.

Photopolymer Composed of a Photosensitive Polymer Binder Bearing a Chalcone Moiety in the Repeating Unit

  • New photopolymers were designed and prepared using the photosensitive polymer binders. Holographic gratings were successfully fabricated in these photopolymer samples by a conventional optical interference method. We also investigated the effect of photocrosslink in the polymer binder on the diffraction behavior of a new photopolymer. The dynamic behavior of the grating formation was monitored by changing exposure intensity in terms of the diffraction efficiency. Particularly, we focused our efforts in observing the variation of diffraction efficiency during a post UV curing process. The surface topographical change of photopolymer layer before and after Vis/UV light exposure was observed by atomic force microscope (AFM). We inscribed the gratings of the glass diffuser on the surface of the photopolymer and investigated their diffusing properties. The diffusers with photopolymer with the main chain polymer binder showed relatively good viewing angle of around ${\pm}30{\circ}$. Two kinds of photopolymer showed similar uniformity of around 47-54%.
