Triterpenoid from Tiarella polyphylla, Regulation of Type 1 Procollagen and MMP-1 in Ultraviolet Irradiation of Cultured Old Age Human Dermal Fibroblasts

  • Although many studies have been performed to elucidate the molecular consequences of ultraviolet irradiation, little is known about the effect of natural products. Ultraviolet irradiation is widely considered to be an environmental stress. Here we investigated the effect of 3,23-dihydroxy- 20(29)-lupen-27-oic acid on the regulation of MMP-1 and type 1 procollagen in Ultraviolet irradiation of cultured old age human dermal fibroblasts. 3, 23-dihydroxy-20(29)-lupen-27- oic acid was isolated from Tiarella polyphylla D. Don (Saxifragaceae). Among them, 3, 23- dihydroxy-20(29)-lupen-27-oic acid induced the regulation of Type 1- procollagen and reduced the regulation of MMP-1 at the protein levels in a dose-dependent manner by ultraviolet irradiation. Taken together, our results suggest that 3, 23-dihydroxy-20(29)-lupen-27-oic acid plays an important role in the induction of Type 1-procollagen and reduction of MMP-1 by ultraviolet irradiation in old age human dermal fibroblasts.

Effect of deletion mutants in the regulatory region of transcriptional regulation of glpD and glpE genes (glpD와 glpE 유전자의 조절영역 결손변이주가 전사조절에 미치는 영향)

  • The glpD genes encoding gly-3-p dehydrogenase is essential for the aerobic growth of E. coli on glycerol or gly-3-p. The glpE gene, the function of which is unknownm is transcribed divergently with respect to glpD gene. Expression of the adjacent but divergently transcribed glpD the glpE genes is positively regulated by the cAMP-CRP complex. In this study, for a precise investigation of the functional elements in the regulatory region for transcription activation by cAMP-CRP, deletion mutation have been introducted into the regulatory region. The effect of the deletion mutant on transcriptional regulation was tested in vivo by $\beta$-galctosidase activity. Deletion mutants in the regulatory region of glpD demonstrated that the presence of the CRP-binding site resulted in an sixfold increase in promoter activity. And also deletion mutants of glpE gene demonstrated that the presence of the CRP-binding site resulted in an eightfold increase in promoter activity. Insertion of 22 bp oligomer in the deletion mutants has shown that the CRP binding site is need for maximal expression of glpD and glpE genes. glpD and glpE gene, cAMP-CRP complex, deletion mutant, transcriptional regulation.

Anti-Inflammatory Activity of Vacuum Distillate from Panax ginseng Root on LPS-Induced RAW264.7 Cells

  • Panax ginseng has been widely applied as an important herb in traditional medicine to treat numerous human disorders. However, the inflammatory regulation effect of P. ginseng distillate (GSD) has not yet been fully assessed. To determine whether GSD can ameliorate inflammatory processes, a GSD was prepared using the vacuum distillation process for the first time, and the regulation effect on lipopolysaccharide-induced macrophages was assessed. The results showed that GSD effectively inhibited nitric oxide (NO) formation and activation of inducible nitric oxide synthase (iNOS) mRNA in murine macrophage cell, but not cyclooxygenase-2 production. The mRNA expression pattern of tumor necrosis factor alpha and IL-6 were also reduced by GSD. Furthermore, we confirmed that GSD exerted its anti-inflammatory effects by downregulating c-Jun NH2-terminal kinase (JNK) phosphorylation, the extracellular signal-regulated kinase phosphorylation, and signaling pathway of nuclear factor kappa B (NF-κB). Our findings revealed that the inflammatory regulation activity of GSD could be induced by iNOS and NO formation inhibition mediated by regulation of nuclear factor kappa B and p38/JNK MAPK pathways.

The Mediated Effects of Self-regulation in the Relationship between Nursing Professionalism and Social Responsibility of the Nursing Students (간호대학생의 간호전문직관과 사회적 책임과의 관계에서 자기조절의 매개효과)

  • The purpose of this study was to identify the mediating effect of self-regulation in the relationship between nursing professionalism and social responsibility of the nursing students. The participants of this study were 179 nursing students in S university of Gyeonggi-do, Korea. Data were collected using structured questionnaires and analyzed using SPSS WIN 25.0 program. As a result of the study, There were statistically significant positive correlation between nursing professionalism, self-regulation and social responsibility. Also, nursing professionalism affected social responsibility and self-regulation had a partial mediating effect in the relationship between nursing professionalism and social responsibility. The results are recommended to be applied by developing various programs to improve nursing professionalism and self-regulation in order to produce nurses with social responsibility in university.

  • Objective : This study aimed to investigate the effect of a 5-minute short mindfulness intervention on emotional regulation and cognitive improvement compared to self-awareness intervention. Methods : A total of 40 participants were randomly assigned and divided into a mindfulness group and a self-aware group. Participants responded to Korean Version of Positive Affect and Negative Affect Schedule (K-PANAS), and Korean version of Toronto Mindfulness Scale (K-TMS) to confirm prior homogeneity. Both groups performed processing according to each group after completing sentences related to themselves for self-focusing. Afterwards, the participants performed the Emotional Attentional Blink (EAB) task as a behavioral measure, and responded to K-PANAS and K-TMS post hoc. Results : The mindfulness group showed lower negative emotions in the K-PANAS than the self-awareness group. The mindfulness group showed higher accuracy than the self-awareness group in negative stimuli presented in the 200 ms condition and neutral stimuli presented in the 800 ms condition. However, there was no difference between groups in K-TMS. Conclusion : The study suggests that mindfulness and self-awareness have different emotion regulation strategies in negative stimuli. Additionally, 5-minute mindfulness intervention was relatively beneficial to improve cognitive function.

  • The aim of this study was to find out the effect of hypoxic condition on the regulation of cyplal gene expression. pcyplal-Luc construct was cloned and transfected into Hepa I cells. When Hepa-I cells containing pcyplal-Luc were treated by DFO (desferrioxamine) which is iron-chelating agent, the stimulatory effect of luciferase by TCDD was decreased. This inhibitory effect of desferrioxamine on the luciferase activity was dose dependent and abolished by concomitant treatment with N$\^$G/-nitro-ι-arginine. And when cobalt chloride which is known as a hypoxia inducing chemical was administrated, the stimulatory effect of luciferase by TCDD was also decreased. This inhibitory effect of cobalt chloride on the luciferase activity was dose dependent and abolished by concomitant treatment with N$\^$G/-nitro-ι-arginine. These data showed that hypoxic condition down regulates cyplal gene expression and this might be through nitric oxide action.

Effect of Self-regulation Group Counseling Program on Self-esteem and Self-control of Adolescents on Probation (자기조절 집단상담 프로그램이 보호관찰소 청소년의 자아존중감과 자기통제력에 미치는 영향)

  • This study aimed to analyse the effects of a self-regulation group counselling program on self-esteem and self-control in adolescents on probation. In respect to sub-areas of the effects the self-regulation group counselling program had on self-esteem in adolescents, there were statistically significant differences in general self-esteem, social self-esteem and family self-esteem. Also, there were statistically significant differences in all the areas including impulse control and emotional control as sub-areas of self-control. Such results experimentally showed that the self-regulation group counselling program had a significant effect on self-esteem and self-control in adolescents on probation. Therefore, it is expected that the probability of repeating their crimes will decrease greatly if the subjects have self-esteem and self-control enhanced through the group counselling program. It is suggested further studies on the self-regulation group counselling program and its practical use are needed to enhance self-esteem and self-control in adolescents on probation.

The Effect of Gongjin-dan on Gliosis in Middle Cerebral Artery Occlusion (MCAO) Rats (공진단이 MCAO모델 흰쥐에서 gliosis 억제에 마치는 영향)

  • Objectives : In conditions of brain infarction, irreversible axon damage occurs in the central nerve system (CNS), because gliosis becomes a physical and a mechanical barrier to axonal regeneration. Reactive gliosis induced by ischemic injury such as middle cerebral artery occlusion is involved with up-regulation of GFAP and CD81. This study was undertaken to examine the effect of the Gongjin-dan (GJD) on CD81 and GFAP expression and its pathway in the rat brain following middle cerebral artery occlusion (MCAO). Methods : In order to study ischemic injuries on the brain, infarction was induced by MCAO using insertion of a single nylon thread, through the internal carotid artery, into a middle cerebral artery. Cresyl violet staining, cerebral infarction size measurement, immunohistochemistry and microscopic examination were used to detect the expression of CD81 and GFAP and the effect on the infarct size and pyramidal cell death in the brain of the rat with cerebral infarction induced by MCAO. Also, c-Fos and ERK expression were measured to investigate the signaling pathway after GJD administration in MCAO rats. Results : Measuring the size of cerebral infarction induced by MCAO in the rat after injection of GJD showed the size had decreased. GJD administration showed pyramidal cell death protection in the hippocampus in the MCAO rat. GJD administration decreased GF AP expression in the MCAO rat. GJD administration decreased CD81 expression in the MCAO rat. GJD administration induced up-regulation of c-FOS expression compared with MCAO. GJD administration induced down-regulation of ERK expression compared with MCAO. Conclusion : We observed that GJD could suppress the reactive gliosis, which disturbs the axonal regeneration in the brain of a rat with cerebral infarction after MCAO by controlling the expression of CD81 and GFAP. The effect may be modulated by the regulation of c-Fos and ERK. These results suggest that GJD can be a candidate to regenerate CNS injury.

Efficiency analysis in the presence of network effect with DEA method (네트워크 효과를 고려한 천연가스산업의 기술적 효율성 분석)

  • This study takes an issue of efficiency analysis in the presence of network effect utilizing the DEA (Data Envelopment Analysis) framework. Network effect has important policy implication for the regulation of local monopolies which undertake their business through physical network, such as electricity, natural gas, local telephony, etc. If the difference in spatial condition between companies is not controlled properly, the performance comparison and associated incentive regulation bear significant bias. In this study, we propose a methodology to measure the true managerial or technical efficiency apart from efficiency difference accruing from the difference in spatial condition. A series of modified DEA efficiency models are combined to investigate the extent of exogenous and endogenous efficiency component in the Korean natural gas distribution companies. Empirical results show that the network effect plays significant role in determining superficial performance difference.

Exploratory Study on Consumer Attitude toward the SSM Regulation Law (유통산업발전법 개정에 따른 소비자 반응 탐색연구)

  • Purpose - Six months have passed since the amendment of the SSM regulation law; however, as yet, there is no confirmed research or report on the effects of this amendment, which are indefinite. Further, there have been no attempts to study the effects of the SSM regulation law from the consumers' viewpoint, which is important because consumers are the main agents that are greatly influenced by the amendment law. Therefore, this study aims to investigate the consumers' attitude toward the SSM regulation law as well as the effects of the SSM regulation law on the changes in purchase behavior. Research design, data, and methodology - This study was initiated from four research problems that are linked to consumer reaction to the SSM regulation law. Research problem 1: What is the consumers' reaction (perception, attitude, and perceived fairness) to the SSM regulation law? Research problem 2: How do the consumers' reactions to the SSM regulation law differ by consumers' characteristics? Research problem 3: Could the SSM regulation law change a consumer's purchase behavior? Research problem 4: Is it necessary to amend the SSM regulation law? This study collected the data through the interview and survey of housewives for the purpose of solving the research problems. The interview was conducted as a pilot study for the field survey. We interviewed three housewives, who were: an employed housewife, a full-time homemaker, and a manager of a housewife club, respectively. We then conducted a field survey of 232 housewives who were housewife club members or elementary school parents in Chunghcheong-do. Results - We verified the reliability and validity of the data, and analyzed it to solve the research problems. The main findings of this study were as follows. First, consumers still have a positive attitude toward large discount chains and SSM, which has been the case since the law was introduced. Second, perceived risk of consumers associated with traditional markets and small neighborhood shops was low. Third, consumers think that amendments of the SSM regulation law are important, and they positively assess the satisfaction, necessity, and propriety of the law. Fourth, although the SSM regulation law caused inconvenience to consumers, this law did not have any influence on the usage frequency and the use-behavior of large discount chains. Finally, consumers reacted very negatively to the toughening up of the SSM regulation law. Conclusions - In short, consumers still have a positive attitude toward the SSM regulation law. However, this act did not have any influence on the use-behavior of large discount chains (General Super Market). Thus, policy making authorities require active communication and promotions to enhance the effect of the SSM regulation law. This study was of the nature of exploratory research, which did not focus on hypothesis testing, but on finding solutions to the research problems. Therefore, this study is no more than a simple data analysis. Future studies should attempt to investigate the actual effects of the SSM regulation law, on the basis of sufficient literature review and real sales data.
