Influence of Controlled- and Uncontrolled-pH Operations on Recombinant Phenylalanine Ammonia Lyase Production in Escherichia coli

  • Effects of controlled- and uncontrolled-pH operations on phenylalanine ammonia lyase (PAL) production by a recombinant Escherichia coli strain were investigated at uncontrolled-pH ($pH_{UC}$) and controlled-pH ($pH_C$) of 5.5, 6.0, 6.5, 7.0, 7.5, 8.0, and 8.5 in bioreactor systems. The results showed that the recombinant PAL activity was improved significantly by controlled pH strategy. Among the $pH_C$ operations, the highest PAL activities were obtained under $pH_C$ 7.5 strategy where cell mass ($OD_{600\;nm}$) and PAL activity was 1.3 and 1.8 fold higher than those of $pH_{UC}$, respectively. The maximum PAL activity reached 123 U/g. The $pH_C$ 7.5 strategy made recombinant plasmid more stable and therefore allowed easier expression of PAL recombinant plasmid, which increased PAL production. It was indicated that the new approach (controlled-pH strategy) obtained in this work possessed a high potential for the industrial production of PAL, especially in the biosynthesis of L-phenylalanine.

Spinach Root-Tissue Based Amperometric Biosensor for the Determination of Hydrogen Peroxide (시금치 뿌리 조직 바이오센서를 이용한 과산화수소의 정량)

  • The response characteristics of the bioelectrode developed by the co-immobilization of spinach root tissue and ferrocene in a carbon paste matrix for the amperometric determination of hydrogen peroxide were evaluated. In the range of electrode potential examined (-0.3~0.0V vs. Ag/AgCl). the response time was relatively short ($t_{95%}=11.8$ sec) and it responded in the wide range of pH. Also, its detection limit was $2.25{\times}10^{-6}M$ (S/N=3) and a relative standard deviation of the measurements which were repeated 15 times using $1.0{\times}10^{-3}M$ hydrogen peroxide was 1.87%. The bioelectrode sensitivity decreased to 40% of the original value in 19 days of continuous use.

Nutritional Intake and Biochemical Status in Blood and Urine of Elderly Women

  • The present paper resents information on the dietary intake and biochemical status of elderly who are living in different types of residence. On hundred and sixty five women(age 65-80 years old) were divided into three groups ; those who are living alone (LA) , living with family (LF) , and living in institutions (LI) within the same general community. Nutrient intake, body fat content, and biochemical measurements in blood and urine were examined statistically and potential environment-related influence are discussed. Elderly women living alone had significantly lower intake of nutrients than elderly women were not living alone LF group showed significantly higher fat consumption than LA and LI groups. Body fat content, plasma albumin and calcium levels in elderly women who lived with other people were significantly higher than those in elderly women living alone, but fat weights and lean body mass between groups were not different. None of the plasma lipid fractions were significantly different among the three groups except LDL-cholesterol (LDL-C) concentration. In the LI group, LDL-C was lower than that of LF and LA groups. From the above results, serious nutritional deficiency has been shown in elderly women that live alone. Therefore, nutritional education and social help should be carried out to improve these situations for elderly people.

A method for discrete event simulation and building information modelling integration using a game engine

  • Building Information Modelling (BIM) and Discrete Event Simulation (DES) are tools widely used in the context of the construction industry. While BIM is used to represent the physical and functional characteristics of a facility, DES models are used to represent its construction process. Integrating both is beneficial to those interested in the field of construction management since it has many potential applications. Game engines provide a human navigable 3D virtual environment in which the integrated BIM and DES models can be visualised and interacted with. This paper reports the experience obtained while developing a simulator prototype which integrates a BIM and a DES model of a single construction activity within a commercial game engine. The simulator prototype allows the user to visualise how the duration of the construction activity is affected by different input parameters interactively. It provides an environment to conduct DES studies using the user's own BIM models. This approach could increase the use of DES technologies in the context of construction management and engineering outside the research community. The presented work is the first step towards the development of a serious game for construction management education and was carried out to determine the suitable IT tools for its development.

Smokeless Tobacco (Shammah) in Saudi Arabia: A Review of its Pattern of Use, Prevalence, and Potential Role in Oral Cancer

  • Background: Shammah is a traditional form of chewing tobacco [smokeless tobacco, (ST)] that is commonly used in the Middle East especially in Saudi Arabia (KSA), Yemen and Sudan. The Substance Abuse Research Centre (SARC) at Jazan University noted that no adequate research and information on the prevalence of shammah use in the province of Jazan, and KSA as well, has been provided in the scientific literature. Materials and Methods: An intensive systematic review of online databases was performed, including AMED (The Allied and Complementary Medicine Database), Biological Abstracts, Cochrane Collection Plus, Dentistry and Oral Sciences Source, E-Journals Database, EBSCO Discovery Service, MEDLINE, PEMSoft, PEP Archive, PsycARTICLES, scopus, Sciencedirect and Google Scholar. Results: Shammah is a mixture of powdered tobacco, lime, ash, black pepper, oils and flavorings. ST in KSA is placed in the buccal or lower labial vestibule of the mouth. The user (or dipper) spits out insoluble debris. The importation of ST products is prohibited in KSA. Accessible information on legislative action to control the use of ST in KSA appeared in 1990. The actual percentage use may be higher, than reported since shammah is illegal in KSA and there may be some reluctance to admit to its use. Conclusions: This review paper is an initial step in a funded research project by SARC to understand the pattern of use of shammah and provide adequate epidemiological data. One goal of this review is to generate further data for public health education.

  • Academic and research computer networks are the essential communications component that can provide scientists and engineers with convenient access to local and remote computing and information resources. Nowadays, academic and research computer networks, providing access to a variety of resources and information services, can significantly increase the productivity of scientists and engineers. In Korea there have been some activities of academic and research networks since early of 1980. This study presents background of development of computer network, summaries the types of services offered by academic and research computer networks and analyses the potential benefits resulting from their availability. Use and need of Korean researchers about academic and research computer networks are also reviewed as well as it prospects Korean Computer Network for Education and Research & Development, a new national-base network being implemented by government.

Antifungal Effect of Amentoflavone derived from Selaginella tamariscina

  • Amentoflavone is a plant biflavonoid that was isolated from an ethyl acetate extract of the whole plant of Selaginella tamariscina (Beauv.) spring. 1D and 2D NMR spectroscopy including DEPT, HMQC, and HMBC were used to determine its structure. Amentoflavone exhibited potent antifungal activity against several pathogenic fungal strains but had a very low hemolytic effect on human erythrocytes. In particular, amentoflavone induced the accumulation of intracellular trehalose on C. albicans as a stress response to the drug, and disrupted the dimorphic transition that forms pseudo-hyphae during pathogenesis. In conclusion, amentoflavone has great potential to be a lead compound for the development of antifungal agents.

A Review on Youth Risk Behavior Surveillance System of U.S.A (미국의 청소년 건강위험행위 감시체계에 관한 고찰)

  • Purpose: This study reviews the Youth Risk Behavior Surveillance System (YRBSS) in the United States. This review focuses on the purposes, questionnaires, survey methods, and use of survey results. The author also discussesthe establishment and use of Korean Youth Risk Behavior Surveillance System. Results:The YRBSS was developed in 1990 to monitor priority health risk behaviors that contribute to the leading causes of death, disability, and social problems among youth and adults in the United States. The purpose of YRBSS is to determine the prevalence of health risk behaviors, to assess the trend of health risk behavior prevalence, provide comparable data, monitor progress toward achieving the Healthy People 2010. The questionnaires inquire about health-related behaviors, including tobacco use, unhealthydietary behaviors, inadequate physical activity, alcohol and drug use, sexual activity, behaviors that contribute to unintentional injuries and violence. The results of YRBS have been used to estimate the prevalence of risk behaviors among youth, to set goals for school health programs, for training and development of health program personnel, for the development of health education curriculum, to support enactment of health-related regulations, and to support funding for futher research. Conclusions: The YRBSS represents a potentially valuable program that can be applied in Korea. Assessment of risk behaviors among Korean youths will provide invaluable insight for many potential purposes.

A Study on Pedagogy Researches in Serious Game Design (기능성 게임 설계에서의 교육학적 연구에 대한 고찰)

  • While there is consensus on the educational potential of serious games, there is still a lack of methodology and tools to support analysis and evaluation as well as design. This paper intends to explore the new methodology or design direction by analyzing pedagogy-related research required for serious games. To do this, we analyzed 4DF(Four Dimensional Framework), GOM(Game Object Model), LM(Learning Mechanics)-GM(Game Mechanics) and a framework for teacher. One of the biggest issues faced by educational games is the inadequate integration of education and game principles. This is due to a variety of factors, including the fact that game designers and educational professionals do not share a common vocabulary or work domain perspective.

The Effects of Delirium Care Training Program for Nurses in Hospital Nursing Units (병동 간호사를 대상으로 한 섬망간호 훈련 프로그램의 효과)

  • Purpose: The purpose of this study was to examine the effects of delirium care training program on the nurses' knowledge of delirium, self-confidence and performance levels in caring patients with delirium. Methods: The study was used a quasi-experimental design. The participants were 131 nurses in general nursing units in B and J hospitals. Sixty four nurses in B hospital were allocated into the experimental group and 67 nurses from J hospital into the control group. The delirium care training program was composed of lecture and clinical practice. Clinical practice of delirium care was applied everyday for 4 consecutive weeks starting from the admission day of the older adults(${\geq}70$ years old), using 'short CAM' and 'nursing checklist for delirium prevention'. Results: The knowledge of delirium, self-confidence in caring patients, and performance levels of nursing care were significantly increased in the experimental group compared to the control group. Conclusion: The findings of this study provided the evidence for the potential utility of the delirium care training program and underscored the needs of broader application of the training program of delirium care for nurses in general nursing units.
