Ecotoxicological Evaluation of Sewage Sludge Using Bioluminescent Marine Bacteria and Rotifer

  • Bioassay using the marine bacteria, Vibrio fischeri and rotifer, Brachionus plicatilis, and chemical analyses were conducted to assess the toxicity of the various sewage sludges, one of the major ocean dumped materials in the Yellow Sea of Korea. Sludge elutriates extracted by filtered seawater were used to estimate the ecotoxicity of the sludge. Chemical characterization included the analyses of organic contents, heavy metals, and persistent organic pollutants in sludge. Bacterial bioluminescent inhibition (15 min), rotifer mortality (24 hr) and rotifer population growth inhibition (48 hr) assay were conducted to estimate the sludge toxicity. EC50 15 min (inhibition concentration of bioluminescence after 15 minutes exposed) values by Microtox(R) bioassay clearly revealed different toxicity levels depending on the sludge sources. Highest toxicity for the bacteria was found with the sludge extract from dyeing waste and followed by industrial waste, livestock waste, and leather processing waste. Clear toxic effects on the bacteria were not found in the sludge extract from filtration bed sludge and rural sewage sludge. Consistent with Microtox(R) results, rotifer neonate mortality and population growth inhibition test also showed highest toxicity in dyeing waste and low in filtration bed and rural sewage sludge. High concentrations of persistent organic pollutants (POPs) and heavy metals were measured in the samples from the industrial wastes, leather processing plant waste sludge, and urban sewage sludge. However, there was no significant correlation between pollutant concentration levels and the toxicity values of the sludge. This suggests that the ecotoxicity in addition to the chemical analyses of various sludge samples must be estimated before release of potential harmful waste in the natural environment as part of an ecological risk assessment.

  • Polycyclic aromatic hydrocarbons (PAHs), butyltins (BTs), nonylphenol (NP), and fecal sterols concentrations in sediments were investigated from Gunsan coast of Korea to evaluate organic pollution from anthropogenic activities. Sediment toxicity was also examined by bacterial bioluminescence toxicity test (Vibrio fischeri). The concentrations of 16 PAHs in sediments ranged from 67.9 to 425 ng/g dry wt; BTs ranged from 2.79 to 14.1 ng Sn/g dry wt; NP ranged from 20.7 to 2171 ng/g dry wt; and coprostanol, a fecal sterol, ranged from 7.60 to 245 ng/g dry wt. Effective concentration 50% ($EC_{50}$) of sediments ranged from 0.38 to 23.8 mg/mL. Most of the chemicals were present at levels lower than or comparable to the previously reported values from Korea. However, NP levels in the present study were in the high range of levels reported from the Korean coast, and 40% of the measured samples exceeded screening and ecotoxicological values of NP suggested by the Netherlands and Canada. This suggests that an ongoing source of NP is a serious concern in the Gunsan coast. High levels of contaminants were found in the proximity of potential sources, such as the outfall of a wastewater treatment plant for NP, an anthracite-fired power plant for PAHs, and ports for BTs, fecal sterols, and sediment toxicity. This indicates that Gunsan coast has various potential sources of marine sediment contaminants.

Risk assessment and evaluation of epidermal growth factor (EGF) transgenic soybean: responses of Cyprinus carpio fed on EGF transgenic soybean

  • The epidermal growth factor (EGF) transgenic soybean was developed and biosynthesis of human epidermal growth factor (hEGF) in soybean seeds was confirmed. Also, EGF transgenic soybean were found to contain a herbicide resistance selectable marker by introduction of phosphinothricin acetyltransferase (PAT) gene from the Streptomyces hygroscopicus. For biosafety assessment, the EGF transgenic soybean expressing the EGF biosynthesis gene EGF and herbicide resistant gene PAT was tested to determine effects on survival of Cyprinus carpio, commonly used as a model organism in ecotoxicological studies. C. carpio was fed 100% ground soybean suspension, EGF soybean or non-genetically modified (GM) counterpart soybean (Gwangan). Gene expression of EGF soybean was confirmed by PCR and ELISA to have EGF/PAT. Feeding test showed that no significant differences in cumulative immobility or abnormal response between C. carpio samples fed on EGF soybean and non-GM counterpart soybean. The 48 h-EC50 values of the EGF and non-GM soybean were 1,688 mg·L-1 (95% confidence limits: 1,585 - 1,798 mg·L-1) and 1,575 mg·L-1 (95% confidence limits: 1,433 - 1,731 mg·L-1), respectively. The soybean NOEC (no observed effect concentration) value for C. carpio was suggested to be 625 mg·L-1. We concluded that there was no significant difference in toxicity for non-target organisms (C. carpio) between the EGF soybean and non-GM counterparts.

Toxicity study of cadmium and copper on riparian collembolan species Yuukianura szeptyckii (Neanuridae) (수변 서식 톡토기 Yuukianura szeptyckii (Neanuridae)에 대한 카드뮴과 구리 독성 연구)

  • The ecosystem provides a diverse array of environmental conditions for organisms, and only those that are capable of successfully adapting to these conditions within their habitats can endure, thrive, and proliferate. Further, the environmental conditions within these habitats can significantly affect the bioavailability of chemicals that are introduced therein, thus resulting in varied adverse impacts on the organisms. The present study aims to evaluate the sensitivity of Yuukianura szeptyckii - a species adapted to riparian - to heavy metals following ISO guideline 11276, with the objective of assessing its potential as an indicator species for ecotoxicological evaluations in riparian habitats. The findings revealed that cadmium and copper both had significant toxic effects depending on their concentrations. For cadmium, the LC50 was 280 mg kg-1, EC50 was 66 mg kg-1, and NOEC and LOEC were 25 and 50 mg kg-1, respectively. For copper, the LC50 was 911 mg kg-1, EC50 was 151 mg kg-1, and LOEC was 50 mg kg-1. Comparative analysis with previous results for the international standard species Folsomia candida and the domestic standard species Allonychiurus kimi indicated that Y. szeptyckii exhibited even greater sensitivity to toxicity values. The adverse effects on survival and reproduction were closely associated with the influx concentration of heavy metals in their bodies. Altogether, the results suggest that Y. szeptyckii is a sensitive species for ecotoxicological assessments in riparian habitats, thus making it suitable as an indicator species, particularly in riparian ecosystems that are characterized by relatively high humidity conditions.

Responses of Misgurnus anguillicaudatus and Cyprinus carpio Fed on Disease Resistant(OsCK1) Rice Variety (병저항성 GM(OsCK1)벼가 미꾸리(Misgurnus anguillicaudatus)및 잉어(Cyprinus carpio)에 미치는 영향)

  • BACKGROUND: The disease resistant (OsCK1) rice was generated by inserting choline kinase (CK1) and phosphinothricin acetyltransferase (PAT) genes isolated from Oriza sativa and Streptomyces hygroscopicus into the genome of rice (Nakdongbyeo). With the potential problems of safety, the non-target organism evaluation is required as an essential element for the environmental risk assessment of genetically modified (GM) crops. In present study, we studied the effects on survival of Misgurnus anguillicaudatus and Cyprinus carpio, commonly used as a model organism in ecotoxicological studies. METHODS AND RESULTS: The M. anguillicaudatus and C. carpio were fed on disease resistant (OsCK1) rice and non-genetically modified (non-GM) rice (Nakdongbyeo) to 0, 10, 100, 1,000 and 5,000 mg/L, as treatment concentration respectively. The OsCK1 rice used for the test was confirmed to have the OsCK1/PAT gene expression by the PCR and ELISA analysis. Feeding test showed that no significant differences in cumulative immobility and abnormal response of M. anguillicaudatus and C. carpio fed on between OsCK1 rice and non-GM rice. The 96hr-$LC_{50}$ values showed no difference between OsCK1 rice (>5,000 mg/L) and non-GM rice (>5,000 mg/L). CONCLUSION(S): The results of this study suggested that there was no significant difference in toxicity for M. anguillicaudatus and C. carpio between OsCK1 rice and non-GM counterparts.

  • This study was carried out to examine the drift of Gammarus sobaegensis by acid stress as pH depression in Oweol creek from April 1996 to October 1996. The behavioral drifting was revealed to the characteristics of G. sobaegensis that is tend to increase as acid stress. And, tolerance level of G. sobaegensis to pH depression was different among the size classes. Individuals belong to small to medium size classes were weak in lower pH. Pattern of response in G. sobaegensis has a lower tolerance to acid stress at below pH 4.0 than above pH 5.0 in the artificial channel and show the possibility as an effective aquatic ecotoxicity test organism. The result of analysis of variance, water temperature (F-ratio : 66.596, p< 0.0005) and the size classes (F-ratio : 71.386, p< 0.0005) except pH level (F-ratio : 353.415, p<0.0005) were showed to the major factor for drift behavior by acid depression. Changes in Susceptibility of Killifish (Oryzias latipes) to Three Pesticides with Growth (농약에 대한 송사리 생육단계별 감수성의 변화)

  • Sensitivity, expressed in the $96h-LC_{50}$ determined with continuous flow system, was compared for eight developmental stages of Oryzias latipes with butachlor, diazinon and fenitrothion. Significance of the laboratory determined $96h-LC_{50}$ in real field situation was evaluated for butachlor using outdoor model agrosystem. In the laboratory tests, the most sensitive stage was post-larva stage IV(7 days after hatching) with $96h-LC_{50}$ of 0.14, 1.4, and 1.6ppm for butachlor, diazinon and fenitrothion, respectively, whereas the least sensitive stage was early embryo stage(1 day after fertilization), and then the susceptibility was somewhat stable after 21days after hatching. In the model agrosystem test, the highest concentration of 0.53ppm of butachlor in water was obtained at the 2nd day after application. The stages IV and VIII showed mortality of over 50% at 96 hours after application. It is concluded that the conventional use of butachlor in the field would cause significant adverse effect on O. latipes. It is suggested that sensitivity test at the early life stages with fish is an important part of the ecotoxicological assessment of chemicals.

  • We developed insect-resistant GM rice(Bt transgenic rice) by inserting the mCry1Ac1 a modified gene from the soil bacterium, Bacillus thuringiensis. The Bt transgenic rice expressing the Bttoxin mCry1Ac1 was tested for the effects on survival of Misgurnus anguillicaudatus and Cyprinus carpio, commonly used as a model organism in ecotoxicological studies. M. anguillicaudatus and C. carpio fed 100% ground rice in suspension, using either Bt rice or non-GM counterpart rice(Nakdong). The Bt rice used for the test were confirmed to have the mCry1Ac1 gene expression by the immuno-strip and ELISA analysis. Feeding test showed that no significant differences in cumulative immobility and abnormal response of M. anguillicaudatus and C. carpio fed on between Bt rice and non-GM counterpart rice. The 96hr-LC50 values showed no difference between Bt rice(>1,000mg/L) and non-GM rice(>1,000mg/L).   • Toxicity and fate assessment is necessary in the evaluation of the environmental, health and safety risks of engineered nanomaaterials (ENMs). Therefore, in order to ensure the reproducibility, reliability and relevance of ENMs toxicity results, stable and monomodal dispersion protocols in toxicity test media are needed. Zinc oxide nanoparticles (nZnO) are widely used in various products such as cosmetic products, paper, paints etc. In this study, nZnO dispersions in ecotoxicity test media were produced by following a series of steps of modified National Institute of Standards and Technology (NIST) Special publication 1200-5. In addition, natural organic matter (humic acid (HA)) was used as a stabilizing agent to disperse nZnO in the test media. The hydrodynamic diameters (HDD) of the nZnO in dispersion ranged between 150 and 200 nm according to the dynamic light scattering (DLS) measurement. Based on these dispersions in ecotoxicity test using ecological species (Oryzias latipes, Daphnia magna, Pseudokirchneriella subcapitata and Chironomusus riparius), dispersion protocol was found to have a considerable potential in ecotoxicity test of ENMs.

Development of Marine Ecotoxicological Standard Methods for Ulva Sporulation Test (파래의 포자형성률을 이용한 해양생태독성시험 방법에 관한 연구)

  • As an aquatic ecotoxicity test method, a bioassay using the inhibition of sporualtion of the green macroalga, Ulva pertusa, has been developed. Optimal test conditions determined for photon irradiance, pH, salinity and temperature were $100\;{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, $7{\sim}9$, $25{\sim}35\;psu$ and $15{\sim}20^{\circ}C$, respectively. The validity of the test endpoint was evaluated by assessing the toxicity of four metals (Cd, Cu, Pb, Zn) and elutriates of sewage or waste sludge collected from 9 different locations. When the metals were assayed, the $EC_{50}$ values indicated the following toxicity rankings: Cu ($0.062\;mg{\cdot}L^{-1}$) > Cd ($0.208\;mg{\cdot}L^{-1}$) > Pb ($0.718\;mg{\cdot}L^{-1}$) > Zn ($0.776\;mg{\cdot}L^{-1}$). When compared with other commonly used bioassays of metal pollution listed on US ECOTOX database, the sporualtion test proved to be the most sensitive. Ulva sporulation was significantly inhibited in all elutriates with the greatest and least effects observed in elutriates of sludge from industrial waste ($EC_{50}=6.78%$) and filtration bed ($EC_{50}=15.0%$), respectively. The results of the Spearman rank correlation analysis for $EC_{50}$ data versus the concentrations of toxicants in the sludge presented a significant correlation between toxicity and four heavy metals(Cd, Cu, Pb, Zn). The method described here is sensitive to toxicants, simple to use, easy to interpret and economical. It is also easy to procure samples and maintain cultures. The present method would therefore probably make a useful assessment of aquatic toxicity of a wide range of toxicants. In addition, the genus Ulva has a wide geographical distribution and species have similar reproductive processes, so the test method would have a potential application worldwide.
