  • The germinatin of e. annuus continued from the middle ofmay to mid-october. The maximum germination occurred on the mid-july. The period bloom of was distingushed amongs the different growth forms ; a orm pr of biennial and a form of pr perennial flowering from the mid-may to mid-september, and a form ps biennial blossom from the beginning of October to earlynovember. the dispersal of seed for(a form pr)occurred from early June to the mid-september. A rotte, germinating from summer to autumn, could classified into several growth forms; individuals without a critical leaf area for bolting until september and October, become a form ps of biennial, whicth did not proceed toreproductive growth unitl the next year, even thought wintering. individuals flowered on 3 years after germination become a form pr of perennial. The growth formular of aform pr of bennial, grown in a pot was w=20.2[1+$3.36{\times}10^3$(-0.062t)]$^{-1}$. The maximum relative growth rate(rgr) was 0.062g/g/day and the maximum net assimlation rate(nar) 0.089g/g/day. Therelative growth among each organ was shown as R=0.12 $T^{1.15}$between the avove-ground part(t) and the below- ground part(r). the relation between the avove-ground part(t) and the ratio of stem weight(wi) was ws/wi=2.56 $T^{0.35}$. n.p.k. was largely distributed on a leaf throughoutthe total growth period. while growing, it tended to decrease on the vegetative organ gut vice versa on the reproductive organ. however, nitrogen was more widely distributed on a leaf then in the reproductive organ.

High plant regeneration and ectopic expression of OsMADS1 gene in root chicory (Cichorium intybus L. var. sativus)

  • Optimal shoot regeneration and transformation conditions of root type chicory (Cichorium intybus L. var. sativus cv Cesare) were studied. Leaf explants were co-cultured with Agrobacterium tumefaciens, which contained NPTII as a selectable marker and a rice homeotic gene, OsMADS1, that encodes a MADS-domain-containing transcription factor. After one day of co-cultivation, explants were transferred to selection media consisting of MS basal medium supplemented with 0.5 mg/L BAP, 0.1 mg/L IAA, 70 mg/L kanamycin, and 250 mg/L cefotaxime. PCR and Southern blot analyses revealed stable integration of the OsMADS1 gene in the chicory genome. Four-teen original transgenic plants ($T_o$ plants) were acclimatized in the greenhouse and examined for their morphological characters. Most of the transgenic plants showed altered morphologies, such as short, bushy, and early-flowering phenotypes with reduced apical dominance. Additionally, half of the transgenic plants exhibited altered leaf shapes, and 4 out of 14 plants were sterile. These phenotypes were inherited by the next generation. Northern blot analysis confirmed expression of the OsMADS1 gene in both floral and vegetative organs.

Effects of soil covering materials on the overwintering and the growth of Cynanchum wilfordii Hemsley

  • This study was performed to select the soil covering materials for winter safety of Cynanchum wilfordii Hemsley and to investigate the effect on the growth after overwinter. Soil covering materials such as vinyl, rice straw, lagging cover, vinyl+lagging cover, and non-covering were investigated how effect on overwintering rate, growth and yield of Cynanchum wilfordii Hemsley. In changes of soil temperature according to the soil covering materials, non-covering treatment was recorded the highest to $26.7^{\circ}C$, and lagging cover and rice straw showed little change of soil temperature with 9.2 and $9.3^{\circ}C$, respectively. Regrowth rates of early growth after overwinter were lowered in the order as rice straw > vinly+lagging cover > lagging cover > vinyl > non-covering. From the above-ground growth by soil covering materials, vine length showed similar growth in all treatments except vinyl covering and stem diameter exhibited the thicker pattern in vinyl and lagging cover, and there was no significant difference among treatments in number of branch. Flowering period is recorded July 30 in all treatments and ripening period is October 16. Yield of seed production is showed the best in vinyl+lagging cover with $25g/m^2$. Root weight and root length for subterranean growth showed no significant differences among all treatments. Yield is produced the highest in vinyl+lagging cover with 433kg/10a and the next to lagging cover with 431kg/10a. In comparison with incomes, lagging cover earned to 9,882 thousand won/10a, showed the effect on income enlargement to 21%, compare to that of the non-covering.

  • Zostera caespitosa Miki has been identified to be distributed along the seacoasts of Korea and Japan. This study was intended to clarify the morphological characters and the habitat characteristics of Korean Z. caespitosa. It was confirmed that Z. caespitosa is distributed along the seacoasts of South, Yellow and East Seas of South Korea. The habitats were located in the bay and port with the depth varying from 2.5 to 5.2 m. The habitats of Z. caespitosa were rather deeper than that of mixed bed with Z. marina. The sediment in the habitats was composed of well-sorted fine sand or muddy sand. Z. caespitosa showed marked differences in several morphological characters among sites. Morphology of Z. caespitosa varied with water depths, grain size and organic contents of sediments. Flowering shoots occurred by water temperature of 6.0∼13.7℃ from January to early April.

The Chloroplast rpl23 Gene Cluster of Spirogyra maxima (Charophyceae) Shares Many Similarities with the Angiosperm rpl23 Operon

  • A phylogenetic affinity between charophytes and embryophytes (land plants) has been explained by a few chloroplast genomic characters including gene and intron (Manhart and Palmer 1990; Baldauf et al. 1990; Lew and Manhart 1993). Here we show that a charophyte, Spirogyra maxima, has the largest operon of angiosperm chloroplast genomes, rpl23 operon (trnⅠ-rpl23-rpl2-rps19-rpl22-rps3-rpl16-rpl14-rps8-infA-rpl36-rps11-rpoA) containing both embryophyte introns, rpl16.i and rpl2.i. The rpl23 gene cluster of Spirogyra contains a distinct eubacterial promoter sequence upstream of rpl23, which is the first gene of the green algal rpl23 gene cluster. This sequence is completely absent in angiosperms but is present in non-flowering plants. The results imply that, in the rpl23 gene cluster, early charophytes had at least two promoters, one upstream of trnⅠ and and another upstream of rpl23, which partially or completely lost its function in land plants. A comparison of gene clusters of prokaryotes, algal chloroplast DNAs and land plant cpDNAs indicated a loss of numerous genes in chlorophyll a+b eukaryotes. A phylogenetic analysis using presence/absence of genes and introns as characters produced trees with a strongly supported clade containing chlorophyll a+b eukaryotes. Spirogyra and embryophytes formed a clade characterized by the loss of rpl5 and rps9 and the gain of trnⅠ (CAU) and introns in rpl2 and rpl16. The analyses support the hypothesis that the rpl23 gene cluster and the rpl2 and rpl16 introns of land plants originated from a common ancestor of Spirogyra and land plants.

  • This experiment was carried out to determine the adequate amount of nitrogen for densely and lately seeded soybean, and the results obtained are as follows, 1) Various fertilization amounts of nitrogen did not effect the date of emergency, but the flowering and the maturing dates were delayed at the plots fertilized the nitrogen heavily. 2) In accordance with an increasing amount of nitrogen fertilization, the length of the stem and the internode was significantly elongated but the stem diameter was thinned down, and the latter was suggested to be the cause of lodging. 3) The 1, 000 grain weight and the number of pods per hill were decreased according to the increasing the amount of nitrogen fertilization. 4) A highly significant difference in grain yield was recognized between varieties but among treatments. However, in case of the extremly late cultivation, the grain yield of the early maturing variety (Choongbukbaeg) was increased at the plot of 6 kg nitrogen.

  • Several experiments were carried out to transfer LEAFY gene to Dendranthema grandiflora 'Shuho-no-chikara' by Agrobacterium LBA4404 carrying pSK109 encoding LEAFY gene. Kanamycin 10mg/L was used in first selection medium, and 20mg/L in the second one. Co-culture for 3 days was more effective in increasing transformation efficiency than that for 7 days. The transformation efficiency by Agrobacterium LBA4404 carrying pSK109 encoding LEAFY gene was about 2.8% until the second selection, but only 0.13% of shoots (two plants) was confirmed as a transgenic plants in Southern analysis. The escape of putative transformants was occured seriously in the process of selections, PCR analysis for confirming of neomycin phosphotransferaseII (npt II), and Southern analysis for LEAFY gene. One transgenic plant appeared 7 days'early flowering in field.

Effects of Polyethylene Film Mulching and Planting Dates on Growth and Yield of Summer Type of Soybean (Glycine max) (하대두(Glycine max)의 파종기 및 비닐 멀칭재배가 생육 및 수량에 미치는 영향)

  • To study the effects of planting dates and mulching on the growth and yield of summer type soybean, a variety, Damamidori introduced from Japan, was planted four times from April 13 with 10 days interval. Experimental plot in each planting date was devided into two sub-plots, polyethylene film mulching and non-mulching. By mulching, days to emergence and first flowering were shortened, especially in the early plantings; e.g. as much as 11 and 12 days, respectively, in the first planting. Compared to non-mulching, plant height, number of nodes of main stem, number of branches, and dry weight of stem and branches per unit area were also much increased by mulching. As the plantings were delayed, plant height, the total number of nodes and dry weight of main stem and branches, and 100 seed weight showed increasing trends, which were mainly due to lower infection rate of soybean seeds to pod and stem blight (Diaporthe phaseolorum) in the later plantings. The higher infection rate of soybean seeds to pod and stem blight, the lower infection rate to purple seed stain (Cercospora kikuchii) was shown in mulching but not in non-mulching.

Environmental Mechanism on Seeding Stage and Ripening Period in Labour Saving-Direct Sowing Rice Culture (벼 성력재배를 위한 담수직파 파종시기와 등숙환경 분석)

  • Lodging and rainfalls during ripening periods and ripening patterns were studied by the different maturing varieties and different seeding dates in labour saving-direct seeding cultivation of rice. Period of emergency in earlier seeding plot was longer than later seeding plot and standing seedlings per unit area and rate of standing seedlings were more and high by the later seeding date. Number of panicles per unit area was decreased by the later seeding dates. On the other hand, the rate of effective tillers was lowest at later seeding plot. Days from seeding to flowering were shortened by the later seeding dates due to the increase of daily growth temperature and reduce of day length. There was much rainfalls during ripening period of early maturing varietal group, however not much rainfalls during ripening period of medium and late maturing varietal groups. The degree of lodging at field was severer by the amount of rainfalls during ripening period. Earlier seeding plots and earlier maturing varietal groups. of used varieties were revealed heavier lodging compared to later seeding plots and later maturing varietal groups. Yield and yield related characteristics were high and good by the earlier seeding dates, especially ripening rate was clear.

Prospect and Situation of Quality Improvement in Oilseed rape (유채 품질 평가 현황과 전망)

  • Rapeseed(Brassica napus L.) is an important oil crop as a vegetable oil, concentrated feed and industrial materials. The name "canola" was registered in 1979 by the Western Canadian Oilseed Crushers Association to describe "double-low" varieties. Double low indicates that the processed oil contains less than 2% erucic-acid and the meal less than 3mg/g of glucosinolates. Today annual worldwide production of rapeseed is approximately 35 million tons on 24 million hectares. China accounts for 33% of the world production and the European Economic Community for nearly 32%. Canola ranks 3rd in production among the world's oilseed crops following soybeans, sunflowers, peanuts and cottonseed. The recent advances in genomics and in gene function studies has allowed us to understand the detailed genetic basis of many complex traits, such as flowering time, height, and disease resistance. The manipulation of seed oil content via transgene insertion has been one of the earliest successful applications of modern biotechnology in agriculture. For example, the first transgenic crop with a modified seed composition to be approved for unrestricted commercial cultivation in the US was a lauric oil, rape-seed, grown in 1995. There were also some significant early successes, mostly notably the achievement of 40% to 60% lauric acid content in rapeseed oil, which normally accumulates little or no lauric acid. The name "$\textrm{Laurical}^{TM}$" was registered in 1995 by Calgene Inc. Nevertheless, attempts to achieve high levels of other novel fatty acids in seed oils have met with much less success and there have been several reports that the presence of novel fatty acids in transgenic plants can sometimes lead to the induction of catabolic pathways which break down the novel fatty acid, i.e. the plant recognizes the "strange" fatty acid and, far from tolerating it, may even actively eliminate it from the seed oil. It is likely that, in the future, transgenic oil crops and newly domesticated oil crops will both be developed in order to provide the increased amount and diversity of oils which will be required for both edible and industrial use. It is important that we recognize that both approaches have both positive and negative points. It will be a combination of these two strategies that is most likely to supply the increasing demands for plant oils in the 21st century and beyond.ant oils in the 21st century and beyond.
