Objectives The purpose of this study was to investigate the effects of AI on AD induced by DNCB in mice. AI has antiallergic property that is useful in treating allergy-related-diseases, such as asthma, anaphylactic shock, acute bronchitis and skin diseases, skin pruritus from gastrointestinal diseases. However, AI has not been studied intensively yet regarding anti-inflammatory effect on AD. Therefore, this study was conducted on 2,4-dinitrochlorobezene (DNCB)-induced mice to investigate effects of AI in AD. Methods In the experiment, we divided mice into four groups: a normal group (NOR), a control group (CON), an AI spread group (AI spread), and an AI spread and feeding group (AI spread & feeding). Then examined the changes in the body weight, weights of spleen and ear, thickness of dorsum skin and ear skin, clinical aspects on dorsum skin, historical assessments, proliferation of splenocytes in vitro and in vivo, and cytokine (TNF-${\alpha}$, IL-10). Results From the experiment, the ear weight of AI spread & feeding group was significantly dropped and the ear thickness of both AI spread and AI spread & feeding were decreased significantly. Dorsum skin thickness was also decreased significantly in both AI spread and AI spread & feeding group. Also, AI treatment improved the symptoms of AD, such as coloration, erythema and desquamation and had a better effect on AI spread & feeding group. In histopathological observation, thickened epidermis, hyperkeratosis, pigmentation, hypergranulosis, parakeratosis were diminished as well in both AI spread and AI spread & feeding group. In vitro, we could observe when AI was increased as proliferation rate of splenocytes were increased, too. Conclusions In conclusion, these data suggest that AI can decrease symptoms of AD and show AI can be useful herbal therapy for AD.
The aim of this study was to compare ultrasonography with video-otoscopy for the evalution of the external ear canal and the tympanum. Fifty dogs with clinical signs ot the bilateral ear diseases were examined using otoscopy under general anesthesia. Ultrasonographic examination was performed with an 11 MHz linear-arrary probe and a 6.5MHz convex probe, with the external ear canal filled with saline. Saline functioned as a contrast media and enhanced image quality of ultrasonography. The images of 100 ears before and after the ear cleansing were obtained at otoscopy and ultrasonography. On the video-otoscopic images, the tympanic membrane was identified in 59 ears before the ear cleansing and 91 ears after cleansing. On the ultrasonography, the tympanic membrane was detected in 98 ears before the ear cleansing and in all ears after cleansing. With these results, it was concluded that ultrasonography with infusing saline into the ear canal, could be a useful diagnostic procedure to identify the tympanic membrane in otitis dogs.
Scutellaria baicalensis Georgi (SB) has been widely studied to treat inflammatory diseases in east Asia. In the recent years, many studies have focused on modifying herbs to increase the pharmacological effects. Roasting alcohol absorbed SB is one of the traditional methods to increase the therapeutic effects. Currently there are no reports on the pharmacological effects of roasted SB. This study investigated the anti-inflammatory effects of roasted 30% ethyl alcohol absorbed SB extract (SR) on mice ear edema. After intra-gastric injection of dexamethasone (for positive control, 2 mg/kg) and SR (50, 100, 400 mg/kg), ear edema was provoked by croton oil (5% v/v in acetone, 10 ul/ear). Ear thickness was measured with a digital caliper to quantify the change in swelling. For histological study, we made paraffin sections and performed Phloxine-Tartrazine staining and Masson's trichrome staining to observe epidermis, dermis and subcutaneous region and collagen fiber of mice ear tissues. Ear thickness decreased dose-dependent manner in SR treated groups. Histological analysis compared with dexamethasone treated group, SR treated groups demonstrated a similar reduction in hypoplasia of epidermis and influx of inflammatory cells. Increase of subcutaneous layer and decrease of collagen fibers were significantly recovered in SR treated group (400 mg/kg) and dexamethasone treated group. In conclusion, treatment with SR ameliorates auricular inflammation induced with croton oil in mice. Experiments are now underway to understand the molecular mechanisms underlying anti-inflammatory activities of SR.
Background: The effect of ginsenoside Rh2 (G-Rh2) on mast cell-mediated anaphylaxis remains unclear. Herein, we investigated the effects of G-Rh2 on OVA-induced asthmatic mice and on mast cell-mediated anaphylaxis. Methods: Asthma model was established for evaluating airway changes and ear allergy. RPMCs and RBL-2H3 were used for in vitro experiments. Calcium uptake, histamine release and degranulation were detected. ELISA and Western blot measured cytokine and protein levels, respectively. Results: G-Rh2 inhibited OVA-induced airway remodeling, the production of TNF-α, IL-4, IL-8, IL-1β and the degranulation of mast cells of asthmatic mice. G-Rh2 inhibited the activation of Syk and Lyn in lung tissue of OVA-induced asthmatic mice. G-Rh2 inhibited serum IgE production in OVA induced asthmatic mice. Furthermore, G-Rh2 reduced the ear allergy in IgE-sensitized mice. G-Rh2 decreased the ear thickness. In vitro experiments G-Rh2 significantly reduced calcium uptake and inhibited histamine release and degranulation in RPMCs. In addition, G-Rh2 reduced the production of IL-1β, TNF-α, IL-8, and IL-4 in IgE-sensitized RBL-2H3 cells. Interestingly, G-Rh2 was involved in the FcεRI pathway activation of mast cells and the transduction of the Lyn/Syk signaling pathway. G-Rh2 inhibited PI3K activity in a dose-dependent manner. By blocking the antigen-induced phosphorylation of Lyn, Syk, LAT, PLCγ2, PI3K ERK1/2 and Raf-1 expression, G-Rh2 inhibited the NF-κB, AKT-Nrf2, and p38MAPK-Nrf2 pathways. However, G-Rh2 up-regulated Keap-1 expression. Meanwhile, G-Rh2 reduced the levels of p-AKT, p38MAPK and Nrf2 in RBL-2H3 sensitized IgE cells and inhibited NF-κB signaling pathway activation by activating the AKT-Nrf2 and p38MAPK-Nrf2 pathways. Conclusion: G-Rh2 inhibits mast cell-induced allergic inflammation, which might be mediated by the AKT-Nrf2/NF-kB and p38MAPK-Nrf2/NF-κB signaling pathways.
Bee venom (BV), well known as a traditional Oriental medicine, has been widely used in the treatment of some immune-related diseases. However, the anti-allergic effect of BV have not been reported. In this study, we investigated the antiallergic effect of BV on triphasic cutaneous reaction (TpCR) and passive cutaneous anaphylaxis (PCA). Our results indicated that BV suppress ear swelling and vascular permeability on IgE mediated type I hypersensitivity response. Increase in ear thickness was significantly inhibited by BV in this model. BV also blocked the infiltration of immune cells into the ear. Moreover, BV suppressed expression of HDAC3, Tryptase, MCP-1 in ear tissue. These results demonstrated that BV has a suppressive effect on allergic reaction.
Objectives In the theory of Korean Medicine, Cicadae Periostracum (CP) has been used to treat skin diseases such as inflammatory dermatitis, tetanus and pruritus. CP can reduce heat and disperse wind. In prior studies, anti-allergic effect and anti-inflammatory effect of CP were reported. However, there has been no report regarding the correlation of CP and allergic contact dermatitis (ACD). This study was performed to show the effects of CP in ACD induced by 2,4-dinitrochlorobenzene (DNCB) in mice. Methods In this experiment, the effects of CP on biological changes were measured, such as changes in ear and spleen weight, ear and dorsum skin thickness, clinical aspect on the dorsum skin and histological changes. The effect of proliferation rate of splenocytes was also investigated in vitro and vivo study. Results In results, CP application (CPA) group and CP application and administration (CPAA) group were significantly restrained in ear weight gain and increase in ear and dorsum skin thickness compared to the control group. In addition, CPA and CPAA group showed diminished erythema, desquamation, bloodstain, and marks. Also, the histological assessment showed that CP treatement diminished thickness of epidermis, hyperkeratosis, pigmentation and parakeratosis. Conclusions In conclusion, these data suggest that CP can decrease symptoms of ACD.
Background: Thymic stromal lymphopoietin (TSLP) acts as a master switch for inflammatory responses. Ginsenoside Rg3 (Rg3) which is an active ingredient of Panax ginseng Meyer (Araliaceae) is known to possess various therapeutic effects. However, a modulatory effect of Rg3 on TSLP expression in the inflammatory responses remains poorly understood. Methods: We investigated antiinflammatory effects of Rg3 on an in vitro model using HMC-1 cells stimulated by PMA plus calcium ionophore (PMACI), as well as an in vivo model using PMA-induced mouse ear edema. TSLP and vascular endothelial growth factor (VEGF) levels were detected using enzyme-linked immunosorbent assay or real-time PCR analysis. Murine double minute 2 (MDM2) and hypoxia-inducible factor 1α (HIF1α) expression levels were detected using Western blot analysis. Results: Rg3 treatment restrained the production and mRNA expression levels of TSLP and VEGF in activated HMC-1 cells. Rg3 down-regulated the MDM2 expression level increased by PMACI stimulation. The HIF1α expression level was also reduced by Rg3 in activated HMC-1 cells. In addition, Rg3-administered mice showed the decreased redness and ear thickness in PMA-irritated ear edema. Rg3 inhibited the TSLP and VEGF levels in the serum and ear tissue homogenate. Moreover, the MDM2 and HIF1α expression levels in the ear tissue homogenate were suppressed by Rg3. Conclusion: Taken together, the current study identifies new mechanistic evidence about MDM2/HIF1α pathway in the antiinflammatory effect of Rg3, providing a new effective therapeutic strategy for the treatment of skin inflammatory diseases.
Background and purpose: The chronic diseases caused by lifestyle are on the increase. This study aims to review the eating habits as etiological factors and related symptoms from the perspective of Korean medicine. In this process, we will make a proposal on the treatment of the lifestyle related diseases. Methods: We studied the sentences about the eating habits and related diseases in Donguibogam Results and Conclusions: 1. The eating habits as etiological factors are overeating, irregular eating, late-night foods, fatty & heavy foods, cold foods, alcohols, etc. 2. The diseases caused by the eating habits are not limited to the gastrointestinal diseases(stomachache, vomiting, diarrhea, etc) but include the non-gastrointestinal diseases(edema, arthralgia, anal diseases, eye diseases, ear diseases, diabetes, cancers, etc.) 3. In the treatment of the diseases caused by the eating habits, the special regard should be paid to the etiological factors.
Recently, demographic studies of veterinary medical database have been conducted to understand patterns of disease occurrence. Understanding incidence of breed-related disease would provide appropriate guidance for future health care strategies and offer useful information for early diagnosis of disease. However, in veterinary medicine, theses research has not yet been investigated in the Republic of Korea. The purpose of this study was to investigate the prevalence of common medical disorders among dog breeds examined at primary-care veterinary clinics in Jeollabuk-Do, Republic of Korea. The data were analyzed based on World Health Organization's International Classification of Disease. A total 13,176 medical records of canine patients were analyzed from six primary veterinary clinics in Jeollabuk-Do from January to December 2016. Results showed that the most common health problems were 'disease of skin' (17.7%); followed by 'diseases of digestive system' (12.26%), 'preventive medicine' (10.08%), and 'diseases of ear and mastoid process' (10.4%). In seven out of ten breeds, the most common medical disorder was skin disease. For poodle such as Pomeranian and Chihuahua, digestive system disease was most prevalent. On the other hand, respiratory system disease was found to be higher in Pomeranian than other breeds; while ear and mastoid process disease was most common for Maltese and Poodle. This study can help owners, breeders, and veterinarians prevent and manage various diseases of popular breeds in Jeollabuk-Do in the future.
