배양 조건에 따른 Vibrio harveyi extracellular products (ECPs)의 특성 (Characteristics of the extracellular products (ECPs) of Vibrio harveyi grown under various conditions)

  • 본 연구에서는 Vibrio harveyi의 주요 병원성 인자로 알려져 있는 ECPs를 분리하는 적정 조건을 제시하고자 하였다. 5 개의 분리균주 (FF8, FF10, FR1, FR2 and FT1 strain)와 2 개의 참조균주 (ATCC 35084 및 ATCC 14126)의 ECPs를 다양한 조건에서 분리해 본 결과, 1.5% NaCl 첨가 TSA 배지에서 25℃, 24 시간 배양하는 것이 가장 좋으며, ECPs 분리시의 buffer system은 pH 8.0이 적정한 것으로 나타났다. 나아가 우리 나라에서 분리한 5 개의 분리 균주들의 주요 ECPs 성분이 serine protease인 것으로 조사되었다.

  • Vibrio harveyi, one of the major causal agent of vibriosis, affects a diverse range of marine vertebrates and invertebrates over a wide geographical area. The aims of this study were to investigate the characteristics of extracellular products (ECPs) from an isolate of the pathogenic non‐luminous V. harveyi and the luminous V. harveyi. And ECPs of V. harveyi were examined the pathogenicity to the black rockfish, Sebastes schlegeli, and histopathological traits of internal organs injected by ECPs. Four strains of V. harveyi cultures produced ECPs showing various enzymatic activites (caseinase, gelatinase, phospholipase, lipase, haemolysin). The ECPs showed strong cytotoxicity on macrophages of black rockfish, Sebastes schlegeli and olive flounder, Paralichthys olivaceus. Especially, the virulence of ECPs from the isolate of V. harveyi FR 2 was higher in the intraperitoneally injected black rockfish. Also, the ECPs of V. harveyi FR 2 caused the expansion of sinusoids in the liver, the activation of ellipsoid in the spleen and the sloughing of the epidermal cell in the intestine. It was suggested that the ECPs from V. harveyi play an important role in the pathogenicity process of the V. harveyi.

여고생들의 응급피임약에 대한 지식, 태도 및 사용의도 (Emergency Contraceptive Pills: Knowledge, Attitude, and Intention of High School Girls in Korea)

  • Purpose: The purpose of this study was to explore the knowledge of, attitude toward, and intention to use Emergency Contraceptive Pills (ECPs) and their relationships among Korean high school girls. Methods: This study is a cross-sectional correlational survey. Data were collected from 191 high school girls by self-report questionnaires from December 10 to 22, 2008. Result: The mean scores of knowledge, attitude, and intention were 4.74 (range 1 to 12), 2.60 (range 1 to 5), and 3.36 (range 1 to 5) respectively. Only 45% of participants had received education on ECPs and most respondents (77.5%) wanted to know more about it. Participants who had a more positive attitude toward ECPs had a greater intention to use it. Participants who had received education on ECPs had more knowledge than those who had not received education. However, there were no differences in attitude or intention between groups. Conclusion: This study indicates that efforts should be made to provide education on ECPs to high school girls to increase awareness and to minimize the misuse of ECPs. Also, educational programs should be designed to improve knowledge as well as promote a more positive attitude toward ECPs.

Edwardsiella tarda extracellular products (ECPs)의 인위투여에 따른 넙치, Paralichthys olivaceus의 생체반응 (Effect of extracellular products (ECPs) of Edwardsiella tarda on olive flounder, Paralichthys olivaceus)

  • 본 연구는 강독성 균주인 E. tarda PoKF-000623가 생산하는 세포외생성물 (ECPs)을 넙치에 투여되었을 때 나타나는 생리학적 및 면역학적 영향을 조사하고자 하였다. E. tarda PoKF-000623 균주가 생성하는 ECPs의 넙치 (평균체중, 5.6g)에 대한 96hr-$LD_{50}$ 은 약 0.714${\mu}g$ g TEX>$fish^{-1}$ 이었으며, ECPs의 주사에 의한 폐사는 고농도 (40${\mu}g$ TEX>$fish^{-1}$) 뿐만 아니라 저농도 (4${\mu}g$ TEX>$fish^{-1}$)에서도 지속적으로 발생하였다. 평균 체중 59.1 g 크기의 넙치에 각각 0, 4 and 40${\mu}g$ ECPs TEX>$fish^{-1}$ 의 농도로 근육 주사한 후, 혈청 중 glucose 농도와 total protein 농도 및 신장 식세포의 활성을 조사하였다. 그 결과 고농도의 ECPs 주사 시험구의 넙치에서 혈중 glucose 농도와 total protein 농도가 대조구에 비하여 유의적으로 높게 나타났다. 그러나 세포성 면역 기구인 신장 식세포의 CL 반응과 세균 살해능은 대조구에 비하여 유의적으로 저하되었다. 따라서 넙치에 고농도의 E. tarda ECPs를 투여하면 생체 지표의 일부에 영향을 미치는 것으로 확인되었고 면역 활성, 특히 신장 식세포 활성의 억제가 edwardsielosis의 발병에 영향을 미치는 것으로 사료된다.

미혼 여성의 응급피임약에 대한 인식 (Perceptions of Emergency Contraceptive Pills among Young Korean Women)

  • Purpose: The purpose of this study was to examine the perceptions regarding the use of Emergency Contraceptive Pills (ECPs) among young Korean women. Method: The study included 14 unmarried women aged 19 to 34 years (mean, 25 years). Data was collected by focus group interviews and an in-depth interview from July to November, 2005. Content analysis was conducted. Result: The four major themes that emerged were control of accessibility, safety concern, lack of knowledge and information, and ambivalence. Control of accessibility: Most of the participants stated a prescription is necessary although they might feel ashamed if they had to ask for an ECP. Safety concerns: Participants thought that taking an ECP would be harmful to their body. Lack of knowledge and information: Most of the participants felt that they did not know enough about ECPs and were willing to learn more. Ambivalence: The participants stated that they would take an ECP when needed and would recommend it to a friend although they were concerned about the risks of ECPs. Conclusion: The results from this study suggest the need for an educational and awareness program to empower women to be able to make informed decisions on ECPs when needed.

클라우드 환경에서 대규모 콘텐츠를 위한 효율적인 자원처리 기법 (ECPS: Efficient Cloud Processing Scheme for Massive Contents)

  • 주요 IT 벤더들은 클라우드 컴퓨팅 기술을 이용하여 설치과정 생략, 운용비용 절감, 서비스품질 등에 중점을 두어 대규모 콘텐츠 서비스를 제공하고 있다. 반면에, 대규모 콘텐츠 데이터의 가공, 분석을 수행하는 데이터 처리 프로세스는 처리 시간의 단축을 위한 방법론이 요구되고 있다. 이에 본 논문에서는 클라우드 환경에서 대규모 콘텐츠를 위한 효율적인 자원처리 기법(Efficient_Cloud_Processing_Scheme : ECPS)을 제안한다. 제안한 기법은 리소스 확장 방안을 CPU 및 스토리지 등의 인프라스트럭처 단계에서 설계한다. 대규모 콘텐츠에 대한 자원 할당 방안을 Hadoop 플랫폼 기반의 MapReduce 프로그래밍 기법과 데이터마이닝 분야에서 숨겨진 패턴을 탐지하는데 사용되는 연관규칙을 이용하여 제시한다. 기존 설정값으로 자원을 할당하여 비교하여 ECPS기법을 적용한 결과, 제안 기법이 20% 이상의 성능 및 속도가 향상되었음을 확인하였다.

조직 괴사 활성을 지닌 Aeromonas hydrophila 의 분비 단백질에 관한 연구 (A Potent Tissue Destructive Activity of Secreted Proteins of Aeromonas hydrophila)

  • Aeromonas hydrophila는 수생계에 흔히 존재하는 병원체로서 출혈성 패혈증, 수종, 궤양 등의 질병을 유발한다. A. hydrophila는 효과적인 감염과 다양한 환경에서 적응하기 위해 세포 외 물질(Extracellular products, ECPs)을 분비한다. 본 연구에서 사용한 A. hydrophila CK257은 매우 독성이 강한 균주로 균이 분비하는 ECP 역시 쥐, 토끼, 금붕어를 포함한 동물 모델에게 조직 손상을 일으키며 독성을 보였다. 이러한 괴사 반응을 일으키는 원인 요소를 찾기 위해 ECP의 특성을 분석하였고 그 결과, 원인 요소가 단백질이라는 것을 확인하여 이후 단백질 분리 및 정제에 초점을 맞추어 실험을 진행하였다. 균체를 제거한 배양액 상의 분비 단백질을 얻기 위하여 ammonium sulfate 침전법을 사용하였고, 이후 겔 거르기 크로마토그래피(Gel filtration chromatography)를 이용하여 단백질을 정제하였다. 정제 후 단백질은 다섯 개의 분획으로 나뉘었고 이 중 한 개의 분획이 동물 모델에 괴사와 같은 피부 손상을 일으키는 것을 확인하였다. 괴사 활성을 가진 분획의 단백질 4개의 밴드를 MALDI-TOF 시스템을 통해 분석한 결과, Peptidase M35와 Peptidase M28로 밝혀졌으며 이들은 각각 금속촉매효소(metalloprotease)임을 확인할 수 있었다. Peptidase M35와 Peptidase M28의 단백질 분해 효소로써의 기능을 확인하기 위해 카제인 분해 활성을 확인하였고, 다양한 금속 이온을 처리함에 따라 그 활성이 달라지는 것을 관찰할 수 있었다. 또한 조직에 존재하는 단백질의 한 종류인 엘라스틴(elastin) 분해능 역시 확인해 보았다. 본 연구에서는 A. hydrophila가 분비하는 물질 가운데 조직 괴사 활성을 일으키는 것으로 추정되는 Peptidase M35와 Peptidase M28의 두 가지 인자를 확인하여 동정하였다. 이후 두 가지 인자를 결손 시킨 돌연변이주를 구축하여 조직 괴사에 이들이 직접적으로 작용하는지 확인 할 예정이다.

Vibrio alginolyticus MviN is a LuxO-regulated Protein and Affects Cytotoxicity Towards Epithelioma Papulosum Cyprini (EPC) Cells

  • Vibrio alginolyticus, a Gram-negative marine bacterium, is one of the causative agents of fish vibriosis. Its virulence factors and pathogenesis mechanism are barely known, except for some extracellular products (ECPs) that are known to be regulated by quorum sensing system. Therefore, the present study used a microarray to analyze the transcription profiles of the wild-type V. alginolyticus and a deletion mutant of luxO, the pivotal regulator in Vibrio quorum sensing systems, which resulted in the identification of a putative virulence factor, MviN. Quantitative real-time reverse transcription PCR confirmed that the transcription of mviN was upregulated in the luxO mutant when compared with wild-type, and down regulated in a luxO-con complemented strain. Furthermore, Western blotting indicated that MviN was greatly induced during the late-exponential and stationary phases of growth, indicating that the expression of MviN was cell-density dependent and quorum sensing regulated in V. alginolyticus. Meanwhile, the mviN null mutant displayed a much slower growth rate than the wild type, signifying the essential role of MviN in V. alginolyticus. Western blotting also revealed that MviN was present as an extracellular protein in V. alginolyticus. When epithelioma papulosum cyprini (EPC) cells were treated with the ECPs of the mviN mutant, no cytotoxicity was observed, whereas EPC cells treated with the wild type exhibited pathological changes, which increased with the ECPs concentration and treatment time. Therefore, the results demonstrated that MviN is a LuxO-regulated ECPs component and involved in the pathogenicity of V. alginolyticus.

Characterization, Antioxidant Capacity and Protective Effect of Peptides from Cordyceps militaris Cultivated with Tussah Pupa on Oxidative Injured HepG2 Cells

  • The antioxidant capacity and protective effect of peptides from protein hydrolysate of Cordyceps militaris cultivated with tussah pupa (ECPs) on H2O2-injured HepG2 cells were studied. Results indicated ECP1 (<3 kDa) presented the strongest antioxidant activity compared with other molecular weight peptides. Pretreated with ECPs observably enhanced survival rates and reduced apoptosis rates of HepG2 cells. ECPs treatment decreased the ROS level, MDA content and increased CAT and GSH-Px activities of HepG2 cells. Besides, the morphologies of natural peptides from C. militaris cultivated with tussah pupa (NCP1) and ECP1 were observed by scanning electron microscopy (SEM). Characterization results suggested the structure of NCP1 was changed by enzymatic hydrolysis treatment. Most of hydrophobic and acidic amino acids contents (ACC) in ECP1 were also observably improved by enzymatic hydrolysis. In conclusion, low molecular weight peptides had potential value in the development of cosmetics and health food.

  • The electrical conductive polymers (ECPs) reported at my research group are introduced in this review, which works are started from the late Professor Su-Moon Park's pioneering research for polyaniline at the University of New Mexico. The electrochemical and spectroelectrochemical properties and their applications to sensor and energy conversion systems are briefly described. At first, the growth and degradation mechanism of polyaniline describes and we extend to polypyrrole, polyazulene, polydiaminonaphthalenes, and polyterthiophene derivatives. In addition, the preparation of monomer precursors having functional groups is briefly described that can give us many exceptional applications for several chemical reactions. We describe the application of these ECPs for the fabrication of chemical sensors, biosensors, biofuel cells, and solar cells.
