Stereoselective Synthesis of L-Deoxyaltronojirimycin from L-Serine

  • (2S,3R)-3-Hydroxy-2-(hydroxymethyl)-3,6-dihydro-2H-pyridine 8, an important precursor for the synthesis of polyhydroxylated piperidine azasugars, has been prepared from L-serine. Highly stereoselective nucleophilic addition to amino aldehyde 5 gave the corresponding allylic alcohol 6 which proceeded to give dihydro-2H-piridine 7a via a Grubbs II catalyzed RCM. Stereoselective H-bond directed epoxidation of allylic alcohol led to the oxiranyl alcohol 9 which was easily converted to L-deoxyaltronojirimycin by regioselective ring opening.

Effects of Exposure to Estradiol Benzoate or Flutamide at the Weaning Age on Expression of Connexins in the Caudal Epididymis of Adult Rat

  • The present research was chiefly designed to determine the effect of the treatment of estrogenic agonist, estradiol benzoate (EB), or antiandrogenic compound, flutamide (Flu), at the weaning age on the expression of connexin (Cx) isoforms in the caudal epididymis of adult male rat. Animals were subcutaneously administrated with a single shot of either EB at a low-dose ($0.015{\mu}g$ of EB/kg body weight (BW)) or a high-dose ($1.5{\mu}g$ of EB/kg BW) or Flu at a low-dose ($500{\mu}g$ of EB/kg BW) or a high-dose (5 mg of EB/kg BW). Expressional changes of Cx isoforms in the adult caudal epididymis were examined by quantitative real-time PCR analysis. The treatment of a low-dose EB caused significant increases of Cx30.3, Cx31, Cx32, and Cx43 transcript levels but reduction of Cx31.1, Cx37, and Cx45 expression. Exposure to a high-dose EB resulted in very close responses observed in a low-dose EB treatment, except no significant expressional change of Cx37 and a significant induction of Cx40. Expression of all Cx isoforms, except Cx45, was significantly increased by a low-dose Flu treatment. Expressional increases of all Cx isoforms were detected by a high-dose Flu treatment. The current study demonstrates that a single exposure to estrogenic or antiandrogenic compound during the early postnatal developmental period is sufficient to disrupt normal expression of Cx isoforms in the adult caudal epididymis.

ebXML 환경에서의 지식기반 검색 시스템 (Knowledge Based Search System In the ebXML Environment)

  • As B2B (Business to business) develops swiftly, at home as well as in other advanced countries, plans for activating Electronic business are made and proceeded in a national viewpoint. However, it is essential task for the construction, advancement and activation of B2B framework to make an efficient search for differently built -up data from B2C and thus to look for optimal business partner suitable for his/her own business. For this, in the last Aug. of 2001, government has also referred to ebXML, the exchange model for electronic business data based on XML, as a suggestion for B2B framework. The purpose of this study is to develop search system for efficient choice of business partner and this will play an important role for data processing and competitiveness strengthening of small and medium enterprises. Meanwhile, this system is built up by using systemic characteristics registered in ebXML Registry/Repository and ‘question-expanding’ searching ways based on the particulars of business profiles for both objectiveness and maximum efficiency of search result.

  • ebXML (Electronic Business using extensible Markup language), sponsored by UN/CEFACT and OASIS, is a modular suite of specifications that enables enterprises of any size and in any geographical location to conduct business over the Internet. Using ebXML, companies now have a standard method to exchange business messages, conduct trading relationships, communicate data In common terms and define and resistor business processes. It is the mission of ebXML that provides an open XML-based infrastructure enabling the global use of electronic business information in an interoperable, secure and consistent manner by all parties for creating a single e1ectronic electronic market. This paper briefly overviews the concept of core component, context, assembly rule and context rule. Then, It designs by standard specifications of ebXML core component commonly used in an industry and among industries, and assembles business messages by using XML schema. Therefore, it suggests the mechanism which effectively exchanges business messages among the trading partners. This paper designs core component by using only three business messages of retail industry : orders, dispatch report, sales report.

Expressional Changes of Connexin Isoform Genes in the Rat Caput Epididymis Exposed to Flutamide or Estradiol Benzoate at the Early Postnatal Age

  • Direct communication between neighboring cells through connexin (Cx)-based gap junction is a crucial biological manner to regulate functions of a tissue consisting of multi-cell types. The present research evaluated expressional changes of Cx isoforms in the caput epididymis of adult rat exposed to estradiol benzoate (EB) or flutamide (Flu) at the early postnatal age. A single subcutaneous administration of EB at a low-dose [$0.015{\mu}g/kg$ body weight (BW)] or a high-dose ($1.5{\mu}g/kg\;BW$) or Flu at a low-dose ($500{\mu}g/kg\;BW$) or a high-dose (5 mg/kg BW) was performed to an animal at 1 week of age. Quantitative real-time PCR analysis was employed to determine expressional changes of Cx isoforms. The transcript levels of Cxs30.3 and 37 were decreased by a low-dose EB treatment, while decreases of Cxs31, 31.1, 32, 40, and 45 transcript levels were observed with a low-dose EB treatment. The treatment of a high-dose EB resulted in expressional reduction of Cxs30.3, 31, 31.1, 37, 40, 43, and 45. The Flu treatment at a low dose caused increases of Cxs26, 37, and 40 transcript levels but decreases of Cxs31.1, 43, and 45 transcript levels. Increases of Cxs30.3, 31, 37, and 40 mRNA amounts were induced by a high-dose Flu treatment. However, exposure to a high-dose Flu produced expressional decreases of Cxs31.1, 32, and 43 in the adult caput epididymis. These observations suggest that exposure to EB or Flu at the neonatal period could lead to aberrant expression of Cx isoforms in the adult caput epididymis.

EB-PVD법과 전기화학방법으로 형성된 Zn-Cr film의 구조와 부식특성 (Corrosion Behavior and Crystal Structure of Zn-Cr Coatings by EB-PVD and Electroplating)

  • It has been investigated corrosion resistance and crystal structure of Zn-Cr coatings fabricated by electroplating method and electron-beam physical vapor deposition method(EB-P VD). The electroplated Zn-Cr alloy consists mainly of η'-Zn phase for the lower Cr content than 7.9 wt% Cr and ${\gamma}$'-ZnCr phase for the higher Cr content. In the Zn-Cr alloy fabricated by EB-PVD the ${\gamma}$'-ZnCr phase appeared clearly at 3 wt% Cr and it became the sole phase at 50 wt%Cr. The amount of η'-Zn phase decreased obviously with increasing Cr content when it exceeded 15 wt% Cr. The electrochemical measurement of the electroplated Zn-Cr film has shown corrosion potential of about -1000 mV. The current density of active region and the amount of dissolved Zn and Cr decreased significantly with increasing Cr content. The electrochemical characteristics of Zn-Cr alloy fabricated by EB-PVD have shown that the alloy of 50 wt% Cr had the highest corrosion potential(-500 mV) and the lowest critical passive current density than that of the electroplated.

EB기법을 이용한 사고잦은 곳 개선사업 우선순위 판정기법 개발 (Development of Evaluation Model for Black Spot Improvement Priorities by using Emperical Bayes Method)

  • 도로 네트워크의 안전 관리는 기본적으로 교통안전 조사를 위한 사이트(교통사고 잦은 지점) 선정, 안전문제에 대한 진단, 잠재적 위험요소들에 대한 가능한 대안 선정, 주어진 예산 제약 하에 대안간의 우선순위 결정과 같은 순서로 이루어진다(Persaud, 2001). 효율적인 안전 조사를 위해 요구되는 지점(교통사고 잦은 지점) 선정에 대한 과정은 매우 중요하다. 이에, 본 연구는 인천시 4지 신호 교차로 중 교통사고 잦은 지점으로 선정된 지점의 3년간(2004~2006년) 사고 자료와 기하구조 자료를 이용하여 EB 기법 이용 시 필요한 사고예측모델을 개발하였다. 또한, 교통사고 잦은 지점을 선정하는데 있어 현재 우리나라에서 적용되고 있는 단순사고건수와 심각도를 고려하여 선정된 우선순위와 단순사고건수 대신 EB 기법을 이용하여 예측된 사고건수를 이용하여 선정된 우선순위 비교를 통해 기존 방법의 한계를 제시하고 합리적인 예측모형 개발 필요성을 보여주고자 하였다. 분석 결과, 총 사고건수 추정 시 기존의 사고 예측 방법인 비선형 회귀모형과 EB 기법의 결과 값 모두 예측력이 높은 것으로 나타났지만 지점별 사고건수 예측력을 함께 고려할 경우엔 EB 기법이 비선형 회귀 모형(포아송)의 결과보다 예측력이 좋은 것으로 나타났다. 또한, 도출된 우선순위 비교 결과 대부분의 지점의 우선순위는 크게 변동이 없었으나, 서해4거리 등 몇 개 지점의 개선우선순위에는 상당한 변동이 발생하는 것으로 나타났다. 이는 실제 사고건수를 이용할 경우 발생하는 RTM문제를 본 연구에서 제안한 기법을 사용할 경우 해결가능하다는 것을 시사한다.

투과증발에 의한 Ester 성분의 소수성막의 투과플럭스 (Permeation Flux of Ester Compounds through Hydrophobic Membrane by Pervaporation)

  • 에스테르(ester)의 모델 수용액으로부터 에스테르 성분을 회수하기 위한 투과증발 공정에서, 공급액의 농도와 온도 변화에 따른 에스테르와 물의 플럭스를 측정하였다. 공급액의 에스테르 농도가 0.15 wt%에서 0.60 wt%로 증가함에 따라 에틸 아세테이트(EA), 프로필 아세테이트(PA), 에틸 프로피오네이트(EP), 부틸 아세테이트(BA), 그리고 에틸 부티레이트(EB)의 플럭스는 증가하였으며, 공급액의 온도가 $30^{\circ}C$에서 $50^{\circ}C$로 증가함에 따라 플럭스가 증가하였다. 에스테르 플럭스의 크기는 (EA) < (PA, EP) < (BA, EB) 순서이었으며, 이는 에스테르와 막 표면과의 친화도에 크게 의존하는 것을 보여준다. 즉, EA는 분자쇄 내에 소수성 기($-CH_2-$)를 1개, (PA, EP)는 2개를 포함하고 있으나, (BA, EB)는 3개를 가지고 있어 가장 소수성이 크기 때문이다. 이러한 에스테르 분자의 소수성이 에스테르 플럭스에 미치는 영향뿐 아니라 막 표면의 소수성이 에스테르 플럭스에 미치는 영향에 관한 연구가 더 필요할 것이다. 온도가 증가함에 따라, EA, PA, EP, BA, and EB 수용액의 물 플럭스는 증가하였으나, 농도 변화에 따른 물 플럭스는 거의 변화가 없었다. 본 투과증발에 대한 실험결과는 기존의 열을 이용하는 증발공정과 증류공정을 대체할 수 있는 공정으로서 향 성분의 회수공정을 개선하는 기초 자료로 활용할 수 있을 것이다.

Genetic parameter estimation for milk β-hydroxybutyrate and acetone in early lactation and its association with fat to protein ratio and energy balance in Korean Holstein cattle

  • Objective: The objective of this study was to estimate the genetic parameters for milk ${\beta}$-hydroxybutyrate (BHBA), acetone (Ac), fat protein ratio (FPR), and energy balance (EB) using milk test day records and investigate the effect of early lactation FPR and EB on milk ketone body concentrations. Methods: Total 262,940 test-day records collected from Korea Animal Improvement Association during the period of 2012 to 2016 were used in this study. BHBA and Ac concentrations in milk were measured by Fourier transform infrared spectroscopy (FTIR). FPR values were obtained using test day records of fat and protein percentage. EB was calculated using previously developed equation based on parity, lactation week, and milk composition data. Genetic parameters were estimated by restricted maximum likelihood procedure based on repeatability model using Wombat program. Results: Elevated milk BHBA and Ac concentrations were observed during the early lactation under the negative energy balance. Milk FPR tends to decrease with the decreasing ketone body concentrations. Heritability estimates for milk BHBA, Ac, EB, and FPR ranged from 0.09 to 0.14, 0.23 to 0.31, 0.19 to 0.52, and 0.16 to 0.42 respectively at parity 1, 2, 3, and 4. The overall heritability for BHBA, Ac, EB and FPR were 0.29, 0.32, 0.58, and 0.38 respectively. A common pattern was observed in heritability of EB and FPR along with parities. Conclusion: FPR and EB can be suggested as potential predictors for risk of hyperketonemia. The heritability estimates of milk BHBA, Ac, EB, and FPR indicate that the selective breeding may contribute to maintaining the milk ketone bodies at optimum level during early lactation.

대두단백질 분해균주 Bacillks subtilis EB464의 선발 및 분해 특성 (Isolation and Characteristics of Soy protein-degrading Strain, Bacillus subtilis EB464)

  • 동물사료로 주로 이용되고 있는 대두박의 이용성을 높히기 위한 방안으로서, 전통발효식품에서 대두단백질 분해능력이 우수한 균주를 분리하여 동정하였으며 Bacillus subtilis EB464로 명명하였다. 분리된 B. subtilis EB464를 대두박에 접종하여 고체배양할 경우, 단백질 분해효소의 최고활성은 배양 24시간 후에 300 PC/g에 달하였으며, 72시간 배양후의 수용성 단백질 함량은 20.6%,유리아미노산의 함량은 22.0%에 달하였다. B. subtilis EB464 균주가 분비하는 단백질분해효소의 최적 pH는 9.0,최적 온도는 $50^{\circ}C$E.확인되었다. 본 효소는 $40^{\circ}C$에서는 안정하였으나 $50^{\circ}C$ 이상에서는 효소활성이 급격히 실활되었으며, pH 6.0에서 10.0가지의 광범위한 pH에서 상당히 안정한 것으로 나타났다.
