감초 약침액의 항암 및 면역활성에 미치는 영향 (Antitumor and Immunomodulatory Effects of Glycyrrhizae Radix Aqua-acupuncture Solution)

  • Glycyrrhizae Radix aqua-acupuncture solution (GRAS) and Glycyrrhizae Radix water-extracted solution (GRWS) were prepared and tested for organ toxicities, antitumor activities, and immunomodulatory effects. The organ-toxicity of GRAS to male ICR mice was studied by the measurements of glutamic oxaloacetic transaminase (GOT), glutamic pyruvate transaminase (GPT), lactate dehydrogenase (LDH), and alkaline phosphatase (ALP-s) activities after injection of GRAS for 7 days. The activities of GOT, GPT, LDH, ALP-s were decreased with GRAS. It was shown to possess considerable toxicity toward various tumor cell lines. Concentration of GRAS at 1.5g/ml and 3g/ml resulted in more than 80% inhibition of growth in Ehrlich ascites tumor cells (EATC), Hepa1c1c7, and HeLa cells. Toxicity of GRAS to A549 revealed that 68% inhibition of growth. GRWS at the concentration of 3g/ml showed more than 80% inhibition of growth with EATC, Hepalclc7, A549 and HeLa. In morphological study, the number of cells were decreased, and the shape of cells was round-form in EATC, Hepalclc7, A549 and HeLa cells with GRAS. Administration of GRAS inhibited the growth of EATC in vivo. Mice given EATC at 1.5g/ml or 0.3g/ml GRAS had 16.7% to 50% survival after 21 days. GRAS increased the proliferation of T and B cells and the cytolytic activity of purified T cell. The biosyntheses of nucleic acid and protein of EATC, Hepalclc7, A549 and HeLa cells were inhibited by GRAS.

  • Gamdutang aqua-acupuncture solution (GAS), Gamdutang water-extracted solution (GWS) and Dae-Gamdutang aqua-acupuncture solution (DGAS) were prepared and tested for antitumor activities. It was shown to possess considerable toxicity toward various tumor cell lines. Concentration of 5 $\times$ and 10$\times$ of GAS resulted in more than 70% inhibition of growth in Ehrlich ascites tumor cells (EATC), Hepa1c1c7 and A549. GAS at concentrations of 5$\times$ and 10$\times$ revealed more than 60% inhibition in HeLa. GWS showed more than 50% inhibition of growth with EATC and HeLa at concentrations of 5$\times$ and 10$\times$, respectively. Toxicity assay with GWS in Hepa1c1c7 and A549 revealed that more than 80% inhibition of growth at the concentration of 5$\times$ and 10$\times$. In morphological study, the number of cells were decreased, and the shape of cells was round-form in EATC, Hepa1c1c7, A549, HeLa with GAS.

감두탕(甘豆湯) 약침액(藥鍼液)이 종양세포(腫瘍細胞)에 미치는 영향(影響) (Effects of Gamdutang Aqua-acupuncture Solution on Tumor Cell Lines in vitro)

It was shown to possess considerable toxicity toward various tumor cell lines. Concentration of $5{\times}\;and\;10{\times}$ of GAS resulted in more than 70% inhibition of growth in Ehrlich ascites tumor cells(EATC), hepa1c1c7 and A549. GAS at the concentration of $10{\times}\;and\;5{\times}$ revealed that more than 60% inhibition in HeLa. GWS showed more than 50% in hibition of growth with EATC, HeLa at the concentration of $5{\times}\;and\;10{\times}$. Toxicity assay with GWS in hepa1c1c7 and A549 revealed that more than 80% inhibition in growth at the concentration of $5{\times}\;and\;10{\times}$. In morphological study, the number of cells were decreased, and the shape of cells was round-form. Most of cells in detached in EATC, Hepa1c1c7, HeLa, and A549 with GAS. These results suggest that GAS has antitumor activity in vitro.

애엽(艾葉) 약침액(藥鍼液)에 의한 Phase II 효소 활성 유도 (Induction of Phase II Enzyme Activity by Artemisia asiatica Nakai Aqua-acupuncture Solution)

  • 애엽으로 약침액을 조제하여 암예방 효과를 살펴 본 결과 다음과 같은 결론을 얻었다. 애엽 약침액으로 10일간 전투여한 후 Ehrlich ascites tumor cell을 이식하여 복수암이 유발된 생쥐의 EATC 성장을 살펴 본 결과 약침액을 투여한 생쥐의 EATC의 성장이 크게 억제되었으며, 복수암이 유발된 생쥐의 비장 무게가 대조군에 비해 증가하였다. 또한 장기간의 관찰에 의해서도 복수암 세포의 성장 저해에 의해 생쥐의 체중변화율은 대조군에 비해 크게 억제되었으며 수명 연장 효과가 나타났다. 이와같은 생체에서의 암예방 효과를 in vitro 상에서 그 기전을 살펴본 결과 애엽 약침액은 phase II detoxification 효소인 QR의 생성을 유도하고 GSH 함량을 증가시켜서 암예방 효과를 나타내었다.

암세포의 종류에 따른 녹차 Catechin의 항암효과 (Antitumor Effects off Green Tea Catechin on Different Cancer Cells)

  • Antitumor effects of various teas have been studied for a long time. Among them, green tea is one of the most popular test and very close to our lives in Korea. However, precise effect and mechanism about antitumor effects of green tea were not estabilished. The present study investigated the antitumor effect of catechin, which is main component of green tea, which was produced in Korea, was used. In each group, morphological changes were observed induced severe cell damage and growth inhibition gradually until 24hours. Then catechin effect was found to be concentration-and time-dependent. EATC was injured abruptly at 100ug/ml catechin treatment for 6 hours and the effect lasted constantly until 24 hours. But in both of cell lines, cell damage and inhibition of proliferation did not show up apparently at concentration of 10 and 1ug/ml. In contrast, catechin led to little or no effect against HepG2 in all of concentrations and periods. These results suggest that catechin extracted from green tea had different effect on cancer cells as cell type, concentration and period. Therefore green tea would be helpful to cancer treatment as well as cancer prevention and this study would be the basic source for further research of green tea.

  • 현재 우리나라에서 널리 이용되고 있는 미생물학적 검사법을 이용하여 TCs 계열의 모화합물과 epimer에 대한 검출 그리고 이를 통한 모화합물과 epimer간의 상대적 생물학적 활성도를 확인하였다. 시험법의 관점에서 낮은 선택성과 낮은 민감도를 나타낸 균주는, 다양한 적용 가능 균주의 발굴 및 적용이 시험법 검증을 통해 이루어질 필요가 있다. 극단적으로 식품 중 epimer만이 잔류하여 미생물 시험에서 의양성이 나올 경우, 기기분석을 통해 확인 정량시 원물질이 검출되지 않는다면, 이는 분석화학과 독성학의 과학적 지식을 바탕으로 볼 때 제도적 맹점으로도 남을 수도 있다. 그리고, 현행 우리나라의 모화합물에 국한 된 잔류허용 기준은 활성이 있는 epimer나 대사산물까지 확대하여 검토 해 볼 필요가 있으며, 이는 최근 의약품류의 환경 중 잔류실태 조사, 환경 중 위해성 평가 및 관리를 위해서도 관리 대상물질을 과학적 근거에 따라 제시해 주는 중요한 자료가 된다.

당귀 약침액의 암예방 효과 (Chemopreventive Potential of Angelicae gigantis Radix Aqua-acupuncture Solution)

  • Angelicae gigantis Radix aqua-acupuncture solution (AGRAS) and Angelicae gigantis Radix water-extracted solution (AGRWS) were prepared and tested for their organ toxicities and chemopreventive potentials. The organ-toxicity of AGRAS to male ICR mice was studied by the measurements of glutamic oxaloacetic transaminase (GOT), glutamic pyruvate transaminase (GPT), lactate dehydrogenase (LDH) and alkaline phosphatase (ALP-s) activities after injection of AGRAS for 7 days. The activities of GOT GPT and LDH were decreased, but the activity of ALP-s was not changed with AGRAS. When AGRAS was administered once daily for 10 days before the tumor implantation, AGRAS exerted antitumor activity by inhibiting the growth of Ehrich ascites tumor cells (EATC) in viva. The inductions of quinone reductase (QR), glutathione (GSH) and glutathione S-transferase (GST) and inhibition of polyamine metabolism were tested for the chemopreventive potentials of AGRAS and AGRWS. AGRAS was potent inducer of QR activity in murine hepatoma Hepalclc7 cells. In cultured rat Ac2F cells, AGRAS was also significantly induced QR activity GSH levels were increased about 1.3 fold with AGRAS. In addition the activity of GST was increased about 2.5 fold with AGRAS at the concentration of $0.1{\;}{\times}{\;}$. The effects of AGRAS and AGRWS were tested on the growth of Acanthamoeba castellanii. Proliferation of Acanthamoeba castellanii in a broth medium was inhibited by AGRAS and AGRWS at the concentration of $1{\;}{\times}{\;}and{\;}5{\;}{\times}{\;}$, respectively: These results suggest that AGRAS has chemopreventive potential by inducing QR activity increasing GSH and GST levels and inhibition of polyamine metabolism.
