Effect of Cnidium officinale Mixture on Recovery Capability of DNCB-induced Allergic Contact Dermatitis in Rat

  • This study is aimed to investigate the skin thickness, plasma components, serum IgE level and antioxidant enzymes activities by mixtures of Cnidium officinale, Saururus chinensis, Houttuynia cordata and Glycyrrhiza uralensis on the allergic contact dermatitis of rat induced by 1-chloro-2,4-dinitrochlorobenzene (DNCB). Mixtures reduced the thickness of the skin and removed the dead skin cells compared to the skin of rats treated with DNCB alone. Also, these mixtures down-regulated the contents of lipid and IgE, and reduced the activities of superoxide dismutase and catalase. These results indicate that the mixtures significantly recovered the contact dermatitis induced by DNCB. In conclusion, it is thought that the mixtures could be useful for the allergic contact dermatitis.

  • Allergy is conditions when a hypersensitivity reaction happens with a certain element, called as an allergen, which is commonly not reactive to ordinary individuals. Allergic diseases involve various organs or systems in the body. The purpose of allergy tests is to make a diagnosis of allergic diseases and to identify the affecting allergens. In vivo tests, more relevant in clinical situation, include skin test, patch test and provocation test. In in vitro tests, there are specific IgE test, histamine releasing assay, and lymphocyte activation test, safer and more objective than in vivo tests. In the view point of clinical practice, skin test, provocation test, total IgE test and specific IgE test were reviewed in depth.

천연 소재 복합물이 항아토피 피부염 및 피부재생에 미치는 영향 (Effects of Natural Herb Mixture on Anti-atopic Dermatitis and Skin Regeneration)

  • Objectives : This study aims to evaluate the effects of natural herb mixutre (NHM) on atopic dermatitis and skin regeneration using in vivo test. Methods : NHM was prepared with DW. 25% of NHM was applied to skin lesion, where atopic dermatitis was induced by DNCB in NC/Nga mice. The levels of cytokines (IL-4, IL-5, IL-6, IL-13, TNF-a, and $IFN-{\gamma}$), and IgE in serum were measured by Luminex. Immune cells (WBC, eosinophil, lymphocyte, and monocyte) in blood were counted by coulter counter. The gross investigation of atopic dermatitis index score test were performed during the NHM treatment period. Also, the histopathological change of dorsal skin was observed by H&E and M&T staining. Results : NHM showed the levels of IL-4, IL-5, IL-6, IL-13, IgE, WBC, eosinophil, lymphocyte, and monocyte in serum or blood were significantly decreased. On the contrary, the productions of FGF, and VEGF were increased in the serum. Also, atopic dermatitis index score in NHM-treated mice were observed in the similar levels to those of normal group. Histological examination demonstrated that NHM suppressed immune cell infiltration and thickening of epidermis, meanwhile the extraction induced collagen production in the dorsal skin. Conclusion : This study demonstrated that NHM is appeared to be effective on atopic dermatitis and skin regeneration efficacy based on the observations with hematologic, gross, and histologic examinations. Therefore, we suggest that NHM could be effectively used as an external therapeutics against atopic dermatitis and a consequence skin damage.

Inhibitory Effects of Camellia sinensis Extract on the Development of Atopic Dermatitis-like Lesions in NC/Nga Mice

  • Atopic dermatitis (AD) syndrome is one of the most common and severe skin diseases in Korea; a large population has this disease. We examined the effects of the extract from the leaf and sprig of Camellia sinensis on the development of AD by using NC mice as a model of atopic dermatitis. Oral administration of the extract to NC/Nga mice treated with 2,4?dinitrochlorobenzene (DNCB) inhibited the development of AD-like skin lesions as shown by a significant decrease in the skin symptoms of the disease and a decrease in ear thickness and levels of immunoglobulin E (IgE) and thymus-and activation-regulated chemokine (TARC) level in the skin. Administration of the extract markedly suppressed the DNCB-induced mRNA expression of interleukin 4 (IL-4) and tumor necrosis factor ${\alpha}$ (TNF-${\alpha}$). The findings suggest that transdermal application of the extract may modulate in the skin of NC/Nga mice. The extract was effective for the prevention and treatment of AD.

Alternatives to In Vivo Draize Rabbit Eye and Skin Irritation Tests with a Focus on 3D Reconstructed Human Cornea-Like Epithelium and Epidermis Models

  • Human eyes and skin are frequently exposed to chemicals accidentally or on purpose due to their external location. Therefore, chemicals are required to undergo the evaluation of the ocular and dermal irritancy for their safe handling and use before release into the market. Draize rabbit eye and skin irritation test developed in 1944, has been a gold standard test which was enlisted as OECD TG 404 and OECD TG 405 but it has been criticized with respect to animal welfare due to invasive and cruel procedure. To replace it, diverse alternatives have been developed: (i) For Draize eye irritation test, organotypic assay, in vitro cytotoxicity-based method, in chemico tests, in silico prediction model, and 3D reconstructed human cornealike epithelium (RhCE); (ii) For Draize skin irritation test, in vitro cytotoxicity-based cell model, and 3D reconstructed human epidermis models (RhE). Of these, RhCE and RhE models are getting spotlight as a promising alternative with a wide applicability domain covering cosmetics and personal care products. In this review, we overviewed the current alternatives to Draize test with a focus on 3D human epithelium models to provide an insight into advancing and widening their utility.

Mitochondria-Targeted Vitamin E Protects Skin from UVB-Irradiation

  • Mitochondria-targeted vitamin E (MVE) is designed to accumulate within mitochondria and is applied to decrease mitochondrial oxidative damage. However, the protective effects of MVE in skin cells have not been identified. We investigated the protective effect of MVE against UVB in dermal fibroblasts and immortalized human keratinocyte cell line (HaCaT). In addition, we studied the wound-healing effect of MVE in animal models. We found that MVE increased the proliferation and survival of fibroblasts at low concentration (i.e., nM ranges). In addition, MVE increased collagen production and downregulated matrix metalloproteinase1. MVE also increased the proliferation and survival of HaCaT cells. UVB increased reactive oxygen species (ROS) production in fibroblasts and HaCaT cells, while MVE decreased ROS production at low concentration. In an animal experiment, MVE accelerated wound healing from laser-induced skin damage. These results collectively suggest that low dose MVE protects skin from UVB irradiation. Therefore, MVE can be developed as a cosmetic raw material.

프랑킨센스 오일의 피부노화 억제 효과 (I) - 육안적 관찰을 중심으로 - (Inhibition Effects of Frankincense Oil on Skin Aging (I) : Focussed on Gross Examination)

  • The purpose of this study was to investigate the effects of frankincense oil in a skin aging animal model. Skin aging was induced by both the irradiation of UVB and the application of squalene monohydroperoxide (Sq-OOH) to the back of experimental animals for 4 weeks. And at the same time experimental materials were applied topically. Six to seven weeks female SHR-1 hairless mice were divided into five groups including normal (N: saline), control (C: UVB+Sq-OOH+saline), vehicle control (VC: UVB+Sq-OOH+jojoba oil), positive control (PC: UVB+Sq-OOH+0.01% retinoic acid) and experimental (E: UVB+Sq-OOH+3% Frankincense oil) groups, five animals each group. The skin erythema index for the PC and E groups were lower than that of the C group. Whereas, both the lipid and water capacities for the PC and E groups were higher than those of the C group. Wrinkles for the C group were formed as a pattern of deep furrows and thick crests. Whereas, wrinkles for the PC and E groups were formed as a pattern of shallow furrows and thin crests which were similar to that of the N group. As for the both absolute and relative weight of the spleen, the PC group were significantly higher than the other groups. In conclusion, frankincense oil can be used practically for the prevention or improvement of skin aging in terms of health promotion and beauty for the people.

연교산(連蘿散)이 아토피 동물 모델에 미치는 영향 (Effect of Yeonkyo-san in Atopic Dermatitis Animal Model)

  • The purpose of this study is to examine closely effect that YKS used to atopic dermatitis disease patient get in atopy eruption control experimentally. We analyzed the expression of IgE, IL-6, IL-4, IL-5 and IL-13's level in serum, and IFN-${\gamma}'$ production by YKS extract. We also analyzed YKS extract get to NC/Nga mice's skin establishment analyzes neck-back skin after biopsy, and H&E method measured about epidermis and dermis part in comparison with control group. In this research YKS extract as treatment result to a NC/Nga mice, IgE and IL-6 content in serum decreased remarkably than control group. And decreased than result control group which measure IL-4, IL-5, IL-13's level in serum, and IFN-${\gamma}'$ production secreted in Th1 cell displayed increase by YKS extract. IL-4 and IFN-${\gamma}'s$ gene revelation amount displayed marked decrease than control group in result that observe effect that get in skin of a NC/Nga mice. Also, In culture supernatant which cultivate for 14 day after separate skin cell, IL-13 and IL-6 production decreased than control group. YKS extract get to NC/Nga mice's skin establishment analyzes neck-back skin after biopsy, and dye by H&E method decreased about epidermis and inflammation of dermis part remarkably than control group. These results suggest that Th1 cell and Th2 cell observe to be shifted by secretion amount of IL-4 and IFN-$\gamma$ by YKS extract could know that YKS extract can use usefully in allergy autoimmune disease.

Chamomile German 오일도포가 아토피성 피부염을 가진 NC/Nga 생쥐모델의 혈청 IgE와 IgG1양 변동에 미치는 영향 (Effect of Chamomile German Oil Application of IgG1 and IgE 1 to Atopic Dermatitis in NC/Nga Mice Model)

  • Atopic dermatitis is one of the most common skin disease in children and a characteristic chronically recurrent form of dermatitis with a hereditary tendency, affecting infants and may extend to the childhood or th the adult age. Environmental factors, stress, and foodstuffs such as milk, egg may cause atopic dermatitis. Nc/Nga mice were used as an animal model for human atopic dermatitis. The divided by 4group such as normal group, BMAC group, FK506 group, MR group for this study raised in conventional conditions. To investigate effect of Chamomile German on atopic dermatitis in NC/Nga mice, the serum IgE and IgG1 level were measured while the severity degree of the skin lesion was examined by the naked eyes of two volunteers who were unaware of the treatment status. The results were the followings. 1. The score on the severity degree of skin dermatitis in FK 506 and MR group was lower than that in control group. 2. The serum IgE level in control group was higher 25% than that in normal group. 3. The serum level of IgE in FK506 and MR group compares to control group was decreased. 4. The serum IgG1 level was decreased more than 3.5 times in FK506 compared to control group while MR group had significantly less the serum IgG1 than control group. From the above results, treatment of Chamomile German oil had the effect on atopic dermatitis in NC/Nga mice. If scientific researches on aroma oil are performed in various way, aroma oil will be used to cure skin dermatitis as a alternative therapy in the future.

The Importance of Food Open Challenge Test in Atopic Dermatitis: The Comparison of Allergy History, Skin-Prick Test, and Specific IgE Detection

  • Food allergies play an important role in Atopic Dermatitis (AD). Dietary manipulation is essential in the management of AD. However, there has been a paucity of data reporting the prevalence of food allergies in AD patients in Korea. In this study, the Food Open Challenge Tests (FOCT) were conducted to investigate food allergies in AD patients. The skin-prick test and the detection of specific IgE, as well as allergy history of patient were used to detect food allergies in all AD patients. Elimination diet was conducted for two weeks prior to FOCTs. The prevalence of food allergies by FOCT is as follows: milk (67.3%); chicken (64.2%); pork (62.8%); eggs (61.0%); beef(55.4%) ; wheat (52.0%) and soybean (45.2%). Allergenic food items in Korean AD patients were different from people in other foreign countries. The seven major foods those tested positively by FOCTs were completely eliminated from the replaced diets for two weeks, and were subsequently reintroduced one at a time. Results from FOCTs were not comparable with allergy history or skin-prick tests or specific IgE detection. The sensitivity and specificity of skin-prick tests and specific IgE detection were lower than FOCTs. Allergy history, skin-prick tests, and specific IgE detection are useful for the identification of food allergen but its clinical significance differed according to food items. Therefore, we conclude that even though a 10-day delay was necessary after food challenge, FOCT is a useful and valid method to confirm food allergies and may be essential for the effective control of food allergies for treatment of AD.
