벼 오갈병 바이러스의 순화와 항혈청 제조에 관한 연구 (Studies on Purification and Serology of Rice Dwarf Virus)

  • 우리나라 중부이남 지역에서 벼 오갈병에 의한 피가 현저히 늘어나고 있는 실정이다. 본시험에서는 매개충을 사용, 순수분리하고 이를 접종하여 증식한 후 Toyoda 등의 순화방법을 개선하여 순화하였다. 그 결과 순화된 바이러스 함량은 ml당 3.12mg 이었다. 순화된 바이러스를 Adjuvant와 함께 토끼에 10-14일 간격으로 5회 주사하여 항혈청을 제조한 결과 1/4,096의 높은 역가를 나타내는 항혈청이 제조되었다.

On the Use of the Number Count of Blue Horizontal-Branch Stars to Infer the Dominant Building Blocks of the Milky Way Halo

  • The formation of the Milky Way stellar halo is thought to be the result of merging and accretion of building blocks such as dwarf galaxies and massive globular clusters. Recently, Deason et al. (2015) suggested that the Milky Way outer halo formed mostly from big building blocks, such as dwarf spheroidal galaxies, based on the similar number ratio of blue straggler (BS) stars to blue horizontal-branch (BHB) stars. Here we demonstrate, however, that this result is seriously biased by not taking into detailed consideration on the formation mechanism of BHB stars from helium enhanced second-generation population. In particular, the high BS-to-BHB ratio observed in the outer halo fields is most likely due to a small number of BHB stars provided by GCs rather than to a large number of BS stars. This is supported by our dynamical evolution model of GCs which shows preferential removal of first generation stars in GCs. Moreover, there are sufficient number of outer halo GCs which show very high BS-to-BHB ratio. Therefore, the BS-to-BHB number ratio is not a good indicator to use in arguing that more massive dwarf galaxies are the main building blocks of the Milky Way outer halo. Several lines of evidence still suggest that GCs can contribute a signicant fraction of the outer halo stars.

A Very Wide-Field Survey of Dwarf Galaxies in the M106 Group

  • We present a very wide-field survey of dwarf galaxies in the M106 Group using the Sloan Digital Sky Survey Data Release 8, covering an area of $10^{\circ}{\times}14^{\circ}$ around M106. We select 18 new members of the M106 group, 10 of which are new findings. Surface brightness profiles of most of these galaxies are fitted well by an exponential law. Twelve of these galaxies are early-types, and the rest are late-types. We produce a master catalog of the M106 Group galaxies by combining these new galaxies with 30 known galaxies. The faint-end of the luminosity function of these galaxies is fitted by a power law with an index ${\alpha}=-1.22{\pm}0.02$. This slope is much flatter than the value predicted by the ${\Lambda}CDM$ models, but is similar to the values for other galaxy groups. The spatial distribution of the dwarf galaxies in the M106 group is quite different from that of the bright members of the group, requiring a further study.

Reverse Transcription Recombinase Polymerase Amplification Assay for Rapid and Sensitive Detection of Barley Yellow Dwarf Virus in Oat

  • Barley yellow dwarf virus (BYDV) is an economically important plant pathogen that causes stunted growth, delayed heading, leaf yellowing, and purple leaf tip, thereby reducing the yields of cereal crops worldwide. In the present study, a reverse transcription recombinase polymerase amplification (RT-RPA) assay was developed for the detection of BYDV in oat leaf samples. The RT-RPA assay involved incubation at an isothermal temperature (42℃) and could be performed rapidly in 5 min. In addition, no cross-reactivity was observed to occur with other cereal-infecting viruses, and the method was 100 times more sensitive than conventional reverse transcription polymerase chain reaction. Furthermore, the assay was validated for the detection of BYDV in both field-collected oat leaves and viruliferous aphids. Thus, the RT-RPA assay developed in the present study represents a simple, rapid, sensitive, and reliable method for detecting BYDV in oats.

Comparison of an Immunochromatographic Assay Kit with DAS-ELISA for Large-Scale Diagnosis and Molecular Discrimination of Satsuma Dwarf Virus Collected from Citrus Orchards

  • Satsuma dwarf virus (SDV) seriously damages citrus production by reducing the quality and yield of fruit. To avoid contamination with SDV, mother trees are checked to be SDV-free in advance of nursery tree distribution. In this study, we compared an immunochromatographic assay (ICA) kit with double-antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA) for large-scale diagnosis of SDV in orchardgrown trees in Shizuoka Prefecture, Japan. The two methods gave conflicting results for 11 of 1,705 samples, all of which were negative by DAS-ELISA but positive by ICA. The samples scored as positive by either DAS-ELISA or ICA were analyzed by reverse transcription polymerase chain reaction and all were confirmed to be positive. These results validate the use of ICA as a screening method for large-scale diagnosis. Strain discrimination revealed that 16 of 22 isolates belonged to SDV, while citrus mosaic virus (CiMV) infection only and co-infection (SDV and CiMV) were in a minority.

PULSATIONAL CHARACTERISTICS OF V1719 CYGNI WITH PECULIAR LIGHT CURVE

  • The light curve and radial velocity curve of multiperiodic dwarf cepheid VI719 Cyg (HD200925) with peculiar light curve have been reanalyzed in order to identify the oscillation modes to confirm the helium settling within the envelope. To do these, through the period search for the photometric and radial velocity data from the literature, two different periods were determined and the oscillation modes corresponding to the first and second periods were identified as the fundamental and first radial overtones. Hence the helium settling within the envelope was confirmed from the period ratio. The color excess, metallicity, effective temperature, and surface gravity corresponding to two different modes were determined and it was found that these parameters almost do not depend upon different oscillation mode. By utilizing the surface brightness method, we investigated the variation of angular diameter and radial displacement and it was found that the angular variation is very peculiar. Also by referring to the stellar models, mass and age were determined as $2.7M_{\bigodot}$ and 0.42 Gyr respectively which make this variable star heavier and younger than other multimode dwarf cepheids. Preliminary spectroscopic CCD observations were carried out and it was found that Mg in VI719 Cygni is nearly solar abundent according to the analysis of $5172.68{\AA}MgI$ line which is inconsistent with the photometric result. It was suggested that VI719 Cyg may be classified as a $\rho$ Pup stars according to the photometric characteristics.

A pilot study on the radio flux variability of dwarf galaxies

  • The black hole occupation fraction in dwarf galaxies can provide an important clue for understanding the black hole seed formation. As a pilot feasibility study, we performed a KVN radio monitoring campaign over 8 months for 4 dwarf galaxies. Two galaxies (IC10 and NGC1569) are detected at 22 GHz, respectively with 39 mJy, 83 mJy. The measured flux (rms) variability is 13% and 8%, respectively for IC10 and NGC1569, while the mean flux uncertainty is 25% and 12%. Thus, the detection of the radio flux variability is at best marginal. Detecting flux variability of faint sources (i.e., 22 GHz flux < 200 mJy) seems challenging with the KVN single dishes. Combining with the 1.4 GHz flux measurements from the NVSS, we find that these two galaxies have a steep spectrum, supporting that the radio sources are AGNs. Instead of a monitoring, single-epoch multi-band observations can be effective for identifying radio AGNs by providing the constraint of the radio continuum slope.

On the origin of blue straggler stars in dwarf galaxies

  • Blue stragglers (BSs) are the objects that are brighter and bluer than the stars at main-sequence turn-off point. In this study, we present the Ca-by and VI photometry for Galactic dwarf spheroidal galaxies using Subaru/Suprime-Cam and investigate the spatial distribution characteristics of BS stars using the hk index as a photometric metallicity indicator. We compare the cumulative radial distribution of the BS stars with those of two groups of red-giant-branch (RGB) stars divided by the hk-index strength, and find that the spatial distribution of all BS stars is closer to that of hk-weak (i.e. metal-poor) RGB stars. We also find that the hk-strong BS stars are more centrally concentrated than the hk-weak ones. We will discuss the use of hk-index as a metallicity indicator for the hot BS stars and suggest possible explanations for the results in terms of the origin of BS stars in the dwarf gal.

SURFACE PHOTOMETRY OF THE DWARF ELLIPTICAL GALAXIES NGC 185 AND NGC 205

  • We present B VRI CCD surface photometry for the central $(6'.35\times6'.35)$ regions of the dwarf elliptical galaxies NGC 185 and NGC 205 in the Local Group. Surface brightness profiles of NGC 185 (R<225') and NGC 205 (R<186') show excess components in the central regions. The colors of NGC 185 get bluer inward at R<25', while they remain constant at $R\geq25'$. The colors of NGC 205 get bluer inward at 1'$\approx10^5\; L_\bigodot$. Distributions of dust clouds in the central regions of the two galaxies are also investigated.

Development and Practical Use of RT-PCR for Seed-transmitted Prune dwarf virus in Quarantine

  • Among imported plants, seeds are the items that have many latent pathogens and are difficult to inspect. Also, they are the import and export items whose market is expected to expand. The biggest problem with seeds is viruses. Prune dwarf virus (PDV) is the virus that is commonly inspected in Prunus cerasifera, P. persica, P. armeniaca, P. mandshurica, P. cerasus, P. avium or P. serotina seeds. In this study, two RT-PCR primer sets, which can promptly and specifically diagnose plant quarantine seed-transmitted PDV, were developed; and nested PCR primers, where products amplify 739 and 673 nucleotides (nt), and an nested PCR-product, 305 nt, can be obtained as these products are amplified again, were developed. Also, a modified-positive control plasmid was developed, where the restriction enzyme XhoI, which can identify the contamination of samples from the control, was inserted. The method developed in this study has detected PDV in 18 cases since 2007, and is expected to continuously contribute to the plant quarantine in Korea.
