  • Although hepatocellular dysfunction occurs during sepsis. the mechanism responsible for this remains unclear. Since Kupffer cells provide signals that regulate hepatic response in endotoxin and inflammation. the aim of this study was to investigate the role of Kupffer cells in the alterations in the hepatic microsomal drug metabolizing function during sepsis. Rats were subjected to polymicrobial sepsis by cecal ligation and puncture (CLP)followed by fluid resuscitation. (omitted)

  • Alcohol extracts of plants widely used in the traditional medicine have been tested to establish if they affect the metabolism of drug in mice. Forteen of the plants tested exhibited prolonging and/or inhibitory effect on hexobarbital sleeping time. Some of them also showed increasing effect on strychnine mortality, whereas piperis retrofracti Fructus reduced the toxicity of strychnine.

Effects of Mahwangsan(麻黃散) Extract and Its Constituent Drug on the Contraction of Isolated Guinea pig Trachea Smooth Muscle (마황산(麻黃散) 및 그 구성약물(構成藥物)이 GUINEAPIG의 기관지(氣管支) 평활근(平滑筋)에 미치는 영향(影響))

  • Mahwangsan is used in the treatment of asthma due to wind and cold(風寒喘). This study was carried out to investigate the effect of Mahwangsan extract and each drug composing it on the contractile force of the isolated guinea pig trachea muscle and elucidate its mechanism. The results were obtained as follows; 1. The isolated trachea smooth muscle of guinea pig was signifi-cantly relaxed by the administration of Mahwangsan extract. 2. The contractile response of the isolated trachea smooth muscle of guinea pig to histamine was significantly inhibited by the administration of Mahwangsan extract. 3. The contractile response of the isolated trachea smooth muscle of guinea pig to histamine was significantly inhibited by the administraction of Herba Ephedrae, Cortex Cinnamoni, Flos Farfarae, Fructus Chebulae and Semen Armeniacae extract. 4. The contractile response of the isolated trachea smooth muscle of guinea pig to acetylcholine was significantly inhibited by the administration of Mahwangsan extract. 5. The contractile response of the isolated trachea smooth muscle of guinea pig to acetylcholine was significantly inhibited by the administration of Herba Ephedrae, Flos Farfarae, Fructus Chebulae and Semen Armeniacae extract. 6. The contractile response of the isolated trachea smooth muscle of guinea pig to 5-hydroxytryptamine was significantly inhibited by the administration of Mahwangsan extract. 7. The contractile response of the isolated trachea smooth muscle of guinea pig to 5-hydroxytryptamine was significantly inhibited by the administration of Herba Ephedrae, Flos Farfarae, Fructus Chebulae and Semen Armeniacae extract.

Study on the Analgesic Effects of Several Herbal Drugs and Their Synergistic Effects (수종 한약재의 진통효과 및 약물의 배합에 따른 Synergy효과에 관한 연구)

  • The current experiment was carried out to investigate the analgesic effects of several herbal drugs in acetic acid - induced pain model. In a single drug group : after administration of herbal drugs(1g/kg or 3g/kg) orally for 30 minutes, 1% acetic acid $(250{\mu}l)$ was administered into abdominal cavity of mouse. And then the number of times of writhing response was measured for 30 minutes. In a combination drug group : after administration of herbal drugs (1g/kg and it's compound 2g/kg) orally for 30 minutes, 1% acetic acid $(250{\mu}l)$ was administered into abdominal cavity of mouse. And then the number of times of writhing response was measured for 30 minutes. The results were summarized as follows; 1. Water extracts of Akebiae caulis(木通) and Stephaniae tetrandrae radix(防己) decreased significantly the number of writhing response. 2. Methanol extracts of Achyranthis bidentatae radix(牛膝), Carthami flos(紅花), Akebiae caaulis(木通), Stephaniae tetrandrae radix(防己), Myrrha(沒藥), Corydalidis tuber(玄胡索) and Persicae semen(桃仁) decreased significantly the number of writhing response. 3. Water extracts of Achyranthis bidentatae radix(牛膝) plus Akebiae caulis(木通), Achyranthis bidentatae radix(牛膝) plus Stephaniae tetrandrae radix(防己) and Achyranthis bidentatae radix(牛膝) plus Ledebouriellae radix(防風) decreased the number of writhing response significantly. 4. Methanol extracts of Achyranthis bidentatae radix(牛膝) plus Myrrha(沒藥), Achyranthis bidentatae radix(牛膝) plus Stephaniae tetrandrae radlx(防己) and Achyranthis bidentatae radix(牛膝) plus Ledebouriellae radix(防風) decreased the number of writhing response significantly.

  • Alnus japonica has been known to exert antioxidative, anti-inflammatory, anti-cancer and immune response inhibitory effects. The aim of this study was to figure out the characteristics of extracts obtained with different extraction solvent such as water, 100% ethanol, 70% ethanol or 70% methanol because characteristic components such as oregonin and hirsutanone extracted from Alnus japonica might be essential for the biological activity. For this purpose, oregonin and hirsutanon of four extracts, index ingredient of Alnus japonica, were analyzed with HPLC and physicochemical studies such as SEM, particle size and zeta potential were conducted. In cell cytotoxicity study, extract of water showed the highest cytotoxicity among four extracts. In case of oregonin, 70% MeOH and water extracts showed high contents and in case of hirsutanone, all extracts showed similar contents except 70% EtOH extracts. The extract of 70% MeOH from Alnus japonica for both oregonin and hirsutanone appeared to have the highest content. Both oregonin and hirsutanone extracted from Alnus japonica using 70% methanol showed stability in pH 1.2.

Poly(L-lysine) Based Semi-interpenetrating Polymer Network as pH-responsive Hydrogel for Controlled Release of a Model Protein Drug Streptokinase

  • With the aim of developing of pH-sensitive controlled drug release system, a poly(Llysine) (PLL) based cationic semi-interpenetrating polymer network (semi-IPN) has been synthesized. This cationic hydrogel was designed to swell at lower pH and de-swell at higher pH and therefore be applicable for achieving regulated drug release at a specific pH range. In addition to the pH sensitivity, this hydrogel was anticipated to interact with an ionic drug, providing another means to regulate the release rate of ionic drugs. This semi-IPN hydrogel was prepared using a free-radical polymerization method and by crosslinking of the polyethylene glycol (PEG)-methacrylate polymer through the PLL network. The two polymers were penetrated with each other via interpolymer complexation to yield the semi-IPN structures. The PLL hydrogel thus prepared showed dynamic swelling/de-swelling behavior in response to pH change, and such a behavior was influenced by both the concentrations of PLL and PEG-methacrylate. Drug release from this semi-IPN hydrogel was also investigated using a model protein drug, streptokinase. Streptokinase release was found to be dependent on its ionic interaction with the PLL backbones as well as on the swelling of the semi-IPN hydrogel. These results suggest that a PLL semi-IPN hydrogel could potentially be used as a drug delivery platform to modulate drug release by pH-sensitivity and ionic interaction.

Naturally Occurring of Sulfur Dioxide in Medicinal Herbs(Crude Drug Materials) and Its Origin (생약재중 천연유래 이산화황 함유량 및 그 출처)

  • Naturally occurring sulfur dioxide in medicinal herbs(crude drug materials) and its origin was investigated. Sulfur dioxide content in 28 kinds (101 samples) of medicinal herbs was studied by two different methods, Monnier-Williams and acid-distillation ion chromatography. Generally, less than 5 ppm of sulfur dioxide was found from the tested herbs. In some herbs, Monnier-Williams method showed higher level of sulfur dioxide than acid-distillation ion chromatography probably due to the presence of volatile organic acid presence in herbs. Naturally occurring sulfur dioxide measured in medicinal herbs may be derived from sulfur compounds of medicinal herbs.
