비정상 층류화염편 모델을 이용한 비예혼합 난류화염 해석 (Unsteady Flamelet Modeling of Turbulent Nonpremixed Flames)

  • The present study is focused on modeling the transient behavior of the local flame structure which is especially important for slow reaction processes, such as NOx formation in the radiating flame field. The recently developed unsteady flamelet model has been applied to analyze a steady, turbulent jet flame. Numerical results are compared with experimental data and numerical results of the conventional steady flamelet model. The numerical result reveals that the unsteady flamelet model correctly predicts the nonequilibrium effect upsteam and the subsequent decay of the superequilibrium radical concentrations the further downstream.

Multi-environment PDF 모델을 이용한 MILD 연소과정 해석 (Multi-environment PDF Modeling for MILD Combustion Processes)

  • In this study, the multi-environment probability density function(MEPDF) approach has been applied to numerically investigate Delft-Jet-in-Hot-Coflow(DJHC) turbulent flames under Moderate or Intense Low-oxygen Dilution (MILD) combustion condition. Computations are made for two different jet velocities(Re = 4100 and 8800). In terms of mean axial velocity, temperature, and turbulent kinetic energy, numerical results are in reasonably good agreements with experimental data even if there exist the noticeable deviations in downstream region. Based on numerical results, the detailed discussions are made for the essential features of the non-visible flame structure and MILD combustion processes.

지우천 유역의 퇴적지형 연구 (Depositional Landforms in Jiwoo Drainage Basin)

  • 본 연구에서는 남강 최상류부의 지우천 유역을 대상으로 침식분지 내 퇴적지형의 형성과정을 고찰하였다. 퇴적 지형의 형태적 특성과 퇴적상 분석을 통하여 밝혀진 형성과정을 요약하면 다음과 같다. 첫째, 사평, 응암, 내동뒷들 지점의 고위퇴적지는 최종빙기 동안의 한랭한 주빙하 환경하에서 젤리플럭션 작용으로 퇴적되었으며, 상류에서 하류방향으로 하상비고가 낮아진다. 고위퇴적지의 범위는 지우천과 남강이 합류하는 곳에서 하류방향 약 1km지점인 것으로 추정된다. 둘째, 장승불들 지점에 분포하는 완경사면의 퇴적물은 심층풍화된 기반암이 완만한 사면을 형성한 이후 최종 빙기 동안 젤리플럭션 작용으로 운반된 것이다. 셋째, 사평, 용추사, 등방들 지점의 저위퇴적지는 후빙기의 온난습윤한 환경하에서 고위퇴적지가 개석되는 동안 퇴적물의 일부가 하상에 남겨지고, 이후 고위퇴적지의 개석이 완료됨에 따라 하천의 저항(퇴적물양)보다 하천의 운반력이 초과함으로써 하상이 다시 침식되어 형성된 것으로 사료된다.

  • The low-emission and high-performance diesel combustion is an important issue in the combustion research community, In order to understand the detailed diesel flame involving the complex physical processes, it is quite desirable to diesel spray dynamics, auto-ignition and spray flame propagation. Dynamics of fuel spray is a crucial element for air-fuel mixture formation, flame stabilization and pollutant formation, In the present study, the diesel RCM (Rapid Compression Machine) and the Electric Control injection system have been designed and developed to investigate the effects of injection pressure, injection timing, and intake air temperature on spray dynamics and diesel combustion processes, In terms of the macroscopic spray combustion characteristics, it is observed that the fuel jet atomization and the droplet breakup processes become much faster by increasing the injection pressure and the spray angle, With increasing the cylinder pressure, there is a tendency that the of spray pattern in the downstream region tends to be spherical due to the increase of air density and the corresponding drag force, Effects of intake temperature and injection pressure on auto-ignition is experimently analysed and discussed in detail.

Potential drug targets in the GPCR-$G{\alpha}_{12}/G{\alpha}_{13}$ signaling pathways

  • GPCRs are large families of cell surface receptors that transmit signals through conformational changes upon ligand activation and an interaction with the heterotrimeric G-proteins. GPCRs regulate the cell-signaling pathways and participate in the regulation of physiological processes through the G-proteins defined by their ${\alpha}$ subunits. A family of 20 G protein-coupled receptors (GPCRs) that provide a large class of tractable drug targets for new anti-inflammatory drugs and, in certain instances, for the treatment of the inflammatory indications such as atherosclerosis, rhinitis, asthma, pulmonary disease and arthritis. In view of the important findings showing that $G{\alpha}_{12}/G{\alpha}_{13}$ regulate the various cellular processes such as actin-stress fiber formation, neurite retraction, platelet aggregation, gene induction, and apoptosis, we became interested in whether, after coupling to the activated GPCRs, the G-proteins and their downstream molecules might be involved in the pathologic processes of chronic inflammatory diseases (e.g., liver fibrosis). In this symposium, the possible link of the G-proteins with the pathophysiology will be discussed with the aim of finding potential new drug targets.

Identification of WDR7 as a Novel Downstream Target of the EphA8-Odin Signaling Complex

  • Eph receptors and their ephrin ligands have been implicated in a variety of cellular processes such as cellular morphogenesis and motility. Our previous studies demonstrated that Odin, one of the Anks family proteins, functions as a scaffolding protein of the EphA8 signaling pathway leading to modulation of cell migration or axonal outgrowth. Here we show that WDR7 is associated with Odin and that it is possibly implicated in the EphA8 signaling pathway. WD40 repeats present in the COOH-terminal region of WDR7 appear to be crucial for its association with Odin, whereas the binding motif of Odin is located in between ankyrin repeats and PTB domain. Co-immunoprecipitation experiments revealed that association of WDR7 with Odin is enhanced by ephrin ligand treatment, possibly through forming large protein complexes including both EphA8 and ephrin-A5. Consistently, immunofluorescence staining experiments suggested that WDR7 constitute a component of the large protein complexes containing Odin, EphA8 and ephrin-A5. Taken together, our results suggest the WDR7-Odin complexes might be involved in the signaling pathway downstream of the EphA8 receptor.

Endocytic Regulation of EGFR Signaling

  • Epidermal growth factor receptor (EGFR) is a member of the ErbB family (ErbB1-4) of receptor tyrosine kinases (RTKs). EGFR controls numerous physiological functions, including cell proliferation, migration, differentiation and survival. Importantly, aberrant signaling by EGFR has been linked to human cancers in which EGFR and its various ligands are frequently overexpressed or mutated. EGFR coordinates activation of multiple downstream factors and is subject of various regulatory processes as it mediates biology of the cell it resides in. Therefore, many studies have been devoted to understanding EGFR biology and targeting the protein for the goal of controlling tumor in clinical settings. Endocytic regulation of EGFR offers a promising area for targeting EGFR activity. Upon ligand binding, the activated receptor undergoes endocytosis and becomes degraded in lysosome, thereby terminating the signal. En route to lysosome, the receptor becomes engaged in activating various signaling pathways including PI-3K, MAPK and Src, and endocytosis may offer both spatial and temporal regulation of downstream target activation. Therefore, endocytosis is an important regulator of EGFR signaling, and increasing emphasis is being placed on endocytosis in terms of cancer treatment and understanding of the disease. In this review, EGFR signaling pathway and its intricate regulation by endocytosis will be discussed.

두경부암 세포주에서 상피성장인자수용체 타이로신 카이네이즈 억제제인 gefitinib의 성장억제에 관한 연구 (Growth inhibition in head and neck cancer cell lines by gefitinib, an epidermal growth factor receptor tyrosine kinase inhibitor)

  • Cell survival is the result of a balance between programmed cell death and cellular proliferation. Cell membrane receptors and their associated signal transducing proteins control these processes. Of the numerous receptors and signaling proteins, epidermal growth factor receptor (EGFR) is one of the most important receptors involved in signaling pathways implicated in the proliferation and survival of cancer cells. EGFR is often highly expressed in human tumors including oral squamous cell carcinomas, and there is increasing evidence that high expression of EGFR is correlated with poor clinical outcome of common human cancers. Therefore, we examined the antiproliferative activity of gefitinib, epidermal growth factor receptor tyrosine kinase inhibitor (EGFR TKI), in head and neck cancer cell lines. SCC-9, KB cells were cultured and growth inhibition activity of gefitinib was measured with MTT assay. To study influence of gefitinib in cell cycle, we performed cell cycle analysis with flow cytometry. Western blot was done to elucidate the expression of EGFR in cell lines and phosphorylation of EGFR and downstream kinase protein, Erk and Akt. Significant growth inhibition was observed in SCC-9 cells in contrast with KB cells. Also, flow cytometric analysis showed G1 phase arrest only in SCC-9 cells. In Western blot analysis for investigation of EGFR expression and downstream molecule phosphorylation, gefitinib suppressed phosphorylation of EGFR and downstream protein kinase Erk, Akt in SCC-9. However, in EGFR positive KB cells, weak expression of active form of Erk and Akt and no inhibitory activity of phosphorylation in Erk and Akt was observed. The antiproliferative activity of gefitinib was not correlated with EGFR expression and some possibility of phosphorylation of Erk and Akt as a predictive factor of gefitinib response was emerged. Further investigations on more reliable predictive factor indicating gefitinib response are awaited to be useful gefitinib treatment in head and neck cancer patients.
