Fibronectin is a glycoprotein found in the extracellular matrix as well as in the serum, and has been known to exert pronouned effed on the myoblast fusion. Our previous studies have suggested that the decrease of fibronectin levels during myogenesis is due to the decreased availability of the receptor for the 28 kDa fragrnent of fibronetin. In the fusion-blocked myoblasts by EGTA, the levels of fibronetin and binding of 28 kDa fragment decreased but far less than the control level. In contrast, the levels of fibronetin and binding of 28 kDa fragment decreased to the control level in the myoblast released from the fusion block. On this account, we suggest that the decrease of fibronetin levels during myoblast fusion is closely associated with the loss or alteration of the receptor for 28 kDa fragment. Mild trypsin treatment decreased the binding of the 28 kDa fragment to the myoblasts significandy. Similarly, the presence of gangliosides in the binding media decreased the binding of the 28 kDa fragment in a dose-dependent manner. Furthermore, gel overlay of 125 I-28 kDa fragment on the SDS-PAGE of the myoblast homogenates revealed that the 28 kDa fragment bound to a 43 kDa protein and to gangliosides as well. These results suggest that myoblast fusion is correlated with decrease of the receptor for the 28 kDa fragment and that the receptor might be a glycoprotein that contains glyco-conjugate found in gangliosides.
In this study, we investigated the quality characteristics of cookies containing mesangi powder (MP). The cookies were stored at 30 $^{\circ}C$ for 28 days. The pH of the dough decreased significantly upon the addition of MP. The spread factor of the cookies also decreased significantly with more MP added to the cookie recipe. The loss rate of cookies was significantly higher in 3% and 5% cookies than in control and 7% cookies. The leavening rate was significantly decreased upon the addition of MP. Hardness was significantly higher in 5% and 7% cookies than in control cookies. The L, a, b values of cookies were significantly reduced at higher MP content. The results of sensory evaluation showed that cookies made with MP did not differ significantly from the control, except in color and moistness. The mineral contents of cookies increased in dose-dependent fashion with MP concentration. The acid and peroxide values were lower in cookies with 5% MP than other cookies.

A parametric sensitivity to the annual exposure dose rate to the farming exposure group has been probabilistically carried out for three principal elements associated with the nuclide transport behavior in the near-field of the pyroprocessed waste repository system. Credit time for both metal and ceramic containers, annual nuclide release rete, and the degree of loss of bentonite buffer around the container are selected as the elements and investigated for important nuclides. All the elements are shown to be sensitive to the results. Methodology studied through this study and the results are expected to make a good feedback to the repository design. As a follow-up study, separated in Part 2, the A-KRS will be deterministically assessed and then compared among each other with the normal, the worst, and the best case scenarios associated with their extreme values these elements could have.
The G-class cement is commonly used in practice for geothermal well cementing in order to protect a steel casing that is designed to transport hot water/steam from deep subsurface to ground surface during operating a geothermal power plant. In order to maintain optimal performance of geothermal wells, physical properties of the cementing material should be satisfactory. In this paper, relevant factors (i.e., groutability, uniaxial compression strength, thermal conductivity and free fluid content) of the G-class cement were experimentally examined with consideration of various water-cement (w/c) ratios. Important findings through the experiments herein are as follows. (1) Groutability of the G-class cement increases by adding a small dose of retarder. (2) There would be a structural defect caused when the w/c ratio is kept higher in order to secure groutability. (3) Thermal conductivity of the G-class cement is small enough to prevent heat loss from hot steam or water to the outer ground formation during generating electricity. (4) The G-class cement does not form free water channel in cementing a geothermal well. (5) The Phenolphthalein indicator is applicable to the distinction of the G-class cement from the drilling mud.

Background: Ginseng has been widely used as a health-promoting tonic. Gintonin present in ginseng acts as a lysophosphatidic acid (LPA) receptor ligand that activates six LPA receptor subtypes. The LPA6 subtype plays a key role in normal hair growth, and mutations in the LPA6 receptor impair normal human hair growth. Currently, human hair loss and alopecia are concerning issues that affect peoples' social and day-to-day lives. Objective: We investigated the in vitro and in vivo effects of a gintonin-enriched fraction (GEF) on mouse hair growth. Methods: Human hair follicle dermal papilla cells (HFDPCs) and six-week-old male C57BL/6 mice were used. The mice were divided into the four groups: control, 1% minoxidil, 0.75% GEF, and 1.5% GEF. The dorsal hair was removed to synchronize the telogen phase. Each group was treated topically, once a day, for 15 days. We analyzed hair growth activity and histological changes. Results: GEF induced transient [Ca2+]i, which stimulated HFDPC proliferation and caused 5-bromo-2'-deoxyuridine (BrdU) incorporation in a concentration-dependent manner. GEF-mediated HFDPC proliferation was blocked by the LPA receptor antagonist and Ca2+ chelator. HFDPC treatment with GEF stimulated vascular endothelial growth factor release. Topical application of GEF and minoxidil promoted hair growth in a dose-dependent manner. Histological analysis showed that GEF and minoxidil increased the number of hair follicles and hair weight. Conclusion: Topical application of GEF promotes mouse hair growth through HFDPC proliferation. GEF could be one of the main components of ginseng that promote hair growth and could be used to treat human alopecia.
The effects of gamma irradiation on the physiochemical and physical characteristics of apples were investigated during post-irradiation storage at $4^{\circ}C\;and\;25^{\circ}C$. The contents of total and reducing sugars were analyzed and the results indicated that apples receiving 1 kGy of gamma irradiation did not show significant differences in sugar contents compared to non-irradiated controls. Important physiological characteristics were evaluated by measurement of total phenolic content and total flavonoid content, reducing power, and radical scavenging ability, and the results indicated that gamma irradiation at a dose of 1 kGy did not affect physiological activities. Changes in physical parameters such as weight loss, strength, cohesiveness and hardness, during post-irradiation storage, were temperature-dependent, whether the apples were irradiated or not The color and sensory acceptance of the apples were not affected by irradiation during cold storage. However, minor deterioration in color quality and sensory acceptance of irradiated apples was noted under ambient temperature storage. We conclude that gamma irradiation(1 kGy) does not affect apple nutritional content stability, functional properties, or physical characteristics, especially upon cold storage after radiation treatment.
Fresh mushroom (Agaricus bisporus L.) was irradiated (0, 1, 2, 3kGy) and kept for 20 days at 9{\pm}1^{\circ}C\;and\;80{\pm}7%$ RH. Parameters of qualities were investigated on the physical and physiological characteristics. The pileus and stipe on nonirradiated mushroom were expanded and elongated from the 3rd day of storage, there by losing the acceptability as edible samples. After 5 days of storage, 2 to 3kGy of gamma irradiation were especially effective for controllong natural maturation and senescence of fresh mushrooms and so irradiated mushrooms were acceptable more than 20days storage. The texture of irradiated samples was superior to that of nonirradiated samples, even though softening of the tissue occurred during storage. Weight loss was greatest in the nonirradiated sample due to evaporation from an increased surface area resulting from expansion and ripending, which were retarded in the 2 to 3kGy irradiated samples after 5days of storage. These results suggest that the irradiation dose of 2 to 3kGy is apparently effective to extend the shelf life of fresh mushrooms stored at the above-mentioned condition.

Corticotropin-releasing factor(CRF), one of the stress driven neuropeptides, was widely proposed to influence hair loss and re-growth. For the development of receptor antagonists, the screening system based on intracellular calcium signal process was developed and optimized. The aequorin parental cells were transfected with CRF1 receptor and alpha 16 promiscuous G protein cDNA to establish HEK293a16/hCRF1, a stable cell line for the human CRF1 receptor. In HEK293a16/hCRF1 cells, the range of sauvagine dose response was 12-fold higher($EC_{50}:15.21{\pm}1.83nM$) than in the transiently expressed cells, hence essential conditions for the antagonist screening experiments such as the robust signals and high solvent tolerance were secured. The standard antagonists for the CRF1 receptor, antalarmin and CP154526, resulted $IC_{50}$ values of $414.1{\pm}5.5$ and $290.7{\pm}1.9nM$, respectively. Similar results were presented with frozen HEK293a16/hCRF1 cells. Finally, our HEK293a16/hCRF1 cells with the aequorin based cellular functional assay can be a model system for the development of functional cosmetics and modulators that can have a clinical efficacy on hair re-growth.
BACKGROUND/OBJECTIVES: Sargassum horneri is an edible brown alga that grows in the subtidal zone as an annual species along the coasts of South Korea, China, and Japan. Recently, an extreme amount of S. horneri moved into the coasts of Jeju Island from the east coast of China, which made huge economic and environmental loss to the Jeju Island. Thus, utilization of this biomass becomes a big issue with the local authorities. Therefore, the present study was performed to evaluate the anti-inflammatory potential of crude polysaccharides (CPs) extracted from S. horneri China strain in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. MATERIALS/METHODS: CPs were precipitated from S. horneri digests prepared by enzyme assistant extraction using four food-grade enzymes (AMG, Celluclast, Viscozyme, and Alcalase). The production levels of nitric oxide (NO) and pro-inflammatory cytokines, including tumor necrosis factor (TNF)-${\alpha}$ and interleukin (IL)-$1{\beta}$ were measured by Griess assay and enzyme-linked immunosorbent assay, respectively. The levels of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2), nuclear factor (NF)-${\kappa}B$, and mitogen-activated protein kinases (MAPKs) were measured by using western blot. The IR spectrums of the CPs were recorded using a fourier transform infrared spectroscopy (FT-IR) spectrometer. RESULTS: The polysaccharides from the Celluclast enzyme digest (CCP) showed the highest inhibition of NO production in LPS-stimulated RAW 264.7 cells ($IC_{50}$ value: $95.7{\mu}g/mL$). Also, CCP dose-dependently down-regulated the protein expression levels of iNOS and COX-2 as well as the production of inflammatory cytokines, including TNF-${\alpha}$ and IL-$1{\beta}$, compared to the only LPS-treated cells. In addition, CCP inhibited the activation of NF-${\kappa}B$ p50 and p65 and the phosphorylation of MAPKs, including p38 and extracellular signal-regulated kinase, in LPS-stimulated RAW 264.7 cells. Furthermore, FT-IR analysis showed that the FT-IR spectrum of CCP is similar to that of commercial fucoidan. CONCLUSIONS: Our results suggest that CCP has anti-inflammatory activities and is a potential candidate for the formulation of a functional food ingredient or/and drug to treat inflammatory diseases.
Twenty five patients with unresectable non-small cell carcinoma of the lung have been treated with hyperfractionated radiotherapy with concomitant boost technique since September, 1989. Those patients with history of previous surgery or chemotherapy, pleural effusion or significant weight loss (greater than $10\%$ of body weight) were excluded from the study. Initially, 27 Gy were delivered in 15 fractions in 3 weeks to the large field. Thereafter, large field received 1.8 Gy and cone down boost field received 1.4 Gy with twice a day fractinations up to 49.4 Gy. After 49.4Gy, only boost field was treated twice a day with 1.8 and 1.4 Gy. Total tumor doses were 62.2 Gy for 12 patients and 65.4 Gy for remaining 13 patients. Follow up period was ranged from 6 to 24 month. Actuarial survival rates at 6, 12, and 18 month were $88\%,\;62\%,\;and\;38\%$, respectively. Corresponding disease free suwival rates were $88\%,\;41\%,\;and\;21\%$, respectively. Actuarial cumulative local failure rates at 9, 12 and 15 month were $36\%,\;43\%,\;and\;59\%$, respectively. No significant increase of acute or late complications including radiation pneumonitis was noted with maximum follow up of 24 month. Although the longer follow up is needed, it is worthwhile to try the prospective randomized study to evaluate the efficacy of hyperfractionated radiotherapy with concomitant boost technique for unresectable non-small cell lung cancers in view of excellent tolerance of this treatment. In the future, further increase of total radiation dose might be necessary to improve local control for non-small cell lung cancer.
