As a bioreactor, bird has proved to be most efficient system for producing useful therapeutic proteins. More than half of the egg white protein content derives from the ovalbumin gene with four other proteins(lysozyme, ovomucoid, ovomucin and conalbumin) present at levels of 50 milligrams or greater. And the naturally sterile egg also contains egg white protein at high concentration allowing for a long shelf life of recombinant protein without loss in activity. In spite of these advantages, transgenic procedures for the bird have lagged far behind because of its complex process of fertilized egg and developmental differences. Recently, a system to transplant mouse testis cells from a fertile donor male to the seminiferous tubules of an infertile recipient male has been developed. Spermatogenesis is generated from transplanted cells, and recipients are capable of transmitting the donor haplotype to progeny. After transplantation, primitive donor spermatogonia migrate to the basement membrane of recipient seminiferous tubules and begin proliferating. Eventually, these cells establish stable colonies with a characteristic appearance, which expands and produces differentiating germ cells, including mature spermatozoa. Thus, the transplanted cells self-renew and produce progeny that differentiate into fully functional spermatozoa. In this study, to develop an alternative system of germline chimera production that operates via the testes rather than through developing embryos, the spermatogonial stem cell techniques were applied. This system consisted of isolation and in vitro-culture of chicken testicular cells, transfer of in vitro-maintained cells into heterologous testes, production of germline chimeras and confirmation of germline transmission for evaluating production of heterologous, functional spermatozoa.
The effect of poly[2-methacryloyloxyethyl phosphorylcholine] (pMPC) on the skin permeation property was investigated by performing in vitro skin permeation study of a model drug, nicotinic acid (NA). Effect of pMPC polymer in donor solution on skin permeation rates was evaluated using side-by-side diffusion cells. Also, the structural alterations in the stratum corneum (SC), inter-lamellar bilayer (ILB) and dermis layers in pMPC-treated and -untreated skin sections were investigated with transmission electron microscopy (TEM). The permeation profile of NA without pMPC in donor solution showed biphasic mode: initial $1^{st} phase and 2^{nd}$ hydration phase. The sudden, more than 10-fold increase in flux from the initial steady state (43.5 $\mu g/cm^2$/hr) to the $2^{nd}$ hydration phase (457.3 $\mu g/cm^2$/hr) suggests the disruption of skin barrier function due to extensive hydration. The permeation profile of NA with 3% pMPC in the donor solution showed monophasic pattern: the steady state flux (10.9 $\mu g/cm^2$/hr) without abrupt increase of the flux. The degree of NA permeation rate decreased in a concentration-dependent manner of pMPC. TEM of skin equilibrated with water or 2% pMPC for 12 h showed that corneocytes are still cohesive and epidermis is tightly bound to dermis in 2% pMPC-treated skin, while wider separation between corneocytes and focal dilations in inter-cellular spaces were observed in water-treated skin. This result suggests that pMPC could protect the barrier property of the stratum corneum by preventing the disruption of ILB structure caused by extensive skin hydration during skin permeation study.
A bioassay technique and organ bath study were performed to analyze the effects of extracellular $Ca^{2+}$ and $Ca^{2+}$-antagonists on endothelium-derived relaxing factor[s][EDRF] released from the endothelial cells of rabbit aorta. Transverse strips with intact endothelium or damaged endothelium were used for the mechanical contraction experiment using organ bath. Long segment including thoracic and abdominal aorta with endothelium [EDRF donor aorta] was perfused with Tyrode solution which was aerated with 95% $O_2-5%$$CO_2$ mixed gas and kept at 35oC. The perfusate was bioassayed with a transverse strip of thoracic aorta with damaged endothelium. The test strip was contracted with nor-epinephrine and acetylcholine was used to stimulate the release of EDRF from endothelial cells. The results obtained were as follows; 1] The endothelium-dependent relaxation[EDR] induced by acetylcholine was biphasic; an initial rapid relaxation followed by a slow relaxation. 2] EDR induced by acetylcholine was reduced gradually with the decrease in the concentration of extracellular $Ca^{2+}$. The effect of extracellular $Ca^{2+}$ on EDR was more prominent in the late slow relaxation phase. 3] EDR to acetylcholine was not altered by acute exposure to organic $Ca^{2+}$-antagonists. Pretreatment with verapamil to the EDRF donor aortic segment did not alter the magnitude of EDR. 4] Among the inorganic $Ca^{2+}$-antagonists $Mn^{2+}$ and $Cd^{2+}$ did not inhibit EDR, whereas $Co^{2+}$ and $La^{3+}$ inhibited EDR. 5] The inhibitory response of $Co^{2+}$ to EDR developed when infused directly on the test strip. That of $La^{3+}$, however, was evoked when added to solution perfusing the donor aortic segment. The above results suggest that $Ca^{2+}$-antagonists do not affect EDR and the inhibitory effect of $Ca^{2+}$ results from influencing the action of EDRF on vascular smooth muscle, whereas that of $La^{3+}$ results from its action on the release of EDRF from endothelial cells.
It was investigated whether an electrode could serve as an electron donor for biological reductive dechlorination of chlorinated phenols in the bio-electrochemical process. There was no dechlorination in the absence of current and scanning electron microscope image showed that the electrode surface was covered with microorganisms. As a result, the electrode attached cells was responsible for reductive dechlorination. Also, initial high chlorinated phenol concentration such as $437mg/{\ell}$ was rapidly reduced within 5 hours. The maximum dechlorination rate using Monod equation was $5.95mg{\ell}$-h($cm^2$ (electrode surface area)) in the bio-electrochemical reactor.

In this study, high-quality Al-N doped p-type ZnO thin films were deposited on n-type Si (100) wafer or Si coated with buffer layer by DC magnetron sputtering in the mixture of $N_2$ and $O_2$ gas. The target was ceramic ZnO mixed with $Al_2O_3$ (2 wt%). The p-type ZnO thin film showed higher carrier concentration $2.93\times10^{17}cm^{-3}$, lower resistivity of $5.349\;{\Omega}cm$ and mobility of $3.99\;cm^2V^{-1}S^{-1}$, respectively. According to PL spectrum, the Al donor energy level depth ($E_d$) of Al-N codoped p-type ZnO film was reduced to about 51 meV, and the N acceptor energy level depth ($E_a$) was reduced to 63 meV, respectively.
We experienced one case of orthotopic cardiac transplantation in a patient with end stage dilated cardiomyopathy. This 50 year-old female recipient was suffered from NYHA functional class IV cardiac failure and dependent upon intravenous inotropic support for 2 months [recipient category 1]. Her preoperative condition was grave with left ventricular ejection fraction of 20% and estimated systolic pulmonary arterial pressure [from Doppler study] was 50mmHg. The brain-dead donor was 31 year-old male with head trauma. The body sizes [weight, height] of the donor/recipient were 70 Kg, 165 cm / 43 Kg, 160 cm and appropriately overmatched. Preoperatively, identical ABO/Rh blood group [A+] and nonreacting HLA crossmatching were confirmed. On November 11 1992 cardiac transplantation was performed without complication. Multiple organ procurement team and heart transplantation team were organized the operation schedule appropriately to minimize the ischemic time. The pump time was 126 minutes and aortic crossclamping time of recipient heart was 73 minutes and, as a result, total ischemic time of the transplanted heart was 75 minutes. Postoperatively, the vital signs were stable with minimal inotropic support. The immunosuppressive therapy was commenced from preoperatively and cyclosporine, azathioprine, and corticosteroid were used as a combination therapy as scheduled and monitored with blood drug concentration, WBC count, renal function and most importantly regular endomyocardial biopsy.Now, 5 months after transplantation, the patient is in NYHA functional class II with minimal cardiac drug support.

Intrinsic layers of homoepitaxial InP grown by the hydride vapor phase epitaxy (VPE) technique were investigated by Fourier-transform photoluminescence(FTPL) and variable temperature Hall measurements. The effect of process variables (i.e., source zone temperature and inlet mole fractions of HCl and $PH_{3}$) on the backgroudn impurity levels was investigated. The background carrier concentration was found to decrease with decreasing source zone temperature and increasing HCl, but was relatively independent of $PH_{3}$ for the range of mole fraction studied. The presence of background donors and acceptors was clearly verified in the FTPL spectra, and the major impurities were tentatively identified as Si donors and Zn acceptors as well as some unidentified acceptors.
SRB(Sulfate Reducing Bacteria) converts sulfate into sulfide using an organic carbon source as the electron donor. The sulfide formed precipitates the various metals present in the AMD (Acid Mine Drainage). This study is the fundamental research on heavy metal removal from AMD using SRB. Two completely mixed anaerobic reactors were operated for cultivation of SRB at the temperature of $30^{\circ}C$ and anaerobic batch reactors were used to evaluate the effects of carbon source, COD/sulfate($SO_4^=$) ratio and alkalinity on sulfate reduction rate and heavy metal removal efficiency. AMD used in this study was characterized by low pH 3.0 and 1000mg/l of sulfate and dissolved high concentration of heavy metals such as iron, cadmium, copper, zinc and lead. It was found that glucose was an organic carbon source better than acetate as the electron donor of SRB for sulfate reduction in AMD. Amount of sulfate reduction maximized at the COD(glucose)/sulfate ratio of 0.5 in the influent and then removal efficiencies of heavy metals were 97.5% of Cu, 100% of Pb, 100% of Cr, 49% of Mn, 98% of Zn, 100% Cd and 92.4% of Fe. Although sulfate reduction results in an increase in the alkalinity of the reactor, alkalinity of 1000mg/1 (as $CaCo_3$) should be should be added continuously to the anaerobic reactor in order to remove heavy metals from AMD.
Recently, antibiotic resistant genes (ARGs) in the environment are emerging as pollutants, since these genetic contaminants can eventually be transferred to human pathogens. The aim of this study was to develop the experimental model of antibiotic resistant gene (ARG) plasmid transfer as a function of various environmental conditions. For this purpose, the multi drug resistant plasmid pB10, which is known to be originally isolated from a wastewater treatment plant, was selected as a model transfer plasmid and Escherichia coli $DH5{\alpha}$ containing pB10 was used as a model donor. Pseudomonas aeruginosa, an opportunistic pathogen, was selected as the recipient for the conjugation experiment. When the donor and recipient were exposed to various stressors including antibiotics and heavy metal as a function of the concentrations (10, 100 and, 1000 ppb), statistically increased plasmid transfer rate was observed at a concentration of 10 ppb of tetracycline and sulfamethoxazole compared to control (no antibiotic exposure). Accordingly, the developed experimental ARG model by various stressor is a promising tool for evaluating the dissemination of ARGs by micro-contaminants in aquatic environment.

Anaerobic reductive dechlorination of tetrachloroethylene(PCE) to ethylene was investigated by performing laboratory experiments using semi-continuous flow two-in-series soil columns. The columns were packed with soils obtained from TCE-contaminated site in Korea. Site ground water containing lactate(as electron donor and/or carbon source) and PCE was pumped into the soil columns. During the first operation with a period of 50 days, injected mass ratio of lactate and PCE was 620:1 and incomplete reductive dechlorination of PCE to cis-DCE was observed in the columns. However, complete dechlorination of PCE to ethylene was observed when the mass ratio increased to 5,050:1 in the second operation, suggesting that the electron donor might be limited during the first operation period. During the degradation of cis-DCE to ethylene, the concentration of hydrogen was $22{\sim}29mM$. These positive results indicate that the TCE-contaminated groundwater investigated in this study could be remediated through biological anaerobic reductive dechlorination processes.
