A Study on Establishment of Technical Guideline of the Installation and Operation for the Biogas Utilization of Transportation and City Gas: Results of the Field Investigation (고품질화 바이오가스 이용 기술지침 마련을 위한 연구(I): 도시가스 및 수송용 - 현장조사 결과 중심으로)

  • Biogasification is a technology that uses organic wastes to reproduce as environmental fuels containing methane gas. Biogasification has attracted worldwide attention because it can produce renewable-energy and stable land treatment with prohibit from landfilling and ocean dumping of organic waste. Biomethane is produced by refining biogas. It is injected into natural gas pipeline or used transportation fuel such as cars and buses. 90 bio-gasification facilities are operating in 2016, and methane gas production is very low due to it is limited to organic wastes such as food waste, animal manure, and sewage sludge. There are seven domestic biomethane manufacturing facilities, and the use of high value-added such as transport fuels and city-gas through upgrading biogas should be expanded. On the other hand, the rapid biogasification of organic wastes in domestic resulted in frequent breakdowns of facilities and low efficiency problems. Therefore, the problem is improving as technical guidance, design and operational technical guidance is developed and field experience is accumulated. However, while improvements in biogas production are being made, there is a problem with low utilization. In this study, the problems of biomethane manufacturing facilities were identified in order to optimize the production and utilization of biogas from organic waste resources. Also, in order to present the design and operation guideline of the gas pretreatment and the upgrading process, we will investigate precision monitoring, energy balance and economic analysis and solutions for on-site problems by facility.

Effect of Areal Mean Rainfall Estimation Technique and Rainfall-Runoff Models on Flood Simulation in Samcheok Osipcheon(Riv.) Basin (면적 강우량 산정 기법과 강우-유출 모형이 삼척오십천 유역의 홍수 모의에 미치는 영향)

  • In terms of flood management, it is necessary to analyze quantitative rainfall and runoff from a spatial and temporal perspective and to analyze runoff for heavy rainfall events that are concentrated within a short period of time. The simulation and analysis results of rainfall-runoff models vary depending on the type and input data. In particular, rainfall data is an important factor, so calculating areal mean rainfall is very important. In this study, the areal mean rainfall of the Samcheok Osipcheon(Riv.) watersheds located in the mountainous terrain was calculated using the Arithmetic Mean Method, Thiessen's Weighting Method, and the Isohyetal Method, and the rainfall-runoff results were compared by applying the distributional model S-RAT and the lumped model HEC-HMS. The results of the temporal transferability study showed that the combination of the distributional model and the Isohyetal Method had the best statistical performance with MAE of 64.62 m3/s, RMSE of 82.47 m3/s, and R2 and NSE of 0.9383 and 0.8547, respectively. It is considered that this study was properly analyzed because the peak flood volume occurrence time of the observed and simulated flows is within 1 hour. Therefore, the results of this study can be used for frequency analysis in the future, which can be used to improve the accuracy of simulating peak flood volume and peak flood occurrence time in mountainous watersheds with steep slopes.

Blood-Brain Barrier Disruption in Mild Traumatic Brain Injury Patients with Post-Concussion Syndrome: Evaluation with Region-Based Quantification of Dynamic Contrast-Enhanced MR Imaging Parameters Using Automatic Whole-Brain Segmentation

  • Objective: This study aimed to investigate the blood-brain barrier (BBB) disruption in mild traumatic brain injury (mTBI) patients with post-concussion syndrome (PCS) using dynamic contrast-enhanced (DCE) magnetic resonance (MR) imaging and automatic whole brain segmentation. Materials and Methods: Forty-two consecutive mTBI patients with PCS who had undergone post-traumatic MR imaging, including DCE MR imaging, between October 2016 and April 2018, and 29 controls with DCE MR imaging were included in this retrospective study. After performing three-dimensional T1-based brain segmentation with FreeSurfer software (Laboratory for Computational Neuroimaging), the mean Ktrans and vp from DCE MR imaging (derived using the Patlak model and extended Tofts and Kermode model) were analyzed in the bilateral cerebral/cerebellar cortex, bilateral cerebral/cerebellar white matter (WM), and brainstem. Ktrans values of the mTBI patients and controls were calculated using both models to identify the model that better reflected the increased permeability owing to mTBI (tendency toward higher Ktrans values in mTBI patients than in controls). The Mann-Whitney U test and Spearman rank correlation test were performed to compare the mean Ktrans and vp between the two groups and correlate Ktrans and vp with neuropsychological tests for mTBI patients. Results: Increased permeability owing to mTBI was observed in the Patlak model but not in the extended Tofts and Kermode model. In the Patlak model, the mean Ktrans in the bilateral cerebral cortex was significantly higher in mTBI patients than in controls (p = 0.042). The mean vp values in the bilateral cerebellar WM and brainstem were significantly lower in mTBI patients than in controls (p = 0.009 and p = 0.011, respectively). The mean Ktrans of the bilateral cerebral cortex was significantly higher in patients with atypical performance in the auditory continuous performance test (commission errors) than in average or good performers (p = 0.041). Conclusion: BBB disruption, as reflected by the increased Ktrans and decreased vp values from the Patlak model, was observed throughout the bilateral cerebral cortex, bilateral cerebellar WM, and brainstem in mTBI patients with PCS.

Effects of High-Molecular-Weight Glutenin Subunits and Agronomic Traits on Bread Wheat Quality Parameters (밀의 고분자 글루테닌 조성과 농업 형질이 품질 특성에 미치는 영향)

  • Improving flour quality is one of the major targets of wheat breeding programs. This study determined the optimum high-molecular-weight glutenin subunits (HMW-GS) to improve flour quality, and analyzed the correlation between agronomic and quality traits in Korea. A total of 180 wheat varieties, including 55 Korean and 125 foreign cultivars, carrying various Glu-1 alleles, were evaluated for their quality and agronomic traits. Results indicated that Glu-A1b, Glu-B1b, and Glu-D1f were the most prevailing alleles for each Glu-1 locus for Korean wheat cultivars. Korean wheat cultivars recorded shorter days to heading (DTH) and longer days to maturity (DTM) compared to foreign cultivars. In addition, an interaction effect was found between Glu-A1 and Glu-B1 alleles on several quality parameters. The combination of Glu-A1c and Glu-B1i showed a higher protein content, dry gluten content, and higher sodium dodecyl sulfate (SDS) sedimentation value than other Glu-A1×Glu-B1 combinations. Cultivars carrying Glu-A1a or Glu-A1b, Glu-B1i or Glu-B1al, and Glu-D1d for each Glu-1 locus exhibited a longer mixing time and stronger mixing tolerance. The DTM positively correlated with the protein content, gluten index and SDS sedimentation value. However, a negative correlation was observed between DTH and quality traits. Owing to the above results, this study suggests that an increase in the frequency of Glu-B1i or Glu-B1al, Glu-D1d coupled with a short DTH and long DTM could significantly improve wheat quality properties.

  • In order to implement a smart factory for manufacturing innovation, more digital twins will be developed and applied gradually. In particular, simulation and optimization of digital twins makes it possible to support critical decision-making like a predictive maintenance of the equipment for manufacturing. In terms of a user perspective, this study suggests the conceptual framework of Digital Twin Management System (DTMS) for supporting the analytical and managerial activities for Digital Twins. We integrate the methods and structure of the area like Manufacturing Engineering, Decision Support Systems, and Optimization for developing the DTMS. The framework suggested in this study shows a typical DSS which consists of dialog management system, model management system and data management system. It also includes Analytical Digital Twins and simulations & optimization module. The framework is being applied in one of the most competitive and complex industrial sector. Also this study is meaningful to suggest a new direction of research.

Medical Certificate as an Evidence of Personal Injury (진단서의 증명력: 상해진단서를 중심으로)

  • Medical certificate is a document to demonstrate a patient's health status, made up and signed by a physician, dentist, or oriental physician who attended the patient. It serves as an evidence in many official process including civil or criminal law suit, especially for one's personal injury. The Korean legal system also acknowledges and protects the evidentiary function of medical certificate by mandating physicians etc. to issue medical certificate in good faith and only when they personally attended the patient, and by criminally punishing them when they do not comply with these legal requirements. There are some reasons, however, that medical certificates often do not reflect the true health status of the patient: When physicians attend the patient and collect information regarding the health status of the patient, their priority is and should be the most cost-effective way to meet the health needs of the patient. It does not necessarily correspond to the accurate examination of the health status of the patient. Even when the patient's report on the history of the illness or the injury seems suspicious, physicians might have to avoid disproving it because that kind of attitude might harm the rapport between the physician and the patient. All these can distort the perception of the physicians and this distortion can be reproduced in the medical certificate they made up. Some of these problems might be resolved or at least enhanced by introducing new form of medical certificate which would guide physicians to reveal the nature, factual and theoretical grounds, and the limit of their findings more accurately. Others, however, would not be able to address, because it stems from the conflict between the physician's primary duty, duty to be loyal to the patient's life and health, and his secondary duty to serve as a public or neutral witness on the health status of the patient, and when both values or duties conflict with each other, they should choose the duty to the patient sacrificing the duty to the public or the court.

A Practical Research for Mode Efficient Utilization of Estuary Reserviors in the South-Western Part of Korea (우리나라 서남부지역 담수호의 효율적 이용방안)

  • The south-western part of Korea is situated in an unbalance of water supply and demand relating to the Keum, Mankyung, Dongjin and Youngsan River and their estuary reservoirs. For example, the Keum River estuary reservoir is discharging the larger amount of yearly runoff into the sea due to the small storage capacity, while Saemankeum estuary reservoir which is under construction, has the smaller runoff comparing with its strorage capacity. And the downstream area of the Youngsan River, such as Youngkwang, Youngam are deficient in water due to larger demand and smaller supply. In order to solve the above unbalanced water supply and demand and also to improve the water use effciency, the Hierarchical Operation Model for Multi-reservoir System(HOMMS) has been developed and applied to analyze the multi-reservoir operation assuming that the above reservoirs were linked each other. The result of this study shows that $2,148{\times}106\;\textrm{m}^3$ of annual additional water requirement for agricultural and rural water demands are required in this region at 2011 of target year, and these demands can be resolved by diverting and reusing $1,913{\times}106\;\textrm{m}^3$ of the released water from the estuary reservoirs into the sea.

Isolation and Characterization of a Novel Calcium/Calmodulin-Dependent Protein Kinase, AtCK, from Arabidopsis

  • Protein phosphorylation is one of the major mechanisms by which eukaryotic cells transduce extracellular signals into intracellular responses. Calcium/calmodulin ($Ca^{2+}/CaM$)-dependent protein phosphorylation has been implicated in various cellular processes, yet little is known about $Ca^{2+}/CaM$-dependent protein kinases (CaMKs) in plants. From an Arabidopsis expression library screen using a horseradish peroxidase-conjugated soybean calmodulin isoform (SCaM-1) as a probe, we isolated a full-length cDNA clone that encodes AtCK (Arabidopsis thaliana calcium/calmodulin-dependent protein kinase). The predicted structure of AtCK contains a serine/threonine protein kinase catalytic domain followed by a putative calmodulin-binding domain and a putative $Ca^{2+}$-binding domain. Recombinant AtCK was expressed in E. coli and bound to calmodulin in a $Ca^{2+}$-dependent manner. The ability of CaM to bind to AtCK was confirmed by gel mobility shift and competition assays. AtCK exhibited its highest levels of autophosphorylation in the presence of 3 mM $Mn^{2+}$. The phosphorylation of myelin basic protein (MBP) by AtCK was enhanced when AtCK was under the control of calcium-bound CaM, as previously observed for other $Ca^{2+}/CaM$-dependent protein kinases. In contrast to maize and tobacco CCaMKs (calcium and $Ca^{2+}/CaM$-dependent protein kinase), increasing the concentration of calmodulin to more than $3{\mu}M$ suppressed the phosphorylation activity of AtCK. Taken together our results indicate that AtCK is a novel Arabidopsis $Ca^{2+}/CaM$-dependent protein kinase which is presumably involved in CaM-mediated signaling.

Studies on the Differences of the Rice Tillering System and Yield Characteristics under the Different Cultivation Methods in Southern region of Korea I. Seedling Characteristics and Growth under the Different Nursing Methods and Time (남부지방에서 벼 재배형태별 분얼체계 및 수량구성형질의 차이에 관한 연구 I. 묘종류와 육묘시기에 따른 묘소질 및 본답 생육의 변화)

  • To study the effects of different nursing methods and transplanting on the growth of the rice plant(Oriza sativa L.hn southern region of Korea, Kumo-byeo, Palgong-byeo and Dong-jinbyeo were transplanted from April 20 to July 20 with 15 days intervals as 8 days seedling (infant rice), 25 days box seedling for machine transplanting and 45 days conventional seedling, respectively. No. of leaves at transplanting were in order of 45 days seedling>25 days seedling>8 days seedling. In 25 days seedling, they were increased as transplanting dates were later from April 20 to June 5, and in 45 days seedling, it showed same tendency until June 5 with Dongjinbyeo, June 20 with Palgong-byeo and Kumo-byeo. Root activity according to the $\alpha$-naphthylamine oxidation activity and rooting activity were higher in order of 8 days seedling>25 days seedling>45days seedling. Panicle formation initiated after maximum tillering stage as transplanting at April 20 with 8 days seedling and 25 days seedling, but it intiated before maximum tillering stage as transplanting at July 20 in Kumo-byeo. and in Palgong-byeo intiations of panicle formation were shown after maximum tillering stage as transplanting until May 20, but June 5 in Dongjin-byeo. However, there was no significant tendency with 45 days seedling. Missing hill ratio were less than 5% as transplanting with 8 days seedling and 25 days seedlingregardless of transplanting periods or varieties.

Genetic Diversity and Molecular Phylogenetic Relationships of the Genus Sarcocheilichthys Fish in Korea (한국산 중고기속(Sarcocheilichthys) 어류의 유전적 다양성과 분자계통학적 유연관계)

  • Using the cytb gene region of the mitochondrial DNA of eight populations of Sarcocheilichthys nigripinnis morii and five populations of S. variegatus wakiyae, which belong to the genus Sarcocheilichthys from Korea, the genetic diversity and molecular phylogenetic relationships of each population were examined. As a result of the analysis, it was confirmed that the S. variegatus wakiyae population had higher genetic diversity than the S. nigripinnis morii population. In the phylogenetic tree of genus Sarcocheilichthys fish in Korea based on the cytb gene, the Yeongsan River (YSR) population of S. variegatus wakiyae forms a clade with the Tamjin River (TJR), Yeongsan River (YSR), and Seomjin River (SJR) population of S. nigripinnis morii, and genetic relationships that do not align with the current classification system were observed. Meanwhile, on the nuclear DNA phylogenetic tree, S. variegatus wakiyae and S. nigripinnis morii could be clearly distinguished, showing mitonuclear inconsistency where mitochondrial and nuclear DNA conflicted on the phylogenetic tree. The Seomjin River (SJR) population of S. nigripinnis morii was translocated to the Dongjin River (DJR) population, haplotype from which crossbreeding was presumed to have occurred was confirmed. Among the rivers flowing into the East Sea, the S. nigripinnis morii population is known to have been introduced and inhabit only the Hyeongsan River (HSR), and it is presumed to be a population formed by translocation from the Han River (HR) population, with a haplotype representing a unique genetic group also confirmed. The Han River (HR), Geum River (GR), and Mangyeong River (MGR) populations of S. nigripinnis morii formed a genetically identical population with S. czerskii and S. soldatovi distributed north of the Yalu River, and accordingly, a taxonomic reexamination was required through morphological and molecular phylogenetic studies by securing various specimens.
