Extraction Conditions and Green Index of Green Pigments from Discolored Garlic (녹변된 마늘의 색소 추출 조건과 Green Index)

  • Extraction conditions, spectrometric, and sensory properties of pigment extracts from green discolored garlic (GDG) were studied for development of green index to evaluate degree of green discoloration of garlic. GDG pigments were extracted using polar solvents, such as methanol, ethanol, and water. Methanol with 1% HCl resulted in highest extraction of green pigments. Extraction of pigments was affected by temperature, and extracted pigments were partially destroyed at high temperatures. Optimum conditions for extraction of green pigments were $20^{\circ}C$ for 30 min. Because green pigments are unstable at room temperature, and no method has been reported to evaluate degree of green discoloration of garlic, color chart was designed by mixing varying amounts of control and GDG. High correlation was observed between sensory scores and "a" value of color chart samples (y=-3.465x - 11.676) with $R^2$ of 0.993. Green index (GI) was developed based on linear regression equation between sensory scores and "a" values. Sensory panel recognized green discoloration of garlic at $GI{\geq}3.0$. GI developed in this study could be utilized to evaluate degree of green discoloration of garlic during cold storage and distribution of garlic.

Effects of Water-Repellent on the Color of Water Paint (발수제의 혼입이 수성페인트의 색에 미치는 영향)

  • Generally surface finishing of building is done with water paint that performs basic water repellency and resisting. When painting the exterior of the building with water paint, application method is easy and inexpensive and it has the advantage of building maintenance. But surface finishing of water paint is easy to discolored or aging due to ultraviolet light so the effect is not satisfying than expected. Therefore this study is to analyze durability of water-repellent paints mixed with water-repellents as outer surface finishing materials, and evaluate its feasibility. Color change and color different due to the accelerated time were tested, and the feasibility was evaluated based on the test results. The experimental results of color different, brightness different and chromaticity suggested that water-repellent paints mixed with water-repellents were suitable for finishing materials. Considering overall general durability performances, stable mixing ratios were 2, 5, and 8%.

Multiple injection of permanganate and hypochlorite for manganese removal by oxidation combined with membrane filtration (산화-막여과에 의한 망간 제거 시 과망간산과 차아염소산 복합 주입의 효과)

  • The water containing soluble manganese may cause problems such as discolored water, unpleasant taste, fouling or scaling of pipes in water distribution system, and so on. Conventional water treatment processes using sand filtration or sedimentation after oxidation, however, cannot often meet manganese standard for drinking water. Two types of oxidants, potassium permanganate ($KMnO_4$) and sodium hypochlorite (NaOCl), were utilized at the same time for manganese oxidation, and then the precipitated manganese oxides were removed by low pressure membrane filtration in this study. In batch experiments, the multiple injection of both oxidants showed more effective manganese removal than did the single injection using either of them. Moreover, the deterioration of manganese removal at low temperature was less serious for the multiple injection than that for the single injection. Manganese removal by the continuous system of oxidation by multiple injection combined with membrane filtration was higher than those by batch experiments at the same oxidation conditions. In addition, less membrane fouling was observed for membrane filtration with oxidation during continuous membrane filtration than membrane filtration without oxidation. These results indicate that the oxidation by multiple injection coupled with membrane filtration was efficient and applicable to actual water treatment for manganese removal.

Occurrence of Damping-off Caused by Pythium spinosum on Cucumis melo in Korea

  • In 2010 and 2012, damping-off symptoms were found on melon seedlings grown in Yeongam and Suncheon, Korea. Water-soaked and discolored lesions appeared on the lower stems of the infected plants. The diseased plants became wilted and stunted, and eventually collapsed. On the basis of morphological characteristics and molecular analyses of internal transcribed spacer (ITS) rDNA and cytochrome oxidase II (cox2) sequences, the causal organism was identified as Pythium spinosum. The isolates were pathogenic to melon under pot conditions. To our knowledge, this is the first report of P. spinosum causing damping-off on melon in Korea.

  • The objective of this study was to find in what color Hanji(Korean hand-made paper) is dyed when it is dyed with a pigment extracted from clove tree using different kinds of mordant, and how the paper is discolored and variety of strength under the condition of accelerated aging test. The results of this experiment are as follows. The Hanji dyed with aluminum acetate mordant was colored yellowish brown at pH 4.82, discoloration after aging was as much as a slight difference of color was recognized, and the decrease of strength after aging test was small. Thus, the method using aluminum acetate mordant was usable in coloring Hanji. The second most useful Hanji dyeing moth of was using distilled water and ferrous sulfate mordant, which dyed Hanji light brown at pH 6.03. However, when pigment was extracted using distilled water and copper sulfate mordant was used, discoloration was satisfactory but strength decreased too much and pale brown was obtained. Thus, this method was not usable.

Changes of Component on Green Discolored Garlic (녹변된 마늘의 성분 변화)

  • Effects of temperature, discoloration, and conditioning on garlic compositions before and after storage were examined. Proximate composition, soluble solids, pH, organic acids, and total sugars of all samples were not significantly different. Sugar contents were sucrose > fructose > glucose, and included such fructooligosaccharide as 1-kestose, 1-nystose, and 1-F-fructosyl nystose. Allicin contents in control, green discolored, and conditioned garlics were 647,9, 613.8, and 648.0 mg%, respectively. Thirty-two flavor compounds were identified by GC/MSD, including 11 sulfide compounds, 5 acids, and 3 aldehydes. Contents of sulfur compounds, including disulfide(di-propenyl), 2-thiophene methanethiol, thiophene, 3,4 dihydro-3-vinyl-1,2-dithiin, and 2-vinyl-4H-1,3-dithin, and water-soluble pectins in low temperature-stored garlic were higher than those in control, and decreased by conditioning or inducing green discoloration. These results show sulfur compounds and water-soluble pectins accumulated in low temperature stored-garlic, and are then converted into green pigments by alliinase in disintegrated garlic tissues.

Effects of Desalinization Management on Rice Yield in Sea Water Flooded Field

  • Over 2,000 ha of rice fields in the western and southern coastal region of Korea were flooded with sea water during the spring tide, on August 19-21, 1997, and the rice plant at heading stage was injured. The field surveys were undertaken at the sea water flooded paddy fields in Chonbuk Province, to identify the injury symptoms and rice yield damage subjected to different flooding condition and desalinization methods. Five days after sea water flooding at heading stage, the flag leaves of rice plants flooded with 30 ㎝ deep sea water withered from the tip, the withering progressed to the lower leaves in deeper flooding. The spikelets were spotted black and discolored from the tip at 50 ㎝ deep flooded rice, and some panicles changed to white at 80 ㎝ deep flooded rice. Most of the rice leaves submerged completely for an hour were withered and most of panicles changed to white. The milled rice yield, percentage of ripened grain, and 1000 grain weight of flooded rice decreased with deeper flooding water, higher water salinity and longer flooding time. Even under the same flooding conditions, the damage of rice yield varied with the growth stage: heading stage>dough stage>booting stage. Rice yield damage was less in the fields on the upper riverside than those of the fields on the estuary and seaside, because of lower water salinity. In a flooded field, the rice yield damages were reduced as the distance increased from the levees where the sea water inflowed and increased as the distance increased from the fresh water irrigation gate. The desalinization treatments consisting of frequent exchange of irrigation water and spraying with fresh water soon after flooding effectively reduced the rice yield damage.

Fruit Rot of Peach (Prunus persica) Caused by Phytophthora cactorum (Phytophthora cactorum에 의한 복숭아 과일역병)

  • A severe brown rot on peach fruit caused by a Phytophthora sp. has occurred at peach orchards in Taegu of Korea from late June to early August in 1997. Infected fruits showed irregularly round or circular water soaking brown regions. In the severe case, fruits were entirely rotten and surface of the fruits were wrinkled. Occasionally, white mycelia and abundant sporangia were developed on the surface of fruit. Inner tissues of the fruits were also discolored to brown. The causal fungus was identified as Phytophthora cactorum based on following characteristics. Sporangia were ovoid, conspicuously papillate, caducous and measured as 28.4~48.1$\times$21.9~37.2 (av. 39.9$\times$30.4) ${\mu}{\textrm}{m}$. Sexuality of the fungus was homothallic. Oogonia were 25.0~34.0 (av. 29.9) ${\mu}{\textrm}{m}$ in size. Most antheridia were paragynous and measured av. 10.5$\times$13.0 ${\mu}{\textrm}{m}$. Optimum temperature for mycelia growth was around 25~3$0^{\circ}C$. However none of the isolates grew under 7$^{\circ}C$ and over 35$^{\circ}C$. The fungus revealed high pathogenicity to fruits, shoots and leaves of peach, apple and pear with different degrees. Phytophthora fruit rot of peach caused by Phytophthora cactorum has not been reported in Korea previously.

TREATMENT OF MINOCYCLINE STAINED NON-VITAL TOOTH (Minocycline에 의해 변색된 실활치의 치료)

  • A mixture of metronidazole, ciprofloxacin and minocycline(3Mix) in distilled water or in a root canal sealer were used to disinfect the infected endodontic and periapical lesions. It has been well acknowledged in recent literature that minocycline, a semisynthetic tetracycline derivative, causes discolorations in adult teeth and various other collagenous tissues. In these cases, pulp treatments were finished successfully using a combination of antibiotics but severe discolorations of the teeth were observed. Discolored non-vital posterior tooth was treated using the prosthodontic procedure. And anterior tooth was successfully treated using the walking bleach technique. Bleaching was performed by temporarily placing a mixture of sodium perborate and distilled water into the pulp chamber. There were significant esthetic improvement.

Change of Water Content and Disease Development on Pinus thunbergii Seedlings Inoculated with Bursaphelenchus xylophilus (소나무재선충 인공접종에 의한 해송묘목의 병 발달 및 수분함량 변화)

  • This study was carried out to provide basic information on physiological changes of 4-year-old seedlings of Pinus thunbergii inoculated with pine wood nematodes by analyzing changes of symptom development, stem and needle water content and nematode populations in stem. Twenty days after inoculation, needles were discolored as an external symptom, and xylem drying and pith browning occurred at the above and below of the inoculation sites as an internal symptom. However, xylem drying began to occur 10 days after inoculation, which was determined by the difference in drying status of xylem and cortex between control and inoculated seedlings. Although population of pine wood nematode increased from 5 to 10 days after inoculation, it has increased dramatically from 10 to 20 days after inoculation when both internal and external symptoms appeared. As the time passed by after inoculation with pine wood nematodes, water content of stem and relative water content in current needles and branch gradually decreased. As the number of nematodes increases, water content of stem and relative water content of current needles and branches decreased significantly. There was a positive relationship between the number of nematodes and xylem drying and/or disease development, but the number of nematodes rapidly decreased as seedlings become severely diseased.
