Protective Effects of the Calyx of Diospyros kaki Thunb. on H2O2-induced Oxidative Stress in C2C12 Myoblasts (C2C12 근육세포에서 과산화수소로 유도된 산화 스트레스에 대한 시체(柿蒂)의 보호효과)

  • Objectives : The calyx of Diospyros kaki Thunb. has usually been used to treat obstinate hiccup. This study investigated the protective effects of Diospyros kaki using C2C12 myoblasts of H2O2-induced oxidative stress. Methods : Cell viability and cytotoxicity were determined by MTT assay. The level of reactive oxygen species (ROS), malondialdehyde (MDA), and reduced glutathione (GSH) were measured by using optical properties. Results : The calyx of Diospyros kaki Thunb. extract showed no toxicity to C2C12 myoblasts until 20 ㎍/mL concentration and increased cell viability compared to the H2O2 treated group. The calyx of Diospyros kaki Thunb. extract inhibited the production of ROS and MDA at all concentrations. In addition, the calyx of Diospyros kaki Thunb. extract increased the concentration of GSH. Conclusion : This study provides that the calyx of Diospyros kaki Thunb. can be used as a potential material that exhibit antioxidative and protective effects on H2O2-induced oxidative stress in C2C12 Myoblasts.

  • To investigate the effect of media and growth regulators in micropropagation of persimmon (Diospyros kaki Thunb.), dormant axillary buds taken from trees of persimmon cultivars such as Ichikikeijiro, Tonawase and Hiratenenashi were used. Shoot tips were successfully cultured in full or half of nitrogen strength of MS medium. The most effective cytokinins for shoot proliferation and elongation of persimmon cv. Ichikikeijiro were 5 mg/L and 2 mg/L zeatin, respectively. Shoots were successfully rooted in 1/2N-MS medium with 1 mg/L IBA.

Preparing Bi-component Dye of Unripe Diospyros kaki THUNB. Fruit and Ecklonia cava and Investigating Its Dyeing Propeties on Fabric (풋감과 감태의 이성분 복합염료 제조와 섬유 염색성 고찰)

  • This paper proposes a bi-component dye, including the unripe fruit of Diospyros kaki THUNB and Ecklonia cava, to substitute for traditional persimmon dyeing because fabrics dyed with persimmon juice become stiffer and natural persimmon is insufficient for dyeing. This study examined the color difference and fabric stiffness depending on the ratio of Ecklonia cava for in a one-bath dye solution with Diospyros kaki THUNB and showed that 6% of Ecklonia cava in the bi-component dye was the optimum for decreasing the fabric stiffness. Based on these results, a bi-component dye constituting of 94% Diospyros kaki THUNB and 6% Ecklonia cava was prepared. The particle size was found to be smaller than both single dyes and it maintained a similar amount of Catechin to Diospyros kaki THUNB dye. Finally, cotton fabric dyed with a bi-component dye was much improved in terms of the fabric hand and the surface color was similar to that of the traditional persimmon-dyed fabric. These results could help to develop the natural persimmon dyeing industry.

Simultaneous Quantitative Analysis of Flavonoids Isolated from the Leaves of Diospyros kaki (감나무 잎으로부터 분리된 플라보노이드의 동시 정량분석)

  • The leaves of Diospyros kaki Thunb. were used to treat heart disease and hypotension in traditionally East Asia. The purpose of this study is to simultaneously quantitiative analyze the content of flavonoids in leaves of D. kaki. The isolated flavonoids from the leaves of D. kaki, and the structure of the isolated flavonoids were analyzed based on 1H and 13C NMR spectrum compared to literature data. Simultaneous quantitative analysis of the isolated flavonoids was validated using high performance liquid chromatography (HPLC). Results showed that the calibration curves of the flavonoid compounds were confirmed that they have a large linearity with a correlation coefficient (R2) of 0.99. In the intra-day and inter-day analysis, accuracy and precision of five compounds were measured that accuracy was 94.39 to 114.47% and precision was less than 3.00%. Content analysis showed hyperoside (1.30 ± 0.09%), astragalin (0.81 ± 0.06%), trifoline (1.58 ± 0.07%), quercetin (0.13 ± 0.02%) and kaempferol (1.33 ± 0.25%).

  • Asiacornococcus kaki (Kuwana) known as a serious pest on Diospyros kaki Thunb. has been reported erroneously as Eriococcus lagerstroemiae Kuwana. These two species were redescribed and illustrated for correct identification by the morphological characters of adult females with known host plants. Asiacornococcus Tang & Hao is a newly known genus from the Korean Peninsula.

Acaricidal Activity and Function of Mite Indicator Using Plumbagin and Its Derivatives Isolated from Diospyros kaki Thunb. Roots (Ebenaceae)

  • Acaricidal effects of materials derived from Diospyros kaki roots against Dermatophagoides farinae and D. pteronyssinus were assessed using impregnated fabric disk bioassay and compared with that of the commercial benzyl benzoate. The observed responses varied according to dosage and mite species. The $LD_{50}$ values of the chloroform extract of Diospyros kaki roots were 1.66 and $0.96{\mu}g/cm^2$ against D. farinae and D. pteronyssinus. The chloroform extract of Diospyros kaki roots was approximately 15.2 more toxic than benzyl benzoate against D. farinae, and 7.6 times more toxic against D. pteronyssinus. Purification of the biologically active constituent from D. kaki roots was done by using silica gel chromatography and high-performance liquid chromatography. The structure of the acaricidal component was analyzed by GC-MS, $^1H-NMR,\;^{13}C-NMR,\;^1H-^{13}C$ COSY-NMR, and DEPT-NMR spectra, and identified as plumbagin. The acaricidal activity of plumbagin and its derivatives (naphthazarin, dichlon, 2,3-dibromo-1,4-naphthoquinone, and 2-bromo-1,4-naphthoquinone) was examined. On the basis of $LD_{50}$ values, the most toxic compound against D. farinae was naphthazarin $(0.011{\mu}g/cm^2)$ followed by plumbagin $(0.019{\mu}g/cm^2),$ 2-bromo-1,4-naphthoquinone $(0.079{\mu}g/cm^2)$, dichlon $(0.422{\mu}g/cm^2)$, and benzyl benzoate $(9.14{\mu}g/cm^2)$. Additionally, the skin color of the dust mites was changed from colorless-transparent to dark brown-black by the treatment of plumbagin. Similar results have been exhibited in its derivatives (naphthazarin, dichlon, and 2-bromo-1,4-naphthoquinone). In contrast, little or no discoloration was observed for benzyl benzoate. From this point of view, plumbagin and its derivatives can be very useful for the potential control agents, lead compounds, and indicator of house dust mites.

Comparison of Radical Scavenging and Immunomodulatory Activities Exhibited by an Aqueous Extract of Diospyros kaki Thunb. Fruit (Persimmon) (청도반시 추출물에 의한 라디칼소거 활성과 면역조절 활성의 상호 비교)

  • To assess the relationship between anti-oxidant and immunomodulatory activities of Diospyros kaki Thunb. fruit, we harvested persimmons on the first Friday of every month from July to October, and conducted a DPPH assay, a T-bet promoter assay, and an ELISA for IL-4 determination. Anti-oxidant activity increased as fruit weight rose. When the fruit was extracted with five different solvents, and the extracts examined for radical scavenging activity, such activity exhibited a pattern similar to that seen when anti-oxidant activity was assessed. T-bet promoter activity decreased on ripening, whereas IL-4 expression increased, as tested in a DNFB-induced animal model. Collectively, the results suggest that inflammation-inhibitory activity, valuable in treatment of some immune diseases, rises as persimmons ripen.

Study on the Anti-allergic Effect using Immature Persimmon (Diospyros kaki Thunb. Sangju-Dungsi) Ethanol Extract (미성숙 감(Diospyros kaki Thunb. Sangju-Dungsi) 에탄올 추출물의 항알레르기 생리활성 연구)

  • This study was conducted to verify the availability of immature persimmon ethanol extract (IPEE) as a natural and functional ingredient in protecting inflammation and allergy of skin based on the mechanism. The major content analysis, antioxidant activities, anti-allergic activity, anti-inflammatory effect, and safety related to irritation of IPEE were evaluated. The gallic acid content per 10 mg/mL of IPEE was 0.522% (5.22 mg/g). The total polyphenol and flavonoid contents were 428.3 mg/g and 31.1 mg/g, respectively. In ABTS+ activity, DPPH ability and SOD-like activity, it showed a concentration-dependent increase, which indicated IPEE has excellent antioxidant activities. As for the anti-allergy test in RBL-2H3 cells, the IPEE showed a decrease in β-hexosaminidase secretion as the concentration increases, and IPEE tended to decrease IL-4 secretion in all RBL-2H3 cells compared to the IgE + HSA group. IPEE showed good anti-inflammatory effect in RAW 264.7 cells by decrease of NO production and inflammation cytokines (TNF-α and IL-6). Also IPEE showed non-irritant in BCOP assay. By the results of this study, the IPEE containing high tannins, had good antioxidant, anti-allergic, and anti-inflammatory effects, which indicated that the immature persimmon is considered to be a useful for the development of related functional ingredients.

Neuroprotective effects of astringency-removed peel extracts of Diospyros kaki Thunb. cv. Cheongdo-Bansi on oxidatively-stressed PC-12 cells (청도반시(Diospyros kaki Thunb. cv. Cheongdo-Bansi) 탈삽 껍질 추출물의 산화스트레스로부터 PC-12 신경세포 보호 효과)

  • Astringent persimmon (Diospyros kaki Thunb. cv. Cheongdo-Bansi) peel with the astringency removed, which is a by-product of dried persimmon (gotgam), was investigated for its antioxidant and neuroprotective properties. A mixture of peel and 40% (v/v) aqueous ethanol was subjected to ultrasonication and then thermal and nonthermal treatments, to produce thermally-treated and nonthermally-treated persimmon peel extracts (TPE and NTPE, respectively). The total phenolic and flavonoid contents and the antioxidant capacity of TPE was approximately 1.3-1.8 times higher than those of NTPE. TPE resulted in the increased viability of neuronal PC-12 cells compared with NTPE. Furthermore, intracellular oxidative stress in PC-12 cells was more decreased by treatment with TPE than NTPE. Cholinesterases, such as acetylcholinesterase and butyrylcholinesterase, were more inhibited by treatment with TPE than NTPE. These results suggest that TPE is useful as a functional material to decrease oxidative stress in neuronal cells and to inhibit cholinesterases.

Evaluation of Genetic Diversity among Persimmon Cultivars (Diospyros kaki Thunb.) Using Microsatellite Markers (초위성 마커를 이용한 감(Diospyros kaki Thunb.)의 유연관계 분석)

  • The genetic diversity among 48 persimmon (Diospyros kaki Thunb.) accessions, indigenous in Korea and introduced from Japan and China, was evaluated by using simple sequence repeat (SSR) markers. From 20 SSR primer sets, a total of 114 polymorphic markers were detected among 12 pollination-constant non-astringent (PCNA), 13 pollination-variant non-astringent (PVNA), 15 pollination-variant astringent (PVA), and 8 pollination-constant astringent (PCA) cultivars. Analysis of pair-wise genetic similarity coefficient (Nei-Li) and unweighted pair-group method with arithmetic averaging (UPGMA) clustering revealed two main clusters and four subclusters for cluster I. The subclustering pattern was in accordance with the classification of persimmon cultivars based on the nature of astringency loss. Phenetic relationships among the subclusters showed a closer relatedness of the PCNA group with the PVNA group, and the PVA with the PCA group. Genetic similarity co-efficiency was 0.499 on average and the highest (0.954) similarity was observed between 'Cheongdo-Bansi' and 'Haman-Bansi'. The similarity was lowest (0.192) between 'Damopan'and 'Atago'. Identification of each cultivar with the execption of 'Cheongdo-Bansi' and 'Gyeongsan-Bansi' was possible based on the SSR fingerprints, suggesting that these SSR markers are a useful tool for protecting intellectual property on newly developed cultivars.
