Performance Responses, Nutrient Digestibility, Blood Characteristics, and Measures of Gastrointestinal Health in Weanling Pigs Fed Protease Enzyme

  • Although exogenous protease enzymes have been used in poultry diets quite extensively, this has not been the case for pig diets. In general, due to their better gut fermentative capacity and longer transit time, pigs have greater capacity to digest dietary proteins than poultry. However, in early-weaned piglets, the stress brought about by weaning adversely affects the digestion of dietary proteins. Therefore, a study was conducted to determine the effects of a commercial protease enzyme in weanling pigs. Indices of growth, nutrient digestibility, blood profiles, fecal microflora, fecal gas emission and fecal scores were measured during the study. A total of 50 weanling pigs ($6.42{\pm}0.12kg$) at 28 d of age were randomly assigned to receive 1 of 2 dietary treatments: i) control diet (corn-soy based) with no supplemental protease (CON), and ii) control diet+200 g/ton protease (PROT) for 42 d. A completely randomized design consisting of 2 treatments, 5 replicates, and 5 pigs in each replicate was used. Growth performance in terms of body weight ($27.04{\pm}0.38kg$ vs $25.75{\pm}0.39kg$; p<0.05) and average daily gain ($491{\pm}7.40g$ vs $460{\pm}7.46g$; p<0.05) in PROT fed pigs were increased significantly, but gain per feed ($0.700{\pm}0.01$ vs $0.678{\pm}0.01$; p>0.05) was similar between treatments at d 42. Relative to CON pigs, PROT fed pigs had increased (p<0.05) apparent total tract digestibility ($84.66%{\pm}0.65%$ vs $81.21%{\pm}1.13%$ dry matter and $84.02%{\pm}0.52%$ vs $80.47%{\pm}1.22%$ nitrogen) and decreased (p<0.05) $NH_3$ emission ($2.0{\pm}0.16ppm$ vs $1.2{\pm}0.12ppm$) in the feces at d 42. Except for a decreased (p<0.05) in blood creatinine level, no differences were observed in red blood cell, white blood cell, lymphocyte, urea nitrogen, and IgG concentrations between treatments. Fecal score and fecal microflora (Lactobacillus and E. coli) were also similar between CON and PROT groups. Overall, the supplementation of protease enzyme in weanling pigs resulted in improved growth rate and nutrient digestibility. Exogenous protease enzyme reduced fecal $NH_3$ emission, thus, potentially serving as a tool in lowering noxious gas contribution of livestock production in the environment.

Change of Solubilization Characteristics of Rice Straw by Different Pre-treatments (전처리 방법에 따른 볏짚의 가용화 특성 변화)

  • In order to find a feasibility of applying an agricultural biomass to the anaerobic digestion system, the effects of pre-treatment of rice straw was investigated by different sizes,temperatures, ultrasonic frequencies, and with/without NaOH treatment on the change of soluble organic matter, which is generated most dominantly in Korea. It was observed that SCOD(soluble chemical oxygen demand) as the index for the decomposition of rice straw increased with the smaller particle size, higher reaction temperature and alkali treatment. With treatment of 5% NaOH, it was shown that the highest concentrations of SCOD were observed at 9,000 mg $L^{-1}$ and 6,000mg $L^{-1}$ at $35^{\circ}C$ and $55^{\circ}C$, respectively. Agitating with ultrasonic irradiation could be enhanced SCOD of rice straw less than 3 cm with 40 kHz of ultrasound. Optimal RPM in this study was at 150 rpm regardless of reaction temperatures.

Extraction, purification and properties of anti-complementary polysaccharide from Arecae Pericarpium (대복피로부터 항보체 활성다당의 추출, 정제 및 그 특성)

  • To examine the characteristics of anti-complementary compounds from Arecae Pericarpium (the pericarps of Areca catechu) which showed the highest activity during our screening procedures, the extraction and purification were performed. AC-1 fraction from Arecae Pericarpium was purified by hot water extraction, methanol reflux, ethanol precipitation, dialysis and lyophilization. This compound had total sugar 48.2%, uronic acid 14.6% and protein 36.8%. Rhamnose, arabinose, mannose and galactose were found in sugar components. By cetavlon (cetyltrimethylammonium bromide) treatment AC-1 was fractionated to AC-2, AC-3 and AC-4. Among them, AC-2 showed the highest activity and yield. By periodate oxidation, AC-2 was deactivated, but had no change in activity by pronase digestion. Moreover active fractions, AC-2-IIIa and AC-2-IIIc isolated from AC-2 by two successive column chromatography using DEAE-Toyopearl $650C(Cl^-form)$ and Sephadex G-100. AC-2-IIIa was mainly made up of rhamnose, mannose, galactose and glucose, and AC-2-IIIc, mannose, galactose and glucose. These both polysaccharides were identified as homogeneous by gel filtration of Sepharose CL-4B and electrophoresis, and molecular weights of them were 120,000 and 15,000, respectively.

Characteristics of Greenhouse Gas Emissions with Different Combination Rates of Activated Rice Hull Biochar during Aerobic Digestion of Cow Manure (왕겨 활성 바이오차 혼합 비율에 따른 우분 호기소화 시 온실가스 발생 특성)

  • BACKGROUND: Among the biomass conversion techniques of livestock manure, composting process is a method of decomposing organic matter through microorganisms, and converting it into fertilizer in soil. The aerobic composting process is capable of treating cow manure in large quantities, and produces greenhouse gas as CO2 and N2O, although it has economical benefit. By using the activated rice hull biochar, which is a porous material, it was intended to mitigate the greenhouse gas emissions, and to produce the compost of which quality was high. Objective of this experiment was to estimate CO2 and N2O emissions through composting process of cow manure with different cooperated biochar contents. METHODS AND RESULTS: The treatments of activated rice hull biochar were set at 0%, 5%, 10% and 15%, respectively, during composting cow manure. The CO2 emission in the control was 534.7 L kg-1, but was 385.5 L kg-1 at 15% activated rice hull biochar. Reduction efficiency of CO2 emission was estimated to be 28%. N2O emission was 0.28 L kg-1 in the control, but was 0.03 L min-1 at 15% of activated rice hull biochar, estimating about 89% reduction efficiency. CONCLUSION: Greenhouse gas emissions during the composting process of cow manure can be reduced by mixing with 15% of activated rice hull biochar for eco-friendly compost production.

Study on Centrocestus armatus in Korea II. Recovery rate, growth and deve]opment of worms in albino rats (Centrocestus armatus에 관한 연구 II. 흰쥐에서의 성장 발육 및 충체회수율)

  • Some biological characteristics of Centrecestus armatui were studied using albino rats as its experimental host. The metacercariae were collected from Zacco Platypus by artificial digestion method. Laboratory rats(Wistar) were fed each 100 or 200 metacercariae and sacrificed on 1, 2, 3, 4, 5, 6, 8, 14 and 28 days after infection to recover worms of various ages. The average recovery rate was 10.7% from 82 rats. The rate decreased rather slowly for the first 8 days but showed a steep decrease thereafter. Of the worms, 35.5% were recovered from the duodenum and 62.5% from the jejunum. At metacercarial stage, body length was $293{\mu\textrm{m}}$ and body width $144{\mu\textrm{m}}$. At adult stage, the length and width reached $382{\mu\textrm{m}}$ and $214{\mu\textrm{m}}$ respectively at 14 days after infection. The testes and Mehlis' gland were recognized at metacercarial stage, whereas the ovarian anlage appeared on the 1st day of infection, seminal vesicle and vitellaria on the 2nd day, and seminal receptacle and uterine eggs on the 3rd day. Until 8 days after infection the genital organs developed continuously and the number of uterine eggs increased. The above results show that albino rats are one of useful experimental hosts for C. armatus and the worms can develop to adults in 3 days after infection.

Effects of Molasses Addition and(or) Pelleting on Nutritional Characteristics of Broiler Litter Processed by Ensiling or Deepstacking and Palatability Improvement by 'Hanwoo' During the Adjustment Period (육계분 혐기 또는 퇴적 발효 사료 제조 시 당밀 첨가 및 펠렛화가 사료영양적 가치 및 사료 적응기의 한우 기호성 개선에 미치는 영향)

  • This study was conducted in order to determine a proper level of molasses addition through the analysis of changes in appearance, nutritive and silage parameters before and after ensiling or deepstacking of broiler litter, to evaluate the effect of pelleting processed broiler litter and to develop methods to enhance palatability of broiler litter and reduce the adjustment period by ‘Hanwoo’ steers. Molasses addition was effective in ensiling and deepstacking of broiler litter and the proper addition level was about 5%. Changes in nutritive values of broiler litter by ensiling and deepstacking with or without molasses treatment were not great. Adding 5% molasses at deepstacking of broiler litter did not affect(P<0.05) in vitro digestion of dry matter and organic matter. Pelleting of broiler litter resulted in significant(P<0.05) moisture evaporation, organic matter reduction and nearly threefold increase of bulk density. Pelleting or molasses addition of broiler litter improved palatability by ‘Hanwoo’ steers and reduced the adjustment period by half(8-9 d).

Screening and characteristics of anti-complementary polysaccharides from Chinese medicinal herbs (한약재로부터 항보체 활성 다당의 검색 및 특성)

  • We conducted screening on Chinese medicinal herbs to examine their anti-complementary activity by hemolytic complementary assay $(TCH_{50})$. Among 55 kinds of herbs, several herbs showed relatively potent anti-complementary activity which decreased $TCH_{50}$, more than 70% in comparison with control. Then, hot water extracts of the following herbs, Curcuma aromatica, Areca catechu, Gleditsiae spina, Euonymus alata, Acanthopanax senticous. Lonicera japonica, Aconitum carmichaeli, Curcuma zedoaria and Cinnamoum cassia, which were shown relatively potent anti-complementary activity were partially purified and analyzed their chemical properties. These activities were resistant to digestion with pronase but decreased by treatment with $NaIO_4$. These results may indicate that the complement activating ability in their herbs is due to polysaccharide. Furthermore, the anti-complementary activity of Areca catechu which was showed the most potent activity, was reduced partially in the absence of the $Ca^{++}\;ion$. After incubation of the normal human serum with partially purified polysaccharide of A. catechu in the absence of $Ca^{++}\;ion$, a cleavage of C3 in the serum was found to have occurred through immunoelectrophoresis using rabbit anti-human C3 serum. These results indicate that the mode of complement activation by polysaccharide of A. catechu is via both the alternative and classical pathway.

Determination of polycyclic aromatic hydrocarbons in processed foods (가공식품 중 다환방향족탄화수소 분석)

  • In this study, the following concentrations of some PAHs (Polycyclic Aromatic Hydrocarbons) were investigated; [benzo(a)anthracene, chrysene, benzo(b)fluoranthene, benzo(k)fluoranthene, benzo(a)pyrene, dibenzo(a,h)anthracene, benzo(g,h,i)perylene, indeno(1,2,3-c,d)pyrene] in processed foods (n=165) and cooked meats (n=45) and established the analytical method by characteristics of processed foods. The methodology involved extraction (alkali digestion, liquid-liquid extraction, microwave extraction), clean-up on Sep-Pak Florisil Cartridges and determination by HPLC/FLD (High Performance Liquid Chromatography/Fluorescence Detector). The recovery of overall method for 8 PAHs spiked into these products ranged from 92 to 103%. The mean level of detected foods was found to be benzo(b)fluoranthene $0.9\;{\mu}g/kg$ in smoked salmon, benzo(b)fluoranthene $1.0\;{\mu}g/kg$, benzo(k)fluoranthene $0.3\;{\mu}g/kg$, benzo(a)pyrene $0.9\;{\mu}g/kg$ in dried banana, benzo(b)fluoranthene $0.2\;{\mu}g/kg$, benzo(k)fluoranthene $0.1\;{\mu}g/kg$, benzo(a)pyrene $0.2\;{\mu}g/kg$ in smoked chicken, benzo(b)fluoranthene $1.3\;{\mu}g/kg$, benzo(k)fluoranthene $0.3\;{\mu}g/kg$, benzo(a)pyrene $0.9\;{\mu}g/kg$ in charcoal grilled pork, respectively.

Quality differences of retorted Samgyetangs as affected by F0-value levels (레토르트 삼계탕의 F0값 수준에 따른 품질 차이)

  • This study was conducted to determine the $F_0$-values of a retort machine at different locations and to evaluate the effects of these $F_0$-values on various quality characteristics of retorted Samgyetang samples. Samples were divided into three groups based on $F_0$-values-T1, 10~20; T2, 20~30; T3, >30. Mineral content in Samgyetang broth and breast meat mostly increased with increasing $F_0$-values. In general, the free amino acid values, hardness, and springiness, except for bone springiness, of Samgyetang decreased significantly at higher $F_0$-values. Protein content of meat and broth of the treated samples were significantly lower than that of the control. An increase in the digestion rate of meat and porridge, as well as the turbidity of the broth was observed in most of the treated samples with increasing $F_0$-values. With increasing $F_0$-values, the $L^*$ and $b^*$ values of meat and the $b^*$ values of broth tended to increase, while the $a^*$ value of broth increased significantly. Electronic nose analysis revealed different flavor patterns for samples treated at different $F_0$-values. For sensory traits, samples treated with higher $F_0$-values tended to receive lower evaluations. Particularly, the color and texture of T3 samples were lower than those of T1 and T2 samples. In conclusion, to improve the quality of Samgyetang, the efficiency and optimization of retort machines as well as the standardization of sterilization techniques are needed.

A Study on Characteristics of Solubilization and Biogas Production for Sewage Sludge using Thermal Pretreatment (열적가용화공정을 이용한 하수슬러지의 가용화 특성 및 바이오가스 생산량 평가)

  • In this study, an applicability of thermal pretreatment for primary and secondary sludge, which are generated in a sewage treatment plant, was evaluated. The efficiency and charateristics was investigated with each sludge after pretreatment under the condition of $100{\sim}220^{\circ}C$ for 30 minutes. As the result, it was found that $SCOD_{Cr}$, $NH_4{^+}$, VFAs concentrations increased as the pre-treatment temperature increased. For COD solubilization, it was also highly dependent on an increase of temperature resulting in acceleration on hydrolysis and acid fermentation. In the BMP (Biochemical Methane Potential) experiment, for the primary sludge, it showed the higher biogas production rate at a temperature of $220^{\circ}C$, however, the effect was insignificant (5.6%). Whereas, for the secondary sludge, the increase on biogass production rate was 38.8% ($180^{\circ}C$) and this means that the secondary sludge is more suitable for an applicability of thermal pretreatment.
