Differential Expression of Nuclear Receptors in T Helper Cells

  • Steroid hormones have long been known to have a profound influence on the immune system. Although the functions of the nuclear receptors in the development of T cells are fairly well studied, the differential expression of these receptors in T helper cells is poorly understood. Here, we investigated the differential expression of nuclear receptors and coregulators in Th1 and Th2 cells by genome-wide micro array analysis. The result showed that several nuclear receptors and coregulators are differentially expressed in these cells. The result was confirmed by RT-PCR. The result showed that $RXR{\alpha}$ is highly expressed in Th2 cells. Overexpression of $RXR{\alpha}$ in a Jurkat human T cell line induced IL4 but not IFN-${\gamma}$ gene expression, suggesting that $RXR{\alpha}$ plays a selective role in Th1 and Th2 differentiation. In summary, these results suggest that Th1/Th2 differentiation is influenced by differential regulation of nuclear receptors and coregulators.

  • Objective: The purpose of this research was to examine the moderating effect of children's negative emotionality on the relationship between maternal attitude toward children's emotional expression based on the differential susceptibility model. Methods: Participants were 216 preschool children including 116 boys and 100 girls from day care centers and preschools located in Chungchung province and Seoul. EAS was used to measure the children's negative emotionality. The children's empathy quotient was used to measure empathy. Mothers reported their attitude toward children's emotional expression. Results: The results revealed that the association between empathy and maternal suppression of children's emotional expression was significant only for the children with high levels of negative emotionality. Moreover, the magnitude of association between empathy and maternal acceptance of children's emotional expression was greater for high levels of negative emotionality. Conclusion/Implications: It was concluded that these findings supported the differential susceptibility model.

  • We propose principal differential analysis based classification methods. Computations of squared multiple correlation function (RSQ) and principal differential analysis (PDA) scores are reviewed; in addition, we combine principal differential analysis results with the logistic regression for binary classification. In the numerical study, we compare the principal differential analysis based classification methods with functional principal component analysis based classification. Various scenarios are considered in a simulation study, and principal differential analysis based classification methods classify the functional data well. Gene expression data is considered for real data analysis. We observe that the PDA score based method also performs well.

통합적 이해의 관점에서 본 고등학교 학생들의 미분계수 개념 이해 분석 (An Analysis on the Understanding of High School Students about the Concept of a Differential Coefficient Based on Integrated Understanding)

  • 본 연구의 목적은 고등학교 상위권 학생들이 미분계수 개념을 통합적으로 이해하고 있는지를 알아보는데 있다. 여기서 미분계수 개념의 통합적 이해란 미분계수의 발생맥락인 접선문제와 속도문제를 미분계수 개념과 연결하여 이해하고, 미분계수 개념, 미분계수의 대수적 기하적 표현, 미분계수를 다루는 응용 상황을 서로 유기적으로 연결하여 이해하는 것을 의미한다. 본 연구를 위하여 청주시에 소재한 S고등학교 2학년 상위권 학생 38명을 연구대상으로 선정하여 미분계수 개념의 통합적 이해 정도를 조사하였다. 통합적 이해의 관점에서 고등학교 수학II 교과서와 여러 책을 참고하여 검사지를 개발한 후 현장 교사들과 전문가의 검토를 받아 수정 보완하였다. 검사지는 총 11개의 문항으로 구성되었으며 문항 1과 2-(1)은 미분계수 개념과 대수 기하 표현의 연결을, 문항 2-(2)와 4는 미분계수 개념의 발생맥락과 미분계수 개념의 연결을, 문항3과 10은 미분계수의 대수 표현과 기하 표현의 연결을 볼 수 있도록 하였다. 문항 5~9는 미분계수의 응용상황들로 구성되었는데 문항 6은 미분계수 개념과 응용의 연결을, 문항 8은 미분계수의 응용과 대수 표현의 연결을, 문항 5와 7은 미분계수의 수학 외에서의 응용과 기하 표현의 연결을, 문항 9는 수학 내에서의 응용과 기하 표현의 연결을 볼 수 있도록 하였다. 연구 결과 미분계수의 개념과 대수 기하 표현의 연결이 잘 이루어진 학생들의 비율은 높게 나타났으나 그 외의 연결이 잘 이루어진 학생들의 비율은 거의 절반이거나 절반에 미치지 못하는 것으로 나타났다.

Relationship between Differential Expression of Estrogen Receptor and Follicle Stimulating Hormone Receptor Genes in Ovary and Heterosis of Egg Number Traits in Chickens

  • In order to understand the molecular mechanism of heterosis of reproduction traits in chickens, we used the quantitative real-time reverse transcriptional polymerase chain reaction (Quantitative real-time RT-PCR) technique to investigate the differential expression of estrogen receptor (ESR) and follicle stimulating hormone receptor (FSHR) genes in 32-week-old ovaries of inbred chickens and their hybrid offspring in $4{\times}4$ diallel crosses, which involved White Plymouth Rock (E), CAU Brown (D), Silkies (C) and White Leghorn (A). We found that there were significant differences in mRNA expression of ESR and FSHR genes not only between hybrids and their parental lines (p<0.01), but also among different crosses (p<0.01). Furthermore, positive correlations between differential expression of both ESR and FHSR in hybrids and heterosis percentages of 32-week-old and 42-week-old egg number traits were significant at p<0.05. Our results suggested that differential expression of ESR and FSHR genes in the ovaries of inbred chickens and their hybrids could play roles in the formation of heterosis of egg number traits to some extent.

Differential Expression of Taste Receptors in Tongue Papillae of DBA Mouse

  • The tongue has 4 kinds of papillae, which are filiform, fungiform (FU), foliate (FO) and circumvallate papilla (CV). Tongue papillae except filiform papilla include taste buds. The papillae differ in taste sensitivities, likely due to differential expression of taste receptors. In this study, we evaluated differences in the expression levels of taste receptors in FU, FO and CV. Male DBA2 mice, 42-60 days old, were used in the study. Messenger RNAs were extracted from the murine epithelial tissues including FU, FO and CV. Cloned DNAs were synthesized by reverse transcription. Quantitative PCRs (qPCRs) were performed to determine mRNA expression levels of taste receptors. Results of qPCR revealed that the relative expression levels and patterns were different among FU, FO and CV. All three type 1 taste receptors were expressed FU, FO and CV at varying relative expression levels. All 35 kinds of type 2 taste receptors showed higher expression in FO and CV than in FU. Tas2r108 and Tas2r137 showed the two highest expression levels in all tested papillae. The differential expression levels and patterns of taste receptors among the three papillae could contribute to the different physiological sensitivities by tongue areas. Additional studies such as in situ hybridization or taste receptor cell activity recording is necessary to elucidate the functional relationship between expression levels of taste receptors and taste sensitivity.

Differential Effects of Nongenotoxic and Genotoxic Carcinogen on Cell Proliferation and c-Jun Expression in the Rat Liver Initiated with Diethylnitrosamine

  • Cell proliferation and c-Jun expression pattern in liver exposed by nongenotoxic carcinogens phenobarbital (PB) and clofibrate, and genotoxic carcinogen 2-amino-3-methylimidazo [4,5-f] quinoline (IQ) were investigated to see whether differential effects of genotoxic and non-genotoxic carcinogens on the development of neoplastic foci may be related to differential effect on cell proliferation. Male F344 rats were initially given a single intraperitioneal injection of diethylnitrosamine (200 mg/kg body weight), and 2 weeks later, animals were fed diets containing 0.03% IQ or 0.5% CE or 0.05% PB or basal diet as a control for 6 weeks. All rats were subjected to the two-thirds partial hepatectomy (PH) at week 3. Sequential sacrifice of rats was performed until 8 weeks. Cell proliferation was examined by immunohistochemical staining of bromodeoxyuridine and c-Jun expression was determined by northern blotting. The increase of cell proliferation rate after PH was significant in the rats fed 0.05% IQ and continued until 8 weeks, while the increase was not significant in the rats fed phenobarbital and clofibrate compared to that in the rats fed control diet. mRNA level of c-Jun in the liver treated with IQ was about 7 fold higher than that of control and peak at 5 hours after rH. In the liver treated with CE, mRNA level of c-Jun was 3-4 fold higher than that of control and the highest level of mRNA of c-Jun was seen at 24 hours after PH. These results show that differential effects of genotoxic and non-genotoxic carcinogens on the development of neoplastic foci may be related to differential effect on cell proliferation pattern.

DIFFERENTIAL EXPRESSION OF RADIATION RESPONSE GENES IN SPLEEN, LUNG, AND LIVER OF RATS FOLLOWING ACUTE OR CHRONIC RADIATION EXPOSURE

  • We analyzed the differential effects of histopathology, apoptosis and expression of radiation response genes after chronic low dose rate (LDR) and acute high dose rate (HDR) radiation exposure in spleen, lung and liver of rats. Female 6-week-old Sprague-Dawley rats were used. For chronic low-dose whole body irradiation, rats were maintained for 14 days in a $^{60}Co$ gamma ray irradiated room and received a cumulative dose of 2 Gy or 5 Gy. Rats in the acute whole body exposure group were exposed to an equal dose of radiation delivered as a single pulse ($^{137}Cs$-gamma). At 24 hours after exposure, spleen, lung and liver tissues were extracted for histopathologic examination, western blotting and RT-PCR analysis. 1. The spleen showed the most dramatic differential response to acute and chronic exposure, with the induction of substantial tissue damage by HDR but not by LDR radiation. Effects of LDR radiation on the lung were only apparent at the higher dose (5 Gy), but not at lower dose (2 Gy). In the liver, HDR and LDR exposure induced a similar damage response at both doses. RT-PCR analysis identified cyclin G1 as a LDR-responsive gene in the spleen of rats exposed to 2 Gy and 5 Gy gamma radiation and in the lung of animals irradiated with 5 Gy. 2. The effects of LDR radiation differed among lung, liver, and spleen tissues. The spleen showed the greatest differential effect between HDR and LDR. The response to LDR radiation may involve expression of cyclin G1.

Involvement of Cytochrome c Oxidase Subunit I Gene during Neuronal Differentiation of PC12 Cells

  • It is becoming increasingly evident that significant changes in gene expression occur during the course of neuronal differentiation. Thus, it should be possible to gain information about the biochemical events by identifying differentially expressed genes in neuronal differentiation The PC12 cell line is a useful model system to investigate the molecular mechanism underlying neuronal differentiation and has been used extensively for the study of the molecular events that underlie the biological actions of nerve growth factor (NGF). In this study, we report an application of the recently described mRNA differential display method to analyze differential gene expression during neuronal differentiation. Using this technique, we have identified several cDNA tags expressed differentially during neuronal differentiation. Interestingly, one of these clones was cytochrome c oxidase subunit I (COX I) gene. The differential expression of COX I gene was confirmed by Northern blot analysis as well as RT-PCR. Southern blot analysis of the genomic DNA of PC12 cells revealed that COX I is a single gene. Induction of the oxidative enzyme might reflect the energy requirement in neuronal differentiation.

Identification of Genes Induced by Low Temperature in Rice

  • Exposure of seedling of rice (Oriza sativa cv.Dongin) to cold stress ($6^{circ}C$, 7day) induced differential gene expression. Differentially expressed polyadenylated RNA induced by low temperature were isolated and identified from the leaves of rice (Oriza sativa cv.Dongin) seedling by using the technique, differential display of reverse transcription through polymerase chain reaction (DDRT-PCR). Four bands of cDNAs were differentially displayed on the PAGE gel through DDRT-PCR, and among them three bands were those of overexpressed genes while one band was of an underexpressed gene One of the overexpressed cDNA was characterized. The size of the DDRT-PCR product was found to be about 200 bp. The sequence of the cloned DNA was compared with those of GenBank through a BLAST E-Mail server, and it was found to have no homologies in the nucleotide sequence with that of any known DNA: therefore, it was designated as RC101 The expression of the cold-stress induced-gene, RC101, was sustained with Northern Blot analysis by using the cloned DDRT-PCR product as a probe.
