Effects of feeding corn naturally contaminated with aflatoxin on growth performance, apparent ileal digestibility, serum hormones levels and gene expression of Na+, K+-ATPase in ducklings

  • Objective: A 14-d trial was conducted to determine the effects of feeding corn naturally contaminated with aflatoxin $B_1$ ($AFB_1$) on growth performance, apparent ileal digestibility, serum hormones levels and gene expression of $Na^+$, $K^+-ATPase$ in ducklings. Methods: A total of 704 ducklings were blocked on the basis of sex and body weight (BW), and then allocated randomly to one of the following two treatments: i) CON, basal diet and ii) $AFB_1$, diets with 100% of normal corn replaced with $AFB_1$ contaminated corn. There were 22 pens per treatment and 16 birds per pen. The concentration of $AFB_1$ was 195.4 and $124.35{\mu}g/kg$ in the contaminated corn and $AFB_1$ diet, respectively. Results: The $AFB_1$ decreased average daily gain, average daily feed intake, d 7 BW, final BW in the whole trial, and feed conversion ratio (FCR) during d 8 to 14 and d 1 to 14 by 10% to 47% (p<0.05), while FCR during d 1 to 7 was increased (p<0.05). $AFB_1$ did not affect mortality to 7 d of age, and then increased to 5.8% from 8 to 14 d of age (p<0.01). Apparent ileal gross energy digestibility was reduced by $AFB_1$, whereas apparent ileal digestibility of dry matter, nitrogen, and amino acid was improved (p<0.01). Feeding $AFB_1$ diets increased serum concentration of leptin and insulin-like growth factors-1 (IGF-1) (p<0.05), but had no effect on neuropeptide Y, ghrelin, cholecystokinin-8 or insulin (p>0.05). Dietary treatments did not influence relative expression of jejunal $Na^+$, $K^+-ATPase$ gene (p>0.05). Conclusion: Taken together, feeding corn naturally contaminated with $AFB_1$ reduced growth performance, improved apparent ileal digestibility, and affected serum leptin and IGF-1 in ducklings from d 1 to 14.

Effects of deoxynivalenol- and zearalenone-contaminated feed on the gene expression profiles in the kidneys of piglets

  • Objective: Fusarium mycotoxins deoxynivalenol (DON) and zearalenone (ZEN), common contaminants in the feed of farm animals, cause immune function impairment and organ inflammation. Consequently, the main objective of this study was to elucidate DON and ZEN effects on the mRNA expression of pro-inflammatory cytokines and other immune related genes in the kidneys of piglets. Methods: Fifteen 6-week-old piglets were randomly assigned to three dietary treatments for 4 weeks: control diet, and diets contaminated with either 8 mg DON/kg feed or 0.8 mg ZEN/kg feed. Kidney samples were collected after treatment, and RNA-seq was used to investigate the effects on immune-related genes and gene networks. Results: A total of 186 differentially expressed genes (DEGs) were screened (120 upregulated and 66 downregulated). Gene ontology analysis revealed that the immune response, and cellular and metabolic processes were significantly controlled by these DEGs. The inflammatory stimulation might be an effect of the following enriched Kyoto encyclopedia of genes and genomes pathway analysis found related to immune and disease responses: cytokine-cytokine receptor interaction, chemokine signaling pathway, toll-like receptor signaling pathway, systemic lupus erythematosus (SLE), tuberculosis, Epstein-Barr virus infection, and chemical carcinogenesis. The effects of DON and ZEN on genome-wide expression were assessed, and it was found that the DEGs associated with inflammatory cytokines (interleukin 10 receptor, beta, chemokine [C-X-C motif] ligand 9, CXCL10, chemokine [C-C motif] ligand 4), proliferation (insulin like growth factor binding protein 4, IgG heavy chain, receptor-type tyrosine-protein phosphatase C, cytochrome P450 1A1, ATP-binding cassette sub-family 8), and other immune response networks (lysozyme, complement component 4 binding protein alpha, oligoadenylate synthetase 2, signaling lymphocytic activation molecule-9, ${\alpha}$-aminoadipic semialdehyde dehydrogenase, Ig lambda chain c region, pyruvate dehydrogenase kinase, isozyme 4, carboxylesterase 1), were suppressed by DON and ZEN. Conclusion: In summary, our results indicate that high concentrations of DON and ZEN suppress the inflammatory response in kidneys, leading to potential effects on immune homeostasis.

Effects of stale maize on growth performance, immunity, intestinal morphology and antioxidant capacity in broilers

  • Objective: This study was conducted to determine the effects of stale maize on growth performance, immunity, intestinal morphology, and antioxidant capacity in broilers. Methods: A total of 800 one-day-old male Arbor Acres broilers (45.4±0.5 g) were blocked based on body weight, and then allocated randomly to 2 treatments with 20 cages per treatment and 20 broilers per cage in this 6-week experiment. Dietary treatments included a basal diet and diets with 100% of control maize replaced by stale maize. Results: The content of fat acidity value was higher (p<0.05) while the starch, activities of catalase and peroxidase were lower (p<0.05) than the control maize. Feeding stale maize diets reduced (p<0.05) average daily feed intake (ADFI) throughout the experiment, feed conversion ratio (FCR) during d 0 to 21 and the whole experiment as well as relative weight of liver, spleen, bursa of Fabricius and thymus (p<0.05) on d 21. Feeding stale maize diets decreased jejunum villus height (VH) and VH/crypt depth (CD) (p<0.05) on d 21 and 42 as well as ileum VH/CD on d 42. The levels of immunoglobulin G, acid α-naphthylacetate esterase positive ratios and lymphocyte proliferation on d 21 and 42 as well as lysozyme activity and avian influenza antibody H5N1 titer on d 21 decreased (p<0.05) by the stale maize. Feeding stale maize diets reduced (p<0.05) serum interferon-γ, tumor necrosis factor-α, interleukin-2 on d 21 and interleukin-6 on d 21 and 42. Broilers fed stale maize diets had lower levels of (p<0.05) total antioxidative capacity on d 42, superoxide dismutase and glutathione peroxidase on d 21 and 42, but higher (p<0.05) levels of malondialdehyde on d 21 and 42. Conclusion: Feeding 100% stale maize decreased ADFI and FCR, caused adverse effects on immunity and antioxidant function and altered intestinal morphology in broilers.

The effect of Sargassum confusum on reduction of body fat in obese women (알쏭이모자반 (Sargassum confusum) 주정추출물이 성인여성 체지방 감소에 미치는 영향)

  • Purpose: This study was conducted in order to investigate the effect of Sargassum confusum extracts on the reduction of body fat for eight weeks in overweight women ($BMI{\geq_-}23kg/m^2$). Methods: Subjects were classified by double-blind randomized trial as the control group (C group, n = 14) and the Sargassum confusum extract supplementation group (SC group, n = 16), which consumed 12 tablets per day. Questionnaires related to their health status were assessed twice (week 0 and week 8). Their dietary intake status was evaluated by 24-recall method and body compositions were measured using a bioelectrical impedance analyzer. In addition, we assessed the anti-obesity effect and the occurrence possibility of health risk factors during the supplementation periods by hematological and clinical analysis of blood. Results: Waist circumference and body fat (%) were significantly decreased in the SC group. Serum leptin level was also significantly decreased in the SC group. Defecation frequency was significantly increased in the SC group. The above results indicate that Sargassum confusum extract supplementation improves overweight on visceral fat and blood leptin level by increasing bowel movement. These results imply a decrease of health risk factors in overweight women. Seven subjects withdrew from the study due to adverse events; however, no differences regarding adverse events were observed between the control and treatment group. Conclusion: Therefore, Sargassum confusum extract is a plausible effective agent for body fat reduction in humans.

Inhibitory activity of Euonymus alatus against alpha-glucosidase in vitro and in vivo

  • The major goal in the treatment of diabetes mellitus is to achieve near-normal glycemic control. To optimize both fasting blood glucose and postprandial glucose levels is important in keeping blood glucose levels as close to normal as possible. ${\alpha}-Glucosidase$ is the enzyme that digests dietary carbohydrate, and inhibition of this enzyme could suppress postprandial hyperglycemia. The purpose of this study was to test the inhibitory activity of methanol extract of Euonymus alatus on ${\alpha}-glucosidase$ in vitro and in vivo to evaluate its possible use as an anti-diabetic agent. Yeast ${\alpha}-glucosidase$ inhibitory activities of methanol extract of E. alatus were measured at concentrations of 0.50, 0.25, 0.10, and 0.05 mg/ml. The ability of E. alatus to lower postprandial glucose was studied in streptozotocin-induced diabetic rats. A starch solution (1 g/kg) with and without E. alatus extract (500 mg/kg) was administered to diabetic rats by gastric intubation after an overnight fast. Plasma glucose levels were measured at 30, 60, 90, 120, 180, and 240 min. Plasma glucose levels were expressed in increments from baseline, and incremental areas under the response curve were calculated. Extract of E. alatus, which had an $IC_{50}$ value of 0.272 mg/ml, inhibited yeast ${\alpha}-glucosidase$ activity in a concentration-dependent manner. A single oral dose of E. alatus extract significantly inhibited increases in blood glucose levels at 60 and 90 min (p<0.05) and significantly decreased incremental response areas under the glycemic response curve (p<0.05). These results suggest that E. alatus has an antihyperglycemic effect by inhibiting ${\alpha}-glucosidase$ activity in this animal model of diabetes mellitus.

Related factors of oral symptoms in adolescents from Korean multicultural families (우리나라 다문화가정 청소년의 구강질환증상과의 관련요인)

  • Objectives: The purpose of this study was to identify the related factors of dental caries and periodontal disease in adolescents from Korean multicultural families, thereby helping to reduce the prevalence rate of oral disease. Methods: The subjects were 710 multicultural adolescents recruited using a web-based survey, National 2015 Korean Youth Risk Behavior, from the Korean Center for Disease Control. A multicultural family was defined in this study as one having an immigrant mother or father. Oral symptoms included dental caries and periodontal disease. Toothache was defined as a symptom of dental caries. Tender or bleeding gums were defined as symptom of periodontal disease. For statistical analysis, Statistical Package for Social Sciences (SPSS) Version 21.0 for Windows was used. Descriptive analysis and a Chi-square test were conducted to determine the factors associated with general characteristics, health behavior, and oral health behavior. Finally, to investigate the associations among oral disease symptoms, logistic regression analysis was performed. Results: Toothache was significantly higher in female 1.52 (95% CI; 1.45-1.60), high school 1.23 (95% CI; 1.18-1.28), women school 1.10 (95% CI; 1.05-1.16), individuals with poor economic status 1.45 (95% CI; 1.30-1.52), and participants who consumed alcohol 1.32 (95% CI; 1.27-1.37). Toothache related to perceived health status was significantly lower in the healthy group 0.69 (95% CI; 0.64-0.75), and was higher in usual stress group 1.65 (95% CI; 1.57-1.74). Gum bleeding was significantly higher in female 1.32 (95% CI; 1.27-1.37), high school 1.15 (95% CI; 1.10-1.19), and individuals with poor economic status 1.38 (95% CI; 1.27-1.50). Gum bleeding related to perceived health status was significantly lower in the healthy group 0.68 (95% CI; 0.63-0.74), and was higher in usual stress group 1.54 (95% CI; 1.46-1.62). Conclusions: Taking into account of social and economic levels, and dietary habits in the multicultural families adolescents, further education and support will be needed for oral disease prevention and early treatment.

Effects of Dietary Feldspar on Productivity and Composition of Eggs in Laying Hens (장석의 첨가가 산란계의 생산성 및 난 성분에 미치는 영향)

  • This study was carried out to investigate the effect of feldspar on laying Performance and e99 qualify in hens. A total of 96 'Tetra Brown' layers were assigned to 4 treatments in a completely randomized design. Each treatment consisted 4 replicates accommodating 6 layers per replication. Experiment diets were a control diet without feldspar and supplemented 0.5, 1.0 and $1.5\%$ feldspar. The egg Production rate was significantly increased in layers 134 diets supplemented 0.5 and $1.0\%$ feldspar compared to that of control (P<0.05). Egg weight, egg mass, feed intake, feed conversion ratio, egg shell strength and egg yolk color were not significantly different among the treatments (P>0.05). The haugh unit and blood spot of the eggs were significantly increased in $1.5\%$ feldspa. diets compared to that of control (p<0.05). However, the albumen index and meat spot in the eggs were not statistically different among treatments (P>0.05). Sensory evaluation traits were not significantly differed in terms of appearance, color, texture flavor and overall acceptability of eggs (P>0.05). The juiciness of boiled eggs was significantly increased in eggs of birds fed $1.0\%$ feldspar diets(P>0.05).

남은 음식물을 첨가한 펠렛사료가 오리의 생산성 및 육질에 미치는 영향

  • This experiment was conducted to investigate the effects of pelleted food waste on performance and meat quality of broiler ducks. One hundred-forty four broiler ducks were divided into 4 groups with 3 replicates per treatment for 5 weeks. Dietary levels of food waste 0%, 25%, 50% and 75% were included in experimental diets Daily weight gain was significantly increased by addition of food waste 25% and .50%, whereas decreased(P<0.05) by addition of 75% food waste. Feed intake were not affected by the pelleted food waste supplementations. Feed efficiency was improved(P<0.05) by the addition 25% food waste, however, there was depressed(P<0.05) by the addition of 75% food waste. Carcass weight was significantly decreased(P<0.05) by the addition 75% food waste. Moisture composition of breast meat were not significantly different by 25% and 50% food waste, however, there was significantly increased by 75% food waste. As the food waste level was increased, lipid and protein composition in beast meat was significantly decreased As the food waste level was increased. pH of beast meat was increased, whereas lightness and redness of meat color were decreased. Cholesterol content was significantly decreased(P<0.05) by addition of food waste. Odor and appearance of sensory evaluation was depressed by addition of food waste, whereas taste of sensory evaluation was improved by addition of food waste. There was no pathological findings from liver, spleen and kidney in slaughtered ducks. The results of this study suggests that pelleted food waste 25% and 50% improved broiler ducks performance and meat quality in broiler ducks.

인공 및 천연제올라이트의 급여가 육계의 생산성 및 장내 미생물에 미치는 영향

  • An experiment was conducted to compare the dietary supplemental influence of artificial zeolite produced from fly ash and natural zeolite on performance and physiological related factor of broiler chicks for five weeks. Diets contained CP 21.5, 19% and ME 3,100, 3,100kcal/kg for starting and finishing period. Three hundred twenty chicks were replaced into five treatments with four replicates. Control, 1.5 and 3.0% artificial(AZ) or natural zeolite(NZ) were supplemented to the basal diets, respectively. Weight gain, feed intake and feed conversion were weekly examined. Blood cholesterol, intestinal microflora, nutrients availability were measured at the end of experiment. Weight gain of chicks fed NZ tended to increase, but was not statistically different. However, the birds of fed 3.0% AZ showed significant decrement of weight gain compared to that of control(P<0.05). Feed intake was no difference in all treatments. Feed conversion was significantly improved in 3.0% NZ treatment relative to that of 3.0% AZ(P<0.05). There were no consistency in intestinal microbes between control and supplemental groups. Blood cholesterol was significantly lower in 3.0% NZ treatments than other treatments(P<0.05). Crude fiber digestibility of birds fed 3.0% NZ was significantly improved compared to that .of other treatments(P<0.05). As the results, artificial zeolite showed no detrimental effect in less than 1.5% addition on performance of broiler chicks.

Effect of Butylated Hydroxytoluene on Experimental Hepatocarcinogenesis Induced by 3'-Methyl-4-Dimethylaminoazobenzene I. Ultrastructural study (실험적인 간암 유발과정에서 Butylated Hydroxytoluene이, 미치는 영향 I : 미세구조적인 연구)

  • A morphologic study on the effect of butylated hydroxytoluene (BHT) on experimental hepatocarcinogenesis induced by 3'-methyl-4-dimethyl aminoazobenzene (3'-MeDAB) was investigated. A total of 110 Sprague-Dowley male rats weighting about 200 g each were used for the experiment, and divided into 4 groups; the 3'-MeDAB, BHT, 3'-MeDAB/BHT treated group, and the control group. Four to eight rats of each group were sacrified on the 4th, 8th, 14th and 16th experimental weeks, with continuous pelletized feeding containing 0.09% 3'-MeDAB and 0.5% BHT. The liver was examined by transmission and scanning electron microscopy. The results were as follows; Electron microscopically, the fine structure of the hepatocytes remained consistently abnormal up to 16 weeks after the 3'-MeDAB treatment. There was no significant difference in the groups observed earlier than in the ones observed later. Many subcellular changes were observed : nuclear change, decreased glycogen, mitochondrial abnormalities, disaggregated rough endoplasmic reticulum, marked proliferation of smooth endoplasmic reticulum, dilatation and distortion of bile canaliculi, increased lysosomes, apoptotic bodies, migration of bile ductule cell. In the BHT treated group, the ultrastructural changes of hepatocytes were not significant, except for the lipid droplets and proliferated smooth endoplasmic reticulum among hepatocytes depending on the experimental duration. The various subcellular changes of 3'-MeDAB/BHT treated groups were simillar to those of the 3'-MeDAB treated group, but the degree of changes in the 3'-MeDAB/ BHT treated group decreased compared with those of the 3'-MeDAB treated group. These results suggest that dietary butylated hydroxytoluene has a protective/inhibitory effect on the hepatocarcinogenesis induced by 3'-methyl-4-dimethyl -aminoazobenzene.
