Effects of Sea Tangle and Its Constituents in Diet on Immune Functions of Diabetic Mice

  • Dieary effects of sea tangle on immune functions were investigated in diabetic mice. Four groups of ICR mice weighing 33.36$\pm$1.01 g were fed either an AIN-76 diet only (control), or with additional sea tangle powder, sea tangle water extract, and alginate at the level of 13.6%, 4%, and 1%, respectively by weight. Cellulose was omitted in sea tangle powder and alginate diets. After 10 days of feeding respective experimental diets, all mice were made diabetic by five consecutive intramuscular injections of streptozotocin (40mg/kg body weight per day) and fed the diets for four more weeks. Plasma IgG concentrations but not those of IgM were significantly higher in mice fed sea tangle powder, extract or alginate than those on the control diet. Plasma TNF$\alpha$ levels were, however, lower in those fed sea tangle power or water extract than control and alginate fed groups. TNF$\alpha$ releases from macro phages isolated from four groups and cultured with 5 $\mu\textrm{g}$/mL LPS for 24 hors showed a similar tendency to the results of plasma concentrations in the respective groups, but IL-1$\beta$ releases were not different among four groups. Lymphocyte proliferation in response to LPS (10$\mu\textrm{g}$/mL) measured using splenocytes cultured for 3 days was highest in the alginate fed group (594$\pm$38%) and those of sea tangle powder (536$\pm$47%) and extract (547$\pm$34%) fed groups tended to be higher than the control (523$\pm$30%). It is concluded that sea tangle contains immunomodulatory components besides alginate that could enhance humoral immunity of itself. The immunomodulatory effects of sea tangle constituents is regarded as beneficial for diabetic subjects.

고지방식이와 STZ 유도 제2형 당뇨병 마우스에서 상엽 추출물의 항당뇨 효과 (Anti-Diabetic Effects of Mori Folium Extract on High-Fat Diet and Streptozotocin-Induced Type II Diabetes Mellitus in Mice)

  • Objectives : The present study was designed to investigate the anti-diabetic effects of Mori Folium (Morus alba L. of Moraceae) extract (MFE) on high fat diet (HFD) and streptozotocin (STZ)-induced type II diabetes mellitus in mice. Methods : The mice (C57BL/6J) were fed HFD for 8 weeks and then was induced with a single injection of STZ (75 mg/kg). The diabetic mice were divided into four groups [(STD, HFD, HFD + MFE and HFD + quercetin (QUR)] and administered with MFE or OUR for 4 weeks. Fasting blood glucose, lipid profile (triglycerides and cholesterol etc.), glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT), insulin and leptin were measured every 2 weeks. Results : Body weight gain was lower in the MFE and QUR groups than HFD group. The fasting blood glucose was lower in the MFE and QUR groups. Oral glucose and insulin tolerance were decreased in the MFE and QUR groups. The levels of serum total cholesterol, triglycerides, and LDL cholesterol were reduced in the MFE and QUR groups. The HDL cholesterol was much higher in the MFE and QUR groups than HFD group. The levels of GOT, GPT and atherogenic index were decreased in the MFE and QUR groups. The serum insulin and leptin concentrations were reduced in the MFE and QUR groups. Conclusions : These results showed that MFE could decrease blood glucose level and lead to an amelioration in dyslipidemia states on HFD/STZ-induced type II diabetes mellitus in mice.

고지방식이와 streptozotocin으로 유도된 당뇨 생쥐에서 β-glucan이 첨가된 보리숙면의 혈당조절과 면역력증진 효과 (Blood Glucose Control and Increase Immunity Effects of β-glucan added Cooked Barley Noodle in High-Fat Diet and Streptozotocin-Induced Diabetic Mice)

  • Purpose : This study was designed to examine the blood glucose control and increase immunity effects of ${\beta}-glucan$ added cooked barley noodle in streptozotocin-induced diabetes mice with a high-fat diet. Method : Forty-eight male ICR mice (6-week-old) were fed AIN-93 diet for 4 weeks. Mice were divided into six groups: normal, diabetic, cooked barley noodle, ${\beta}-glucan$ (5 %) control and two experimental groups (${\beta}-glucan$ 2.5 % and 5 %, cooked barley noodle contained diet with ${\beta}-glucan$ 2.5 % and 5 % w/w). Diabetes mellitus was induced by intraperitoneal injection of streptozotocin (150 mg/kg). Result : Blood glucose level was significantly decreased in groups consuming cooked barley noodles, but no significant difference was exhibited in diabetic and ${\beta}-glucan$ control group. These results were in accordance with the result of oral glucose tolerance test. Blood interfereon $(IFN)-{\gamma}$ was measured in order to identify increase immunity effect of ${\beta}-glucan$ in diabetic mice. Inhibited $IFN-{\gamma}$ concentration was recovered in cooked barley noodle and ${\beta}-glucan$ control group. Moreover, $IFN-{\gamma}$ concentration was dramatically elevated in ${\beta}-glucan$ contained cooked barley noodle groups in a dose dependent manner. Streptozotocin induced AST and ALT activities were decreased in ${\beta}-glucan$ contained cooked barley noodle groups with a strong lipid lowering effect. Conclusion : Although addition of ${\beta}-glucan$n did not give any significant synergistic effect on cooked barley noodle in blood glucose regulation, suppressed $IFN-{\gamma}$ production by STZ was dramatically enhanced by ${\beta}-glucan$ supplementation in a dose dependent manner. Liver function and blood lipid profile were also in accordance with the increase immunity effect of ${\beta}-glucan$. Consequently, ${\beta}-glucan$ added cooked barley noodle can be consumed as good diets for patients with chronic diseases with reduced immunity.

  • 번행초는 주요 화합물인 수용성 다당류와 cerebroside의 작용에 기인하여 기대되는 항궤양성 약용작물이다. 스트렙토조토신 유도 당뇨마우스에서 번행초의 다당 추출물(TPE)의 항당뇨 효과는 이전에 보고된 바가 없다. 본 연구는 체중 1 kg 당 65 mg의 스트렙토조토신을 복강에 주사하여 확립된 당뇨 마우스에서 TPE의 항당뇨 효과를 측정하기 위해 실시하였다. 100 mg 농도의 TPE를 3주간 투여했을 때, 당뇨 마우스의 체중은 유지되었으며, 혈당, 중성지방, 그리고 lactate dehydrogenase, alkaline phosphatase 및 glutamic pyruvic transaminase가 낮은 수준으로 감소함이 관찰되었다. 또한 내당능 실험에서도 TPE의 투여는 대조군인 당뇨 마우스에 비해 혈당 수준을 현저하게 감소시켰으며, 실험 가능한 거의 모든 생화학적 요인을 당뇨마우스에 비해 낮은 수준으로 회복시켰다. 따라서 전통적인 약용작물인 번행초의 잠재적인 항당뇨 효능을 본 연구에서 처음으로 증명하였다.

흑삼농축액을 이용한 흑삼두부 제조 및 품질 특성 (Preparation and Characterization of Black Ginseng Tofu)

  • This study was performed to process Tofu as an hypolipidemic food using black ginseng. Before processing we performed the ameliorating effect of black ginseng on the serum levels of triglyceride and total cholesterol in diabetic mice (db/db mice). As a result, the diabetic mice, whose diet was supplemented with black ginseng has a reduced level of serum lipid total cholesterol ($124.58{\pm}10.59mg/dL$) compared to non-supplemented diabetic mice, The supplemented mice exhibited a significant decrease (p<0.05) in serum lipid triglycerides ($80.32{\pm}35.40mg/dL$), which provided the efficacy of black ginseng in reducing hyperlipemia, thus indirectly proving the prevention and treatment of obesity. Then we processed Tofu as a hypolipidemic food using 0~8% black ginseng extract. We evaluated the quality characteristic after producing black ginseng Tofu. For color value, as the addition level of color increases, the value of L (lightness) decreased and a (redness), b (yellowness) increased. With increase in black ginseng concentrate additional level increases, the hardness, gumminess, and chewiness values increased (p<0.05), but the springiness and cohesiveness showed no significant differences. In case of sensory evaluation, Tofu with the addition of 2% black ginseng concentrate (BGT2) showed the highest preference overall. To sum up, black ginseng demonstrated pharmacological effects in treating diabetic complications like hyperlipidemia and reducing body deposit fat.

Micro-CT analysis of LPS-induced Alveolar Bone Loss in Diabetic Mice

  • Periodontal disease induces an increased incidence of tooth loss, particularly in cases with an associated loss of alveolar bone and periodontal ligaments. In this study, alveolar bone loss was detected by micro-computed tomography (CT) following exposure to E. coli lipopolysaccharide (LPS) in a streptozotocin (STZ)-induced diabetic mouse model. A 10 mg/ml dosage of E. coli LPS was applied between the first, second and third molars of the mice three times a week for 10 weeks. The loss of periodontal ligaments and alveolar processes was then evaluated by micro-CT using two and three dimensional microstructure morphometric parameters. In the diabetic mice, E. coli LPS induced the destruction of periodontal ligaments and loss of alveolar process spaces. The distances between periodontal ligaments were significantly widened in the STZ-LPS group compared with the untreated STZ group. The 10 mg/ml exposure to E. coli LPS in the STZ mice also resulted in a significant decrease in the alveolar bone volume fraction. The results of our study suggest that alveolar bone loss can be readily detected by volumetric micro-CT analysis as an increase in the distance between periodontal ligaments and in the alveolar process length.

오가피와 죽력 배합약물이 Streptozotocin으로 유발된 당뇨 생쥐에 미치는 영향 (Effects of Mixed Extracts with Acanthopanax chiisanensis and Bambusae Caulis in Liquamen on the Blood Sugar of Diabetic mice induced with Streptozotocin)

  • This study was carried out to investigate the optimal mixed extract with Acanthopanax Chiisanensis Nakai in order to strengthen anti-diabetic effects on the hyperglycemia induced by streptozotocin in mice. The original Bambusae Caulis in Liquamen filtered and refined. The effects of Acanthopanax Chiisanensis Nakai with Bambusae Caulis in Liquamen were administered to mice for 4weeks and its anti-diabetic effect examined. Mice used in this experiment were divided into two groups and distrilled water(control), Acanthopanax Chiisanensis Nakai mixed with refined Bambusae Caulis in Liquamen(AC+BCL.D) were given orally for 28days respectively. And then, experimental groups were observed in terms of blood sugar, creatinine, BUN and GPT. The amount of glucose was significantly decreased compared with the control group(P < 0.01). The amount of creatinine was decreased compared with the control group(P < 0.05). The amount of GPT did not show any differences between two groups.

당뇨 KK마우스에서 비타민 E 보강식이가 신장 당화단백질 생성에 미치는 영향 (Effects of Vitamin E Supplementation on Renal Glycosylation Products in Diabetic KK Mice)

  • KK마우스에 고지방 식이를 섭취시켜 당뇨를 유발하고, Maillard 반응의 초기산물인 Hb $A_{IC}$와 후기 산물인 신장조직의 CLF가 증가되는지를 확인하고. 당뇨병에 의해 증가되는 Maillard 반응이 비타민 I의 투여로 억제되는지 관찰하였다. 또 신장조직의 광학 현미경 관찰을 통하여 신장병변을 검사하였다. 이유즉시 KK마우스를 1개월간 pellet diet로 적응시킨 후 저비타민 E 식이(corn oil 20%, wt/wt)를 먹여 당뇨를 유도하였다. 비타민 E 보강은 2080IU/kg diet 수준으로 하였으며. 생후 4개월 , 6개월 . 9개월(당뇨 0개월. 2개월, 5개월)에 희생시켰다. 정상 KK-마우스에서는 월령이 증가하였을 때. 혈당과 Hb $A_{IC}$는 유의적인 변화를 보이지 않았으나 신장조직의 CLF는 점진적으로 증가되었다. 당뇨군에서 Hb $A_{IC}$는 6개월에서 증가되기 시작하였고 9개월군과 차이가 없었으며 . 신장조직의 CLF는 9개월 군에서 급격히 증가하였다. 또 고비타민 E 식이를 섭취시킨 경우 당뇨군에서 초기 및 후기 Maillard 반응생성물의 감소가 관찰되었다. 이상의 결과로부터 KK마우스에서는 월령이 증가함에 따라 초기 및 후기 당화산물의 축적과 신장 병변의 변화가 관찰되었고, 또한 당뇨 유발에 의해 이 변화들은 더욱 촉진되었으나, 2080IU/kg diet 수준의 고비타민 E 식이를 섭취시켰을 때 이런 변들이 억제되었음을 확인하였다.확인하였다.

db/db mice에 대한 약용 식물추출 혼합물의 항당뇨 상승효과 (Antidiabetic Synergistic Effects of Medicinal Plant Extract Mixtures on db/db Mice)

  • 본 연구는 db/db mice에 대한 구아바 잎(Pg), 바나바 잎(Ls) 추출물 그리고 혼합물 A (바나바, 구아바, 뽕, 솔잎 그리고 가시오가피 뿌리 추출물)의 효과를 조사하였다. 4주간 db/db mice에 이들 추출물을 섭취시킨 결과 Pg, Ls 그리고 혼합물 A의 섭취는 당뇨 대조군에 비해 체중, 혈당, 인슐린을 감소시켰으며 이들 중에서 혼합물 A의 감소효과가 가장 높았다. 이들 추출물은 당뇨 대조군에 비해 총 콜레스테롤, 중성지방, 유리지방산을 감소시켰으며 이들 중에서 혼합물 A의 항고지혈증 효과가 가장 뛰어났다. 또한 혼합물 A는 당뇨 대조군에 비해 Langerhans' islets의 손상을 유의하게 감소시켰다. 따라서 혼합물 A는 포함된 구성물질의 상호보완적인 약리작용에 의해 체중증가의 부작용이 없이 고혈당을 개선시키는 유익한 상승효과를 발휘한 것으로 나타났다.

소맥엽의 섭취가 Streptozotocin 유발 당뇨 흰쥐에서 혈중 포도당 및 지질에 미치는 영향 (Effects of Triticum aestivum sprout on Blood Glucose and Lipid Levels in the Streptozotocin-Induced Diabetic Mice)

  • This study investigated the functional effect by diets of Triticum aestivum sprout (TA) on the levels of blood glucose and lipid profiles in streptozotocin(STZ)-induced diabetic mice. Diabetes mellitus were induced by STZ intraperitoneal injection (50 mg/kg) into the male Balb/c mice. All mice fed AIN-93 diet for 3 weeks. Mice were divided to 4 groups: normal, diabetic control and two experimental groups (TA 5.0, diet with Triticum aestivum sprout 5.0% w/w; TA 2.5, diet with Triticum aestivum 2.5% w/w). The blood glucose level was decreased by TA intake in diabetic mice in the dose-dependant manner; 21.34% and 35.77% in TA 2.5 and TA 5.0 groups. The weight gain and feed efficiency ratio also were improved by TA intake compared with DM group. The weight gain was increased 1.28 and 1.09 folds in TA 5.0 and TA 2.5 groups, respectively. The weight of organs, including liver and kidney, was reduced but that of spleen and abdominal fat was increased by TA intake. Moreover, TA treatment increased the level of HDL but decreased the level of total cholesterol and triglycerides in the blood of diabetic mice. The blood of HbA1c also was significantly decreased without changing hematocrit by TA treatment compared with DM group. These results indicate the TA intake has the functional effects attenuating blood glucose and plasma lipid levels in insulin-dependent diabetes.
