Pulmonary Aspergillosis Secondary to Open Fracture of Pneumatic Bone in a Brown Hawk Owl (Ninox scutulata) (솔부엉이의 함기골 개방골절에 의한 속발성 폐 아스퍼질러스 감염 증례)

  • A flightless brown hawk owl (Ninox scutulata) weighing 180 g was rescued and referred to the teaching veterinary hospital with humerus open fracture. On physical examination and radiography, open fracture of the left humerus was found. After 3 days, orthopedic surgery was operated with intramedullary pins and wires. The bird died 2 days after surgery with anorexia. On necropsy, multiple green nodules with 2-3 mm in diameter were observed at the surface and inside of the left lung. Numerous conidial heads and spores were seen in the center of foci in the histopathological examination. The mycelia penetrated the surrounding pulmonary parenchyma, showing inflammation and necrosis. The fungus was isolated from the lung and cultured on Sabouraud Dextrose Agar at $30^{\circ}C$ for 7 days. The colony was blue-green color with a powdery surface. The fungus was identified as Aspergillus fumigatus by DNA analysis, including the internal transcribed spacer region, partial ${\beta}$-tubulin, and the calmodulin gene. This case was diagnosed as pulmonary aspergillosis secondary to open fracture of pneumatic bone in a brown hawk owl.

Optimizing Culture Conditions to Maximize the Production of Laccase from Pholiota highlandensis (Pholiota highlandensis 유래 laccase 생산을 위한 배양조건의 최적화)

  • The culture conditions needed to maximize the production of laccase from Pholiota highlandensis mycelia were investigated. Among the tested media for laccase production, Coriolus versicolor medium (CVM; 2% dextrose, 0.4% peptone, 0.6% yeast extract, 0.046% KH2PO4, 0.1% K2HPO4, 0.05% MgSO4·7H2O) showed the highest activity for the enzyme. Then, to optimize culture conditions for laccase activity, the influences of various carbon, nitrogen, phosphorus, and inorganic salt sources in CVM were investigated. The optimum culture medium was 2% fructose, 0.4% peptone with 0.6% yeast extract, 0.05% NaH2PO4, and 0.05% MgSO4·7H2O as carbon, nitrogen, phosphorus, and inorganic salt sources, respectively. Several aromatic compounds in the medium enhanced laccase activity to varying degrees. Guaiacol induced maximum laccase production, yielding 114.1 U/ml laccase activity after cultivation for 11 days at 25℃. The optimum pH and temperature for laccase production were 8.0 and 35℃, respectively. Native polyacrylamide-gel electrophoresis (PAGE) followed by laccase-activity staining with 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) as the substrate was performed to identify the presence of laccase under the optimum conditions studied. Zymogram analysis of the supernatant culture showed an enzymatic band with a molecular mass of about 90 kDa.

Development of selective media for Enterococci (장구균 검출 배지 개발)

  • An enterococci selective medium was developed to detect the presence of enterococci for use as a fecal contamination indicator. Among several media which have been known to detect enterococci, the following 9 different kinds of media were selected: Enterococci Confirmatory agar, Azide dextrose agar, Bromocresol-purple azide agar, Esculin bile agar, Citrate azide tween carbonate agar, KF Streptococcus agar, BROLACIN agar, Kanamycin esculin azide agar, and Membrane filter Enterococcus selective agar. Various components from the nine media were mixed to develop a more effective enterococcus selective medium. The newly developed medium named as 'Enterococcus Mixed medium' was more effective than the previous 9 media. Enterococci strains (Enterococcus avium KACC 10788, Enterococcus faecium KACC 11954, Enterococcus saccharolyticus KACC 10783, Enterococcus durans KACC 10787, Enterococcus faecalis KACC 11304, and Enterococcus hirae KACC 10779) and non-enterococci strains (Escherichia coli KACC 10005, Staphylococcus aureus subsp. aureus KACC 10768, and Bacillus subtilis KACC 10111) were used to test the new medium. As a result, the enterococci strains grew well on the Enterococcus Mixed medium whereas the non-enterococci strains did not grow well on it. Additionally, growth of enterococci with freshwater and seawater samples was observed to be good on the Enterococcus Mixed medium. The result of this study confirmed that the Enterococcus Mixed medium was effective in detecting the target enterococci.

Blind and Ultrasonography-guided Injection Therapy for Calcific Tendinitis of Supraspinatus (극상건 석회화 건염에 대한 맹검 주사요법과 초음파하 주사요법)

  • Purpose: To evaluate the differences between blindly and ultrasonography (US)-guided during multiple needling and dextrose injection technique for calcific tendinitis of shoulder. Materials and Methods: We chose 36 symptomatic calcific tendinitis patients, whose age ranged from 27 to 69. Our procedures were multiple needling and injection of dextrose over the lesion of calcific deposits. The bind injection group were 19 patients whose age ranged from 27 to 64-year-old (mean 52.2), and the US-guided injection group were 17 patients ranged from 31 to 69-year-old (mean 49.0). We compare these groups by VAS (visual analogue scale) and range of motion before and after procedures. Results: There is no difference between two groups in VAS and ROM before procedure (p>0.05). Two groups revealed significant improvement without limitation of shoulder function, however, the group under US-guided revealed better results than under blind (VAS:p=0.001, Flexion:p=0.000, Abduction:p=0.000, External rotation: p=0.016). Conclusion: Ultrasonography-guided procedure showed better results than blind, so the use of ultrasonography is more promising procedure.

Effect of Ultrasonic treatment on the Isolation of the Chlamydospores of Cylindrocarpon destructans Causing Root rot of Panax ginseng (초음파처리에 의한 인삼 뿌리썩음병균 Cylindrocarpon destructans 후막포자의 분리)

  • Chlamydospores were isolated from hyphae of Cylindrocanon destmctans by homogenization and/or ultrasonic treatment. Rate of the isolated chlamydospores by the homogenization with glass tissue grinder were 9.8% of all total chlamydospores formed in the culture of C. destructans. The length of mycelial fragments after the homogenization was about 400㎛ They were, however, formed in clusters of the chlamydospores and the mycelia The rate of the isolated chlamydospores from additional ultrasonic treatment after the homogenization of the mycelia were 74.3%. The length of mycelial fragments with the ultrasonic treatment was about 20 fm and chlamydospores seemed to be isolated from the mycelial mats and dispersed evenly in the culture. The numbers of chlamydospore in a catena were 1 to 8 cells after the homogenization on potato dextrose agar (PDA). Meanwhile the numbers of them after added ultrasonic treatment were 1 to 4 cells. Germination percentages of the isolated chlamydospores from the ultrasonic treatment were 46.8% after incubation of 2 days on PDA at 20。C and 60.7% after incubation of 13 days at 5。C, respectively. Germination rate of chlamydospores to the total chlamydospores produced by the ultrasonic treatment was 55.8%. However, it was increased to 74% when it was measured in the germinated catenae to the total catenae.

Antifungal effect of electrolyzed hydrogen water on Candida albicans biofilm (Candid albicans 바이오필름에 대한 전기분해 수소수의 항진균 효과)

  • Purpose: Candida albicans can cause mucosal disease in many vulnerable patients. Also they are associated with denture-related stomatitis. Electrolyzed water is generated by electric current passed via water using various metal electrodes and has antimicrobial activity. The aim of this study was to investigate antifungal activity of electrolyzed water on C. albicans biofilm. Materials and Methods: C. albicans was cultured by sabouraud dextrose broth and F-12 nutrient medium in aerobic and 5% $CO_2$ condition to form blastoconidia (yeast) and hyphae type, respectively. For formation of C. albicans biofilm, C. albicans was cultivated on rough surface 6-well plate by using F-12 nutrient medium in $CO_2$ incubator for 48 hr. After electrolyzing tap water using various metal electrodes, the blastoconidia and hyphal type of C. albicans were treated with electrolyzed water. C. albicans formed blastoconidia and hyphae type when they were cultured by sabouraud dextrose broth and F-12 nutrient medium, respectively. Results: The electrolyzed water using palladium electrode (EWP) exhibited antifungal effect on blastoconidia of C. albicans. Also, the EWP significantly has antifungal activity against C. albicans biofilm and hyphae. In the electrolyzed water using various metal electrodes, only the EWP have antifungal activity. Conclusion: The EWP may use a gargle solution and a soaking solution for prevention of oral candidiasis and denture-related stomatitis due to antifungal activity.

Growth Characteristics of Paecilomyces tenuipes by the Passage in Liquid Media (액체배지에서 눈꽃동충하초균(Paecilomyces tenuipes)의 계대증식에 따른 증식 특성)

  • The growth characteristics of Paecilomyces tenuipes according to the passage in the two kind of liquid media were investigated by comparing the mycelium and conidium formation degrees. The potato dextrose broth medium and the silkworm larvae medium containing the silkworm powder were used as the liquid media, and the potato dextrose agar medium and the brown rice medium containing the powder of silkworm pupa were used as the solid media. The conidium formation degree in liquid media differed by the passages but that in solid media was not. This suggested that the passage in liquid media did not affect significantly the conidium formation in solid media. When the brown rice media were inoculated with the concentration of $1{\times}10^{10}$ conidia/ml, $1{\times}10^8$ conidia/ml and $1{\times}10^6$ conidia/ml, respectively, the conidium formation degrees were similar. This indicated that the optimal inoculation concentration of conidium to the brown rice media is $1{\times}10^6$ conidia/ml.

Mycelial characteristics for the artificial cultivation of Inonotus obliqua (Pers.) Pilot (차가버섯(Inonotus obliqua) 인공재배를 위한 균사 배양적 특성)

  • The results of examining characteristics of mycelial growth and culture condition for determining the condition of artificially culturing Inonotus obliqua (chaga) are as follows. 1) Mycelial growth and density oft, oblique were the highest in the medium of BDA (Birch Dextrose Agar; 66.3mm/10d) followed by the order of GDA, PDA, CDA, PODA, ODA, YM, MCM, MEA (pH 7.0), CHA, and MEA (pH 4.7). 2) Optimal temperature for the mycelial growth and density of 1. obliqua were shown to be 30$^{\circ}C$, but the mycelia were dead at 40$^{\circ}C$. the mycelial growth and density of KNAC3005 strains was the highest at 30$^{\circ}C$(66.3mm/10d) followed by the order of 25, 20, 15, 35, 10, and 5$^{\circ}C$. 3) Optimal pH for the mycelial growth and density were revealed to be 6.0 (88.4mm/10d). Above or below pH 6.0, the mycelial growth and density were shown to be retarded. 4) Optimal carbon, nitrogen and organic acid sources for the spawn growth of 40 were maltose (331mg/25$m\ell$/15d), peptone (347mg/25$m\ell$/15d), and glutamic acid (357mg/25$m\ell$/15d), respectively. Optimal level of biotin was 370mg/15d and optimal C/N ratio was 40.

  • Rhodosporidium fluviale 201-C-1 and Kluyveromyces thermotolerans 150-JE-2(1) were isolated from wild flowers in Geumsan-gun, Chungcheongnam-do and Wando-gun, Jeollanam-do, respectively. Rhodosporidium paludigenum 86-J-1 and Candida sp. 80-J-3 were also isolated from flowers in Jeju island, Korea. Characteristics for these yeasts were not reported before. Therefore, morphological and physiological characteristics for these yeasts were investigated in this study. R. fluviale 201-C-1 was halophilic asporogenous yeast and formed pseudomycelium. It also grew at $30^{\circ}C$ and pH 4.0-6.0. K. thermotolerans 150-JE-2(1) was sugar-tolerant and halotolerant asporogenous yeast. It formed pseudomycelium and showed positive urease activity. Candida sp. 80-J-3 and R. paludigenum 86-J-1 were asporogenous yeasts, and grew well in yeast extract-peptone dextrose(YPD) medium, potato-dextrose medium as well as vitamin-free medium and YPD medium containing 5% NaCl.

First Report of Summer Patch Caused by Magnaporthiopsis poae on Cool Season Grass (Magnaporthiopsis poae에 의한 한지형 잔디의 여름잎마름병 보고)

  • Symptoms of summer patch were observed on Kentucky bluegrass (Poa pratensis L.) cv. "Midnight II" from mid-June in 2015 in Seoul, Korea. The symptoms appeared as leaf blight, root rot, and frog-eye patch, which are typical of summer patch. To identify the causal agent of these symptoms, a pathogen was isolated from diseased leaves and roots, and the cultural, morphological, and phylogenetic characteristics were analyzed. The isolate reached 50-60 mm on potato dextrose agar (PDA) after 10 days as a white-grey mycelium with septa, and became olive-green or brown from the center. Phialide-like structures were observed at the ends of hyphae, and conidia were rarely observed. A phylogenetic analysis was conducted based on large subunit (LSU) and RNA polymerase II large subunit (RPB1) sequences. According to this analysis, the isolated pathogen was confirmed to be Magnaporthiopsis poae. In a pathogenicity test, summer patch symptoms were observed at 20 days after inoculation using the same grass cultivar. This is the first report of summer patch disease caused by M. poae on cool season grass in Korea.
