Persimmon Gray Mold Caused by Botrytis cinerea (Botrytis cinerea에 의한 감나무 잿빛곰팡이병)

  • Botrytis cinerea was isolated from a gray mo이 leaf lesion on persimmon in fields of Kyeonsangnam-do from 1996 to 1998, and etiological study was conducted including physiological characteristics. It formed gray mold lesion with light green color on leaves of persimmon (Diospyros kaki). The temperature range for mycelial growth was between $5^{\circ}C$ and $30^{\circ}C$ with the optimum temperatures of $20^{\circ}C$ to $25^{\circ}C$. Conidia were oviod, cylindric, and colorless and their dimensions in culture were $8.4~11.5\times7.0~8.9\mu\textrm{m}$. The optimum temperature of conidial germination was $25^{\circ}C$. Sclerotia on potato dextrose agar medium were well formed and brownish condiophores were observed with their size of $18.5~64.9\times4.5~8.0\mu\textrm{m}$. Symptoms on artificially inoculated plants were similar to those of gray mold disease on persimmon caused by Botrytis cinerea in fields.

Culture conditions for mycelial growth of Poria cocos

  • This study was carried out to determine the basic mycelial culture conditions for Poria cocos growth. According to colony diameter and mycelial density, suitable media for mycelial growth were Malt yeast extract, Potato dextrose agar, Yeast extract agar, and Yeast malt agar. The optimum temperature for mycelial growth was between 25 and $35^{\circ}C$, and the optimum pH value was between 4 and 7. Carbon and nitrogen sources were fructose and yeast extract. The optimum C/N ratio was about 10 to 1 with 2% glucose. Other minor components for optimal growth were thiamine-HCl and nicotinamide as vitamins, acetic and lactic acid as organic acids, and $MgSO_4{\cdot}7H_2O$ and $FeSO_4{\cdot}7H_2O$ as mineral salts.

  • The cuclodextrin glucanotransferase (CGTase) gene of Bacillus macerans was subcloned at the downstream of yeast ADH1 promoter, and then the resulting plasmid pVT-CGTM(9.15 kb) was introduced into the yeast host strain, Saccharomyces cerevisias 2805. The transformed yeast, S. cerevisiae 2805/pVT-CGTM, showed the starch-hydrolyzing activity on the starch-azure plate. The optimal conditions for the CGTase expression were found to be 2% dextrose, initial pH5.5, 3$0^{\circ}C$, and 48hr cultivation. Under this condition, the extracellular CGTase activity reached at 0.53 U/mL, whereas the intracellular activity was about 0.03U/mL. This result indicates that the signal peptide of Bacillus CGTase functioned well in S. cerevisiae.

Calcium Absorption by the Fruitbody of Saesongi (Pleurotus eryngii) Mushroom

  • Saesongi (Pleurotus eryngii) was cultivated in both potato dextrose agar (PDA) and sawdust media supplemented with Ca salts. The addition of Ca phosphate and Ca carbonate to sawdust media did not affect the growth, whereas Ca sulfate addition suppressed the mycelial growth appreciably. The efficiencies of Ca accumulation in the fruiting were studied based on mycelial growth experiments on Ca-supplemented sawdust media. Supplementation with 0.1 to 5% Ca phosphate increased the Ca content in the fruiting body by 4.5-6.5 fold, to a level of $314.6{\pm}22.7$ to $449.7{\pm}29.3$.

Sooty Mold of Persimmon (Diospyros kaki) Caused by Cladosporium cladosporioides

  • In 2002, sooty mold was observed on persimmon (Diopyros kald) from the Jinju Agricultural Products Wholesale Market in Gyeongnam, Korea. The lesion of the sooty mold usually appeared as scars or wounds formed on the surface of the fruits. The symptom started with water soaking lesion, then the fruit softened rapidly. Colony of the causal pathogenic fungus was dark green in color on potato dextrose agar. The diameter of growing hyphae was 1-2 $\mu\textrm{m}$. The mostly one-celled conidia were ovoid, lemon or cylinder in shape, blastophores, and sized 46-132 $\mu\textrm{m}$. Erected conidiophores were long-branched chains and their lengths varied from 24 to 346 $\mu\textrm{m}$ with 2-5 $\mu\textrm{m}$ width. The optimum temperature for mycelial growth was about 2$0^{\circ}C$. Based on their mycological characteristics, the fungus was identified as Cladosporium cladosporioides. This is the first report in Korea of the sooty mold of persimmon caused by the said fungus.

Occurrence of Anthracnose on Indian Fig Cactus Caused by Glomerella cingulata and Colletotrichum gloeosporioides

  • Anthracnose symptoms were frequently found on stems of Indian fig cactus in Cheju island of Korea in 1998 and 1999. Typical symptoms were gray to black dry rot of stems with concentric arrays of dot-like spots. A Glomerella sp. or a Colletotrichum sp. was frequently isolated from the symptoms, both of which were identified as Glomerella cingulata and its anamorph, Colletotrichum gloeosporioides based on their morphological and cultural characteristics. Out of 31 isolates obtained from the symptoms, 12 isolates were the anamorph producing only conidia, four isolates the telemorph producing only ascospores, and 15 isolates the holomorph producing both spores on potato dextrose agar. Stem rots similar to the original anthracnose symptoms were induced by wound inoculation of conidia and ascospores but not by non-wound inoculation. The anamorphic isolates caused more extensive stem rot than the telemorphic and holomorphic ones.

Production and Exudation of Botryosphaeria dothidea conidia Using Cucumber Disks and Cereal Media (오이 절편과 배지를 이용한 Botryosphaeria dothidea 분생포자의 생성과 분출)

  • A method for inoculum production of Botryosphaerisa dothidea was developed using cucumber disks and cereal media. Disks of cucumber fruits, and cereal media of barley, wheat, and rice seeds were inoculated with mycelial plugs of B. dothidea and incubated at 27$^{\circ}C$. Pycnidia were produced on the surface of cucumber disks and seeds after 5 days of inoculation. When the inoculated barley seeds were immersed in sterilized distilled water for 5 minutes, abundant conidia of B. dothidea were exuded from mature pycnidia. Conidia were held together by mucilage as they were released from an ostiole. Compared with the conventional method for inoculum preparation using agar media, such as potato-dextrose agar and oatmeal agar, this method could minimize the tedious work required for inoculum preparation within a shorter period of time.

Trichophyton mentagrophytes infection in an African lion cub (Panthera leo) and transmission to a zookeeper (동물원 새끼 사자의 Trichophyton mentagrophytes 감염과 사육사에 전파된 원형피부백선 증례)

  • Dermatophytosis was found on the right front leg of a 4-month-old female African lion cub (Panthera leo) kept at a zoo with locally marginal alopecia. For diagnosis, culturing on sabouraud dextrose agar was performed and skin scrapings from the lesion were analyzed. The ones from the culture and skin scrapings were identified as Trichophyton mentagrophytes. A zookeeper that had been in contacted with the lion for artificial rearing developed skin lesions with well-defined erythematous plaques on the right arm about 1 month after the lesion in the lion was observed. The ringworm was probably transmitted from the lion through continuous contact.

The release of cellular constituents of Lactobacillus lactis by freezing and freeze-drying

  • Lactobacillus lactis cell was markedly damaged when shocked by freezing and freeze-drying. The supernatant of shocked cells in 1% phosphate buffer (pH7.3) showed a maximum u.v. absorption spectra at 260nm after further incubation for 90min at $37-^{\circ}C.$ The leakage of cellular constituents by shocking could be prevented by dimethyl sulfoxide, glycerol, dextrose and $MgCl_2.$ Among them, $MgCl_2$ showed the preventing effect before and after freezing wh-ereas the rests, only before the treatment. This leakage was proved not to be related to the cellular repairing mechanism.

  • We sampled the fungi on the grounds of the floor and books in the library for the purpose of attack the fungi in the traditional library. We cultured it on the Sabouraud Dextrose Agar(Baltimore Biological Laboratory) and was obtained the fungi of six species, Here irradiated the ultra violet lamp(100V, 15W, 253Å), which was occurred to the colony after 24 hours. And irradiation by 2hrs/day for 2days here. The other method, we were sterilized test by the methanal fumigation The following results were obtained 1. All rearing is suspensed the result of the ultra violet rays irradiation, but the colony that is gained from the grounds of the floor in the library is not sterilized. 2. The result of air tight box to methanal 14ℊ/㎥ for the 24hours fumigation, each test group(the fungi of six species) is all sterilized.
