Exploitation of the Dose/Time-Response Relationship for a New Measure of DNA Repari in the Single-Cell Gel Electrophoresis (Comet) Assay

  • The comet assay (also called the single-cell gel electrophoresis assay) has been widely used for detecting DNA damage and repair in individual cells. Since the conventional methods of evaluating comet assay data using frequency statistics are unsatisfactory we developed a new quantitative measure of DNA damage/repair that is based on all information residing in the dose/time-response curves of a comet experiment. Blood samples were taken from 25 breast cancer patients before undergoing radiotherapy. The comet assay was performed under alkaline conditions using isolated lymphocytes. Tail DNA, tail length, tail moment and tail inertia of the comet were measured for each patient at four doses of $\gamma$-rays (0, 2, 4 and 8 Gy) and at four time points after irradiation (0, 10, 20 and 30 min) using 100 cells each. The resulting three-dimensional dose-time response surface was modeled by multiple regression, and the second derivative, termed 2D, on dose and time was determined. A software module was programmed in SAS/AF to compute 2D values. We applied the new method successfully to data obtained from cancer patients to be assessed for their radiation sensitivity. We computed the 2D values for the four damage measures, i.e., tail moment, tail length, tail DNA and tail inertia, and examined the pairwise correlation coefficients of 2D both on the log scale and the unlogged scale. 2D values based on tail moment and tail DNA showed a high correlation and, therefore, these two damage measures can be used interchangeably as far as DNA repair is concerned. 2D values based on tail inertia have a correlation profile different from the other 2D values which may reflect different facets of DNA damage/repair. Using the dose-time response surface, other statistical models, e.g., the proportional hazards model, become applicable for data analysis. The 2D approach can be applied to all DNA repair measures, Le., tail moment, tail length, tail DNA and tail inertia, and appears to be superior to conventional evaluation methods as it integrates all data of the dose/time-response curves of a comet assay.

Synthesis, Crystal Structure, Spectra Characterization and DFT Studies on a Di-Cycle Pyrazoline Derivative

  • A dicycle pyrazoline derivative, 1-phenyl-5-(p-fluorophenyl)-3,4-($\alpha$-p-fluoro-tolylenecyclohexano) pyrazoline, was synthesized and characterized by elemental analysis, IR, UV-vis, fluorescence spectra and X-ray single crystal diffraction. Density function theory (DFT) calculations were performed by using B3LYP method with 6-$311G^{**}$ basis set. The optimized geometry can well simulate the molecular structure. Vibrational frequencies were predicted, assigned and compared with the experimental values, which suggest that B3LYP/6-$311G^{**}$ method can well predict the IR spectra. Both the experimental electronic absorption spectra and the predicted ones by B3LYP/6-$311G^{**}$ method reveal three electron-transition bands, with the theoretical ones having some red shifts compared with the experimental data. Natural bond orbital analyses indicate that the absorption bands are mainly derived from the contribution of n $\rightarrow\pi^*$ and $\pi\rightarrow\pi^*$ transitions. Fluorescence spectra determination shows that the title compound can emit blue-light at about 478 nm. On the basis of vibrational analysis, the thermodynamic properties of title compound at different temperature have been calculated, revealing the correlations between $C^0_{p,m}$, $S^0_m$, $H^0_m$ and temperature.

Sensitivity Analysis of Linear Elastic Problem due to Variations of the Traction Boundary Conditions (하중경계조건의 변화에 대한 선형탄성문제의 민감도 해석)

  • A shape design sensitivity of the elastic deformation due to a change of traction boundary condition is presented. The solution of governing equations for a linear elasticity problem is obtained by finite element method and the traction boundary is defined by design variables. The performance functional to be considered involves both the domain and boundary integral. Variations of geometry can be defined as design velocity. Using material derivative concept and adjoint equations, the design sensitivity is derived by Lagrange multiplier method. For a given geometry of a structure, the change of traction boundary is described by the tangential component of the design velocity only. The final result for the shape design sensitivity is formulated as the boundary integral form, the integrand is defined by tangential component of design velocity and first order derivatives of parameters. Numerical implementation of design sensitivity is discussed and is compared with the difference of the actual values.

Acute and Subacute Toxicity Studies of Water Soluble Dimethyl Dimethoxy Biphenylate Derivative in Rats (수용성 DDB유도체의 주사제 개발을 위한 급성독성 및 아급성독성시험연구)

  • The acute and subacute toxicity of water soluble dimethyl dimethoxy biphenylate derivative (new DDB), hepatitis therapeutics, were investigated in SD rats. In the acute toxicity study, body weights and clinical signs were observed for 7 days after the intravenous injection of new DDB at doses of 140, 182, 236, 307 and 400 mg/kg(r=1.3). Death. Severe convulsion, tremor and decrease motor activity were observed in almost treated groups (except the 140 mg/kg treated group). Changes of body weight in treated groups were not significantly different from control group. Autopsy of survived animals revealed no abnormal gross findings related to new DDB. As a results, the $LD_{50}$ values of new DDB were 244.1 mg/kg for male and 232.5 mg/kg for female. In subacute toxicity study, body weights and clinical signs were observed after intravenous injection of new DDB at doses of 57, 75 and 100 mg/kg/day for 28 days. Death, decrease motor activity and tremor were observed above 75 mg/kg treated groups. Statistically significant changes were observed in hematological and biochemical parameters of new DDB-treated groups; however, these changes were within normal range and had no relationship with dosage. Several abnormal findings were observed in microscopic examination of tissue; however, these findings were not caused by new DDB but environmental factor. The no toxic dose level of new DDB were estimated to be 57 mg/kg/day in this study.

  • The numerous applications surface interpolation include the modeling and visualization phenomena. A tetrahedrization is one of pre-processing steps for 4-D space interpolation. The quality of a piecewise linear interpolation 4-D space depends not only on the distribution of the data points in $R^2$, but also on the data values. We show that the quality of approximation can be improved by data dependent tetraheadrization through visualization of 4-D space. This paper discusses Delaunary tetrahedrization method(sphere criterion) and one of the data dependent tetrahedrization methods(least squares fitting criterion). This paper also discusses new data dependent criteria:1) gradient difference, and 2) jump in normal direction derivative.

Splines via Computer Programming

  • Traditionally, polynomials have been used to approximte functions with prescribed values at a number of points(called the knots) on a given interal on the real line. The method of splines recently developed is more flexible. It approximates a function in a piece-wise fashion, by means of a different polynomial in each subinterval. The cubic spline gas ets origins in beam theory. It possessed continuous first and second deriatives at the knots and is characterised by a minimum curvature property which es rdlated to the physical feature of minimum potential energy of the supported beam. Translated into mathematical terms, this means that between successive knots the approximation yields a third-order polynomial sith its first derivatives continuous at the knots. The minimum curvature property holds good for each subinterval as well as for the whole region of approximation This means that the integral of the square of the second derivative over the entire interval, and also over each subinterval, es to be minimized. Thus, the task of determining the spline lffers itself as a textbook problem in discrete computer programming, since the integral of ghe square of the second derivative can be obviously recognized as the criterion function whicg gas to be minimized. Starting with the initial value of the function and assuming an initial solpe of the curve, the minimum norm property of the curvature makes sequential decision of the slope at successive knots (points) feasible. It is the aim of this paper to derive the cubic spline by the methods of computer programming and show that the results which is computed the all the alues in each subinterval of the spline approximations.

Evaluation of the Function exp$(x^2)$ erfc(x) to Higher Precisions for Higher Order Derivative Polarography of CE-type Electrode Process

  • The function exp$(x^2)$erfc(x), which is often encountered in studies of electrode kinetics, is evaluated to an extended precision with 32 significant decimal digits in order to find theoretical relationships used in derivative polarography/voltammetry for a chemically-coupled electrode process. Computations with a lower precision are not successful. Evaluation of the function is accomplished by using three types of expansions for the function. Best ranges of arguments are selected for each equation for particular precisions for efficiencies. The method is successfully applied to calculate higher-order derivatives of the current-potential curves in all potential ranges for a reversible electron transfer reaction coupled with a prior chemical equilibrium (i.e., a CE type process). Various parameters that characterize the peak asymmetry (such as ratios of peak-heights, ratios of half-peak-widths, and separations in peak-potentials) are analyzed to find how kinetic and thermodynamic parameters influence shapes of the derivatives. The results from the CE process is compared with those from an EC process in which a reversible electron transfer is coupled with a follow-up homogeneous chemical reaction. The two processes exibit quite contrasting differences for values of the parameters.

Effect of Capsaicin and Its Novel Derivative on the Isolated Guinea Pig Bronchi (캡사이신과 그 합성유도체의 기니픽 기관지 평활근에 대한 작용)

  • In the present study we investigated the peripheral function of capsaicin and KR-25018, a newly synthesized capsaicin derivative, which was demonstrated to have a potent analgesic activity through different mechanism from morphine and nonsteroidal antiinflammatory drugs. Capsaicin (10-8~10-5 M) and KR-25018 (10-8~10-5 M) produced concentration-dependent contractions of the isolated guinea pig bronchi. There were no significant differences in the maximum response and the EC50 values (EC50: 0.137$\pm$0.025 $\mu$M and 0.097$\pm$0.031 $\mu$M for capsaicin and KR-25018, respectively, P>0.05). Phosphoramidon (10 $\mu$M) and indomethacin (10 $\mu$M) had no significant effect on contractile response to the submaximal concentration range of capsaicin and KR-25018 (3$\times$10-9~3$\times$10-7 M). The response to KR-25018, like that to capsaicin, was significantly inhibited by ruthenium red with reduction in the maximum response, which is indicative of non-competitive antagonism. A further common feature of the responses to capsaicin and KR-25018 in the guinea pig bronchi was their sensitivity to capsazepine. Capsazepine caused a rightward parallel shift in concentration-response curves obtained by capsaicin and KR-25018. the pA2 values of capsazepine were 5.90 and 5.99 against capsaicin and KR-25018 response, respectively. In conclusion, KR-25018 and capsaicin exert their contractile effects in the isolated guinea pig bronchial muscle by common mechanisms, probably via the activation of a specific receptor.

Examination of Bioconcentration of a New Algicide, Thiazolidinedione Derivative (TD49) to Marine Organisms (신규 살조제 Thiazolidinedione 유도체 (TD49)의 해양생물에 대한 생물 농축도 조사)

  • In this study, a newly synthesized thiazolidinedione derivative, TD49 with a highly selective algicide to red tide, was examined in order to evaluate the bioconcentration on aquatic organisms of coast. BAF (accumulation of TD49 by aquatic food chain) and BCF (accumulation of TD49 by sea water) were examined employing the shrimp (Fenneropenaeus chinensis) as the feed organism, and the olive flounder Paralichthys olivaceus as a consumer in marine ecosystem. Bioconcentration degree in sea water showed that the order in P. olivaceus was viscera > gill > muscle. The average BCF values of TD49 were 67.70, 63.32 and 20.25 at viscera, gill and muscle, respectively. Bioaccumulation degree using feed showed that the order in the organs of P. olivaceus was viscera > gill > muscle. The average BAF values of TD49 were 175.89, 114.88 and 32.59 at viscera, gill and muscle, respectively. When compared with two results, the accumulation by the food and water was higher than that by water. After the elimination experiment in sea water, the TD49 concentration was 2.81 nmole/g in the viscera and were not found in the gill and the muscle. More than 50% of the accumulated TD49 were eliminated from viscera in 7 days and all the accumulated TD49 were eliminated from gill and muscle in 7 days. On the other hand, the octanol/water partition coefficient (log $K_{ow}$) was measured to be 3.66 and experimental BCF of this study was 67.7.

The Singular Position Detection Method from the Measured Path Loss Data for the Cellular Network (이동 통신 망에서 측정하여 계산된 경로 손실의 급격한 변동 위치 추출 방법)

  • The path loss data was re-calculated according to the distance between the base station and a mobile station in the mobile telecommunications network. In this paper, the averaged path loss data was plotted with the conventional path loss models(free space, plane earth, Hata model ${\ldots}$). The standard deviations for the 2 Km, 1 Km, 0.5 Km-interval averaged path loss were 2.29 dB, 3.39 dB, 4.75 dB, respectively. Additionally, the derivative values for the 2 Km, 1 Km, 0.5 Km-interval averaged path loss were evaluated to find the positions with more than 1 times or 2times of the standard deviation. The situations with the sharply fluctuated path loss were calculated to 5 positions in the 2 Km interval, to 7 positions in the 1 Km interval, to 19 positions in the 0.5 Km interval, respectively. And, the exact distances between the base station and a mobile station were found with the sharply fluctuated path loss.
