소형루프 전자탐사의 감도분석 및 가탐심도 추정 (Sensitivity Analysis and Estimation of the Depth of Investigation in Small-Loop EM Surveys)

  • 소형루프 전자탐사의 가탐심도를 추정하기 위하여 주파수영역에서 2층구조에 대한 감도를 해석적으로 유도하였다. 이를 기초로 송수신 간격 2m 내외의 다중주파수 전자탐사 기기의 감도를 분석하고, 반응의 크기를 기준으로 가탐심도를 추정하였다. 먼저 감도 계산 결과는 하부층에 대한 감도는 동상성분이 이상성분에 비해 매우 높고 상부층의 두께가 20m에 이르더라도 저주파수 대역에서는 상부층의 감도에 비해 절대적으로 크다는 것을 나타낸 반면, 이상성분은 하부층에 대한 감도가 매우 약함을 보여준다. 따라서 다중 주파수를 이용한 소형루프 전자탐사에서는 동상성분의 정확한 측정이 가탐심도의 증대에 필수적임을 입증하였다. 전기비저항이 100ohm-m인 상부층 밑에 10ohm-m의 전기비저항을 갖는 하부층이 존재할 경우에는, 동상성분의 정확한 측정을 통하여 잡음 수준을 고려하더라도 10m까지의 가탐심도를 충분히 확보할 수 있으며 따라서 매립장의 침출수 분포 범위 영상화 등을 위해서 유용할 것으로 보인다. 그러나 전기비저항이 1,000ohm-m로 높은 기반암이 존재할 경우에는 비록 하부층의 감도는 상부층에 비해 매우 높으나 반응의 절대값이 매우 미약하여 탐지가 어려우며 이상성분 자료와의 복합적인 해석을 통해서도 기기의 정확도를 고려할 때, 가탐심도가 5 m를 넘기 힘든 것으로 나타났다. 따라서 전기비저항이 높은 지역에서는, 송수신 간격이 2m 내외로 짧은 다중주파수 소형루프 전자탐사법이 금속성 매설물의 탐지를 위해서는 유용하지만 기반암의 심도 규명에는 적절치 않다.

위선암종에서 핵의 Retinoic Acid Receptor (RAR) 및 cAMP Response Element Binding Protein (CREB)의 면역조직화학적 발현양상 (The Immunohistochemical Expression of Nuclear Retinoid Receptor and CREB in Gastric Adenocarcinoma)

  • 목적: 인체 내 여러 조직에서 상피세포의 분화 및 증식에 중요한 역할을 담당한다고 알려진 retinoic acid (RA)와 여러 유전자들에서 전사조절인자로 성장관여 유전자들의 활성화에 관여하며 세포증식 및 분화에 매우 중요한 세포내 조절인자인 CREB의 발현정도와 위선암종간의 상호 연관성 및 병리학적 인자들과의 관계를 관찰하였다. 대상 및 방법: 중앙대학교 의과대학 용산병원에서 1998년 1월부터 2007년 12월까지 위절제술을 시행 받고 위선암종으로 진단받은 환자의 위조직표본 중 보존상태가 양호한 파라핀 포매괴 150예를 연구대상으로 조직 표본에서 면역 조직화학적 염색을 통해 관찰하였다. 결과: 1. RAR의 발현은 장형 위선암종(72.2%)에서 미만형 위선암종(40.5%)보다 높게 나타났으며(P<0.01), 림프절 전이가 있는 경우(74.7%)가 림프절 전이가 없는 경우(49.2%)보다 의미 있는 발현양상을 나타냈다(P<0.01). 2. cAMP response element binding protein (CREB)의 발현은 장형 위선암종(69.4%)에서 미만형 위선암종(38.1%)보다 높게 나타났으며(P<0.01), 림프절 전이가 있는 경우(71.1%)가 림프절 전이가 없는 경우(47.8%)보다 높은 발현양상을 나타냈다(P<0.01). 3. 총 150예의 위선암종에서 RAR은 63.3% (95/150), CREB은 60.7%(91/150)에서 발현을 나타냈다(P<0.01). 결론: 이상의 결과로 RAR과 CREB은 조직학적 분화도 및 종양의 전이와 관련이 있고, 이들의 발현이 장형 위선암종에서의 생물학적 악성도에 관한 예후인자로서 관련이 있으나 이들의 발현이 위선암종에 미치는 생물학적 기전에 대한 추가 연구가 필요하다.

다큐멘터리 인터뷰 영상표현의 정량적 분석 (Quantitative Analysis about Visual Image Expression of Documentary Interview)

  • 미디어의 영상은 우리의 생활 속에 일부가 되어있으며, 사람들은 다양한 영상으로부터 정보를 얻기도 하고 판단이나 행동의 근거로 삼기도 한다. 특히 시사다큐멘터리는 사회적으로 중요한 이슈에 대해 정보를 제공할 뿐만 아니라, 찬성과 반대의 의견을 형성시키기도 한다. 영상을 통해서 전달되는 인터뷰는 얼굴 표정, 몸짓 등을 직접 보여줌으로써 신뢰감과 사실성이 뛰어나 강력한 전달 효과를 가지게 된다. 따라서 인터뷰의 영상 표현법의 차이에 따라 수용자에게 전달되는 메시지의 영향력이 다르게 나타날 수 있으므로, 인터뷰 영상의 표현법에 대한 연구는 중요하다. 본 연구는 인터뷰의 영상표현에 대한 실증적 연구를 위하여 국내 3개 방송사의 대표적인 시사다큐멘터리 프로그램에서 각각 5편씩 총 15편을 선택하여, 다큐멘터리에 나오는 인터뷰 샷 전체에 대한 정량적 분석을 시도하였다. 영상표현의 분석틀로 선행연구에서 공통으로 사용된 형태, 색, 움직임, 깊이를 인터뷰영상에 적합하게 유형화하였다. 분석결과 시사다큐멘터리의 인터뷰는 취재원보호를 위한 영상표현이 과반수에 가까웠으며, 샷의 형태는 버스트샷과 함께 클로즈업을 많이 사용되었다. 즉흥적 인터뷰가 많아서 현장 조명을 이용하여 색을 만들었고, 고정된 촬영보다는 핸드헬드 기법을 이용한 움직임이 나타났다. 또한 와이드렌즈의 사용으로 화면의 깊이감은 나타나지 않았다.

네이티브 광고의 광고 표시 및 규제 방법에 대한 탐색적 연구 - 제작 실무자들의 전문가 조사를 중심으로 - (An Exploratory Study on the Advertising Display and Regulation Method of Native Advertising - Focus on Expert research by production practitioners -)

  • 본 연구는 다양한 플랫폼에 나타나고 있는 네이티브 광고의 표현과 형식에 대한 문제점을 살펴보고 이를 규제할 수 있는 방안을 마련하고자 하였다. 이를 위하여 광고 실무 담당자들에게 심층 면접을 실시하여 이를 살펴보았다. 첫 번째 네이티브 광고의 표현과 형식에 대한 문제점으로 다양한 플랫폼에 나타나고 있는 무분별한 네이티브 광고에 대한 혼잡성과 기만성으로 인해 소비자들에게 부정적인 인식을 가져올 수 있다고 보았다. 두 번째 이용자들의 상호작용을 위해 플랫폼별 타겟팅을 통한 맞춤형 광고 표현이 제작되어야 할 것이라고 보았다. 세 번째 네이티브 광고에 대한 규제와 관련하여 소비자 보호를 위한 규제방안이 마련되어야 한다는 의견과 시장 자율에 맡겨야 한다는 의견을 제시하였다. 다양한 플랫폼에 따른 네이티브 광고의 전략적인 운영방안이 마련되어야 할 것이다.

토노메트리 측정 관점에서의 부침맥 고찰 (Review on Floating Pulse and Sinking Pulse in the View Point of Tonometric Measurement)

  • In pulse diagnosis, floating pulse and sinking pulse are frequently used for diagnosis about where disease is located and how much severe they are. However, in what mechanism floating pulse and sinking pulse arise is not known well. There are two point of views on substantial of floating pulse and sinking pulse. The first one is the floating and sinking degrees is the expression on the depth of pulsation. And, the second one is floating and sinking pulse is based on the response of pulsation to the indent pressure on radial artery. In this paper, we discussed these two opinions in the view point of tonometric measurement. The process for diagnosis on floating pulse and sinking pulse is similar to the tonometric measurement for non invasive blood pressure or intraocular pressure. We modelled the degrees of depth of pulsation with different indent pressures for initial pulsation feeling and different slopes of indent pressure lines. From this modelling, we can confirm the effect of pulsation depth on P-H curve, that is, in the model where lower pulsation is assumed, the shift of optimal indent pressure to the right was observed. The response of pulse pressure to the indent pressure was tried to be modelled with the degrees of mean blood pressure. Consequently, we tried to model the phenomenon of floating and sinking pulse for the first. And, from this modelling, we can get abundant understanding on how floating and sinking pulse can be caused. In the further study, we want to prove the suitability of this tonometric measurement based modelling with various studies including ultrasound measurement for the depth of pulsation in different EMI subjects.

위선암에서 Matrix Metalloproteinase-2, Cathepsin D 및 E-cadherin의 발현 (Expression of Matrix Metalloproteinase-2, Cathepsin D and E-cadherin in Human Gastric Adenocarcinomas)

  • Purpose: The prognosis of gastric cancer depends on the depth of invasion, lymph-node metastasis, invasion to adjacent tissues, and distant metastasis. Recently, it is known that tumor-associated proteases and adhesion molecules have been shown to play a relevant role in the process of progression and metastasis. The purpose of our study was to demonstrate the value of MMP-2 (matrix metalloproteinase), cathepsin D and E-cadherin as prognostic factors. Materials and Methods: In this study, formalin-fixed, paraffin-embedded tissue blocks from 69 patients with gastric cancer were immunohistochemically studied using antibodies to MMP-2, cathepsin D, and E-cadherin, and their expressions were analyzed according to the pathologic stage, lymph-node metastasis, histological differentiation, and patient survival. The medical records of these patients were retrospectively reviewed. Results: Increased expression of MMP-2 significantly correlated with advanced pathologic stage (P=0.026). Patients with lymph-node metastasis also had increased expression of MMP-2. Those patients with increased expression of MMP-2 showed a poorer survival; nevertheless, it was not statistically significant. Increased expression of cathepsin D significantly correlated with advanced pathologic stage (p=0.029). However, no correlation was observed between advanced pathologic stage and either lymph-node status or histological differentiation. Patients with increased expression of cathepsin D had a poorer survival, but that result was not statistically significant. No association was found between reduced expression of E-cadherin and pathologic stage, lymph-node status, or histological differentiation. Also, no correlation was found between the expression of E-cadherin and survival. In addition, when a combination of MMP-2 and cathepsin D expressions was analyzed, if both were negative, the survival seems to be longer, but it was not statistically significant. Conclusions: In patients with gastric cancer, expressions of MMP-2 and cathepsin D correlated with tumor stage; therefore, they may be considered as prognostic factors.

Changes in the components of salivary exosomes due to initial periodontal therapy

  • Purpose: Exosomes are membrane vesicles that are present in body fluids and contain proteins, lipids, and microRNA (miRNA). Periodontal tissue examinations assess the degree of periodontal tissue destruction according to the probing depth (PD), clinical attachment loss (CAL), bleeding on probing, and X-ray examinations. However, the accurate evaluation of the prognosis of periodontitis is limited. In this study, we collected saliva from patients before and after initial periodontal therapy (IPT) and compared changes in the clinical parameters of periodontitis with changes in the components of salivary exosomes. Methods: Saliva was collected from patients with stage III and IV periodontitis at the first visit and post-IPT. Exosomes were purified from the saliva, and total protein and RNA were extracted. Changes in expression levels of C6, CD81, TSG101, HSP70, and 6 kinds of miRNA were analyzed by western blots and real-time polymerase chain reaction. Results: Patients with increased C6 expression after IPT had significantly higher levels of periodontal inflamed surface area (PISA), miR-142, and miR-144 before and after IPT than patients with decreased C6 expression after IPT. Patients with decreased and unchanged CD81 expression after IPT showed significantly higher PD, CAL, and PISA before IPT than after IPT. Patients with decreased and unchanged TSG101 expression after IPT had significantly higher PD before IPT than after IPT. Patients with increased HSP70 expression after IPT had significantly higher PD and PISA before and after IPT than patients with unchanged HSP70 after IPT. The expression levels of miR-142, miR-144, miR-200b, and miR-223 changed with changes in the levels of C6, CD81, TSG101, and HSP70 in the salivary exosomes of periodontitis patients before and after IPT. Conclusions: The expression levels of proteins and miRNAs in salivary exosomes significantly changed after IPT in periodontitis patients, suggesting that the components of exosomes could serve as biomarkers for periodontitis.

방광암 환자의 요세포 검사에서 p53 단백 발현의 의의 (Immunohistochemical Detection of p53 Gene Mutation in Urine Samples in the Patients with Bladder Cancer)

  • Although bladder cancers are very common, little is known about their molecular pathogenesis. It is known that p53 alteration is found in about 60% of muscle-invasive bladder cancer, necessiating aggressive therapy and poor outcome. We examined the nuclear expression of p53 protein, using D07 monoclonal antibody in the urine samples from 31 patients with transitional cell carcinoma of the bladder to investigate the correlation of p53 overexpression with histologic grades and depth of invasion. The positive rate of p53 protein was 27% in superficial bladder tumor, but increased up to 71% in the invasive bladder carcinomas. The overexpression of p53 protein increased according to Mostofi grading system from 18% in grade I, 45% in grade II, and up to 100% in grade III. The p53 expression tended to be higher in the invasive and high grade bladder cancers than in the superficial and low grade ones(p<0.05). These results suggest that immunohistochemical analysis of the urine specimen in the bladder cancer patients could be a useful method of screening for the presence of p53 mutant protein. The mutant p53 protein expression may be an indicator of bladder cancer with more proliferative potential and/or aggressive biologic behavior.

Expression of Intercellular Adhesion Molecule-1 and E-Selectin in Gastric Cancer and Their Clinical Significance

  • Purpose: Among cell adhesion molecules, serum levels of intercellular adhesion molecule-1 and E-selectin are known to be correlated with the metastatic potential of gastric cancer. In the present study, the authors investigated the expression of intercellular adhesion molecule-1 and E-selectin in gastric cancer tissues and cultured gastric cancer cells, and examined their clinical value in gastric cancer. Materials and Methods: The protein was extracted from gastric cancer tissues and cultured gastric cancer cells (MKN-28 and Kato-III) and the expression of intercellular adhesion molecule-1 and E-selectin was examined by western blotting. The clinical significance of intercellular adhesion molecule-1 and E-selectin was explored, using immunohistochemical staining of specimens from 157 gastric cancer patients. Results: In western blot analysis, the expressions of intercellular adhesion molecule-1 in gastric cancer tissues and cultured gastric cancer cells were increased, however, E-selectin in gastric cancer tissues and cells were not increased. Among 157 gastric cancer patients, 79 patients (50%) were intercellular adhesion molecule-1 positive and had larger tumor size, an increased depth of tumor invasion, lymph node metastasis and perineural invasion. The intercellular adhesion molecule-1 positive group showed a higher incidence of tumor recurrence (40.5%), and a poorer 3-year survival than the negative group (54.9 vs. 85.9%, respectively). Conclusions: Intercellular adhesion molecule-1 is overexpressed in gastric cancer tissues and cultured gastric cancer cells, whereas E-selectin is not overexpressed. Increased expression of intercellular adhesion molecule-1 in gastric cancer could be related to the aggressive nature of the tumor, and has a poor prognostic effect on gastric cancer.

Indole-3-Carbinol Promotes Goblet-Cell Differentiation Regulating Wnt and Notch Signaling Pathways AhR-Dependently

  • Using an in vitro model of intestinal organoids derived from intestinal crypts, we examined effects of indole-3-carbinol (I3C), a phytochemical that has anticancer and aryl hydrocarbon receptor (AhR)-activating abilities and thus is sold as a dietary supplement, on the development of intestinal organoids and investigated the underlying mechanisms. I3C inhibited the in vitro development of mouse intestinal organoids. Addition of ${\alpha}$-naphthoflavone, an AhR antagonist or AhR siRNA transfection, suppressed I3C function, suggesting that I3C-mediated interference with organoid development is AhR-dependent. I3C increased the expression of Muc2 and lysozyme, lineage-specific genes for goblet cells and Paneth cells, respectively, but inhibits the expression of IAP, a marker gene for enterocytes. In the intestines of mice treated with I3C, the number of goblet cells was reduced, but the number of Paneth cells and the depth and length of crypts and villi were not changed. I3C increased the level of active nonphosphorylated ${\beta}$-catenin, but suppressed the Notch signal. As a result, expression of Hes1, a Notch target gene and a transcriptional repressor that plays a key role in enterocyte differentiation, was reduced, whereas expression of Math1, involved in the differentiation of secretory lineages, was increased. These results provide direct evidence for the role of AhR in the regulation of the development of intestinal stem cells and indicate that such regulation is likely mediated by regulation of Wnt and Notch signals.
