Background: Currently, cationic liposome has become the commonly used vehicles for gene transfection. Furthermore, one of the most significant steps in microRNAs expression studies is transferring microRNAs into cell cultures successfully. In this study we aim to approach the feasibility of transfection of cervical cancer cell lines mediated by liposome and to obtain the optimized transfection condition for cervical cancer cell lines. Materials and Methods: $Lipofectamine^{TM}2000$ as the carrier, miR-101 mimic was transfected into Hela cells and Siha cells. Using green fluorescent protein as reporter gene, to set different groups according to cell seeding density, the amount of miRNA, miRNA and the proportion of Liposomes, Whether to add serum into medium to study their impact on the liposomal transfection efficiency. Finally, MTT assay was used to analyze the relative minimal cell toxicity of liposome reagents. Results: The seeding density of Hela cell line and Siha are $1.5{\times}10^4$ (per well of 24 well plates), miRNA amount is 1ul of both, the ratio of miRNA and liposome is 1:0.5 of Hela cell line; 1:0.7 of Siha cell line respectively, after 24 hours we can get the highest transfection efficiency. Compared with serum medium, only Siha cells cultured with serum-free medium obtained higher transfection efficiency before transfection (P<0.01). MTT assay showed that according to the above conditions which has the lowest cytotoxicity. Conclusions: The method of Liposome to transfected is a suitable way and it can be an efficient reagent for miRNA delivery for Hela cells and Siha cells in vitro. It may serve as a reference for the further research or application.
Background: Nowadays, the encapsulation of cytotoxic chemotherapeutic agents is attracting interest as a method for drug delivery. We hypothesized that the efficiency of helenalin might be maximized by encapsulation in ${\beta}$-cyclodextrin nanoparticles. Helenalin, with a hydrophobic structure obtained from flowers of Arnica chamissonis and Arnica Montana, has anti-cancer and anti-inflammatory activity but low water solubility and bioavailability. ${\beta}$-Cyclodextrin (${\beta}$-CD) is a cyclic oligosaccharide comprising seven D-glucopyranoside units, linked through 1,4-glycosidic bonds. Materials and Methods: To test our hypothesis, we prepared ${\beta}$-cyclodextrin-helenalin complexes to determine their inhibitory effects on telomerase gene expression by real-time polymerase chain reaction (q-PCR) and cytotoxic effects by colorimetric cell viability (MTT) assay. Results: MTT assay showed that not only ${\beta}$-cyclodextrin has no cytotoxic effect on its own but also it demonstrated that ${\beta}$-cyclodextrin-helenalin complexes inhibited the growth of the T47D breast cancer cell line in a time and dose-dependent manner. Our q-PCR results showed that the expression of telomerase gene was effectively reduced as the concentration of ${\beta}$-cyclodextrin-helenalin complexes increased. Conclusions: ${\beta}$-Cyclodextrin-helenalin complexes exerted cytotoxic effects on T47D cells through down-regulation of telomerase expression and by enhancing Helenalin uptake by cells. Therefore, ${\beta}$-cyclodextrin could be superior carrier for this kind of hydrophobic agent.
Nuclear factor erythroid 2-related factor 2 (Nrf2) has been recognized as a transcription factor that controls mechanisms of cellular defense response by regulation of three classes of genes, including endogenous antioxidants, phase II detoxifying enzymes and transporters. Previous studies have revealed roles of Nrf2 in resistance to chemotherapeutic agents and high level expression of Nrf2 has been found in many types of cancer. At physiological concentrations, luteolin as a flavonoid compound can inhibit Nrf2 and sensitize cancer cells to chemotherapeutic agents. We reported luteolin loaded in phytosomes as an advanced nanoparticle carrier sensitized MDA-MB 231 cells to doxorubicin. In this study, we prepared nano phytosomes of luteolin to enhance the bioavailability of luteolin and improve passive targeting in breast cancer cells. Our results showed that cotreatment of cells with nano particles containing luteolin and doxorubicin resulted in the highest percentage cell death in MDA-MB 231cells (p<0.05). Furthermore, luteolin-loaded nanoparticles reduced Nrf2 gene expression at the mRNA level in cells to a greater extent than luteolin alone (p<0.05). Similarly, expression of downstream genes for Nrf2 including Ho1 and MDR1 were reduced significantly (p<0.05). Inhibition of Nrf-2 expression caused a marked increase in cancer cell death (p<0.05). Taken together, these results suggest that phytosome technology can improve the efficacy of chemotherapy by overcoming resistance and enhancing permeability of cancer cells to chemical agents and may thus be considered as a potential delivery system to improve therapeutic protocols for cancer patients.

This review explores recent works involving the use of the self-assembled nanoparticles of bile acid-modified glycol chitosans (BGCs) as a new drug carrier for cancer therapy. BGC nanoparticles were produced by chemically grafting different bile acids through the use of l-ethyl-3-(3-dimethylaminopropyl)-carbodiimide (EDC). The precise control of the size, structure, and hydrophobicity of the various BGC nanoparticles could be achieved by grafting different amounts of bile acids. The BGC nanoparticles so produced formed nanoparticles ranging in size from 210 to 850 nm in phosphate-buffered saline (PBS, pH=7.4), which exhibited substantially lower critical aggregation concentrations (0.038-0.260 mg/mL) than those of other low-molecular-weight surfactants, indicating that they possess high thermodynamic stability. The SOC nanoparticles could encapsulate small molecular peptides and hydrophobic anticancer drugs with a high loading efficiency and release them in a sustained manner. This review also highlights the biodistribution of the BGC nanoparticles, in order to demonstrate their accumulation in the tumor tissue, by utilizing the enhanced permeability and retention (EPR) effect. The different approaches used to optimize the delivery of drugs to treat cancer are also described in the last section.
최근 효과적인 암 치료 방법으로 광역학치료(photodynamic therapy)와 광열치료(photothermal therapy)가 주목받고 있다. 본 연구에서는 광열치료에 필요한 광열인자로써의 역할을 할 수 있는 골드 나노로드(AuNR)를 합성하고, 그 표면에 광역학치료를 위한 광증감제(photosensitizer)를 결합하였다. 즉, 골드 나노로드를 체내에 오래 머무르도록 하기 위해 PEG(polyethylene glycol) 및 효과적인 암 표적지향성을 위해 FA (folic acid) 리간드를 도입하였고, FA-PEG와 poly-${\beta}$-benzyl-L-aspartate (PBLA)로 이루어진 블록 공중합체를 3,4-dihydroxy hydrocinnamic acid (HCA) linker를 사용하여 골드 나노로드의 표면개질을 하였다. 또한 $AgNO_3$의 feeding ratio 변화를 통해 다양한 aspect ratio를 갖는 골드 나노로드를 합성하였고, UV-visible spectrophotometer, $^1H$-NMR, XPS, TEM 분석을 통해 FA-PEG-$P(Asp)_{50}$-HCA-AuNR100의 물리 화학적 특성과 morphology를 분석하였고, 성공적인 표면 개질을 확인할 수 있었다. 골드 나노로드의 표면 개질을 통한 생체 적합성 약물전달체의 합성은 효과적인 암 진단 및 다양한 광역학/광열치료 분야에 응용이 될 수 있을 것으로 기대된다.
콜로이드와 계면화학은 표면적과 표면에너지의 학문이다. 계면상의 위치에 따라서 분자밀도, 분자간의 상호작용력, 분자 배향성 그리고 반응성이 달라진다는 것은 흥미있는 주제가 되고 있다. 이러한 계면에너지가 중요하게 작용하는 시스템으로서 회합체, 에멀젼, 입자분산, 거품, 2차원적 표면이나 필름을 들 수 있다. 특히 나노 입자에 관련된 생체 적합성 재료를 사용하여 약물 전달체와 화장품 나노 소재로 이용하는데 관심이 고조되고 있다. 나노 입자는 수 nm에서 수백 nm 크기를 갖는, 넓은 표면적을 가진 콜로이드 상의 불균일 분산 입자의 일종이다. 지금까지 나노 입자의 제조, 특성 규명, 나노입자를 이용한 약물 봉입에 관한 연구가 활발히 이루어져 약물 전달체로서의 가능성이 충분히 입증되었다. 또한 난용성분 가용화 나노소재, 피부 흡수 증진용 나노소재, 자외선 차단용 나노소재, 안정화용 나노소재, 서방형 나노소재 등의 화장품 연구에 생체적합 나노전달체를 이용한 예가 보고되었다. 나노/마이크로 입자 시스템은 제조방법과 형태에 따라 나노/마이크로 스피어, 나노/마이크로 캡슐, 나노/마이크로 에멀젼, 폴리머 마이셀, 리포좀 등으로 구분된다. 수용액상에서 자기 회합체를 구성하는 나노수준의 폴리머 마이셀입자, 고농도, 고활성 물질에 대하여 농도 및 활성을 일정하게 제어할 수 있는 나노/마이크로 캡슐, 단일 이중층 또는 다층(100~800 nm)을 형성하여 여러 생리 환성 물질의 전달체로 이용되는 리포솜(liposome)에 대하여 제조방법과 산업의 응용에 대해 소개하였다.
The contribution of endogenous transport systems to the blood-brain barrier (BBB) transport of basic and acidic drugs was studied by using a carotid injection technique in rats and an isolated bovine cerebrovascular disease state were compared between the normotensive rats (WKY) and stroke-prone spontaneously hypertensive rats (SHRSP) which have been well established as an animal model with pathogenic similarities to humans. Basic drugs such as eperisone, thiamine and scopolamine inhibited, in a concentration dependent manner the in vivo uptake of $[{^3}H]choline$ through BBB, whereas amino acids and acidic drugs such as salicylic acid and valproic acid did not inhibit the uptake. The uptake of $[^3H]choline$ by B-CAP increased with time and showed a remarkable temperature dependency. The uptake of $[^3H]choline$ by B-CAP showed the very similar inhibitory effects as observed in the in vivo brain uptake, and was competitively inhibited by a basic drug, eperisone. The in vivo BBB uptakes of $[^3H]acetic$ acid and $[^{14}C]salicylic$ acid were dependent on pH of the injectate and the concentration of drugs. Several acidic drugs such such as salicylic acid, benzoic acid and valproic acid inhibited the in vivo uptake of $[^3H]acetic$ acid, whereas amino acid, choline and a basic drug such as eperisone did not inhibit the uptake. The uptake of acetic acid by B-CAP was competitively inhibited by salicylic acid. The permeability surface area product (PS) through BBB for $[^3H]choline$ in SHRSP was significantly lower than that in WKY. The concentration of choline in the brain dialysate in SHRSP was about half of that in WKY, while no significant difference was observed in the plasma concentration of choline between SHRSP and WKY. No significant difference was observed in the transport of monocarboxylic acids, glucose and neutral amino acid through BBB between SHRSP and WKY. From these results, it was concluded that BBB transport system of choline contributes to the transport of basic drugs through BBB, that acidic drugs can be transported via a moncarboxylic acid BBB transport system and that the specific dysfuntion of the BBB choline transport in SHRSP was ascribed to the reduction of the maximum velocity of choline concentration in the brain interstitial fluids.
(hfac)Cu(vtmos) [$C_{10}H_{13}O_{5}CuF_{6}$Si: 1,1,1,5,5,5-hexafluoro-2,4- pentadionato (vinyltrimethoxysilane) copper (I)] 구리원을 액체분사법으로 공급하여 반응성 스퍼터 증착된 PVD-TiN과 급속열처리 변환된 RTP-TiN 기판상에 구리를 유기금속 화학증착법으로 성장시키고, 증착조건과 기판 종류가 박막의 증착율, 결정구조 및 미세조직, 전기비저항 등에 미치는 영향을 분석하였다. 구리원 유량 0.2ccm에서 증착반응은 Ar 유량 200sccm까지 물질전달 지배과정과 전압 1.0Torr 이상에서 기화기에서의 공급율속을 보였다. 전압 0.6Torr일 때 활성화에너지는 155~225$^{\circ}C$의 표면반응 지배영역에서 12.7~14.1kcal/mol의 값을 나타내었으며, 225$^{\circ}C$ 이상의 기판온도에서는 $H_2$ 첨가에 따른 증착율 개선이 간응한 것으로 판단되었다. 증착층은 기판온도 증가에 따라 3차원 island 양식으로 성장하였으며, 증착초기 구리 핵생성밀도가 큰 RTP-TiN상 증착층이 PVD-TiN상보다 현저한 (111) 우선방위와 낮은 전기비저항값을 나타내었다. 구리박막의 전기비저항은 결정립간 연결성이 양호한 165$^{\circ}C$에서 가장 낮았으며, 증착온도에 따른 박막 미세구조 변화로 인해 그 거동은 3개의 영역으로 구분되어 나타났다.

지난 2010년 제정되어 2011년 8월과 10월에 개정된 미국 교통부의 행정입법 '항공 여객의 보호증진에 관한 법규(Enhancing Airline Passenger Protections, EAPP)'는 항공 소비자의 보호를 위한 연방정부의 입법으로 우리에게 시사하는 바가 크다. 특히 오늘날 장시간 활주로 지연과 관련한 항공사의 의무사항, 항공사의 고객 서비스강화, 항공권 광고와 판매 시에 발생하는 문제점 인식 등을 감안할 때 항공업계의 현 시류를 잘 반영하고 있다. 먼저, 장시간 활주로 지연과 관련하여 국내선 3시간, 국제선 4시간을 초과하는 활주로 대기 금지, 활주로에서 2시간 이상 대기 시 승객에게 물과 음식 제공, 화장실과 의료서비스 상시 제공, 지연발생 최초 30분과 이후 매 30분 간격으로 진전사항 공지 및 항공기 출입문이 열려있을 때 승객의 하기 가능성 안내 등이 규정되어 있다. 그 밖에도 고객서비스에 관한 절차와 계획의 수립을 비롯하여 그러한 계획을 활주로 지연에 관한 비상계획절차, 운송약관 등과 함께 항공사의 인터넷 웹사이트에 공시할 의무도 부과 하였다. 또한 고객 불만사항 처리를 위해 각 항공사는 모니터링 전담자를 배치하여야 하고, 고객 불만사항 처리시한도 법으로 마련되었다. 그리고 최근 이용자가 많은 SNS를 통한 고객 불만사항 처리에 관한 규정도 도입되었다. 또한 항공권 초과판매로 인한 비자발적 탑승거부 상황에 대한 규정도 마련하였다. 즉, 자발적 탑승 유예자 모집 시에 지급 대가를 명확히 제시하고, 비자발적 탑승거부승객에 대한 배상기준도 마련하였다. 또한, 항공권 판매광고 시에 총액운임 표시제도를 도입하였다. 이에 따르면 항공권 광고에서 항공운임은 실제 소비자가 구매할 때 지급하여야 하는 총액운임을 표시하여야 하며, 항공권 구매를 조건으로 기타 여행상품과의 묶음판매도 규제하고 있다. 이와 같은 법규가 시행된 이후 여러 건의 법적 쟁송이 있었다. 그 주요한 사건으로 우선, 스피릿 항공사건을 들 수 있다. 이 사건은 원고 항공사들이 법 내용 중 총액운임 표시제, 환불규정, 구매 후 추가 지불금지 규정이 자사의 헌법상 권리인 표현의 자유를 침해할 뿐만 아니라 그 법규의 내용이 지나치게 자의적이고 일관성이 없으므로 무효라고 주장하였다. 이 밖에도 상당수 법규 위반 사례가 확인되었는데 대표적인 사건이 유나이티드 항공사건이라 할 수 있다. 본 사안에서 유나이티드 항공은 장시간 활주로 지연상황에서 승객들에게 하기가능 사항을 적절히 고지하지 않았고, 그러한 지연상황을 DOT에 보고하면서 일부 사항을 누락시켜 문제가 되었다. 한편, 이러한 시대적 요구에 발맞추어 최근 우리 항공법도 항공 소비자 보호에 관한 규정을 추가하였다. 하지만 활주로 지연과 관련한 다양한 피해구제책의 마련, 항공권 초과판매에 따른 비자발적 탑승거절에 따른 보상기준 제시, 항공권 광고 시에 운임표시에 관한 총액 운임제의 기준 마련, 인터넷을 활용한 항공운송계약 체결과 관련한 소비자 보호제도 기반조성 등에 관한 사항들은 향후 추가 논의를 거쳐 도입될 필요가 있을 것으로 본다.

제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
